このように、本明細書において使用される「生物学的に活性な分子」とは、スフィンゴイド−ポリアルキルアミン抱合体と組み合わせて投与したときに、被検体の免疫系に対して影響を及ぼす任意の物質を表す。生物学的に活性な分子は、好ましくは、抗原性タンパク質、抗原性ペプチド、抗原性ポリペプチド又は抗原性炭水化物である。

本発明は、被検体の免疫応答を調節するのに有効である、生物学的に活性な分子を運搬するための捕捉剤としての、スフィンゴイド−ポリアルキルアミン抱合体の使用に関する。
スフィンゴイド−ポリアルキルアミン抱合体は、個別の脂質様分子の形態とし、又は集合物の形態とすることができる。適切な集合物の一例には、ミセル又は小胞、特に、リポソームの形成が含まれる。集合物の他の例には、逆相、立法相などのミセルの形成が含まれる。明らかに、スフィンゴイド−ポリアルキルアミン抱合体は、組み合わされた小胞/ミセルの形態とすることができ、又は集合物の他の任意の組み合わせとすることができる。
R1は、水素、分枝又は直鎖のアルキル、アリール、アルキルアミン、又は−C(O)R5基を表し;

スフィンゴイド−ポリアルキルアミン抱合体と生物学的に活性な物質の好ましい重量比は、1000:1から1:1の重量比である。
脂質集合物中に含まれることができ、同様の構造物中に使用されることが知られている他の成分は、立体安定化物質(steric stabilizer)である。一般的に使用される立体安定化物質の一例は、リポポリマーのファミリー、例えば、ポリエチレングリコール誘導体化脂質(PEG−脂質抱合体)である。化合物のこのファミリーは、とりわけ、脂質の循環時間を増加(延長する)ことが知られている。
本発明のスフィンゴイド−ポリアルキルアミン抱合体は、好ましくは、ワクチンの調製のために使用される。
本発明は、生物学的に活性な物質とともに、スフィンゴイド−ポリアルキルアミン抱合体で被検体を処置することを含む、被検体の免疫応答を調節する方法にも関する。
生物学的に活性な物質と会合されたスフィンゴイド−ポリアルキルアミン抱合体を含む本発明の組成物は、様々な方法で投与することができる。投与経路の非限定的な例には、経口、皮下(s.c.)、静脈内(i.v.)、動脈内(i.a.)、筋肉内(i.m.)、腹腔内(i.p.)、直腸内(i.r.)及び鼻内(i.n.)投与を含む非経口、並びに、目への注入技術、眼内が含まれる。好ましくは、投与の様式は、鼻内又は筋肉内投与である。
生理的に許容される担体の非限定的な例には、単独の、又は、微量な(最大10%)アルコール(エタノールなど)を加えた、水、生理的食塩水、5%デキストロース(グルコース)、10%スクロースなどが含まれる。
HN抗原が搭載された陽イオン性リポソームの性質決定
コレステロール(Chol)を加えて、又は加えずに、様々な脂質/タンパク質 w/w比(3/1−300/1)で、様々な陽イオン性リポソーム調合物上に搭載されたHN(インフルエンザウイルス由来のヘマグルチニンとノイラミニダーゼの市販の調製物)のカプセル化効率を調べた。表1は、陽イオン性脂質DOTAPとCCSを用いた、このような実験の結果を示している。
これらの知見を総合すると、全ての調合物で、簡易且つ迅速な(5分)手順を用いて、極めて高いローディング効率(>60%)が得られることが示唆される。さらに、水性懸濁液中に予め形成されたリポソームでさえ、インフルエンザウイルスの表面抗原と効果的に会合できた。
第一の実験では、若い(2ヶ月齢)BALB/cマウスに、HNが負荷された中性、陰イオン性又は陽イオン性リポソームを鼻内ワクチン接種した後の、HI、IgG1及びIgG2a抗体の血清レベルを測定した(表2A)。HN抗原は、A/New Caledonia(H1N1)株に由来する一価サブユニットワクチンであった。同じ実験において、HI、IgG1、IgG2a及びIgA抗体の肺及び鼻のレベル、脾臓細胞によって産生されたINFγレベルも調べた(表2B及び2C)。
