Description
本発明は、免疫療法的な組成物および方法に関し、また、改良された予防用および治療用のワクチン、および、抗原に対する免疫応答を予防および/または治療に導入するための改良された方法に関する。

本明細書において、”免疫調節タンパク質”という用語はヒトIL-12、GM-CSF、IL-1、TNF-α、TNF-β、IL-2、IL-15、IL-18、IL-4、IL-5およびIL-10およびBL-1と名付けられた新規の分子のうちの一つについて言及することを意味する。
本発明の側面は、顆粒球-マクロファージコロニー刺激因子(GM-CSF)をコードする核酸分子の使用を含む。GM-CSFは好中球、単核球/マクロファージ、および好エオシン球のコロニー形成を刺激する造血性増殖因子である。これは、赤血球および巨核球前駆細胞の分化も誘導する。GM-CSFは、好中球、好エオシン球およびマクロファージの抗体依存性の細胞を介した細胞傷害性も増大させるが、in vivoでのCTL応答の形成において直接的な役割を果たしているとは報告されていなかった。本発明は個体へのGM-CSFをコードする核酸配列を含む遺伝子構築物の投与により免疫応答を引き起こす組成物および方法を提供する。核酸分子は、細胞により取り込まれ、GM-CSFをコードする核酸配列が発現し、即ち細胞により生産される。GM-CSFは生物学的に活性があり、その活性は個体により形成される免疫応答の誘導および/または増強を引き起こす。いくつかの好ましい態様においてはGM-CSFをコードする核酸分子はプラスミドである。
本発明のいくつかの態様によれば、IL-12のそれぞれのサブユニットをコードするヌクレオチド配列は単一のプラスミド、非プラスミド核酸分子、または、それぞれのサブユニットをコードするヌクレオチド配列がそれ自身の調節因子群の制御下にあるウィルスまたは微生物ゲノム上に存在し得る。いくつかの好ましい態様においては、IL-12の両方のサブユニットをコードする配列は、それぞれのコード配列がそれ自身の調節因子群に作動可能なように連結されて、単一のプラスミド上に存在する。いくつかの態様においては、作動可能なように調節因子に連結された標的免疫原性タンパク質をコードする配列は、両方のサブユニットをコードする配列を含むプラスミド由来の別々のプラスミド上に存在し、二つのプラスミドが単一個体に投与される。
発現可能な状態のIL-12タンパク質をコードするヌクレオチド配列を提供することで、DNAワクチン、組換えベクターワクチンおよび弱毒化ワクチンのような、ワクチン投与された個体の細胞において標的抗原を発現することで機能するワクチンが、改善される。
本発明のいくつかの側面は、IL-2、IL-5、IL-10、IL-18、TNF-αまたはTNF-βを同時に投与することによる、Tヘルパー細胞の抗原特異的な増殖の増大を提供する。
即ち、T細胞を介した応答が優勢だが、体液性応答は必要ではなく有害でさえある場合、特異的なDNA免疫原と共に投与される免疫調節物質としてIL-12遺伝子が好ましい。一方、細胞外の細菌を標的とするワクチンの構築のためには、例えばIL-4、IL-5またはIL-10遺伝子がともに注入され得る。さらに、CD4+ Tヘルパー細胞および抗体が防御においてより重要な役割を果たす場合は、IL-12およびGM-CSFが共に投与され得る。最後に、3系統の免疫応答が全て決定的である場合は、抗体、Tヘルパー細胞およびCTL応答の組み合わさった増幅を起こすために、TNF-αが共に注入され得る。
本発明は遺伝物質によりコードされる、タンパク質及びペプチドに対する免疫応答を誘導するために、個体の細胞に遺伝物質を導入する方法に関連する。その方法は、標的タンパク質をコードするヌクレオチド配列および免疫調節タンパク質をコードするヌクレオチド配列を含む単一の核酸分子、または標的タンパク質をコードするヌクレオチド配列を含むものおよび免疫調節タンパク質をコードするヌクレオチド配列を含むものの、二つの核酸分子を含む組成物の、前述の個体の組織への投与の段階を含む。核酸分子はプラスミドDNA、組換えベクターの核酸分子または弱毒化ワクチン中の遺伝物質の一部として提供され得る。
本発明は標的タンパク質、即ち病原または個体自身の"異常な"細胞に特異的に関連するタンパク質、に対する広範な免疫応答を引き起こすために有用である。
本発明は過剰増殖細胞に特異的に関連する標的タンパク質に対する免疫応答を引き起こすことにより、癌のような過剰増殖性の疾患および損傷を排除するために有用である。本発明は自己免疫状態に関与する細胞に特異的に関連する標的タンパク質に対する免疫応答を引き起こすことにより、自己免疫疾患および損傷を排除するために有用である。
本発明によれば、標的タンパク質をコードする、発現可能な形のヌクレオチド配列を含む遺伝子構築物、および免疫調節タンパク質をコードする、発現可能な形のヌクレオチド配列を含む遺伝子構築物の組み合わせが提供される。生きた細胞への、遺伝子構築物の組み合わせを含むDNAまたはRNA分子の導入はDNAまたはRNAの発現および標的タンパク質および免疫調節タンパク質の生産をもたらす。標的タンパク質に対する増幅された免疫応答が起きる。
特にヒトのための遺伝子ワクチンの生産において本発明を実行するために有用なプロモーターの例は、シミアンウィルス40(SV40)、マウス乳ガンウィルス(MMTV)プロモーター、HIV末端長反復プロモーター(LTR)のようなヒト免疫不全症候群ウィルス(HIV)プロモーター、CMV最初期プロモーターのようなCMVプロモーター、エプシュタインバーウィルス(EBV)、ルース腫瘍ウィルス(RSV)および、ヒトアクチン、ヒトミオシン、ヒトヘモグロビン、ヒト筋クレアチンおよびヒトメタロチオネインのようなヒトの遺伝子由来のプロモーターを含むが、それらに限定されない。

