Abstract
Description
プロテインキナーゼは、細胞増殖を制御することが示されてきた。プロテインキナーゼの阻害が、増殖性疾患特に癌の新しい治療剤の開発の可能性を有する研究領域として浮かび上がってきている。
1.この疾患の分子的基礎
慢性骨髄性白血病(CML)は、特異的な染色体異常(即ち、フィラデルフィア又はPh1染色体)と関連付けらた最初の疾患である。分子レベルにおいて、最も顕著な特徴は、プロトオンコジーンc−ablの、第9染色体長腕から第22染色体上のブレークポイントクラスター領域(bcr)への転座であり、これは、bcr−ablハイブリッド遺伝子の形成を生じる。c−ablプロトオンコジーンは、通常、チロシンキナーゼ活性を有するタンパク質(ABL)をコードする。bcr−ablハイブリッド遺伝子を有する細胞において、オンコジーンBCR−ABLが発現される。BCR−ABLのチロシンキナーゼ活性は、ABLのチロシンキナーゼ活性と比較して相当増大されている。
イマチニブ耐性は、キナーゼのイマチニブ結合部位における突然変異から生じうる。突然変異は、いわゆる「ゲートキーパー」残基、Thr315及び他のイマチニブと結合中に接触する残基(例えば、Phe317及びPhe359)を含むキナーゼの触媒ドメインで起こりうる。他の変異は、ATP結合ドメイン(p−ループ)及び活性化ループ(イマチニブ結合に際して起きるコンホメーション変化に関与するキナーゼ構造の領域)において起こる。
血小板由来成長因子レセプター(PDGFR)は、別のチロシンキナーゼである。BCR−ABL癌タンパク質を生成する染色体転座と類似の分子機構が、イマチニブ感受性のPDGFR融合タンパク質PDGFRα(SEQ ID NO:2)及びPDGFRβ(SEQ ID NO:3)を生じることが示されている。PDGFRα及びPDGFRβは、特発性好酸球増加症候群及び慢性骨髄性単球性白血病にそれぞれ関係している(Apperley等、N. Engl. J. Med., 347, 481-487 (2002), 及び Cools等、N. Engl. J. Med, 348, 1201-14 (2003))。好酸球増加症候群において、好酸球の過剰増殖は、しばしば、FIP1L1とPDGFRαとの融合から生じるキメラのFip1様(FIP1L1)−PDGFRタンパク質の構成的チロシンキナーゼ活性から生じる。
胃及び小腸の間充織腫瘍である胃腸間質腫瘍(GIST)は、KITチロシンキナーゼレセプター(SEQ ID NO:4)を発現する。GIST細胞は、しばしば、KITキナーゼドメイン中に、構成的キナーゼへと導く活性化点突然変異を含む。マウスにおける活性化KIT変異の発現は、ヒトのGISTに類似の腫瘍を引き起こすことが示された(Somsmer等、Proc.Natl.Acad.Sci., 100, 6706-11 (2003))。c−kit発現は又、小脳の高度に侵襲性の未分化神経外胚葉性腫瘍であり、最も一般的な子供の悪性の中枢神経系の腫瘍である髄芽細胞腫にも関連している。10の髄芽細胞腫の腫瘍試料の研究において、すべては、c−kitを発現した(Chilton-Macneill等、Pediatr. Dev. Pathol., 7, 493-8 (2004))。c−kitは又、多くの子宮平滑筋肉腫において発現されることも示されている(Raspollini等、Clin. Cancer Res., 10, 3500-3 (2004))。
上皮成長因子レセプター(EGFR、SEQ ID NO:5)は、他のチロシンキナーゼである。ゲフィチニブ(EGFRインヒビター)は、確立された化学療法養生法に応答しない進行した非小細胞肺癌(NSCLC)の治療につき承認されている(Muhsin等、Nature Rev. Drug Discov., 2, 515-516 (2003)及びCohen等、Clin. Cancer Res., 10, 1212-18 (2004))。イマチニブ不従順T315I BCR−ABL変異体へと導くCからTへの単一ヌクレオチドの変異は又、対応するゲートキーパー残基Thr766をメチオニン残基で置き換えること(T766M)によりゲフィニチブに対してEGFRを劇的に不安定化させる(Blenke等、Chem. Biol., 11, 691-791 (2004)及びBlenke等、J. Biol. Chem, 278, 15435-40 (2003))。
