Abstract
【解決手段】(1)ヒトα−gal Aを過剰発現し分泌するように遺伝的に修飾されたヒト細胞、または(2)遺伝的に修飾された培養ヒト細胞から得られる精製ヒトα−gal Aのいずれかを用いて治療する、治療用組成物。

Description
本発明は、α−ガラクトシダーゼA及びα−ガラクトシダーゼ欠損症の治療に関する。
ファブリー病はX−連鎖で遺伝的に受け継がれるリソソーム蓄積症であって、それは重篤な腎障害、角化血管腫、及び心室拡張や僧帽弁不全などの心血管異常のような症状に特徴がある。またこの疾患は末梢神経系にも影響を及ぼし、激痛、即ち極限状態の強烈な痛みを引き起こす。ファブリー病は酵素のα−ガラクトシダーゼA(α−gal A)が欠乏することに原因があり、結果的に中性のグリコスフィンゴ脂質の異化作用が妨害されるとともに、細胞内や血流において酵素基質であるセラミドトリヘキサイドの蓄積が起こる。
培養されたヒトの細胞でヒトα−gal AをコードするDNAを発現させると、適当にグリコシル化されたポリペプチドを、酵素的に活性であって、ファブリー病で蓄積するグリコスフィンゴ脂質の基質に作用する能力があるだけでなく、この疾患で必要とされているところ、即ち作用細胞、特に患者の血管の内層の内皮細胞のリソソーム区画に正確にターゲティングさせる細胞表面の受容体を経て細胞によって効率的に中に取り入れられるように生成することが判明している。この知見については下記により詳細に説明されているが、ファブリー病のようなα−gal A欠損症であることが疑われる個体を、(1)ヒトα−gal Aを過剰発現して分泌するように遺伝的に修飾されたヒト細胞、または(2)遺伝的に修飾された培養ヒト細胞から得られる純粋なヒトα−gal Aのいずれかを用いて治療することができる。
細胞に関して本明細書で用いられる「遺伝的に修飾された」という用語は、特定の遺伝子産物および/またはコード配列の発現をコントロールする調節要素をコードするDNA分子を導入したあとで、その特定の遺伝子産物が発現する細胞が包含されていることを意味する。DNA分子の導入は遺伝子の標的化(即ち、特定のゲノム部位へのDNA分子の導入)によって成し遂げられ、更に相同組換えによって欠損遺伝子自体の置換が可能になる(欠損したα−gal A遺伝子またはその一部分が、ファブリー病の患者自身の細胞において、遺伝子全体またはその一部分と置き換えられうる)。
「シグナルペプチド」とは、さらなる翻訳後処理および/または分布のために小胞体に付着した新しく合成されたポリペプチドに指向するペプチド配列を意味する。
「第1クロマトグラフィー工程」という用語は、クロマトグラフィーカラムへのサンプルの最初の適用を意味する(サンプルの調製に関与する全ての工程は除く)。
α−gal Aのようなリソソームの酵素は、マンノース−6−ホスフェート(M6P)受容体、即ち、リソソームの区画のためと運命づけられた酵素のオリゴサッカライド部分に存在するM6P残基に結合する受容体との相互作用により細胞のリソソーム区画を標的として指向する(Kornfeld, S. and Mellman, I, Ann. Rev. Cell Biol. 5: 483-525, 1989)。一次の相互作用はゴルジ体で起こり、そこではゴルジのM6P受容体に結合した酵素がリソソームに移行するために分離される。二次的なタイプの相互作用は、細胞表面にて細胞外のα−gal AとM6Pとの間で起こると考えられている。細胞によってインターナリゼーションされた細胞外の基質はエンドサイトーシス小胞において細胞質を通って移行し、それは最初のリソソームと融合し、そのリソソームに内容物を移す。この過程で細胞表面のM6P受容体もエンドサイトーシス小胞に導入され、リソソームに移される。
二つの発現プラスミドであるpXAG−16及びpXAG-28を構築した。これらのプラスミドには、α−gal A酵素の398のアミノ酸をコードしているヒトα−gal A酵素(α−gal Aシグナルペプチドを欠いている)、ヒト成長ホルモン(hGH)遺伝子の第1イントロンによって割り込まれたhGHシグナルペプチドのゲノムDNA配列、及びポリアデニル化のシグナルを含むhGH遺伝子の3’非翻訳配列(UTS)が含まれる。プラスミドpXAG−16はヒトサイトメガロウイルスの即時型−初期(CMV IE)プロモーターおよび第1イントロン(非コードエクソン配列に隣接)を有しており、これに対してpXAG-28はコラーゲンIα2プロモーターによって推進され、またβ−アクチン遺伝子の第1イントロンを持つβ−アクチン遺伝子の5’UTSも含んでいる。