ブピバカイン-HClは化学的にデザインされて、2-ピペリジンカルボキシアミド、1-ブチル-N-(2,6-ジメチルフェニル)-一塩酸塩、一水塩を生じ、薬学用途にAstra Pharmaceutical Products Inc.(Westboro,MA)およびSanofi Winthrop Pharmaceuticals(New York,NY)、Eastman Kodak(Rochester,NY)を含む多くの供給源から商業的に入手可能である。ブピバカインは商業的にメチルパラベンとともにおよびそれ無しに、およびエピネフリンとともにまたはそれ無しに製剤される。任意のそのような製剤が用いられ得る。0.25%、0.5%および0.75%の濃度で製剤のための用途で商業的に入手可能であり、それらは本発明に用いられ得る。望ましい効果を引き起こす代替的な濃度、特に0.05%-1.0%の間の濃度が、望ましければ調製され得る。本発明によれば、約250gから約10mgのブピバカインが投与される。いくつかの態様においては、約0.05mgから約5.0mgが投与される。いくつかの態様においては、約0.5mgから約3.0mgが投与される。いくつかの態様においては、5から50gが投与される。例えばいくつかの態様においては約50μlから約2mlまでの、好ましくは50μlから約1500μl、およびより好ましくは約1mlの0.25%-0.50%の等張の製剤学的担体中のブピバカイン-HClおよび0.1%のメチルパラベンがワクチンと同一の部位に、ワクチンの投与の前、同時、またはその後に投与される。同様に、いくつかの態様においては約50μlから約2mlの、好ましくは50μlから約1500μlの、およびさらに好ましくは約1mlの0.25-0.50%の、等張の製剤学的担体中のブピバカイン-HClがワクチンと同一の部位に、ワクチンの投与の前、同時またはその後に、投与される。ブピバカインおよびその他の任意の同様な働きを持つ化合物、特に局所麻酔剤に関連する種類のものが細胞による遺伝子構築物の取り込みの望ましい促進を提供する濃度において投与され得る。
ここで用いられる場合、「過剰増殖性疾病」とは、細胞の過剰増殖によって特徴づけられる疾病および障害を指す。過剰増殖性疾病の例は癌および乾癬の全ての形を含む。
本発明の方法はヒトおよび獣医学の両方の分野において有用である。それゆえ、本発明は、哺乳類、鳥類および魚類の遺伝学的免疫化に関する。本発明の方法は、特にヒト、ウシ、ヒツジ、ブタ、ウマ、イヌ、およびネコの各種を含む哺乳類に有用である。
新しい免疫改変剤であるヒトBL1がすでに発見されている。DNAおよび予測されるアミノ酸配列は図14に述べられる。このタンパク質およびその断片は個体に免疫原を導入し得るワクチン組成物と共に投与される際、免疫応答を強化させる。
BL1をコードする単離されたcDNAは、BL1またはその免疫改変断片を生産し得る組換え発現ベクターの製造における出発材料として有用である。cDNAは発現ベクターを含むベクターに組込まれ、そのベクターは宿主細胞に導入され、宿主細胞は組換えで生じたタンパク質を発現する。核酸分子およびその断片は、プローブとして、BL1コーディング配列の存在を検出するのに用いられ得る。このようなプローブはBL1のコーディング配列に特異的にハイブリッド形成する。ここで使用される場合、「特異的BL1配列」の語は、BL1に固有の配列を指す。BL1をコードする特異的cDNA断片に相補するヌクレオチド配列を含む核酸分子は、アンチセンス分子およびプライマーとして、各々mRNAの翻訳を阻害するため、および遺伝配列を増幅するために用いられ得る。
このようにして生産された本発明のタンパク質は、培地から、溶菌によって、または適当な、および当該技術分野で知られている培養培地から、回収される。通常の当業者は、よく知られた技術を用いて、このような発現系を用いて生産されるBL1を単離し得る。上に記載されたようなBL1に特異的に結合する抗体を用いて自然源からBL1を精製する方法は、同じように組換えDNA技術によって生産されたBL1を精製するのに応用され得る。
遺伝子構築物の例は、上に述べたようにヒトにおいて、または構築物がトランスフェクションされる細胞系において機能するプロモーターに作動可能なように連鎖したBL1コーディング配列を含む。構造性プロモーターの例は、巨細胞ウイルスまたはSV40由来のプロモーターを含む。導入可能なプロモーターの例は、マウス哺乳類白血病ウイルスまたはメタロチオネインプロモーターを含む。
本発明による薬学組成物は、核酸分子と組み合わされた輸送成分を含み、それらはさらに、薬学上受容可能な担体または媒体、例えば塩水を含む。核酸の首尾よい輸送を可能にするいかなる媒質も用い得る。通常の当業者は、本発明で使用しうる薬学上受容可能な多くの媒質を即座に理解するであろう。適当な薬学的担体は、この分野の標準的参照物であり、また本明細書に参照によって編入されるRemington's Pharmaceutical Science,A.Osolに記述されている。本発明の薬学組成物は、活性剤が標的細胞に到達することを可能にするいかなる方法によっても投与され得る。ペプチドは経口的に投与された場合消化されやすいため、吸収を効果的にするため、非経口的投与、すなわち、静脈注射、皮下注射、皮膚を通した投与、筋肉内注射が一般に用いられるであろう。静脈注射は、点滴ポンプに補助されて行われ得る。本発明の薬学組成物は、乳濁液として処方され得る。あるいはそのかわりに、それらの組成物は、鼻腔内または吸入法による投与のためのエーロゾル薬品として処方され得る。いくつかの場合では、局所的投与が望ましい。
