Abstract
【解決手段】 本発明に係る、インプラント可能で生体親和性を有するスカフォールドは、生体親和性を有する多孔質のポリマー基質と、このポリマー基質によりカプセル封入され、その内部に配された、生体親和性を有する、多孔質の、繊維からなるマットと、組織スカフォールド内に播種された、複数の哺乳動物細胞とを含む。このスカフォールドを使用して、哺乳動物の疾病または構造欠陥を治療する。

Description
90/10のPGA/PLA繊維から構成されたニードルパンチ不織マット(厚さ2mm)を下記のようにして作製した。PGA/PLA(90/10)コポリマーを溶融押し出しし、ヤーンを造る慣用的方法により、連続マルチフィラメントヤーンとし、ついで、配向して、強度、伸度および破断に要するエネルギーを高めた。このヤーンは直径が約20μmのフィラメントからなっていた。これらのヤーンを一様な2インチ(換算値は5.08cm)の長さに切断、捲縮して、2インチの長さのステープルファイバーとした。
凍結乾燥により発泡体としたポリマーが、バーミンガムポリマーズ社、バーミンガム、アラバマ州が製造した、I.V.が1.45dL/gの60/40のPLA/PCLコポリマーである以外は実施例1の処理と同様にして、生分解性複合スカフォールドを作製した。この複合スカフォールドの孔サイズは、水銀ポロシメトリー分析法で決定した。孔サイズは1μm〜300μmの範囲にあり、メジアン孔サイズは45μmであった。
凍結乾燥により発泡体としたポリマーが、バーミンガムポリマーズ社、バーミンガム、アラバマ州が製造した、I.V.がそれぞれ1.50dL/gと1.45dL/gの、60/40のPLA/PCLコポリマーと35/65のPCL/PGAコポリマーとの50:50ブレンド物である以外は実施例1の処理と同様にして、生分解性複合スカフォールドを作製した。
凍結乾燥により発泡体としたポリマーが、I.V.が1.50dL/gの60/40のPLA/PCL(バーミンガムポリマーズ社、バーミンガム、アラバマ州)とI.V.が1.78dL/gの85/15のPLA/PGA(ピュラック社(Purac),リンコルシャイン(Lincolshine),イリノイ州)との70:30ブレンド物である以外は実施例1の処理と同様にして、生分解性複合スカフォールドを作製した。
凍結乾燥により発泡体としたポリマーが、I.V.が1.50dL/gの60/40のPLA/PCL(バーミンガムポリマーズ社、バーミンガム、アラバマ州)とI.V.が1.78dL/gの85/15のPLA/PGA(ピュラック社,リンコルシャイン,イリノイ州)との30:70ブレンド物である以外は実施例1の処理と同様にして、生分解性複合スカフォールドを作製した。
凍結乾燥により発泡体としたポリマーが、I.V.が1.50dL/gの60/40のPLA/PCL(バーミンガムポリマーズ社、バーミンガム、アラバマ州)とI.V.が1.78dL/gの85/15のPLA/PGA(ピュラック社,リンコルシャイン,イリノイ州)との50:50ブレンド物である以外は実施例1の処理と同様にして、生分解性複合スカフォールドを作製した。
ドライレイのニードルパンチ不織マットをPDO繊維から構成した以外は実施例1の処理と同様にして、生分解性複合スカフォールドを作製した。
ドライレイのニードルパンチ不織マットをPGA繊維から構成した以外は実施例1の処理と同様にして、生分解性複合スカフォールドを作製した。
ドライレイのニードルパンチ不織マットをPGA繊維から構成した以外は実施例4の処理と同様にして、生分解性複合スカフォールドを作製した。