ヒトα−gal AのcDNAは、次のように構築したヒト繊維芽細胞のcDNAライブラリーからクローニングした。Poly−A+mRNAは全RNAから単離し、cDNAの合成は製造指示書にしたがってラムダザップII(lambda ZapII:登録商標)システムの試薬を用いて行った(Stratagene Inc., LaJolla, CA)。簡単に述べると「第1鎖」のcDNAは、内部XhoI制限エンドヌクレアーゼ部位を含むオリゴ−dTプライマーの存在下で逆転写を行うことで生成した。続いてRNase Hを用いて処理し、そのcDNAを二本鎖のcDNAを生成するためにDNAポリメラーゼIを用いてニック−トランスレーションした。このcDNAはT4DNAポリメラーゼを用いて平滑末端を形成し、EcoRIアダプターに連結した。この連結の生成物をT4DNAキナーゼで処理してからXhoIを用いて切断した。そのcDNAをセファクリル−400(Sephacryl-400:登録商標)クロマトグラフィーによってフラクションに分けた。大量で中間の大きさのフラクションを集め、そのcDNAをEcoRIとXhoI−切断性のラムダザップII(lambda ZapII)アームに連結した。次にこの連結反応の生成物を包装し、力価を測定した。主なライブラリーは力価が1.2×107pfu/mlであり、平均的な挿入物の大きさは925bpであった。
ヒトコラーゲンIα2プロモーターを次のようにα−gal A発現構築物pXAG-28で利用すべく単離した。ヒトコラーゲンIα2プロモーターの部分を含むヒトゲノムのDNAからなる408bpのPCRフラグメントを次のオリゴヌクレオチド、即ち5’−TTTTGGATCCGTGTCCCATAGTGTTTCCAA−3’(オリゴ72;配列番号:9)及び5’−TTTTGGATCCGCAGTCGTGGCCAGTACC−3’(オリゴ73;配列番号:10)を用いて単離した。
繊維芽細胞においてα−gal Aを発現させるために、公開された方法にしたがって二次の繊維芽細胞を培養し、トランスフェクションした(SeldenらのWO 93/09222)。
プラスミドpXAG−13、pXAG−16及びpXAG-28をエレクトロポレーションによってヒトの包皮の繊維芽細胞にトランスフェクションすることで、安定してトランスフェクトされたクローン細胞株を発生させることができ、そして得られたα−gal Aの発現レベルを実施例IDにおいて記載したようにモニターした。正常の包皮繊維芽細胞によるα−gal Aの分泌は2〜10ユニット/106細胞/24時間の範囲内であった。対照的にトランスフェクトされた繊維芽細胞は、表1に示されているような平均の発現レベルを示した。
α−gal A活性を、水溶性の基質である4−メチルウンベリフェリル−α−D−ガラクロピラノシド(4-MUF-gal; Research Products., Inc.)を用い、Ioannouら(J. Cell Biol. 119:1137-1150, 1992)により記載されたプロトコールの変形法によって測定した。この基質は、基質緩衝液(0.1Mのクエン酸塩−燐酸塩、pH4.6)に溶解して濃度を1.69mg/ml(5mM)にした。典型的には、10μlの培養液の上清を75μlの基質溶液に添加した。そのチューブを覆い、37℃の水浴中、60分間インキュベートさせた。このインキュベーションの終わりに2mlのグリシン−炭酸塩緩衝液(130mMのグリシン、83mMの炭酸ナトリウム、pH10.6)を用いることによって、その反応を止めた。それぞれのサンプルの相対蛍光度を、365nmの固定励起波長を持っていて460nmの固定放出波長を検出するモデルTK0100蛍光度測定計(Hoefer Scientific Instruments)を用いて測定した。サンプルの判断は、1μMのメチルウンベリフェロン(Sigma Chemical Co.)のストックから準備した標準と比較し、加水分解された基質の量を計算した。α−gal Aの活性はユニットで表し、即ち1ユニットのα−gal A活性は37℃で1時間あたりに加水分解された基質の1ナノモルに等価である。細胞発現のデータは一般に、分泌されたα−gal A活性ユニット/106個の細胞/24時間として表した。またこのアッセイは、細胞溶菌液及び下記に記載したように種々のα−gal精製工程から得られるサンプルにおけるα−gal活性の量を測定するためにも用いた。