実施例1
以下は、本発明の方法に用いられる構築物のリストである。下に記述された構築物の多くを生産するための出発材料として使用されるベクターpBabe.puroは、もともとは、参照により編入されているMorgenstern,J.P.and H.Land,1990Nucl.Acids Res.18(12):3587-3596により構築され報告された。pBabe.puroプラスミドは、特に哺乳類細胞での細胞膜外遺伝子の発現に役立つ。発現するべきDNA配列はMoloneyネズミ白血病ウイルス(MOMuLV)長末端反復(LTR)プロモーターの制御下でクローニング部位に挿入される。プラスミドはピューロマイシン耐性のための選択マーカーを含む。
nef領域を欠失させる。HIV標的タンパク質をコードするこれらのHIVウイルス遺伝子およびCMVプロモーターおよびSV40ポリA配列に作動可能なように連鎖したIL-12コーディング配列を含むプラスミドは、HIV感染およびAIDSに対する免疫化および治療に役立つ。HIV-1/3BのDNA配列は、参照により本明細書に編入されたFisher,A.,1985 Nature 316:262によって発表されている。この配列は参照によって本明細書に編入されたGenbank No.:K03455から入手可能である。
のL1キャプシッドタンパク質をコードするPCRで生産した断片、およびCMVプロモーターおよびSV40ポリA配列に作動可能なように連結されたIL-12コーディング配列を含むプラスミドである。このプラスミドはHPV感染およびそれによってひきおこされる癌に対する免疫化に有用である。DNA配列は参照により本明細書に編入されたGenebank No.:M15781に開示されている。Howley,1990 Fields Virology,Volume 2,Eds.:Channock,R.M.etal.Chapter 58:1625;および、Shar,K.andP.Howley,1990 Fields Virology,Volume 2,Eds.:Channock,R.M.et al.Chapter 59も参照のこと。これら両者は参照により本明細書に編入されている。
プラスミドpCSIL-12は、CMVプロモーターおよびSV40ポリA配列に作動可能なように連結されたIL-12コーディング配列を含む。
本発明のいくつかの態様に従った組成物は、プラスミドpCSIL-12を次のプラスミドのいずれかと組み合わせることにより調製し得る。
ワクチンを改良するために免疫応答を特異的に指示することが大切であろう。
この種類の免疫応答(Th1対Th2)は、感染性疾病、自己免疫疾病、およびアレルギーを含む様々な疾病モデルにおいて重要であることが報告されている。HIV感染性疾病に対する強く安定な細胞媒介免疫応答を誘導するために、免疫化と共に、免疫学的補助的手段(アジュバント)およびサイトカインなどの免疫改変剤の使用は細胞抗原反応を強化し、Th2からTh1タイプの抗原依存性免疫応答を指示し得るであろう。
生後6−8週のBalb/c雌マウスがHarlan Spraque Dawley,Inc.,(Indianapolis,Indiana)より購入された。このマウスは制御された温度、光で囲まれた部屋に入れられた。彼らの世話はNational Institute of Healthおよびペンシルバニア大学のガイドラインに従って行われる。
DNAワクチン処方pCMN160,pCGN160,pCGag/Polが調製された。IL-12およびGM-CSF遺伝子はクローニングされ、CMVプロモーターと共に発現ベクターに挿入された。組換えワクシニア(vMN462,vVK1,VV:gag,およびvSC8)はNIH AIDS Research and Reference Reagent Programより得られた。
生体内でプラスミド由来遺伝子からの増加された生体内タンパク質発現をひきおこす簡易化されたDNA接種プロトコルが使用された。特異的に、BALB/cマウスの大腿四頭筋が0.25%ブピバカイン-HCl(Sigma,St.Louis)を含む溶液100μlと共に27内径の針を用いて注射された。二日後に、リン酸塩緩衝液中の50gのDNA構築物がブピバカイン注射と同じ筋肉領域に注射された。様々な遺伝子発現カセットの共投与は、注射前の既定プラスミド混合を含んだ。
細胞(1×105)がFACS緩衝液(1% BSAおよび0.1%アジ化ナトリウムを含むPBS)で3回洗浄され、FITCおよび/またはPE結合モノクローン抗体とともに氷上で飽和条件で30分保温された。FACS緩衝液で3回洗浄された後、細胞はFACScan(Becton Dickinson)を用いて解析された。
0.1M炭酸-二炭酸緩衝液(pH9.5)中の2μg/mlのgp120またはgp41(Intracel Corp.,Cambridge,MA)が、以前に記載されたようにマイクロタイタープレートの穴に4℃で一晩吸着された。プレートはPBS-0.05% Tween-20で洗浄され、3% BSAを含むPBS-0.05% Tween-20で37℃で1時間ブロッキングされ、次に製造者推奨のように希釈されたHRP結合ヤギ抗マウスIgGまたはIgA(Sigma,ST Louis,MO)とともに保温された。プレートは洗浄され、TM blue緩衝液(Sigma)で発色された。450nm吸光が、Dynatech MR5000プレート読取装置で読みとられた。