ピリド[2,3−d]ピリミジン例えばPD180970(表1)は、BCR−ABL活性化ループ変異体例えばH396Pの癌促進活性を抑制することが示されている(La Posee等、Cancer Res., 62, 7149-53 (2002)及びvon Bubnoff等、Cancer Res., 9, 1267-73 (2003))。これらの化合物は又、臨床的に一般的なTyr253又はGlu255と関係するp−ループ変異を含むBCR−ABLをも阻害する。PD180970は又、M351T変異体(この変異は、イマチニブ耐性CMLの症例の約15〜20%を説明する)を含むBCR−ABLを阻害することも示されている(Kantarjian等、Blood, 101, 473-475 (2003))。
ある種のα,β−不飽和スルホン、スルホキシド及びスルホンアミド、特に、スチリルベンジルスルホン、スチリルベンジルスルホキシド及びスチリルスルホンアミドは、抗増殖性、放射線防護的及び化学防護的活性を有することが示されている。米国特許第6,599,932号、6,576,673号、6,548,553号、6,541,475号、6,486,210号、6,414,034号、6,359,013号、6,201,154号、6,656,973号及び6,762,207号を参照されたい。しかしながら、これらのα,β−不飽和スルホン、スルホキシド及びスルホンアミドの何れも、ATP競合性キナーゼインヒビターを用いる治療に耐性のキナーゼ依存性の増殖性疾患の治療において活性を有することは報告されていない。
この発明の一具体例により、個人における、ATP競合性のキナーゼインヒビターを用いる治療に耐性の、キナーゼ依存性の増殖性疾患特に癌を治療する方法を提供する。この方法は、かかる治療の必要な個人に、有効量の少なくとも一種の下記式Iの化合物を投与することを含む:
Xは、N又はCHであり;
nは、1又は2であり;
Rは、−H又は−(C1〜C8)ヒドロカルビルであり;そして
*は、XがCHで且つRが−H以外の場合には、Xの炭素原子上の置換基のコンホメーションが、R−、S−又はR−とS−の任意の混合物であり;又はその塩であることを示し;

図1は、化合物1、イマチニブ又は塩溶液の、32D/BCR−ABLT315I細胞の、該細胞1×106を静脈注射した後の、ヌードマウスにおけるイン・ビボでの成長に対する効果のグラフである。データは、10フィールド当たりの32D/BCR−ABLT315I細胞の平均数±SEM(n=10)としてプロットしてある。
図2は、化合物1(100mg/kg)、イマチニブ(100mg/kg)又は塩溶液を投与された個々のマウスの体重のプロットである。これらの3投与グループの各マウスの体重は、毎日測定された。平均体重を、出発体重のパーセントとしてプロットした。
図3は、塩溶液又は塩溶液に溶解させた化合物1(100mg/kg)を静脈注射(尾静脈注射)したCD−1マウスにおける造血コロニー形成のグラフである。
図4(a)は、変化する濃度の化合物1又はイマチニブとの72時間のインキュベーション後に残っている生存可能なCML K562細胞のパーセントのプロットである。
図4(b)は、変化する濃度の化合物1又はイマチニブとの72時間のインキュベーション後の生存可能なマウス32Dcl3.BCR−ABL細胞のパーセントのプロットである。
図5(a)は、変化する濃度の化合物1又はイマチニブとの72時間のインキュベーション後に残っている生存可能なBCR−ABL T315L発現細胞のパーセントのプロットである。
図5(b)は、変化する濃度の化合物1又はイマチニブとの72時間のインキュベーション後に残っている生存可能なBCR−ABL E255K発現細胞のパーセントのプロットである。