生体分布実験では、インフルエンザHN抗原が負荷されていない、又は負荷されている、脂質集合物の三つの調合物:DMPC/DMPG(陰イオン性), DOTAP/Chol(陽イオン性)及びCCS/Chol(陽イオン性)を、BALB/cマウスに鼻内投与した(マウス当たり200μg脂質、2μg抗原)。次いで、24時間にわたって(投与から1時間後、5時間後及び24時間後の時点で)、様々な組織のホモジネート中で、蛍光標識された脂質を追跡した。
毒性(局所、全身)は、筋肉内及び鼻内ワクチンに伴う主要な懸念であるため、予備的な毒性研究を行った。インフルエンザ抗原ヘマグルチニン+ノイラミニダーゼ(HN)が負荷された陽イオン性脂質調合物(DMTAP、DOTAP、CCSベース)を、マウス(n=4/グループ)に鼻内(1週間の間隔を置いて、2回)投与し、血球数(総数、差)、血液の化学検査および組織学的検査(鼻、肺切片)を72時間後に実施した。マウスは、明瞭な毒性の徴候を一切示さなかった。血球数及び血液化学は、正常な範囲内にあり、予想通り、陽イオン性調合物で処置されたマウスの鼻及び肺には、最小限ないし弱い炎症応答が見られた。程度はさらに低いが、同様の炎症応答が、生理的食塩水のみ又は非カプセル化抗原で処置されたマウスの一部にもみられた。
これらの実験では、様々なリポソーム調合物(DMPC、DMPC/DMPG、DOTAP/Chol、CCS/Cholから構成される。)、0.5から1mgの脂質を、HN抗原あり又はなしで、マウスに腹腔内注射した。マウスには、処置を施さないか、又はリポソーム調合物の注射から2日前に、チオグリコレート(TG)、マクロファージ産生を増加させるため)とともに腹腔内注射した。リポソームの投与から24ないし48時間後に、腹腔細胞を採集し、そのまま使用するか、又はプラスチック皿に、37℃で、4時間吸着させ、非付着性細胞を除去した後に使用した。別の実験では、TG処置されたマウスから腹腔細胞を採集し、リポソーム調合物とともに、24から48時間インキュベートした。MHC II並びに同時刺激分子CD40及びB7の発現について、細胞をフローサイトメトリーによって調べた。サイトカイン:インターフェロンγ(IFNγ)、腫瘍壊死因子α(TNFα)及びインターロイキン12(IL−12)並びに一酸化窒素(NO)について、上清を検査した。
インフルエンザに加えて、鼻内(i.n.)及び直腸内(i.r.)投与されたHAVワクチンについても、CCS脂質集合物の免疫増強能を調べた。
2週間隔で、HAVワクチン(Aventis Pasteur)、10EU(約1.5μgタンパク質)を2回投与し、2度目のワクチン投薬から3週後に、ELISPOT技術によって、応答を調べた。粘膜アジュバントとして使用されるCpG−ODNを、1回の投薬当たり10μg与えた。HAV−CCS脂質集合物は、インフルエンザワクチンについて上述したように調製した(表1)。
さらなる実験では、0.4μg用量の市販のC.botulinumトキソイド(バイオテロ剤のモデル、ウルグアイ、アルミナなし)を用いて、マウスの鼻内に免疫し、2回目のワクチン投薬から4週後に、ELISAによって、抗体力価を調べた。
C.botulinumトキソイドを用いた実験の結果は表18に要約されており、鼻内への滴下注入後に、特に小腸及び糞便中のIgAレベルに関して、CCS−トキソイド調合物が標準的なワクチンより優れていることを示している。このような抗体は、経口曝露時に、毒素を中和することが予想される。ワクチンを単独で鼻内に免疫化したマウスは、IgAを産生しなかった。
化学
N−パルミトイル D−エリスロスフィンゴシル−1−カルバモイルスペルミン(CCS)の合成