回収されたマウス脾臓由来のリンパ球が調製された。単離細胞懸濁液は、1×106細胞/mLの濃度に再懸濁された。1×105細胞を含む100μL分画が直ちに96穴マイクロタイター平底プレートの各穴に加えられた。10μlのタンパク質が20μg/mLで3つ独立に穴に加えられた。細胞は5% CO2で37℃3日間保温された。1μCiのトリチウム化チミジンが各穴に加えられ、細胞は37℃で12-18時間保温された。プレートから細胞が回収され、取り込まれたチミジンがベータプレートリーダー(Wallac,Turku,Finland)で測定された。細胞が健康であることを確認するため、10g/mlのPHAがポリクローン刺激性の正の対照として用いられた。
5時間のクロミウム51放出CTL試験が行われた。脾臓からリンパ球が回収され、赤血球の除去、および新鮮な培地による数回の洗浄によってエフェクター細胞として調製された。3×106p815細胞を37℃16時間感染させることによって、ワクシニアに感染した標的が調製された。標的細胞は100μCi/mlのNa2 51CrO4で90分間標識され、刺激を受けた脾臓細胞を37℃で4-6時間保温するために用いられた。CTLは、エフェクター:標的(E:T)比50:1から12.5:1で試験された。上清が回収され、LKBガンマ計数器で計測された。特異的溶解の割合は、式: 100×(実験上の放出-自発放出)/(最大放出-自発放出)で決定された。最大放出は、1% Triton X-100を含む培地中での標的細胞の溶解によって決定された。自発放出の計数値が最大放出の20%以上である場合は、試験は無効とされた。
共接種の後、個々の実験集団から回収された脾臓は外見上異なることが観察された。従って、全ての免疫化された動物から回収された脾臓は計量され、視覚的に検査された。それらの動物の脾臓重量を図1Aに示す。対照の単一の調剤を注射されたマウスの脾臓が非免疫化対照マウスの脾臓と同様の重量(約100mg)だったのに対し、Gag/Pol+IL-12遺伝子を注射されたマウスの脾臓は対照の脾臓の3倍もの重量であった。一方、Gag/Pol+GM-CSFで免疫化されたマウスの脾臓は肥大していなかった。注目すべきは、Gag/Polのみ、またはIL-12のみで免疫化されたものの脾臓が有意に肥大しなかったことである。抗原およびIL-12遺伝子カセットが共注射された時にのみ脾臓肥大が生じることから、これが両方の遺伝子産物の複合効果であることが示唆される。さらに、図1Bに示すように、Gag/Pol+IL-12の脾臓由来のリンパ球数は、対照の脾臓由来の数の3倍以上である。ここでも、Gag/Polのみ、またはIL-12のみで免疫化されたマウスのリンパ球数は、対照の脾臓細胞数以上に有意に上昇することはなかった。代表的な脾臓の写真を図2に示す。
IL-12のp35鎖が多くの型の細胞で構成的に発現されていることが以前に報告されている。即ち、p40一本鎖およびDNA免疫原のみが、これらの効果の原因であり得る。これを調べるために、2つのIL-12ヘテロ二量体遺伝子(p35およびp40)の各々がGag/Polとともに共投与された。図3に示すように、どちらの場合にも脾臓の肥大は観察されなかった。これらのデータから、DNAワクチンおよびp35とp40 IL-12遺伝子との共注射によって、脾臓が肥大し、脾臓細胞数がそれに対応して増大することが示唆される。これらのデータにより、in vivoで複数のプラスミドが同一の細胞に入り、p35鎖、p40鎖および特異的な抗原の3組成物全ての転写が同調して起こり、観察された生物的変化をもたらしたことが支持される。
肥大した脾臓の細胞内組成の特性をさらに調べるために、FACS解析が行われた。表3に、CD3に対する抗体と、B220、CD4、およびCD8に対する抗体による脾臓細胞の二重染色によるFACSの結果を示す。示されたように、エンベロープ+IL-12またはGag/Pol+IL-12構築物で免疫化されたグループでは、対照の非免疫化脾臓中のB細胞の割合(25.43%)に比べて、B220陽性B細胞の割合が僅かに減少する(それぞれ17.46%、21.62%)ことが観察された。さらに、エンベロープ+IL-12またはGag/Pol+IL-12構築物で免疫化されたグループでは、対照の非免疫化グループ中のCD8+T細胞の割合(13.69%)に比べて、CD8+T細胞の割合がある程度上昇する(それぞれ21.72%、16.88%)ことが観察された。
免疫化されたマウス由来の抗血清が回収され、HIV-1抗原に対する特異的な抗体応答についてELISA解析された。図4に、免疫後28日目に回収された試料のELISAの結果を示す。1:100の希釈では、pCEnv+pCGM-CSFによる免疫化グループ由来の血清は、pCEnvのみによる免疫化グループよりも大きいHIV-1 gp120タンパク質に対する抗体応答を示した。一方、pCEnv+pCIL-12による免疫化グループは、同一の期間有意に低い体液性応答を示した。複数回行われた実験において、IL-12は大体において特異的抗体応答を10-20%抑圧し、GM-CSFは見かけ上逆の効果を有した。この体液性効果は、FACS解析で同定された脾臓細胞中のB細胞数の変化の観察結果に関連し得る。