この例は、複合スカフォールド中のポリマー発泡体またはドライレイのニードルパンチ不織マットの構成が、軟骨細胞のインビトロの反応に影響を与えたことを示すものである。
この例は、複合スカフォールド中のポリマー発泡体またはドライレイのニードルパンチ不織マットの構成が、セルトリ細胞のインビトロの反応に影響を与えたことを示すものである。
(1)前記スカフォールドが生分解性を有する、請求項1に記載のスカフォールド。
(2)前記ポリマー基質が生分解性のポリマーからなる群から選ばれたポリマーを含み、前記繊維からなるマットが、リン酸カルシウムを含む生分解性ガラスとセラミックスおよび生分解性ポリマーからなる群から選ばれた材料を含む繊維を含有する、請求項1に記載のスカフォールド。
(3)前記ポリマー基質および前記繊維からなるマットが、生分解性ポリマーを含む、実施態様(2)に記載のスカフォールド。
(4)前記生分解性ポリマーが、脂肪族ポリエステル、ポリアルキレンオキサレート、ポリアミド、ポリカーボネート、ポリオルトエステル、ポリオキサエステル、ポリアミドエステル、ポリアンハイドライドおよびポリフォスファゼンのホモポリマーおよびコポリマーからなる群から選ばれたものである、実施態様(3)に記載のスカフォールド。
(5)前記繊維からなるマットが90/10のポリグリコリド/ポリ乳酸のコポリマーを含む、実施態様(4)に記載のスカフォールド。
(7)前記ポリマー基質が、ポリ乳酸/ポリグリコリドのモル比が約95/5〜約85/15の範囲のポリ乳酸とポリグリコリドのコポリマーを含む、実施態様(4)に記載のスカフォールド。
(8)前記多孔質のポリマー基質が、ポリカプロラクトン/ポリグリコリドのモル比が約35/65〜約45/55の範囲のポリカプロラクトンとポリグリコリドのコポリマーを含む、実施態様(5)に記載のスカフォールド。
(9)前記多孔質のポリマー基質が発泡体を含む、実施態様(8)に記載のスカフォールド。
(10)前記多孔質のポリマー基質が、ポリ乳酸/ポリカプロラクトンのモル比が約35/65〜約65/35の範囲のポリ乳酸とポリカプロラクトンのコポリマーを含む、実施態様(4)に記載のスカフォールド。
(12)前記哺乳動物細胞が、骨髄細胞、平滑筋細胞、間質細胞、幹細胞、間葉幹細胞、滑膜由来の幹細胞、胚幹細胞、血管細胞、軟骨細胞、骨芽細胞、脂肪組織に由来する前駆細胞、骨髄由来の前駆細胞、腎臓細胞、腸細胞、膵島、β細胞、セルトリ細胞、末梢血前駆細胞、線維芽細胞、グロムス細胞、ケラチン生成細胞、髄核細胞、線維輪細胞、線維軟骨細胞、成人の組織から分離した幹細胞、卵円形細胞、神経幹細胞、グリア細胞、マクロファージおよび遺伝学的に形質転換した細胞からなる群から選ばれたものである、請求項1に記載のスカフォールド。
(13)前記細胞が、膵島とセルトリ細胞とからなる群から選ばれたものである、実施態様(12)に記載のスカフォールド。
(14)前記細胞が、成人の神経幹細胞、胚幹細胞およびグリア細胞からなる群から選ばれたものである、実施態様(12)に記載のスカフォールド。
(15)さらに、生物学的因子を含む、請求項1に記載のスカフォールド。
(17)さらに、治療薬を含む、請求項1に記載のスカフォールド。
(18)前記治療薬が、抗拒絶反応剤、鎮痛剤、酸化防止剤、抗細胞消滅剤、エリスロポエチン、抗炎症剤、抗腫瘍壊死因子α、抗CD44、抗CD3、抗CD154、p38キナーゼ抑制因子、JAK−STAT抑制因子、抗CD28、アセトアミノフェン、細胞増殖抑制剤、ラパマイシンおよび抗IL2剤からなる群から選ばれたものである、実施態様(17)に記載のスカフォールド。