図6は、BCR−ABLの野生型又はイマチニブ耐性型を発現するようにトランスフェクトされて化合物2で処理された32Dcl3細胞の成長のグラフである。
図7は、野生型BCR−ABL、又はBCR−ABL変異G250E、T315I又はM351Tを発現する生存可能な細胞の、変化する濃度の化合物4との72時間のインキュベーション後のパーセントのプロットである。
本発明によれば、ある種のα,β−不飽和スルホン スルホキシド及びスルホンアミド、又は製薬上許容しうるそれらの塩は、ATP競合性キナーゼインヒビターによる治療に耐性のキナーゼ依存性の増殖性疾患における標的細胞の増殖の阻止において有効である。
イマチニブで治療した慢性骨髄性白血病(CML)におけるBCR−ABLキナーゼドメイン内の突然変異は、CMLにおけるイマチニブ耐性の獲得の主要な機構である。同様に、他のキナーゼをコードする遺伝子のキナーゼドメイン内の突然変異例えばPDGFR、KIT、EGFR、SRC及びP38は、非変異キナーゼに対して抗増殖活性を有するプロテインキナーゼインヒビターに対する獲得された耐性の主たる機構として関係してきた。
これらの化合物は、経口投与及び非経口投与を含む任意の経路によって投与することができる。非経口投与には、例えば、静脈投与、筋肉内投与、動脈投与、腹腔内投与、鼻内投与、直腸投与、膣内投与、嚢(例えば、膀胱)内投与、皮内投与、局所投与又は皮下投与が含まれる。やはりこの発明の範囲内で企図されるのは、遅れて起きる薬物の全身又は局所的放出を用いる、制御された配合での患者の身体における薬物の点滴である。例えば、循環への又は腫瘍成長の局所部位への制御された放出のために薬物をデポー剤中に局在化させることができる。
式Iの化合物は、製薬上許容しうるキャリアーと組合わせた医薬組成物の形態で投与することができる。かかる配合物中の活性成分は、0.1〜99.99重量パーセントを含むことができる。「製薬上許容しうるキャリアー」とは、配合物の他の成分と適合性で且つレシピエントに有害でないの任意のキャリアー、希釈剤又は賦形剤を意味する。
式Iの化合物は、塩の形態をとりうる。用語「塩」は、この発明の化合物である遊離酸又は遊離塩基の付加塩を包含する。用語「製薬上許容しうる塩」は、医薬適用において有用性を与える範囲内の毒性を有する塩を指す。
α,β−不飽和スルホンD1(式中、Ar1、Ar2、R及び*は、式Iの化合物について定義した通りである)を、芳香族アルデヒドのベンジルスルホニル酢酸C1とのクネーベナーゲル縮合によって製造することができる。その手順は、Reddy等、Acta. Chim. Hung. 115:269-71 (1984);Reddy等、Sulfur Letters 13:83-90 (1991);Reddy等、Synthesis No. 4, 322-323 (1984);及びReddy等、Sulfur Letters 7:43-48 (1987)により記載されており、これらの全開示を、参考として本明細書中に援用する。クネーベナーゲル縮合に従う一般的合成を、下記のスキーム3に描いた。
A.32Dcl3細胞株
32Dcl3細胞(Rovera等、Oncogene, 1, 29-35 (1987))を、10% FBS、1U/mL ペニシリン−ストレプトマイシン及び10% WEHI−3B調整培地(IL−3源として)(Ymer等、Nature, 317, 255-258 (1985))を補ったIscove改変ダルベッコ培地中に維持した。
イマチニブ耐性と関係する公開されたbcr−abl突然変異(Shah等、Oncogene 22, 7389-85 (2003))に対応するオリゴヌクレオチドを利用して、これらの変異を、完全長のbcr−abl cDNA中に、PCRベースの位置指定突然変異導入法(Myers等、PCR Technology, Erlich, H.A.編、Stockton Press, London (1989))を用いて導入した。すべての構築物を、配列分析により確認した。野生型及びイマチニブ耐性型のBCR−ABLをコードするpCDNA3ベースの発現プラスミドを、活発に増殖している32Dcl3細胞に、エレクトロポレーションによって以前に記載された(Kumar等、Mol. Cell. Biol., 23, 6631-45 (2003))ようにして導入して、細胞をIL−3の非存在下で選択した。BCR−ABLタンパク質の発現を、下記のように、ウエスタンブロット及びキナーゼアッセイにより測定した。