Tヘルパーリンパ球の増殖の活性化は、抗原により活性化されるB細胞の増殖を介する体液性免疫応答と、CD8+細胞傷害性Tリンパ球の増殖を介する細胞性免疫応答の両者を誘導するために重要な役割をしている。DNA免疫化後2週間で、免疫化マウスから脾臓が回収され、リンパ球が単離された。これらの細胞は次に上に記載したようにT細胞増殖について試験された。図5に、HIV-1 gag/pol(pCGag/Pol)をコードするDNAワクチンで免疫化されたマウスと、pCGag/PolとIL-12またはGM-CSFで共免疫化されたマウスの増殖試験の結果を示す。レクチンPHAの組換えp55タンパク質20g/mlが、ポリクローン刺激性の正の対照として用いられた。示したように、無処理のマウス脾臓由来のグループからは増殖のバックグラウンド値が低いことが観察され(1:2希釈で活性化指数1.2)、pCGag/Polのみで免疫化されたグループからは中程度の増殖が観察された(1:2希釈で活性化指数9.2)。pCGag/PolとIL-12遺伝子で共免疫化されたグループおよび、pCGag/PolとGM-CSF遺伝子で共免疫化されたグループからは、増殖の劇的な加速が観察された(それぞれ活性化指数17.1、15.6)。
細胞活性の増強をさらに調査するために、直接的CTL試験が行われた。CTL試験は、脾臓細胞にin vitro刺激を誘導せずに、免疫化されたマウスから回収した脾臓に対して上に記載したように行われた。試験は脾臓回収の日に、特定のワクシニア感染標的および不特定のワクシニア感染標的からのクロミウムの放出を測定することによって行われた。Gag/Pol+IL-12で免疫化された脾臓細胞では、特異的CTL活性の劇的な上昇が観察された(図6)。IL-12遺伝子カセットのみで免疫化された対照グループでは、標的細胞の特異的な溶解はバックグラウンド値以上には起きなかった。さらに、エフェクター:標的比50:1のGag/Polのみの免疫化では、特異的な溶解レベルは低かった(3%)。対照的に、エフェクター:標的比50:1のGag/Pol+IL-12共投与試料では62%の特異的な溶解が観察され、エフェクター:標的比12.5:1では9%に下がった。一方、Gag/PolおよびGM-CSFプラスミドで免疫化されたものでは、検出可能なCTL活性は見られなかった。同様な結果が、HIV-1エンベロープ構築物とサイトカイン遺伝子で共免疫化されたマウスで観察された(図7)。
DNAの接種を用いたin vivoでの免疫応答の誘導は、種々の治療標的および導入法を用いて報告されている。
例えば自然界での感染において、抗HIV-1 CTL応答は非常に初期に見られ、ウィルスの設定値の確立と一時的に相関するように見える。CTL T細胞はウィルスに感染した細胞を標的として破壊することで、ウィルスの排除に重要な役割を担っている。特異的CTL応答の誘導によりウィルスタンパク質に対して免疫応答を行うことによって、ウィルス中の複数の抗原性標的に対するより広範な免疫応答の誘導が可能になると考えられる。ウィルスに対するCTL活性は、AIDS患者に比べて、健康な感染者においてより良く測定され、疾患の病原が増大するに従って特異的CTLが減少することが報告され、好ましい臨床状態とCTL応答が関係付けられている。この観点から、高い特異的CTLと低い抗体応答を有するカニクイザルはキメラSIV/HIV(SHIV)の攻撃から守られ、低い特異的CTLと高い抗体応答を有する動物はウィルスの複製は制御したが、その防御は完全ではなかった。特異的CTL応答は、HIV感染の無症候期の維持に貢献していると考えられる。即ち、DNA免疫化によるin vivoでの強いHIV-1特異的CTLの誘導は、HIV感染の進行から究極的に宿主を防御するために重要な役割を果たし得る。
in vivoでの免疫応答をさらに制御するために、HIV-1 DNA免疫原構築物と広範なサイトカイン遺伝子の共導入による免疫応答の誘導および制御が調査された。
DNAプラスミド
HIV-1エンベロープタンパク質を発現するDNAワクチン構築物(pCEnv)およびgag/polタンパク質を発現するDNAワクチン構築物(pCGag/Pol)が、標準的な方法および直ちに入手可能な原材料を用いて調製された。ヒトIL-1α(IL-2、IL-5、IL-10、IL-15、TNF-α、TNF-β、およびマウスIL-4およびIL-18が、標準的な方法および直ちに入手可能な原材料を用いてpCDNA3発現ベクター(Invitrogen,Inc.,San Diego,CA)にクローン化された。ヒトIL-1α、IL-2、IL-5、IL-10、IL-15、TNF-αおよびTNF-βは、マウス細胞中で活性を有することが報告されている。プラスミドDNAは細菌中で生産され、Qiagen Maxi Prepキット(Qiagen,Santa Clara,CA)を用いて精製された。
ヒト横紋筋肉腫(RD)およびマウス肥満細胞腫p815細胞系列がATCC(Rockville,MD)から入手された。HIV-1エンベロープ(vMN462)、gag/pol(vVK1)、およびβ-ガラクトシダーゼ(vSC8)を発現する組換えワクシニアが、NIH AIDS Research and Refence Reagent Programから入手された。HIV-1エンベロープペプチド(RIHIGPGRAFYTTKN)が標準的な方法および直ちに入手可能な原材料を用いて合成された。
IL-1α、IL-2、IL-5、IL-10、IL-15、TNF-αおよびTNF-βに対する抗体は、R&D Systems(Minneapolis,MN)から入手された。