(i)N−パルミトイルスフィンゴシン(1.61g、3mmol)を、加熱しながら、無水THF(100mL)中に溶解した。透明な溶液を室温とし、N,N’−ジスクシニミジルカーボネート(1.92g、7.5mmol)を添加した。攪拌しながら、DMAP(0.81g、7.5mmol)を添加し、この反応をさらに16時間攪拌した。減圧下で溶媒を除去し、n−ヘプタンから再結晶された残留物から、融点73−76℃の白色粉末として、ジスクシニミジルセラミジルカーボネート1.3g(68%)を得た。
CCSの構造は、1H及び13C−NMR分光法によって確認した(データは示さず。)。分析の詳細な説明は、同時係属の国際特許出願第 号に記載されている。
上記手順と同様に、以下の手順を適用することができる。

対応する1,3−ジ−O−スクシニミジル誘導体を得るために、DMAPの存在下で、1当量のセラミドを、2.5当量のジスクシニミジルカーボネートと反応させる。
このようにして得られたジスクシニミジル誘導体を、触媒量のDMAPを用いて、室温で、当量のスペルミンと反応させて、図1Bの3−一置換−セラミド−スペルミン抱合体を得る。
上述したように調製された一当量の1,3−ジ−O−スクシニミジルスフィノギド誘導体を、触媒量のDMAPの存在下において、80°で、2.5当量のスペルミンと反応させる。1,3−二置換CCSが、このようにして得られる。
上述のように調製された一当量の1,3−ジ−O−スクシニミジルセラミド誘導体を、触媒量のDMAPの存在下において、80°で、2.5当量のα−ω−ジプロテクトスペルミンと反応させる。
プロテクションを除去し、1,3−「分枝」二置換セラミド−スペルミン抱合体を得る。
上述のように調製した一当量の1,3−ジ−O−スクシニミジルセラミド誘導体を、触媒量のDMAPの存在下において、80℃で、0.75当量のスペルミンと反応させる。
インフルエンザA/ニューカレドニア/20/99類似(H1N1)株由来の一価サブユニット抗原調製物は、Drs. Gluck及びZurbriggen, Berna Biotech, Bern、Switzerlandから頂いた。この調製物(本明細書ではHNと表記する。)は、80から90%のヘマグルチニン、5から10重量%のノイラミニダーゼ並びに微量のNP及びMIタンパク質から構成された。A/ニューカレドニア/20/99(H1N1)株、A/パナマ/2007/99(H3N2)及びB/山東/7/97由来のHNを含有する、2003/2004シーズン向けの市販の三価サブユニットワクチン(Fluvirin(R))は、Evans Vaccines Ltd.,Liverppol.UKから購入した。このワクチンは、カプセル化の前に、約8倍に濃縮した(Eppendorf Concentrator 5301, Eppendorf AG, Hamburg, Germany)。インビトロ刺激のために、幾つかの実験では、完全な不活化ウイルスを用いた。
リン脂質(PL) ジミリストイルホスファチジルコリン(DMPC)、ジミリストイルホスファチジルグリセロール(DMPG)、及びジオレオイルホスファチジルエタノールアミン(DOPE)は、Lipoid GmbH, Ludwigshafen, Germanyから購入する。DMPC(中性)およびDMPC/DMPG(9/1モル比、陰イオン性)リポソームに加え、陽イオン性リポソーム/脂質集合物の6つの調合物を調製した。モノ陽イオン性脂質ジメチルアミノエタンカルバモイルコレステロール(DC−Chol)、1,2−ジステアロイル−3−トリメチルアンモニウム−プロパン(塩化物塩)(DSTAP)、ジオレオイル−3−トリメチルアンモニウム−プロパン(塩化物塩)(DOTAP)およびジミリストイル−3−トリメチルアンモニウム−プロパン(塩化物塩)(DMTAP)は、Avanti Polar Lipids(Alabaster, AL, USA)から購入する。一陽イオン性脂質ジメチルジオクタデシルアンモニウム・ブロミド(DDAB)およびコレステロール(Chol)は、Sigmaから購入する。新しい、独自のポリ陽イオン性スフィンゴ脂質N−パルミトイル D−エリスロスフィンゴシルカルバモイル−スペルミン(酢酸塩)(セラミドカルバモイル−スペルミン、CCS)は、Biolab Ltd., Jerusalem, Israelから購入する。表記されている場合には、1/1から4/1のモル比の脂質/ヘルパー比で、ヘルパー脂質(DOPE、Chol)を使用した。