雌の無胸腺ヌードマウス(ner/ner)に、1×106の変異型BCR−ABLT315Iを発現する32Dcl3細胞を尾静脈から静脈注射した。治療(投薬群ごとに10匹のマウスよりなる3群)を、細胞注射の24時間後に、塩溶液(ビヒクル)、化合物1(100mg/kg)又はイマチニブ(100mg/kg)の日々の腹腔内注射によって開始した。イマチニブ注射を、毒性のために、10日後に停止した。
試験動物の体重を、2日ごとに測定した。この体重データを、図2に示してある。化合物1で治療したマウスは、毒性の徴候例えば体重減少、もつれた毛、嗜眠又は異常な糞を示さなかった。定常的に、10日にわたるイマチニブの投与は、20%を超える体重の減少から判断して重い毒性を生じ、これは、薬物投与の停止を生じた。
治療したマウスにおける血液塗抹標本作成を、治療開始後7日及び14日に、行なった。各マウスからの一滴の血液を、スライド硝子上に塗って、風乾し且つ改変Wrightステイン(Sigma)でしみを付けた。血液中のT315I細胞は、それらのブルー染色及びサイズの差異のために容易に見ることができた。10フィールド当たりのT3151の数を、オリンパス製の直立顕微鏡で同密度の赤血球を含む10視野を40倍対物レンズを用いて計数することにより測定した。この研究におけるT315I細胞のイン・ビボでの成長を図1に示した。化合物1で治療したマウスの血液中のT315I細胞の数は、7日目及び14日目において、ビヒクル及びイマチニブ処理した群において見出された細胞数と比較して有意に減少した。
イン・ビボで正常な造血細胞集団に対する化合物1の効果を調べるために、この化合物の正常塩溶液(又は対照としての塩溶液のみ)中の100mg/kgの投与量を、CD−1マウス(1群当たり10匹)に静脈注射(尾静脈注射)した。これらのマウスに由来する正常骨髄細胞のイン・ビトロでの造血コロニー形成に対する効果を、投与の24時間後に測定した。24時間後に骨髄細胞をマウスから抽出して、2×105細胞を、系統特異的なコロニー形成に適当なサイトカインを含むメチルセルロース上にプレートした。5〜14日のインキュベーションの後にコロニーを計数した。骨髄コロニー形成アッセイについてのデータは、図3に示してある。赤血球、骨髄又はリンパ系統のコロニー形成には、化合物1の投与による減少はない。
A.細胞株
K562細胞を、10% FBS及び1U/ml ペニシリン−ストレプトマイシンを補ったRPMI(Roswell Park Memorial Institute)培地中に維持した。
K562細胞及びマウス32Dcl3.BCR−ABL細胞(野生型p210BCR-ABL癌タンパク質を異所的に発現)を、変化する濃度の化合物1と共に72時間にわたってインキュベートした。次いで、細胞生存力を、トリパンブルー排除により測定した。生存可能な細胞の、ビヒクルで処理した対照と比較したパーセントを測定した。両細胞株において、化合物1とのインキュベーションは、図4a及び4bに示したように、10〜15nMのLD50での、生存力の急速な喪失を生じた。この細胞生存力アッセイを、イマチニブを用いても実施した。イマチニブのIC50は、100nMより大きいことが見出された(図4a及び4b)。このイマチニブについてのデータは、公表されたデータ(O'Dwyer、同書)と一致する。
文献(Shah等、Oncogene, 22, 7389-7395 (2003))に公開されたものに対応する突然変異を、bcr−abl cDNA中に導入してから、活発に増殖している32Dcl3細胞(Rovera等、Oncogene 1, 29-35 (1987))中に下記の手順に従って導入した。