サイトカイン遺伝子構築物の発現は、RD細胞へのトランスフェクション後に免疫沈降またはサイトカインELISAによって確認された。細胞はPBSで2回洗浄され、血清、メチオニンおよびシステインを含まないDMEM中で1時間飢餓状態におかれ、次に200μCi/ml(1200Ci/mmole)の35Sタンパク質標識反応液(NEN/DuPont)で標識された。標識された細胞は0.5mlのRIPA緩衝液(50mM Tris-HCl(pH7.6);150mM NaCl;0.2% Triton X-100;0.2%デオキシコール酸;0.1%SDKs;1mM PMSF)中で氷上で溶解され、次に1分15000rpmで遠心され浄化された。浄化された溶解液は個々の抗体(R&D System)とともに氷上で90分保温された。プロテインAセファロースが抗原-抗体複合体に加えられ、4℃で90分振とうして混合された。BSAを含む高塩濃度の緩衝液で念入りに洗浄された後、タンパク質沈殿物は50μlの1×試料緩衝液に再懸濁され、100℃で3-5分熱された。タンパク質試料の一部がSDS 12%-PAGEによって解析された。蛍光写真のために、ゲルは10%グリセロールを含む1Mサリチル酸ナトリウムに15分浸され、乾燥され、コダックX-omat-ARフィルムを用いて放射線写真を撮影された。トランスフェクションしたRD細胞由来の上清は回収され、サイトカインELISAキット(Pharmingen,San Diego,CAおよびR&D System)を用いて発現を調べられた。
生後6から8週齢のbalb/cマウス(Harlan Sprague Dawley,Inc.,Indianapolis,IN)の四頭筋に、0.25%ブピバカイン-HClを含むりん酸塩緩衝液(PBS)中に調製された目的の各DNA構築物50μgが注射された。種々の遺伝子発現カセットとの共投与には、注射に先だって選択されたプラスミドを総量100μg/注射で混合することが含まれた。
0.1M炭酸-二炭酸緩衝液(pH9.5)中に2μg/ml濃度で希釈されたp24またはgp120タンパク質50μlが、マイクロタイタープレートの穴に4℃一晩吸着された。プレートはPBS-0.05% Tween-20で洗浄され、3% BSAを含むPBS-0.05% Tween-20で37℃で1時間ブロッキングされた。マウス抗血清が0.05% Tween-20で希釈され、37℃1時間保温され、次にHRP結合ヤギ抗マウスIgG(Sigma,ST Louis,MO)とともに保温された。プレートは洗浄され、3'3'5'5'TMB緩衝液(Sigma)で発色された。プレートの450nm吸光が、Dynatech MR5000プレート読取装置で読みとられた。
脾臓からリンパ球が回収され、赤血球の除去、および新鮮な培地による数回の洗浄によってエフェクター細胞として調製された。単離細胞懸濁液は、5×106細胞/mLの濃度に再懸濁された。5×105細胞を含む100μL分画が直ちに96穴マイクロタイター平底プレートの各穴に加えられた。組換えPr55またはgp120タンパク質が最終濃度5μg/mLおよび1μg/mLで3つ独立に穴に加えられた。細胞は5%CO2で37℃3日間保温された。1μCiのトリチウム化チミジンが各穴に加えられ、細胞は37℃で12-18時間保温された。プレートから細胞が回収され、取り込まれたトリチウム化チミジンの量がBeta Plate reader(Wallac,Turku,Finland)で測定された。活性化指数(SI)は式: 活性化指数(SI)=計測カウント/自発カウントで決定された。自発カウントウエルは、非特異的タンパク質の対照となる10%ウシ胎児血清を含む。さらに、pCEnvまたは対照の免疫化動物は通常、gp120タンパク質に対してSI値1である。細胞が健康であることを確認するため、PHAまたはCon A(Sigma)がポリクローン刺激性の正の対照として用いられた。PHAまたはCon Aの対照試料は、20-40のSIを有する。
ワクシニアに感染した標的、またはペプチドで処理した標的を用いて、5時間の51CR放出CTL試験が行われた。アッセイは、in vitroエフェクター刺激存在下および非存在下の両者について行われた。in vitro刺激アッセイでは、エフェクターは特定のワクシニア(エンベロープにはvMN462、gag/polにはvVK1)に感染した細胞による刺激を受け、0.1%グルタルアルデヒド、または濃度1mMのエンベロープ特異的ペプチド(RIHIGPGRAFYTTKN)で5×106細胞/mlでCTL培地中で4-5日間固定された。CTL培地は、10%ウシ胎児血清1640(Gibco-BRL)および10% RAT-T-STIM without Con A(Becton Dickinson Labware,Bedford,MA)を含む、1:1のIscove's Modified Dulbecco培地(Gibco-BRL,Grand Island,NY)およびHank's Balanced Salt溶液(Gibco-BRL)からなる。3×106p815細胞を10から20の複合感染(MOI)で37℃で5から12時間感染させることによって、ワクシニアに感染した標的が調製された。ペプチドで処理した標的は、p815細胞を1μM濃度のペプチドと保温することによって調製された。標準的なクロミウム51放出CTL試験が行われ、標的細胞は100μCi/mlのNa2 51CrO4で60から120分間標識され、刺激を受けたエフェクター脾臓細胞とともに37℃で4-6時間保温するために用いられた。CTLは、エフェクター:標的(E:T)比50:1から12.5:1で決定された。上清が回収され、LKB CliniGammaガンマ計数器で計測された。特異的溶解の割合は、式: 100×(実験放出-自発放出)/(最大放出-自発放出)で決定された。最大放出は、1% Triton X-100を含む培地中での標的細胞の溶解によって決定された。自発放出の計数値が最大放出の20%以上である場合は、試験は無効とされた。