(19)前記スカフォールドが生分解性を有する、請求項2に記載の方法。
(20)前記ポリマー基質が生分解性のポリマーからなる群から選ばれたポリマーを含み、前記繊維からなるマットが、リン酸カルシウムを含む生分解性ガラスとセラミックスおよび生分解性ポリマーからなる群から選ばれた材料を含む繊維を含有する、請求項2に記載の方法。
(22)前記生分解性ポリマーが、脂肪族ポリエステル、ポリアルキレンオキサレート、ポリアミド、ポリカーボネート、ポリオルトエステル、ポリオキサエステル、ポリアミドエステル、ポリアンハイドライドおよびポリフォスファゼンのホモポリマーおよびコポリマーからなる群から選ばれたものである、実施態様(21)に記載の方法。
(23)前記繊維からなるマットが90/10のポリグリコリド/ポリ乳酸のコポリマーを含む、実施態様(22)に記載の方法。
(24)前記ポリマー基質が、ポリカプロラクトン/ポリグリコリドのモル比が約35/65〜約45/55の範囲のポリカプロラクトンとポリグリコリドのコポリマーを含む、実施態様(23)に記載の方法。
(25)前記ポリマー基質が発泡体を含む、実施態様(24)に記載の方法。
(27)前記疾病が糖尿病である、請求項2に記載の方法。
(28)前記スカフォールドがセルトリ細胞と膵島とを播種したものである、実施態様(27)に記載の方法。
(29)前記デバイスがさらに生物学的因子を含む、実施態様(27)に記載の方法。
(30)前記スカフォールドが生分解性を有する、請求項3に記載の方法。
(31)前記哺乳動物細胞が、骨髄細胞、平滑筋細胞、間質細胞、幹細胞、間葉幹細胞、滑膜由来の幹細胞、胚幹細胞、血管細胞、軟骨細胞、骨芽細胞、脂肪組織に由来する前駆細胞、骨髄由来の前駆細胞、腎臓細胞、腸細胞、膵島、β細胞、セルトリ細胞、末梢血前駆細胞、線維芽細胞、グロムス細胞、ケラチン生成細胞、髄核細胞、線維輪細胞、線維軟骨細胞、成人の組織から分離した幹細胞、卵円形細胞、神経幹細胞、グリア細胞、マクロファージおよび遺伝学的に形質転換した細胞からなる群から選ばれたものである、請求項3に記載の方法。
(32)前記構造欠陥が、関節軟骨、半月板および骨からなる群から選ばれた組織中のものである、請求項3に記載の方法。

Claims (3)
- インプラント可能で生体親和性を有するスカフォールドにおいて、
生体親和性を有する多孔質のポリマー基質と、
当該ポリマー基質によりカプセル封入され、その内部に配された、生体親和性を有する、多孔質の、繊維からなるマットと、
当該組織スカフォールド内に播種された、複数の哺乳動物細胞と
を含むスカフォールド。 - 哺乳動物に生体親和性を有するスカフォールドを埋植することを含む、哺乳動物の疾病を治療する方法において、当該スカフォールドが、
生体親和性を有する多孔質のポリマー基質と、
当該ポリマー基質によりカプセル封入され、その内部に配された、生体親和性を有する、多孔質の、繊維からなるマットと、
当該組織スカフォールド内に播種された、複数の哺乳動物細胞と
を含む、哺乳動物の疾病を治療する方法。 - 哺乳動物に生体親和性を有するスカフォールドを埋植することを含む、哺乳動物の構造欠陥を治療する方法において、当該スカフォールドが、
生体親和性を有する多孔質のポリマー基質と、
当該ポリマー基質によりカプセル封入され、その内部に配された、生体親和性を有する、多孔質の、繊維からなるマットと、
当該組織スカフォールド内に播種された、複数の哺乳動物細胞と
を含む、哺乳動物の構造欠陥を治療する方法。