表2に列記した細胞株を、250nM 化合物2又はイマチニブの存在下で培養した。その結果を、図6に示した。これらの細胞株のすべては、化合物2の成長阻害活性に敏感であった。
慢性骨髄性白血病と診断されたある成人の患者は、イマチニブ耐性であることが見出されている。白血病細胞により発現されるbcr−abl遺伝子の配列決定は、そのbcr−ablキナーゼドメインにおける突然変異の存在を確認する。この患者の病気は、慢性期にある(血液又は骨髄試料において、10%未満の芽細胞、又は合わせて20%の芽細胞及び前骨髄細胞)。この患者を、次の投与養生法を用いて、化合物1で治療する:200ミリグラムの化合物1を毎日一回経口投与する。治療を、病気の進行の証拠又は許容しえない毒性のない限り継続する。病気の促進期への進行の出現(骨髄又は血液試料が、10%より多くの芽細胞を有する(又は合わせて20%の芽細胞及び前骨髄細胞)が、30%未満の芽細胞及び前骨髄細胞)の場合には、化合物1の毎日の投与量を、300ミリグラムに増加させる。もし必要であれば、更なる投与量の増加を(許容しえない毒性が現れなければ)行なって、所望の治療結果を達成する。

慢性骨髄性白血病と診断されたある成人の患者は、先ず、イマチニブで治療される。時間が経つと、この患者の病気は、イマチニブ耐性となる。白血病細胞により発現されるbcr−abl遺伝子の配列決定は、そのbcr−ablキナーゼドメイン内の突然変異の存在を確認する。この患者の治療養生法は、次の組合せ養生法に変えられる:イマチニブを、患者が許容する最大投与量まで増加させて、毎日2回、400ミリグラムの投与量で投与する。300ミリグラムの投与量の化合物1も、毎日一回、経口投与する。治療を、病気の進行の証拠又は許容しえない毒性のない限り継続する。病気の進行が現れた場合には、化合物1の量を、所望の治療結果を達成するのに必要なだけ増大させる。
ある成人の患者が、新たに、慢性骨髄性白血病と診断される。イマチニブへの感受性は、未知である。第一線の治療として、この患者には、イマチニブと化合物1の組合せを、次の投与養生法を用いて投与する:400ミリグラムのイマチニブと100ミリグラムの化合物1を含む一日一回の経口投与を施与する。治療を、病気の進行の証拠又は許容しえない毒性のない限り継続する。病気の進行が現れた場合には、イマチニブの投与量を、600ミリグラムに増加させて、化合物1の投与量を、200ミリグラムまで増加させる。もし必要であれば、イマチニブ(最大800ミリグラム)及び化合物1の一方又は両方において、更なる投与量の増加を(許容しえない毒性が現れなければ)行なう。
表3に列記した細胞株を、様々な濃度の化合物3((E)−2−(5−((2,4,6−トリメトキシスチリルスルホニル)メチル)−2−メトキシフェニルアミノ)−2−メチルプロパン酸ナトリウム塩)の存在下で培養した。この野生型は、野生型p210BCR-ABL癌タンパク質を異所的に発現した32Dcl3.BCR−ABL細胞を含む。生存可能な細胞の総数を、処理後72時間で、トリパンブルー排除により測定した。ビヒクルで処理した対照と比較した生存可能な細胞のパーセントを測定した。これらの細胞株の、GI50で表した感受性を表3に列記した。これらの細胞株のすべては、化合物3の成長阻害活性に対して感受性であった。
野生型p210BCR-ABL癌タンパク質(32Dcl3.BCR−ABL)又はBCR−ABL変異G250E、T315I又はM315Tを発現する細胞を、変化する濃度の化合物4((E)−1−(2−(4−ブロモベンジルスルホニル)ビニル)−2,4−ジフルオロベンゼン)と共にインキュベートした。生存可能な細胞の総数を、処理後72時間で、トリパンブルー排除により測定した。ビヒクルで処理した対照と比較した生存可能細胞のパーセントを、測定した。その結果を、図7に示した。これらの細胞株のすべては、化合物4の成長阻害活性に感受性であった。
Claims (32)