実施例IIA−IIEには、α−gal Aを産生するように安定してトランスフェクトされた培養のヒト細胞株の条件付けられた培地からほとんど等質性にその酵素を精製できる点が示されている。
低温条件を整えた培地(1.34リットル)を遠心分離によって不純物を除去し、ガラス繊維製のプレフィルターを用いた0.45μmの酢酸セルロースフィルターを通して濾過した。攪拌しながら低温の濾過済み培地のpHを1NのHClを滴下して添加することによって5.6に調節し、続いて3.9Mの超純粋の硫酸アンモニウムのストック溶液(室温)を滴下により添加することで最終濃度が0.66Mになるまで硫酸アンモニウムを加えた。この培地をさらに4℃で5分感攪拌し、濾過し、次にブチルセファロース(登録商標)の4高速カラム(4 Fast Flow column;カラム容積は81ml、2.5×16.5cm; Pharmacia, Uppsala, Sweden)、即ち0.66Mの硫酸アンモニウムを含む10mMのMES−トリス、pH5.6(緩衝液A)で平衡化されたカラムにかけた。このクロマトグラフィーは、全蛋白質濃度と塩濃度をそれぞれ評価するためのライン上に設けたUV(280nm)と導電率モニターを装着したグラジ−フラク(Gradi-Frac;商品名)システム(Pharmacia, Uppsala, Sweden)にて4℃で行った。流速10ml/分でサンプルを適用してからそのカラムを、カラム容積の10倍の緩衝液Aで洗浄した。α−gal Aは、緩衝液A(硫酸アンモニウムを含む)から10mMのMES−トリス、pH5.6(硫酸アンモニウムを含まない)まで直線状の勾配をつけた14カラム容積の溶媒を用いてブチルセファロース(登録商標)カラムより溶出した。フラクションを4−MUF−galアッセイによってα−gal A活性について評価し、適当な酵素活性を含むフラクションを集めた。図6及び精製の概要(表3)に示すように、この工程によって約99%の不純物としての蛋白質が除去されている(カラムに通す前のサンプル=全蛋白質は8.14g、カラムを通った後のサンプル=全蛋白質は0.0638g)。
ブチルセファロース(登録商標)カラムのピークフラクションを、10mMのMES−トリス(4リットル)、pH5.6(一度交換)に対して4℃で透析した。透析物の導電度を、必要に応じてH2OまたはNaClを添加することによって4℃で1.0mMHOに調節した。その後でサンプルを、9mMのNaClを含んだ10mMのMES−トリス、pH5.6(緩衝液B)で平衡化したヘパリンセファロース(登録商標)6高速のカラム(Pharmacia, Uppsala, Sweden; 29mlのカラム容積、2.5×6cm)にかけた。これは10ml/分の流速で4℃にて行った。ライン上のUV(280nm)と導電度モニターによって全蛋白質と塩の濃度を測定した。そのサンプルを適用した後でそのカラムを10カラム容量の緩衝液Bで洗浄し、続いて3カラム容量の8%緩衝液C/92%緩衝液B(ここでの緩衝液Cは、250mMのNaClを含む10mMのMES−トリス、pH5.6である)までの線形勾配をつけた溶媒、さらに10カラム容量の8%の緩衝液Cを用いて洗浄した。この後、1.5カラム容量の29%までの線形勾配をつけた緩衝液Cを用いて、続いて10カラム容量の35%までの線形勾配をつけた緩衝液Cを用いてα−gal Aの溶出を行った。フラクションはα−gal A活性についてアッセイし、適当な活性を含むフラクションを集めた。
