ソーム
ラグメント(「イムノリポソーム」)を調製するための方法、イムノリポソーム
を使用するインビトロトランスフェクションのための方法、およびインビボにお
ける全身性遺伝子送達のための方法を提供するものである。本発明のリポソーム
は、標的化遺伝子送達を行うのに有用であり、全身投与後の効率的な遺伝子発現
のために効果的である。送達系の特異性は、標的化抗体フラグメントから誘導さ
れる。

り有望ではあるが、一方でこの有望性を実現し得る前に指向すべき多くの問題を

存在する場所はどこでも腫瘍細胞を選択的に標的化し得るものである。

スの諸方式に対して多くの利点を提供する。最も重要なことは、リポソームは、

。リポソームにより腫瘍細胞を標的化することは、その成分が腫瘍細胞に選択的
に送達するようにリポソームを修飾することによって達成し得る。癌細胞の外側
表面に存在する特異的分子に関する重要な知識基盤がここに存在する。かかる細
胞表面分子は、腫瘍細胞の外表面に存在する該分子は通常細胞上にあるものとは
異なるので、リポソームを腫瘍細胞に標的化するために使用することができる。

ある領域においては不十分である(Ledley FD. et al. Hum. Gene Ther(1995) 6

なカチオンリポソームの形態は以下を包含する:調製の簡易性;大量のDNAを
複合体化する能力;いかなるタイプおよびサイズのDNAまたはRNAの使用の
際にも融通性があること;非分割細胞を含む多くの異なるタイプ細胞をトランス
フェクションする能力;免疫原性もしくは生物学的危険物質活性の欠如(Felgne
r PL, et al., Ann NY Sci.(1995)772:126-139;Lewis JG, et al.,Proc.Natl. A
cad. Sci USA(1996)93:3176-3181)。ヒト腫瘍治療に関する知見から、より重要
性があるものは、カチオンリポソームは、インビボ遺伝子送達に関して安全かつ
効率的であることが証明されてきた(Aoki K et al., Cancer Res.(1997)55:381
0-3816; Thierry AR, Proc. Natl. Acad. Sci.USA(1997)92:9742-9746)。30
以上の臨床試験が、現在遺伝子治療に関してカチオンリポソームを用いて行われ
ており(Zhang W et al.,Adv. Pharmacology (1997)32:289-333;RAC Committee
Report : Human Gene Therapy Protocols-December 1998)、小分子治療物質(
例えば、抗菌および従来の化学療法)を送達するためのリポソームはすでに市販
されている(Allen TM, et al., Drugs(1997)54 Suppl 4:8-14)。

によるリガンドとの初期結合に続く結合複合体の内在化によって細胞へのDNA
導入を助ける。トランスフェリンおよび葉酸を含む多様なリガンドが、そのリポ
ソーム標的化能力に関して試験されてきた(Lee RJ, et al., J. Biol Chem.(19
96)271:8481-8487)。トランスフェリン受容体(TfR)レベルは、前立腺癌を
含む種々の腫瘍タイプの腫瘍細胞において上昇するが、ヒトリンパ節および骨の
転移から派生した前立腺癌細胞系でもそうである(Ke HN et al.,J. Urol(1990)
143:381-385); Chackal-Roy M et al., J.Clin. Invest.(1989)84:43-50;Rossi
MC, et al., Proc. Natl. Acad Sci. USA(1992)89:6197-6201;Grayhack JT, et
al., J. Urol.(1979)121:295-299)。また、上昇したTfRレベルは、腫瘍細胞
の促進または増殖能と相関する。そのため、TfRレベルは、予後の腫瘍マーカ
ーとして有用であると考えられ、TfRは悪性腫瘍細胞の治療の際に薬物送達に
関する有力な標的である(Miyamoto T et al., Int J. Oral Maxillofac. Surg.(
1994)23:430-433;Thorstensen K. et al., Scand J. Clin. Lab. Invest.Suppl(
1993)215:113-120)。我々の研究室では、リガンドを持たないカチオンリポソー
ムではわずか5〜20%であるのと比較して、SCCHNで腫瘍細胞トランスフ
ェクション効率が60〜70%であるトランスフェリン複合体カチオンリポソー
ムを調製した(Xu L, et al., Hum. Gene Ther.(1997)8:467-475)。
も、それらを特異的な腫瘍細胞表面抗原(これは受容体に制限するものではない
)(Allen TM, Biochim. Biohpys. Acta (1995)1237:99-108)を指向しうるリポ
ソーム表面へ結合することができる(Allen TM et al.,(1995) Stealth Liposom es ,pp.233-244)。これらの「イムノリポソーム」、特に立体的に安定化したイ
ムノリポソームは、特定の標的細胞群に治療薬物を送達することができる(Alle
n TM et al.,(1995) Stealth Liposomes,pp.233-244)。Parkら(Park JW, et a
l., Proc. Natl. Acad.Sci.USA(1995)92:1327-1331)は、リポソームに接合した
抗HER-2モノクローナル抗体(Mab)Fabフラグメントが、肺癌細胞系S
K-BR-3を重複発現するHER-2特異的に結合し得ることを明かにした。イ
ムノリポソームは、被覆ピット経路を介する受容体介在エンドサイトーシスによ
って、また膜融合によっても効率的に内在化することが見出された。さらに、抗
HER−2Fabフラグメントのアンカリングによって、その阻害効果は増強し
た。ドキソルブチンを有する抗HER−2イムノリポソームは、インビボおよび
インビトロにおける標的細胞に対して重要な特異的細胞毒性を示した(Park et
al.,Proc. Natl Acad.Sci.USA(1995)92:1327-1331)。さらに、Suzuki et al.,(
Suzuki S, et al.,J.Cancer (1997)76:83-89)は、インビトロにおいてヒト白血
球細胞においてさらに効率よいドキソルブチンを送達するために抗トランスフェ
リン受容体モノクローナル抗体接合イムノリポソームを用いた。Huwyler J, ら(
Huwyler J, et al., Proc.Natl. Acad. Sci. USA (1996) 93:14164-14169)は、
インビボにおいてラットグリオーマ細胞(RT2)に、ダウノマイシンを送達す
るために抗TfRモノクローナル抗体イムノリポソームを用いた。このペグ化(P
EGlated)イムノリポソームは、通常の組織および臓器において薬物の濃度低下を
生じる。これらの研究は、腫瘍標的化薬物送達のためのイムノリポソームの有用
性を示すものである。SuzukiらおよびHuwylerらによって使用されたイムノリポ
ソーム複合体は、彼らが、アニオンリポソームを使用したこと、および彼らの方
法では核酸を送達することができない点で、本発明に記載のものとは異なってい
ることに注意すべきである。