- 個人における、ATP競合性のキナーゼインヒビターを用いる治療に耐性のキナーゼ依存性の増殖性疾患特に癌を治療する方法であって、該方法は、かかる治療の必要な個人に、有効量の少なくとも一種の下記式Iの化合物を投与することを含む当該方法:
Xは、N又はCHであり;
nは、1又は2であり;
Rは、−H又は−(C1〜C8)ヒドロカルビルであり;そして
*は、XがCHで且つRが−H以外の場合には、Xの炭素原子上の置換基のコンホメーションが、R−、S−又はR−とS−の任意の混合物であり;又は製薬上許容しうるその塩であることを示し;
但し、XがNである場合には、nは2である)。 - 置換されたアリール及び置換されたヘテロアリールAr1上の置換基を、独立に、ハロゲン、−(C1−C8)ヒドロカルビル、−C(=O)R2、−NR2 2、−NHC(=O)R3、−NHSO2R3、−NH(C2−C6)アルキレン−C(=O)R6、−NHCR2R4C(=O)R6、−C(=O)OR2、−C(=O)NR2 2、−NO2、−CN、−OR2、−OC(=O)R3、−OSO2R3、−O(C2−C6)アルキレン−C(=O)R6、−OCR2R4C(=O)R6、−P(=O)(OR2)2、−OP(=O)(OR2)2、−O(C2−C6アルキレン)N(C1−C3)アルキル)2、−NHC(=NH)NHR2、−(C1−C6)ハロアルキル、−O(C1−C6)ハロアルキル;−SO2NH2;及び−N=CH−R7よりなる群から選択し;そして
置換されたアリール及び置換されたヘテロアリールAr2上の置換基を、独立に、−(C1−C8)ヒドロカルビル、−C(=O)R2、ハロゲン、−NO2、−CN、−OR2、−C(=O)OR2、−NR2 2、−(C1−C6)ハロアルキル;−SO2NH2;及び−O(C1−C6)ハロアルキルよりなる群から選択する、請求項1に記載の方法

各R3は、独立に、−(C1−C8)ヒドロカルビル、−O(C1−C8)ヒドロカルビル、置換された及びされてないアリール、置換された及びされてないヘテロシクリル(C1−C3)アルキル、置換された及びされてないヘテロアリール(C1−C3)アルキル、−(C2−C10)へテロアルキル、−(C1−C6)ハロアルキル、−CR2R4NHR5、−NR2 2、−(C1−C3)アルキレンNH2、−(C1−C3)アルキレンN((C1−C3)アルキル)2、−(C1−C3)ペルフルオロアルキレン−N((C1−C3)アルキル)2、−(C1−C3)アルキレン−N+((C1−C3)アルキル)3、−(C1−C3)アルキレン−N+(CH2CH2OH)3、−(C1−C3)アルキレン−OR2、−(C1−C4)アルキレン−CO2R2、−(C1−C4)アルキレン−C(=O)ハロゲン、ハロ(C1−C3)アルキル−、−(C1−C3)アルキレン−C(=O)(C1−C3)アルキル、及び−(C1−C4)ペルフルオロアルキレン−CO2R2よりなる群から選択され;
各R4は、独立に、−H、−(C1−C6)アルキル、−(CH2)3−NH−C(NH2)(=NH)、−CH2C(=O)NH2、−CH2CO2R2、−CH2SH、−(CH2)C(=O)−NH2、−(CH2)CO2R2、−CH2−(2−イミダゾリル)、−(CH2)4−NH2、−(CH2)2−S−CH3、フェニル、−CH2−フェニル、−CH2−OH、−CH(OH)−CH3、−CH2−(3−インドリル)、及び−CH2−(4−ヒドロキシフェニル)よりなる群から選択され;

各R6は、独立に、−OR2、−NR2 2、及び1〜3アミノ酸を含むアミノ末端結合したペプチジル残基よりなる群から選択され、該ペプチジル残基の末端カルボキシル基は、−CO2R2及び−C(=O)NR2 2よりなる群から選択する官能基として存在し;そして
各R7は、独立に、置換された及びされてないアリール及び置換された及びされてないヘテロアリールよりなる群から選択され;