ヘパリンのプールを濾過し、1mMの燐酸ナトリウム、pH6.0(緩衝液D)で平衡化したセラミックハイドロキシアパタイトHC(40μm;American International Chemical, Ntick, MA;12mlのカラム容量、1.5×6.8cm)からなるカラムに直接かけた。そのクロマトグラフィーは、ライン上にUV(280nm)と導電度モニターを装着したハイブリッドグラジ−フラク(Gradi-Frac:商品名)/FPLC(登録商標)システム(Pharmacia, Uppsala, Sweden)にて室温で行った。サンプルを適用した(5ml/分)後でそのカラムを10カラム容量の緩衝液Dで洗浄した。α−gal Aを7カラム容量の42%緩衝液E/58%緩衝液D(ここでの緩衝液Eは、250mMの燐酸ナトリウム、pH6.0である)までの線形勾配をつけた溶媒、さらに10カラム容量の52%までの勾配をつけた緩衝液Eを用いて溶出した。フラクションはα−gal A活性についてアッセイし、適当な活性を含むフラクションを集めた。
ハイドロキシアパタイトのプールをH2Oを用いて約1.5倍に希釈して、最終的な導電度が室温で3.4〜3.6mMHOにした。濾過してからそのサンプルを、10%の緩衝液G/90%の緩衝液Fで平衡化したQセファロース(登録商標)HP(Pharmacia, Uppsala, Sweden;5.1mlのカラム容量、1.5×2.9cm)からなるカラムに直接かけた。ここで緩衝液Fは25Mの燐酸ナトリウム、pH6.0であり、また緩衝液Gは25mMの燐酸ナトリウム、pH6.0、250mMのNaClである。このクロマトグラフィーは、グラジ−フラク(Gradi-Frac:商品名)/FPLC(登録商標)ハイブリッドシステム(Pharmacia, Uppsala, Sweden)にて室温で行い、全蛋白質と塩の濃度をライン上に設けたモニターで測定した。そのサンプルに5ml/分の流速を適用してから、次の工程を行った。即ち、(1)10%の緩衝液Gで5カラム容量の洗浄、(2)12%の緩衝液Gで7カラム容量の洗浄、(3)3カラム容量の50%までの線形勾配をつけた緩衝液G、(4)10カラム容量の53%までの線形勾配をつけた緩衝液G、(5)3カラム容量の100%までの勾配をつけた緩衝液G、及び(6)10カラム容量の100%の緩衝液Gで洗浄工程。α−galは工程3及び4の間におもに溶出した。適当な活性を含むフラクションを集めた(「Qプール」)。
QプールをCentriprep(登録商標)−10遠心濃縮機ユニット(Amicon, Beverly, MA)を用いて約5倍に濃縮し、スーパーデックス(登録商標)200(Pharmacia, Uppsala, Sweden;189mlのカラム容量、1.6×94cm)からなるカラムにかけた。このカラムを、150mMのNaClを含む25mMの燐酸ナトリウム、pH6.0を用いて平衡化し、溶出した。このクロマトグラフィーは、蛋白質の溶出を追跡するためにライン上のUVモニター(280nm)を用いたFPLC(登録商標)システム(Pharmacia, Uppsala, Sweden)にて室温で行った。そのカラムにかけたサンプルの容量は≦2ml、流速は0.5ml/分、そしてフラクションの量は2mlであった。多数回カラムの作動を遂行し、フラクションをα−gal A活性についてアッセイして適当な活性を含むフラクションを集めた。
高度に精製したα−gal Aは、精製された蛋白質(≦1mgの蛋白質/ml)の希釈溶液として保存した場合、長い期間は安定していない。したがって配合剤を長く保存する間、即ち数週間から少なくとも数カ月間不変のまま保存を行う間、安定性を改善できるように工夫した。精製された酵素を、遠心濃縮機(25mMの燐酸ナトリウム(pH6.0)及び150mMのNaClからなる酵素緩衝液中で)を用いて少なくとも1mg/mlに濃縮した。ヒト血清アルブミン(HSA;ブミネート(Buminate:登録商標、Baxter-Hyland)を添加して最終濃度を2.5mg/mlにした。それからこの蛋白質溶液を、シリンジに取り付けた0.2μmの酢酸セルロースフィルター(Schleicher and Schuell)を用いて滅菌濾過した。α−gal A溶液を、滅菌した発熱体フリーのガラス溶液に分配し、テフロン(登録商標)製の蓋で密閉し、スナップ冷却を行ってから−20℃で保存した。
本発明により調製した精製ヒトα−gal Aの構造的及び機能的特徴を、この明細書に記載されたDNA分子とトランスフェクトしたヒト細胞株によって産生された対応する発現糖蛋白質とがそれぞれ遺伝子治療または酵素置換治療で利用可能であることを証明するために調べた。