胎児性抗原に対して指向したscFv、HER-2、CD34、黒色腫関連抗原
およびトランスフェリン受容体を提示するように作製したレトロウイルスベクタ
ーが開発された(Jiang A, et al.,J. Virol(1998)72:10148-10156, Konishi H.
et al., Hum Gene Ther.(1994)9:235-248:Martin F, et al., Hum. Gene Ther.
(1998)9:737-746)。ウイルスを指向したこれらのscFvは、特別な抗原を発
現する細胞のタイプに特異的に結合および感染する標的を示している。さらに、
少なくとも癌胎児性抗原の場合、scFvは、親の抗体として同じ細胞特異性を
持つことが示されている(Nicholson IC, Mol. Immunol.(1997)34:1157-1165)
。
標的化遺伝子送達のための有望なシステムであると考えられる。

に基に、これらのTfRscFvを組込んだイムノリポソームDNA複合体は、
完全なTf分子を持つ同じリポソームDNA複合体以上にトランスフェクション
効率のより高いレベルのものを作ることができる。そのため、本発明の一態様に
おいて、本発明のイムノリポソームは、トランスフェリン受容体を発現する多様
な哺乳動物細胞型の高いトランスフェクション効率のためのキットを作成するた
めに使用することが可能である。本発明の一態様において、我々は、リピドタグ
を有するscFvタンパク質を構築したが、これはリピドが細菌細胞によって自
然に加えられ、scFvを不活性化させ得る化学反応を回避しながらリポソーム
へのscFvの組込みを容易にするようにした。
脂質膜可溶化法およびダイレクト・アンカー法である。この脂質膜可溶化法は、
界面活性剤の透析法を修飾したもので、Laukkanen ML, et al., (Laukkanen ML,
et al., Biochemistry (1994)33:11664-11670)およびKruf et al.,(de Krurif
et al.,FEBS Lett.(1996)399:232-236)によって、中性またはアニオンリポソー
ムに関して記載されており、この両方法は出典明示により本明細書の一部とする
。この方法は、リピドタグscFvとカチオンリポソームとを結合するために適
当である。脂質膜可溶化法において、クロロホルム中の脂質は、減圧下蒸発し、
丸底ガラスフラスコ内に乾燥脂質膜を得る。次いで、脂質膜0.5〜4%、好ま
しくは1%のリピド修飾scFvを含むβ-D-グルコシド(OG)を用いて可溶
化し、撹拌にかける。滅菌水で希釈後、該溶液を簡単に音波処理し、透明にする
。
ダイレクト・アンカー法であり、これはE. Coliリポタンパク質N末端の9個の
アミノ酸とscFv(lpp-scFv)または他のリピド修飾抗体またはフラ
グメントを結合するため、これを予め形成したリポソームに結合するために特異
的に有用である。scFvと予め形成したリポソームを結合するために、1:3
〜1:10の体積比で、1%のOG中リピド修飾scFvは撹拌にかけながら予
め形成したリポソームに添加される。該混合物は、さらに5〜10分間撹拌にか
けられ、scFvイムノリポソームの透明溶液を得る。残存OGおよび非複合体
化scFvは、クロマトグラフィーで除去することができるが、後の過程で干渉
しない。分離実験、即ちCentricon-100(Amicon)を用いる限外濾過、Ficoll−
400濾過(Shen DF, et al., Biochem. Biophys. Acta(1982)689:31-37)また
はSepharose CL-4B(Pharmacia)クロマトグラフィーにより、カチオンリポソー
ムに、添加された全リピドタグscFv分子を結合または接続されていることを
示す。これは、中性またはアニオンリポソームに対するlpp-scFvの非常