但し、Ar1及びAr2上の置換基の最大数は、それらの置換基が結合している環内の置換可能な水素原子の数に等しい)。 - 式1に従う化合物を、(E)−2−(5−((2,4,6−トリメトキシスチリルスルホニル)メチル)−2−メトキシフェニルアミノ)酢酸;(ラセミ)−(E)−2−(5−((2,4,6−トリメトキシスチリルスルホニル)メチル)−2−メトキシフェニルアミノ)プロパン酸;(R)−(E)−2−(5−((2,4,6−トリメトキシスチリルスルホニル)メチル)−2−メトキシフェニルアミノ)プロパン酸;(S)−(E)−2−(5−((2,4,6−トリメトキシスチリルスルホニル)メチル)−2−メトキシフェニルアミノ)プロパン酸;(E)−2−(5−((2,4,6−トリメトキシスチリルスルフィニル)メチル)−2−メトキシフェニルアミノ)酢酸;(E)−2−(5−((2,4,6−トリメトキシスチリルスルフィニル)メチル)−2−メトキシフェニルアミノ)プロパン酸;(E)−2,4,6−トリメトキシスチリル−N−(3−カルボキシメチルアミノ−4−メトキシ−フェニル)スルホンアミド;(S)−(E)−2−(5−((2,4,6−トリメトキシスチリルスルホニル)メチル)−2−メトキシフェニルアミノ)−2−フェニル酢酸;(R)−(E)−2−(5−((2,4,6−トリメトキシスチリルスルホニル)メチル)−2−メトキシフェニルアミノ)−2−フェニル酢酸;(ラセミ)−(E)−2−(5−((2,4,6−トリメトキシスチリルスルホニル)メチル)−2−メトキシフェニルアミノ)−2−フェニル酢酸;及び製薬上許容しうるこれらの塩よりなる群から選択する、請求項2に記載の方法。
- 式Iに従う化合物を、(E)−2−(5−((2,4,6−トリメトキシスチリルスルホニル)メチル)−2−メトキシフェニルアミノ)−2−メチルプロパン酸;(E)−1−(2−(4−ブロモベンジルスルホニル)ビニル)−2,4−ジフルオロベンゼン;及び製薬上許容しうるこれらの塩よりなる群から選択する、請求項2に記載の方法。
- 式Iに従う化合物が、反対のエナンチオマーを実質的に含まない単離された光学異性体を含む、請求項2に記載の方法。
- 単離された光学異性体が、*により示された原子における(R)−絶対配置を有し、実質的に(S)−エナンチオマーを含まない、請求項5に記載の方法。
- 単離された光学異性体が、*により示された原子における(S)−絶対配置を有し、実質的に(R)−エナンチオマーを含まない、請求項5に記載の方法。
- キナーゼ依存性の増殖性疾患が、PD180970、BMS−354825、イマチニブ、SU5416、SU6668、SU11248、AP23464、ゲフィチニブ、エルロチニブ、PD153035、及びSB203580よりなる群から選択する少なくとも一つの化合物による治療に耐性である、請求項1に記載の方法。
- 増殖性疾患が、BCR−ABLのATP競合性インヒビターに耐性である、請求項1に記載の方法。

- 突然変異が、His396の変化を含む、請求項14に記載の方法。
- 突然変異が、F317L、H396R、M351T、H396P、Y253H、M244V、E355G、F359V、G250E、Y253F、F311L、T315I、E255V、Q252H、L387M、及びE255Kよりなる群から選択する少なくとも一つの変異を含む、請求項10に記載の方法。
- 増殖性疾患を、慢性骨髄性白血病、急性リンパ芽球性リンパ腫、特発性肺線維症、特発性好酸球増加症候群、慢性骨髄性単球性白血病、悪性線維性組織球腫、前立腺癌、アンドロゲン依存性前立腺癌、皮膚線維肉腫、子宮内膜癌、子宮乳頭漿液性癌、脊索腫、グリオーマ、悪性星状細胞腫、グリア芽細胞腫細胞、胃腸間質腫瘍、髄芽細胞腫、子宮平滑筋肉腫、及び非小細胞肺癌よりなる群から選択する、請求項1に記載の方法。
- 増殖性疾患が、KITのATP競合性インヒビターに耐性である、請求項1に記載の方法。
- ATP競合性KITインヒビターに対する耐性が、KITの少なくとも一つのアミノ酸残基の突然変異から生じる、請求項18に記載の方法。
- 突然変異を、T670I、Y823D及びD816Vよりなる群から選択する、請求項19に記載の方法。
- 増殖性疾患が、上皮成長因子レセプターのATP競合性インヒビターに耐性である、請求項1に記載の方法。