α−gal Aの分子質量をMALDI−TOF質量分光測定法で見積った。これらの結果から、ダイマーの分子質量が102,353Daであり、これに対してモノマーの分子質量が51,002Daであることが証明された。アミノ酸組成に基づいてモノマーについて予測された分子質量は45,400Daである。したがって、酵素の炭水化物含有量は5,600Daまでの分子量であるとの計算が推測できる。
精製した蛋白質に対して行った標準的なアミノ酸分析の結果は、トランスフェクトしたヒト細胞によって産生された蛋白質がヒト組織から精製した蛋白質にアミノ酸レベルで同一であるという推断と一致している。
ヒトα−gal AのcDNAヌクレオチド配列は429個のアミノ酸をコードする。31個のN末端アミノ酸はシグナルペプチド配列を構成し、それは形成されつつある蛋白質が内部細胞質の網状質を通過するように開裂する(LeDonneらのJ. Biol. Chem. 262: 2062, 1987)。α−gal Aが異型のシグナルペプチド配列(例えば、ヒト成長ホルモンのシグナル配列)と関連していて、トランスフェクトしたヒト繊維芽細胞において発現する場合にα−gal Aが適当に処理されることを確認するために、分泌された蛋白質の10個のN末端アミノ酸をミクロシークエンシングした。サンプルをSDS−PAGEによって電気泳動し、10mMのCAPS(pH11.0)、10%のメタノール緩衝液システムを用いてプロブロット(ProBlott:登録商標、ABI, Foster City, CA)に転写した。プロブロット上の蛋白質はクマージー染色によって可視化し、そして適当サイズ(50kDa)のバンドを削り取った。N末端の配列は、自動化エドマン分解を行うアプライドバイオシステムズ(Applied Biosystems)パルス−液相アミノ酸配列同定装置を用いて得た。得られたN末端配列、LDNGLARTPT(配列番号:28)は、シグナルペプチドの適切な開裂と一致しており、分泌された蛋白質に対して予測されているN末端配列と適合する。
本発明により産生された分泌α−gal AのC末端アミノ酸残基は、自動化ヒューレットパッカードC末端配列決定装置(Hewlett Packard C-terminal sequencer)を用いて同定した。この結果はC末端がロイシン残基であることを示しており、それはこのDNA配列によって予言されたC末端アミノ酸と一致している。
本発明により産生されたα−gal Aのグリコシル化パターンも評価した。適当にグリコシル化されることは、α−gal Aがインビボで最適な活性を示すために重要である。即ち非グリコシル化システムで発現したα−gal Aは不活性であったりまたは不安定であったりする(HantzopolousらのGene 57: 159, 1987)。またグリコシル化は、α−gal Aが目的とする標的細胞にインターナリゼーションされるためにも重要であり、インビボでの酵素の循環血中半減期に影響を与える。α−gal Aのそれぞれのサブユニットには、アスパラギンに結合する炭水化物鎖の付加にとって役立つ4つの部位があり、そのうち3つの部位だけが占められている(DesnickらのIn The Metabolic and Molecular Bases of Inherited Disease, pp2741-2780, McGraw Hill, New York, 1995)。
精製されたα−gal Aの能力または比活性を、酵素の触媒活性(4−MUF−galアッセイを用いて)と、蛋白質濃度の両方を測定することによって計算する。蛋白質濃度は、BCAシステム(Pierce)を用いるようなどれかの標準法によって、または280nmでの吸収を測定し、吸光度係数2.3mg/ml(アミノ酸分析から決定した)を用いて値を計算することによって決定することができる。これらの手法を用いると、トランスフェクトしたヒト繊維芽細胞の条件を整えた培地から精製されたα−gal Aの比活性は2.2〜2.9×106ユニット/蛋白質のmgであり、それはヒト組織から精製されるα−gal Aの比活性に匹敵する(Bishopらの、J. Biol. Chem. 256: 1301, 1981)。
