Abstract
【解決手段】アミノ酸総数に対するRGD配列の割合が少なくとも0.4であり、RGDエンリッチゼラチンが350以上のアミノ酸を含む場合には、350のアミノ酸の各ストレッチが少なくとも1つのRGDモチーフを含む。好ましくは、RGDエンリッチゼラチンは、組換え技術によって調製され、ヒトゼラチン又はコラーゲンアミノ酸配列に由来する配列を有する。

Description
ことが必要である。
i)容積に対する表面積の高い割合。これは、マイクロキャリアの密度を変化させることにより、変動可能であり、単位容積あたり高い細胞密度となり、高密度の細胞製品を得られる可能性がある。ii)多数の低生産性容器を使用せずに、単一の高生産性容器で細胞を伝播することができる。その結果、培地を有効に使用することができ、著しく節約することができる。iii)マイクロキャリア培養はよく混合されているため、pH、pO2、pCO2等の様々な環境条件を容易にモニター、制御することができる。iv)細胞のサンプリングを容易に行うことができる。v)ビーズを容易に設置できるので、細胞の回収及び製品のダウンストリーム処理を容易に行うことができる。vi)適切に改善された例えば醗酵槽等の従来の装置使用して、マイクロキャリア培養を比較的容易にスケールアップすることができる。
i)ビーズの表面の特性は、細胞が迅速に付着し、増殖できなくてはならない。外形は均等であることが好ましい。ii)容易に単離できるように、ビーズの密度は、培養培地より若干高くなければならない。従来の天然の培養培地は水性であり、密度は1.03〜1.09g/ccである。しかし、密度は、1.03〜1.045g/mlである適切な範囲を一定以上超えてはならない。剪断に感受性を有する細胞を傷つけないような緩やかな攪拌で、懸濁状態を維持することができ、ビーズが配置された場合には、細胞の増殖が抑制される。iii)ビーズのサイズの分布は、全てのマイクロキャリアの懸濁が達成され、細胞が同時にコンフルエントになるように、小さくなければならない。さらに、溶液内のマイクロキャリアのクラスター形成を防止しなければならない。iv) 光学特性は、顕微鏡で、容易に観察できなければならない。v)細胞の生存と増殖のためだけでなく、獣医学的又は臨床目的で使用される細胞培養製品のためにも、毒性であってはならない。vi)ビーズのマトリックスは、培養の攪拌中に起きる衝突が、ビーズを断片化しないようなものでなければならない。

平均直径が100マイクロメートルのポリスチレンビーズを用いた。ポリスチレンビーズ上にゼラチンをコバレントに固定するためにヘテロ二機能性クロスリンク剤であるBBA−EAC−NOSを用いた。BBA−EAC−NOSをポリスチレンビーズに添加し、吸着させる。次に、ゼラチンを添加し、NOS合成ポリマーと反応させ、スペーサーにコバレントなカップリングを産出した。スペーサー(及びコバレントにカップリングしたゼラチン)をコバレントにポリスチレンビーズに固定するために、感光(320nm)した。最後に、接着の弱いゼラチンを、リン酸緩衝液(pH7.2)内で弱い漂白剤Tween 20で一晩洗浄して取り除いた。
細胞数の計数には、細胞を、0.1Mのクエン酸内にクリスタルバイオレット(0.1%w/w)を等量混合して染色し、血球計数器で計数した。培地からの細胞の減少は、ビーズに付着した細胞の指標として用いた。
Claims (10)
- アミノ酸総数に対するRGDモチーフの割合が少なくとも0.4であり、RGDエンリッチゼラチンが350以上のアミノ酸を含む場合に、350のアミノ酸の各ストレッチが少なくとも1つのRGDモチーフを含むRGDエンリッチゼラチンを含有する血小板凝集予防剤。
- アミノ酸総数に対するRGDモチーフの割合が、少なくとも0.6、好ましくは少なくとも0.8、さらに好ましくは少なくとも1.0、さらに好ましくは少なくとも1.2、最も好ましくは少なくとも1.5であることを特徴とする、請求項1記載の血小板凝集予防剤。
- RGDエンリッチゼラチン内のRGDモチーフの数が、250のアミノ酸あたり少なくとも4であることを特徴とする、請求項1又は2記載の血小板凝集予防剤。
- RGDエンリッチゼラチンが、RGDモチーフを少なくとも4、好ましくは6、さらに好ましくは8、より好ましくは12以上16以下含むことを特徴とする、請求項1〜3いずれかに記載の血小板凝集予防剤。
- RGDエンリッチゼラチンの分子量が約30kDa〜約200kDaであることを特徴とする、請求項1〜4いずれかに記載の血小板凝集予防剤。
- RGDエンリッチゼラチンの分子量が、最大10kDa、好ましくは最大5kDaであることを特徴とする、請求項1〜4いずれかに記載の血小板凝集予防剤。
- RGDエンリッチゼラチンが、5%未満、好ましくは3%未満のヒドロキシプロリン残基を含むことを特徴とする、請求項1〜6いずれかに記載の血小板凝集予防剤。
- RGDエンリッチゼラチンが、pH7〜7.5において正味の正電荷を有することを特徴とする、請求項1〜7いずれかに記載の血小板凝集予防剤。
- RGDエンリッチゼラチンの少なくとも80%が、天然ヒトコラーゲン配列の1又は複数の部分からなり、該天然ヒトコラーゲン配列の部分が少なくとも30アミノ酸の長さを有することを特徴とする、請求項1〜8いずれかに記載の血小板凝集予防剤。
- RGDエンリッチゼラチンが、1又は複数の天然ヒトコラーゲン配列の1又は複数の部分からなることを特徴とする、請求項9記載の血小板凝集予防剤。