特定病原体除去(SPF)のメスのBALB/cマウス、6から8週齢、及びC57BL/6マウス、18月齢を使用した(5から10匹/グループ)。動物は、SPF条件下で飼育した。
インフルエンザ抗原のリポソーム/脂質集合物中へのカプセル化
巨大な(平均直径0.1から5μm)異種(標準的なサイズでない)小胞中に、HN抗原(上記参照)をカプセル化した。全てのワクチン調合物の調製のために、以下の手順を一般に使用した。1mLの第3ブタノール中に脂質(10から30mg)を溶かした後、ろ過(GF92, Glasforser, Vorfilter no. 421051, Schleicher & Schuell, Dassel, Germany)によって滅菌した。無菌脂質溶液を−70℃で凍結した後、24時間凍結乾燥して、乾燥を完了した。乾燥された脂質は、有意な(<5%)脂質分解又は「カプセル化」能を失わずに、2年超、4℃で保存することができた。必要となった時点で、3/1から800/1の脂質:抗原(タンパク質)w/w比で、脂質粉末を抗原溶液(PBS pH7.2中)で水和した。20から50μLずつ増量しながら、抗原溶液を段階的に添加し、0.5から1mLの最終容量になるまで、各添加後に激しくボルテックス攪拌を行った。一部の実験では、乾燥された脂質をPBSで水和し、あらかじめ形成された「空の」脂質集合物を、抗原溶液と混合した。この混合物を1から2分間ボルテックス攪拌し、30から60分以内に使用した。
遊離(F−HN)および集合された/リポソームの(Lip−HN)ワクチン(0.25−4μg抗原/株/投薬および0.075−1.2mg脂質/投薬)を、筋肉内に一回(i.m.、30μL中)、3、7若しくは14日の間隔を置いて鼻内に一回若しくは二回(i.n.、5−50μL/鼻孔)、又は1週間の間隔を置いて経口に二回(50μL中)投与した。全ての事例において、PBS中の4%抱水クロラール0.15mLを腹腔内に与え、マウスに軽い麻酔をかけた。経口ワクチン接種の場合、ワクチン接種の30分前に、0.5mLの酸中和剤溶液(8部のHanks平衡塩類溶液+2部の7.5%炭酸水素ナトリウム)をマウスに経口的に処置した。比較のために、標準的な粘膜アジュバントとして、全ての実験で、1μgの/投薬のコレラ毒素(CT, Sigma, USA)を使用した。2つの実験では、遊離およびリポソームのCpG−ODN(ODN 1018,Dr. E. Raz, University of California, San Diego, CA, USAから頂いた。)、10μg/投薬を、 アジュバントとして使用した。
1/10又は1/20試料希釈から開始し、ワクチン接種から4ないし6週後に、個別に又はプールして、血清、肺ホモジネートおよび鼻洗浄液を検査した。血球凝集反応阻害抗体は、1/10の試料希釈から始めて、標準的な血球凝集阻害(HI)アッセイによって測定した。40以上のHI力価(ヒトにおいて防御力価と考えられる。)を有するマウスを、血清変換されたものと定義した。抗原特異的なIgG1、IgG2a、IgA及びIgEレベルは、ELISAによって測定した。対照(抗原+正常なマウス血清、OD<0.1)を0.2 OD上回る吸光度を与える最高の試料希釈を、ELISA抗体力価と考えた。