安定したトランスフェクトがなされた細胞によって産生されるα−gal Aがα−gal A欠損症の有効な治療薬となるためには、その酵素は影響を受けた細胞によってインターナリゼーションされなくてはならない。α−gal Aは生理学的なpHレベルでは活性でなく、また血液や小腸液では有効となりそうではない。リソソームの酸性環境でインターナリゼーションされる場合にのみ、蓄積した脂質基質は最適に代謝される。このインターナリゼーションは、細胞表面に現れていて、エンドサイトーシス経路によってリソソームにそのα−gal Aを供給するマンノース−6−ホスフェート(M6P)受容体にα−gal Aが結合することによって仲介される。M6P受容体はいたるところに現れており、ほとんどの体細胞はそれをある程度まで発現する。糖蛋白質上にある露出したマンノース残基に特異的なマンノース受容体はほとんど行き渡ってはいない。後者の受容体は一般に、マクロファージ及びマクロファージ様の細胞にのみ見られるもので、これらの細胞型にα−gal Aが入る別の手段を提供する。
α−gal A活性(ユニット/全蛋白質のmg)
遺伝子治療では、α−gal Aを産生する自家細胞の移植片が適当に修飾された形態でその酵素を産生することによって、標的細胞のα−gal A欠損症を「修正」できなくてはならない。ファブリー細胞におけるトランスフェクトしたヒト繊維芽細胞によるα−gal Aの産生の効果を評価するために、ファブリー病の患者から採集した繊維芽細胞(NIGMS Human Genetics Mutant Cell Repository)を、トランスウェルズ(登録商標、Costar, Cambridge, MA)でα−gal A−産生細胞株(BRS−11)とともに共培養した。その実験スキームが図8に図示されている。ファブリー細胞は12ウェルの組織培養皿にて培養し、そのうちのいくつかは、細胞が成長できる表面を持つインサート(トランスウェルズ(Transwells、登録商標、0.4μmの孔径)を含んでいた。このインサートの成長マトリックスは多孔性であり、巨大分子が上方から下方の環境へと通ることを可能にする。1セットのインサートは、最小レベルのα−gal Aを分泌するヒトの包皮(HF)の繊維芽細胞を含んでおり、これに対して別のセットは大量のα−gal Aを分泌する安定にトランスフェクトしたヒト繊維芽細胞株、BRS−11を含んでいた。α−gal A−産生細胞とともに共培養したウェルでは、α−gal Aがファブリー細胞を浸している培地に入ることができ、ファブリー細胞によってインターナリゼーションされる可能性を秘めている。
α−gal A活性(ユニット/全蛋白質のmg)
他の細胞型をこの明細書に記載された方法で利用できる。この細胞はさまざまな組織から得ることができ、またそれには培養して維持できる全ての細胞型が含まれる。例えば、本発明によってトランスフェクトすることのできる一次細胞及び二次細胞には、ヒト繊維芽細胞、ケラチン生成細胞、上皮細胞(例えば、乳房または小腸の上皮細胞)、内皮細胞、神経膠細胞、神経細胞、血液を形成する要素(例えばリンパ球や骨髄細胞)、筋肉細胞、及びこれらの体細胞型の前駆体が含まれる。繊維芽細胞は特に関心がもたれる。一次細胞は、患者の免疫システムによってそれらが拒絶されることのないように、トランスフェクトした一次または二次の細胞を投与すべき患者から得ると好ましい。しかしながら、免疫拒絶(下記に記載したような)を阻止したり抑制したりするために適当な注意が払われるならば、患者以外のヒトドナーの細胞をうまく用いることができる。このことは、標準化され、確立され、安定してトランスフェクトされた細胞系の使用を、全ての患者において可能にするであろう。
上記に記載した細胞は、それらが例えば腎臓の被膜下、皮下の区画部分、中枢神経系、脊髄内空間、肝臓、腹膜腔内空間、または筋肉内などに存在するように、種々の標準化された投与経路を経てヒトに導入することができる。またこの細胞は、ヒトの血流で循環するように静脈内または動脈内に注入してもよい。一旦ヒトに移植すると、そのトランスフェクトされた細胞は治療用産物であるグリコシル化ヒトα−gal Aを産生しかつ分泌する。