抗CD8モノクローン抗体(Pharmingen,San Diego,CA)処理によってCD8+ T細胞が脾臓細胞から単離され、次にウサギ補体(Sigma)とともに37℃で45分保温された。
サイトカイン遺伝子カセットの発現
サイトカイン遺伝子は各々pCDNA3プラスミド発現ベクターにクローン化された(図10)。これらのサイトカイン発現カセットは、挿入断片全長のシークエンス解析によって確認された(5'側と3'側の両方)。さらに材料と方法に記載されたように、これらのサイトカイン遺伝子はRD細胞へトランスフェクションされ、特定の抗体を用いた免疫沈降またはサイトカインELISAによって発現を確認された。
pCGag/Pol免疫化マウスの抗血清が回収され、ELISAによってHIV-1抗原に対する特異的抗体応答について解析された。
Tヘルパーリンパ球の活性化および増殖は、B細胞を介した体液性免疫応答およびCD8+細胞傷害性T細胞を介した細胞性免疫応答の両者の誘導において重要な役割を果たす。マウスは2週間おきに2回のDNA免疫化を受けた(各50μg)。追加注射の1週間後にマウスは屠殺され、脾臓が回収され、リンパ球が単離されてTヘルパー細胞増殖を検査された。
pCEnvまたはpCGag/polと炎症誘発性サイトカインIL-1α、TNF-αおよびTNF-βを共注射されたマウス由来の脾臓を用いて増殖試験が行われた。
Th1サイトカインIL-2、IL-15およびIL-18の共注射の効果が調査された。pCEnv+IL-18およびpCGag/pol+IL-18共注射グループはそれぞれ活性化指数4.4および10.0であった(5μg/mlの各タンパク質濃度)。INF-γ誘導性Th1サイトカインIL-12およびIL-18の共注射に伴うTヘルパー細胞増殖応答を評価する実験では、方法は以下の通りであった。pCEnv(50μg)による最初のDNA免疫化の2週間後、マウス(4匹/グループ)は同量で追加刺激された。さらに1週間後、免疫化マウスから脾臓が回収され、リンパ球が単離されて組換えgp120タンパク質(最終濃度5および1μg/ml)に対して試験された。pCGag/pol(50μg)による最初のDNA共注射の2週間後、マウス(4匹/グループ)は同量で追加刺激された。さらに1週間後、免疫化マウスから脾臓が回収され、リンパ球が単離されてPr55タンパク質(最終濃度5および1μg/ml)に対するT細胞増殖性応答を検査された。これらの実験は2回繰り返され、結果は同様であった。以下のデータが得られた。
炎症誘発性およびTh1サイトカインの試験に加えて、Th2サイトカインIL-4、IL-55およびIL-10と、pCEnvおよびpCGag/polとの共注射の効果が同様に調査された。pCEnv+IL-18およびpCGag/polのみを注射されたグループに比べて、IL-4およびIL-5の共注射グループはそれぞれ中程度のTヘルパー細胞の増殖を示した。Th2サイトカインIL-5およびIL-10の共注射に伴うTヘルパー細胞増殖応答を評価する実験では、方法は以下の通りであった。pCEnv(50μg)による最初のDNA免疫化の2週間後、マウス(4匹/グループ)は同量で追加刺激された。さらに1週間後、免疫化マウスから脾臓が回収され、リンパ球が単離されて組換えgp120タンパク質(最終濃度5および1μg/ml)に対して試験された。pCGag/pol(50μg)による最初のDNA共注射の2週間後、マウス(4匹/グループ)は同量で追加刺激された。さらに1週間後、免疫化マウスから脾臓が回収され、リンパ球が単離されてPr55タンパク質(最終濃度5および1μg/ml)に対するT細胞増殖性応答を検査された。これらの実験は2回繰り返され、結果は同様であった。以下のデータが得られた。
細胞性免疫の増強をさらに調査するために、pCEnvおよびpCGag/polで免疫化されたマウスの脾臓を用いて細胞傷害性Tリンパ球(CTL)試験が行われた。マウスは2週間おきに2回のDNA免疫化を受けた(各50μg)。追加注射の1週間後にマウスは屠殺され、脾臓が回収され、リンパ球が単離されてCTL応答を検査された。試験は、特異的および非特異的ワクシニアに感染した標的、またはペプチドで処理した標的からのクロミウムの放出の測定に先だつエフェクター脾臓細胞に対する刺激の存在下および非存在下の両者について行われた。標的の特異的溶解を測定するために、非特異的標的(vSC8感染)の溶解度が、特異的(vMN462またはvVK1感染)標的の溶解度から差し引かれた。
細胞傷害性Tリンパ球応答を評価するために、種々のサイトカインの共注射後にデータが得られた。以下のデータが得られた。
炎症誘発性サイトカイン、IL-1α、TNF-αおよびTNF-βとpCEnvまたはpCGag/polとの共注射を受けたマウスのCTL試験のデータについては、特定の細胞破壊の一定のバックグラウンド値が対照動物でも観察され、pCEnvのみで免疫化した動物は低レベルのCTL応答を示した。pCEnv+IL-1αまたはpCEnv+TNF-βの共注射によって、CTL活性は中程度上昇した。一方、pCEnv+TNF-αの共注射に伴って、ワクシニア(vMN462)に感染した、HIV-1を発現する標的細胞の特異的な破壊が観察された。エフェクター:標的(E:T)比50:1で、pCEnv+TNF-αの共注射に伴って、標的細胞の30%以上の特異的溶解が観察された。同様に、pCGag/pol+TNF-αで免疫化されたマウスでは、抗原特異的CTLによる、ワクシニア(vMN462)に感染した、HIV-1 gag/polを発現する標的細胞の溶解の有意な増強が観察され(E:T比50:1で29%溶解)、pCGag/pol+IL-1αまたはpCGag/pol+TNF-βの共注射によるCTL活性の上昇は小さかった。