ワクチン接種から5ないし6週後に取得した脾細胞を、抗原でインビトロ刺激した後の、増殖性応答、IFNγ及びIL−4産生、並びに細胞毒性活性について、検査した。5×10−5Mの2−メルカプトエタノールを加えて(細胞毒性の場合)、又は加えずに、5%(増殖、サイトカインの場合)又は10%(細胞毒性の場合)のウシ胎児血清(FCS)を補充した濃縮RPMI 1640又はDMEM培地中において、37℃で培養を行った。細胞培養は、以下のように行った。(i)増殖:0.2mLの最終容量中、抗原を加え又は加えずに(0.5から5μg/ウェル)、三つ組みで、0.5×106細胞/ウェルをU字型96ウェルプレート中においてインキュベートした。72から96時間後、1μCiの3Hチミジンで、培養物を16時間標識した。結果は、Δcpm=(抗原を加えて培養された細胞の平均カウント/分)−(抗原を加えずに培養された細胞の平均カウント/分)で表す。(ii)サイトカイン:1mLの最終容量中、抗原を加え又は加えずに(5から10μg/ウェル)、二つ組みで、2.5×106から5×106個の細胞/ウェルを24ウェルプレート中においてインキュベートした。48から72時間後に上清を集め、Opt EIA Set(Pharmingen, USA)を用いて、マウスのIFNγとIL−4についてELISAによって調べた。(iii)細胞毒性:応答している脾細胞(2.5×106)を、(ii)と同様に、X/127(H1N1)インフルエンザウイルスを感染された同数の刺激性BALB/c脾細胞とともに、7日間インキュベートした(下記参照)。感染のために、150血球凝集ユニット/1×106脾細胞のウイルスを加えたRPMI 1640培地(FCSなし)中にて、3時間37℃で、時折攪拌しながら、脾細胞をインキュベートした後、洗浄した。続いて、10IU/mLのrhIL−2の存在下、1/4のエフェクター/刺激細胞比で、感染された放射線照射(3,000rad)脾細胞を用いて、初回刺激を受けたエフェクター細胞を、5日間、再度刺激した。標準的な4時間の51Cr放出アッセイを用いて、100/1のエフェクター/標的細胞比で、細胞毒性を測定した。使用した被標識標的細胞は、非修飾P815及びHA2 189−199ペプチド(IYSTVASSLVL、20μg/1x106細胞)で、90分間37℃でパルスされたP815であった。
マウスを麻酔し、マウスに対して感染性であり、A/ニューカレドニアと交叉反応する、再集合体ウイルスX−127(A/北京/262/95(H1N1)×X−31(A/香港/1/68×A/PR/8/34)を用いて、25μLの生ウイルス懸濁液/鼻孔(約107 EID 50(egg−infectious dose 50%))を投与した。第4日目に肺を取り出し、冷PBS中で三回洗浄し、PBS(1.5mL/肺/マウス、1/10希釈と称する。)中でホモジナイズした。各グループのホモジネートをプールし、2000rpmで、30分間、4℃にて遠心し、上清を集めた。段階的な10倍希釈を行い、11日齢の孵化したニワトリの卵の尿嚢中に、二つ組みで、各希釈0.2mLを注射した。37℃で48時間後及び4℃で16時間後、0.1mLの尿膜腔液を取り出し、ニワトリ赤血球(0.5重量%、0.1mL)を用いた血球凝集(室温で30分)によって、ウイルスの存在をチェックした。肺のウイルス力価は、尿膜腔液中にウイルスを産生する(陽性の血球凝集)肺ホモジネートの最高希釈として求める。
空の、又は三価のサブユニットインフルエンザワクチン(HN)と会合したリスサミン−ローダミン標識脂質集合調合物を、20μLの容量でマウスに接種した。1、5又は24時間後にマウスを屠殺し、様々な臓器を取り出した。−20℃で、臓器を一晩保存し、翌朝、溶解緩衝液中でホモジナイズした。得られたホモジネート0.2mLを、エッペンドルフチューブに移し、0.8mLのイソプロパノールを添加し、蛍光プローブを上清中に放出させるために、15分間回転させた。50μLの上清を、384ブラックプレート上に載せ、蛍光を読み取った(Em:545、Ex:596)。