安定してトランスフェクトした(または別の方法で遺伝的に修飾した)ヒト細胞によって発現して分泌され、そして本明細書に記載したように精製されるα−gal A蛋白質を、α−gal A蛋白質産生が不十分または欠損している患者に投与することができる。この蛋白質は、薬学的に受容可能なキャリアに入れて、例えば実施例IIIに記載されたような配合剤に入れてpH6.5以下で投与することができる。配合剤で含まれていてもよい賦形剤の例は、クエン酸塩緩衝液、燐酸塩緩衝液、酢酸塩緩衝液、もしくは重炭酸塩緩衝液のような緩衝液、アミノ酸、ウレア、アルコール、アスコルビン酸、リン脂質、血清アルブミンもしくはゼラチンのような蛋白質、EDTA、塩酸ナトリウム、リポソーム、ポリビニルピロリドン、マンニトール、ソルビトール、グリセロール、プロピレングリコール及びポリエチレングリコール(例えば、PEG−4000、PEG−6000)である。投与経路は例えば静脈内、動脈内、皮下、腹腔内、脳内、筋肉内、肺内、または粘膜経由であるとよい。投与のルート及び分配される蛋白質の量は、技術熟練者の評価能力に充分入るファクターによって決定することができる。さらに技術熟練者は、治療用蛋白質の投与経路及び用量を、治療用量レベルが得られるまで対象の患者において変化させるとよいことがわかっている。典型的には、α−gal Aは0.01〜100mg/体重1kgの用量で投与されるであろう。この蛋白質を調節しながら繰り返し投薬することは、患者の生涯にわたって必要であろう。
α−gal A以外のリソソーム貯蔵酵素の欠損症によって引き起こされる他の状態は、本明細書に記載した方法に匹敵する方法による治療に影響を受けるであろうと見込まれる。これらの場合では、欠乏した酵素の機能的な型をコードするDNAを、本明細書に開示された発現構築物でのα−gal AをコードするDNAと置換すればよい。確認されている酵素欠損症候群、及び本明細書に記載されたような治療に影響される可能性のある酵素欠損症候群の例が表8に示されている。この表における情報はNeufeld(Ann. Rev. Biochem. 60:257-280, 1991)から得られるものであり、それは参照文献として本明細書に組み入れられる。
本明細書に記載された本発明は、トランスフェクション、即ち遺伝子産物をコードするとともに、コード配列の発現をコントロールする調節要素を持っている構築物の導入後に特定の遺伝子産物を発現する細胞を用いる治療方法によって部分的に説明した。またこれらの方法は、他の工程、即ち遺伝子の標的化及び遺伝子の活性化によって遺伝的に修飾された細胞を用いて行ってもよい(参照文献として本明細書に組み入れられるTrecoらのWO 95/31560参照、またSeldenらのWO 93/09222も参照)。

Claims (35)
- α−ガラクトシダーゼA欠損症であると推測される患者を同定する段階と、ヒトα−gal Aを過剰発現し、かつ分泌するように遺伝的に修飾されたヒト細胞を該患者に導入する段階とを含む治療方法。
- 細胞が、ヒトα−gal A(配列番号:26)を含むポリペプチドをコードするコード配列を有するDNA分子でインビトロにてトランスフェクトされた請求項1記載の方法。
- ポリペプチドが異種シグナルペプチドをさらに含む請求項2記載の方法。
- 異種シグナルペプチドがヒト成長ホルモン(hGH)シグナルペプチド(配列番号:21)である請求項3記載の方法。
- DNA分子が、前記シグナルペプチドをコードする配列にイントロンを含む請求項3記載の方法。
- DNA分子が、コード配列の終結コドンの3’に隣接して存在する少なくとも6個のヌクレオチドからなる非翻訳配列を含み、該非翻訳配列がポリアデニル化部位を含む請求項3記載の方法。
- hGHシグナルペプチド(配列番号:21)をコードし、イントロンを含む第1配列と、
該第1配列の3’末端に結合し、ヒトα−gal Aを含むポリペプチドをコードする第2配列とを含むDNA分子。 - ポリアデニル化部位を含む3’非翻訳配列をさらに含む請求項7記載のDNA分子。
- 請求項7記載のDNA分子を含み、ヒト細胞での発現に適当である発現構築物。
- (a)異種シグナルペプチドに結合されたヒトα−gal Aを含むポリペプチドをコードし、かつ(b)細胞における該ポリペプチドの発現を可能にするDNA分子を含む培養されたヒト細胞。