Th1サイトカインをDNAワクチン構築物と共導入する効果が調査された。Th1サイトカインIL-12およびIL-18により免疫化されたマウス、およびpCEnvにより免疫化されたマウスのCTL試験のデータについては、IL-12の共投与とは異なり、IL-18共注射によってはCTL応答がより緩やかに上昇した。IL-2の共投与によっても、CTL応答は中程度上昇した。一方、pCEnv+IL-15による免疫化に伴って、46%特異的溶解という劇的なCTL応答の上昇が観察された。同様に、pCGag/pol+IL-15を注射されたマウスでは、抗原特異的溶解の有意な増強(30%)が起きた。
炎症誘発性サイトカインおよびTh1サイトカインの共注射の効果の調査に加えて、Th2サイトカインIL-4、IL-5およびIL-10がCTL応答に与える効果が調査された。これらのサイトカインの共注射によりTヘルパー細胞増殖応答が上昇したが、pCEnvまたはpCGag/polのいずれとも、IL-4、IL-5およびIL-10の共注射でCTL応答の特定の上昇をおこさなかった。
TNF-αおよびIL-15の共注射を介するCTL応答の上昇が、CD8+ MHCクラスI制限性刺激によるかどうか決定するため、balb/cマウスについて、MHCクラスI制限性CTLに特異的なエピトープであるHIV-1エンベロープペプチド(RIHIGPGRAFYTTKN)を用いてCTL試験が行われた。マウスは各DNA構築物(50μg)による2回の免疫化を2週間おきに受け、2回目の免疫化の1週間後、脾臓が回収された。in vitro刺激後、エンベロープ特異的ペプチドを用いて脾臓のCTL検査が行われた。IL-15およびTNF-αの共注射後、E:T比50:1でそれぞれ25%および32%の特異的細胞破壊を伴う有意なCTL応答の上昇が観察された(図11A-11E)。この観察結果は、補体溶解によってエフェクター細胞集団からCD8+ T細胞を取り出した後のCTL活性を測定することによって確認された。マウスは各プラスミド(50μg)による2回の免疫化を2週間おきに受けた。CTL試験が行われ、エフェクター細胞の1つのグループが上のように処理され、別のグループからCD8+ T細胞が取り出された。図11F-110に示すように、CD8+ T細胞の除去によって、IL-15およびTNF-αの共注射後に観察される抗原特異的CTL増強が抑制された。これらの結果から、細胞溶解活性の増強が抗原特異的であり、クラスIに制限され、CD8+細胞依存性であることが示唆された。
IL-15またはTNF-αの共注射グループでは一定の高いレベルの直接的CTL応答(エフェクターをin vitro刺激しない場合)が観察されたため、それらによって誘導される直接的CTL応答のレベルが調査された。クロミウム放出試験が、脾臓細胞が単離された日に行われた。IL-12共注射の場合の直接的CTLを観察した場合と異なり、pCEnvとTNF-αまたはIL-15の共注射後には直接的CTL活性は誘導されなかった(図12)。
いかなる免疫化の方法でも、総体的な目標は、最少数の免疫化を用いて、有効で持続的な病原体特異的免疫応答を誘導することである。しかし、防御の相関現象は病原体ごとに異なり得、方法の改善は免疫応答を制御することによって達成され得る。例えば、B型肝炎ウィルスの感染に対する防御には高レベルの特異的抗体応答が重要であると考えられ、リンパ細胞choriomeningitisウィルス(LCMV)感染に対する防御は主にT細胞を介した応答に媒介される。DNA種痘化法の設計は、誘導される免疫応答の方向と強度を調節して、各標的病原体に対する防御相関現象に適合するようにすることで改善され得る。
さらに臨床的に有効なワクチンが、免疫応答の方向と強度を調節する戦略によって生産され得る。ワクチンによる免疫応答および免疫治療の特定の型および方向を得る際により精密な調節は、サイトカインおよび共活性化分子のような免疫学的に重要な分子の遺伝子を共導入することによって達成され得る。
DNA免疫化による宿主の免疫応答の誘導における種々の免疫学的に重要なサイトカインの役割が調査された。図13に要約するように、特異的防御性免疫応答の誘導は、サイトカイン遺伝子の共投与によって設計され得る。このサイトカイン遺伝子アジュバントの系は、疾病ごとに異なる防御の相関現象により正確に合わせて種痘化プログラムを設計するための、特異的免疫応答の誘導における新たなレベルの制御が特徴である。ワクチンおよび免疫治療のこの型の精密な制御は、以前には得られなかった。この結果、免疫応答の方向および強度の調節は種々の種痘化法において有益である。例えば、T細胞を介する応答が好ましいが体液性応答が不要であるか有害である場合、特定の免疫原とともに共導入される免疫調節因子としてIL-12遺伝子が選択され得る。一方、細胞外細菌を標的とするワクチンを生産する場合、例えばIL-4、IL-5またはIL-10遺伝子が共注射され得る。
PCR反応および図15に示す適当な制限酵素を用いて、図14に示すBL1挿入断片がライゲーションによってPCR3真核生物発現ベクターおよびpBBKanベクターにクローン化された。BL1構築物は異なるHIV-1抗原とともに共投与され、マウス中での免疫活性化効果が測定された。図16および17A、17B、17C、および17Dに示す結果から、HIV-1抗原で共免疫化された場合に、BL1のDNA断片が免疫応答を増強することが示唆された。観察された異なる効果は、脾臓サイズの上昇および抗体およびCTL応答の上昇である。