Claims (75)
- 被検体の免疫応答を調節するための薬学的組成物を調製するための、スフィンゴイド−ポリアルキルアミン抱合体の使用。
- 前記スフィンゴイド−ポリアルキルアミン抱合体が、カルバモイル結合を介して、少なくとも一つのポリアルキルアミン鎖を有するスフィンゴイド骨格を含む、請求項1に記載の使用。
- 前記調節が、免疫応答の刺激又は増強を含む、請求項1又は2に記載の使用。
- 生物学的に活性な分子と組み合わせた、請求項1から3の何れか1項に記載の使用。
- 前記生物学的に活性な分子が、生理的pHにおいて、正味負の双極子モーメント若しくは正味負の電荷を有するか、又は正味負の電荷を有する少なくとも一つの領域を含有する、請求項4に記載の使用。
- 前記生物学的に活性な分子が、免疫調節アミノ酸分子、核酸分子又は低分子量化合物である、請求項4又は5に記載の使用。
- 前記生物学的に活性な分子が、抗原性タンパク質、抗原性ペプチド、抗原性ポリペプチド、炭水化物又は免疫賦活薬である、請求項6に記載の使用。
- 前記核酸分子が、オリゴデオキシヌクレオチド(ODN)である、請求項6に記載の使用。
- 免疫賦活薬と組み合わせた、請求項1から8の何れか1項に記載の使用。
- 前記スフィンゴイド−ポリアルキルアミン抱合体が脂質集合物を形成する、請求項1から9の何れか1項に記載の使用。
- 前記スフィンゴイド−ポリアルキルアミン抱合体が、小胞、ミセル又はそれらの混合物を形成する、請求項10に記載の使用。
- 前記スフィンゴイドが、セラミド、ジヒドロセラミド、フィトセラミド、ジヒドロフィトセラミド、セラミン、ジヒドロセラミン、フィトセラミン、ジヒドロフィトセラミンから選択される、請求項1から11の何れか1項に記載の使用。
- 前記スフィンゴイドがセラミドである、請求項12に記載の使用。
- 前記ポリアルキルアミンが、スペルミン、スペルミジン、ポリアルキルアミン類縁体又はそれらの組み合わせから選択される、請求項1から13の何れか1項に記載の使用。
- 前記スフィンゴイド−ポリアルキルアミン抱合体が、生物学的に活性な分子とともに凍結乾燥されるか、又は前記生物学的に活性な物質が、予め形成されたスフィンゴイド−ポリアルキルアミン抱合体集合物と混合される、請求項4から14の何れか1項に記載の使用。
- 前記スフィンゴイド−ポリアルキルアミン抱合体が、N−パルミトイル D−エリスロスフィンゴシルカルバモイル−スペルミン(CCS)である、請求項1から15の何れか1項に記載の使用。
- ワクチンを調製するための、請求項1から16の何れか1項に記載の使用。
- インフルエンザワクチンを調製するための、請求項17に記載の使用。
- 前記生物学的に活性な物質が、インフルエンザウイルスに由来するか、又はインフルエンザウイルスに由来する分子の類縁体である、請求項18に記載の使用。
- 前記生物学的に活性な物質が、ヘマグルチニンとノイラミニダーゼ(HN)の組み合わせである、請求項19に記載の使用。
- 鼻内又は筋肉内ワクチン接種を調製するための、請求項1から20の何れか1項に記載の使用。
- 被検体のインフルエンザウイルスに対する免疫応答を増強又は刺激するための薬学的組成物を調製するための、N−パルミトイル D−エリスロスフィンゴシルカルバモイル−スペルミン(CCS)の使用。
- 生物学的に活性な分子とともに、治療的有効量のスフィンゴイド−ポリアルキルアミン抱合体を被検体に与えることを含む、被検体の免疫応答を調節する方法。
- 前記スフィンゴイド−ポリアルキルアミン抱合体が、カルバモイル結合を介して、少なくとも一つのポリアルキルアミン鎖を有するスフィンゴイド骨格を含む、請求項23に記載の方法。
- 前記調節が、免疫応答の刺激又は増強を含む、請求項23又は24に記載の方法。