- 繊維芽細胞である請求項10記載の細胞。
- 上皮細胞、内皮細胞、骨髄細胞、神経膠細胞、肝細胞、ケラチン生成細胞、筋細胞、神経細胞、またはこれらの細胞型の前駆体からなる群より選択される請求項10記載の細胞。
- シグナルペプチドが、hGHシグナルペプチド(配列番号:21)である請求項10記載の細胞。
- (a)異種シグナルペプチドに結合されたヒトα−gal Aを含むポリペプチドをコードし、かつ(b)細胞における該ポリペプチドの過剰発現を可能にするDNA分子を用いてトランスフェクトされた複数の培養ヒト細胞から本質的になるクローン細胞株。
- 細胞が繊維芽細胞である請求項14に記載のクローン細胞株。
- (a)異種シグナルペプチドに結合されたヒトα−gal Aを含むポリペプチドをコードし、かつ(b)細胞における該ポリペプチドの過剰発現を可能にするDNA分子を用いてトランスフェクトされた複数の不死化ヒト細胞から本質的になるクローン細胞系。
- 細胞が、Bowesメラノーマ細胞、Daudi細胞、Hela細胞、HL-60細胞、HT1080細胞、Jurkat細胞、KB癌細胞、K-562白血病細胞、MCF-7乳癌細胞、MOLT-4細胞、Namalwa細胞、Raji細胞、RPMI 8226細胞、U-937細胞、WI-38VA13(サブ系統2R4)細胞、及び2780AD卵巣癌細胞からなる群より選択される請求項16記載のクローン細胞系。
- (b)ヒトα−gal Aにペプチド結合によって結合された(a)hGHシグナルペプチド(配列番号:21)を含む蛋白質。
- 前記DNA分子からヒトα−gal Aを発現させ、細胞の培養培地にグリコシル化ヒトα−gal Aが分泌されるような条件下で請求項11記載の細胞を培養する段階と、
該培養培地からグリコシル化ヒトα−gal Aを単離する段階とを含むグリコシル化ヒトα−gal Aを製造する方法。 - 請求項19記載の方法によって製造された精製グリコシル化ヒトα−gal A。
- サンプルが疎水性の相互作用樹脂を通過する第1クロマトグラフィー工程を含む、サンプルからヒトα−gal Aを精製する方法。
- 前記樹脂上の機能的部分がブチル基を含む請求項21記載の方法。
- 前記DNA分子からヒトα−gal Aを発現させ細胞の培養培地にグリコシル化ヒトα−gal Aが分泌されるような条件下で請求項16記載の細胞を培養する段階と、
前記培養培地からグリコシル化ヒトα−gal Aを単離する段階とを含む、グリコシル化ヒトα−gal Aを製造する方法。 - 固定化ヘパリン樹脂、ハイドロキシアパタイト、アニオン交換樹脂、及びサイズ排除樹脂からなる群より選択される第2樹脂にサンプルを通す段階をさらに含む請求項21記載の方法。
- α−gal A欠損症であると推測される患者を同定する段階と、
請求項20記載の精製グリコシル化ヒトα−gal Aを該患者に投与する段階とを含む治療方法。 - 薬学的に許容されるキャリア中にN−結合マンノース−6−ホスフェートを有する薬学的に許容される精製ヒトα−gal Aを含む治療用組成物。
- 請求項20記載の精製ヒトα−gal Aと薬学的に許容される賦形剤とを含む治療用組成物。
- pH6.5またはそれ以下で処方された請求項27記載の治療用組成物。
- 薬学的に許容される賦形剤がヒト血清アルブミンである請求項28記載の治療用組成物。
- hGH以外のポリペプチドに結合されたhGH(配列番号:21)のシグナルペプチドを含む蛋白質をコードするDNA分子。
- シグナルペプチドをコードする配列にイントロンを含む請求項30記載のDNA分子。
- (a)ヒトα−gal Aを含むポリペプチドをコードし、かつ(b)細胞における該ポリペプチドの過剰発現を可能にするDNA分子を含む培養されたヒト細胞。
- 前記DNA分子からヒトα−gal Aが発現されるような条件下で、ヒトα−gal Aを含むポリペプチドをコードするDNA分子を含有するヒト細胞を培養する段階を含む、グリコシル化ヒトα−gal Aを製造する方法。
- 薬学的に許容される賦形剤をさらに含む請求項26記載の治療用組成物。
- 機能的部分としてブチル基を含む疎水性相互作用樹脂に前記サンプルを通す段階を含む、サンプルからヒトα−gal Aを精製する方法。