に低い結合率を改善する。そのためこの改善は、結合scFvを除去するための
さらなる精製工程を不必要とした。
メントまたは別のペプチド/タンパク質リガンドは、本発明において有用である
。他のリピド修飾方法は、Liposome Technology, 2nd Ed.Gregorisdis, G., Ed
., CRC Press, Boca Raton, FL, 1992に記載の、リピド鎖と抗体もしくはフラグ
メントとの直接的接合を包含する。
該タンパク質は、E. Coliの封入体で発現し、活性scFvを生成して再折りた
たみが生じる。C末端システインは、scFvとリポソームの接合を助ける遊離
メルカプト基を提供する。2つの方法が、接合過程において使用し得る。1)前
結合法:第一段階は、マレイミドイル(maleimidyl)基または別のメルカプト反
応基を含むカチオンリポソームとのscFv-SHを接合し、scFvリポソー
ムを生成する。次いで、核酸はscFvリポソームに添加し、scFv-リポソ
ーム-DNA複合体を形成する。2)後結合法:この方法は、はじめに、核酸と
カチオンリポソームを複合体化し、縮合構造(condensed structure)を形成す
る。次いで、scFv-SHは、DNAリポソーム複合体の表面上で結合し、s
cFv-リポソーム-DNAを生成する。後結合法は、scFvがDNA複合体化
後に結合するように設計される。そのため、この方法は、標的化リガンドscF
vの使用および制御された複合体の内部構造を良好にしうる。

る。好ましい複合体は、注目とする部位、好ましくは癌細胞を示す細胞、より好
ましくはトランスフェリン受容体に結合する細胞を標的にする。標的化剤は、ト
ランスフェリン受容体に好ましく結合する該抗体または該抗体フラグメントであ
る。核酸は、治療薬であり、好ましくはDNA分子であり、より好ましくは野生
型p53分子をコードする核酸である。核酸イムノリポソーム複合体は、好まし
くは治療用組成物は、全身投与、好ましくは静脈注射することが可能である。

用する方法を目指すものである。様々なタグをつけたイムノリポソームおよびs
cFvをリポソームに結合する多様な方法を包む多様な態様を開示する。イムノ
リポソームは、リピドタグを含むか、または還元基、好ましい態様においては遊
離メルカプト基と結合してもよい。
性の前立腺癌を含めて、ヒト腫瘍の50%以上と関連がある。また、p53にお
ける異常は、多様なタイプの悪性腫瘍における貧困な予後に関連する。そのため
、全身投与し、wtp53の機能を効果的に修復させるように腫瘍に対して効果
的に遺伝子治療の標的とする能力は、癌治療における重要な治療態様であろう。
即ち、本発明の方法によって生成されたイムノリポソームは、wtp53機能の
修復のみならず他の治療薬剤遺伝子に関して腫瘍標的化全身性送達ビヒクルとし
ても効率的な新規治療態様として有用であろう。 本発明は次の実施例で説明する。
(ssLPP)と該scFvクローニング部位(de Kruif et al. FEBS Lett. (19
96)399:232-236)との間のアミノ酸リンカー配列を含むベクターpLP1を使用
した。このベクターは、発現したscFvの精製および検出のために使用し得る
c−mycとHis6タグ配列の両方を含む(図1)。
発現ベクターは、DNA結合タンパク質と結合する5E9抗体に関する単鎖フラ
グメントを含み(Haynes, et al. J. Immunol(1981) 127:347-351)、ヒトトラ
ンスフェリン受容体(TfR)を認識する。このベクターは、DNA結合タンパ
ク質に関する配列を含み、pDFH2T−vecOK中のscFv配列をフランキ
ングする独特な制限酵素部位は存在しない。さらに、我々は、NotIを含む3’
プライマー
クローニングした。プライマーRB551およびRB552を用いるPCR増幅
によって、81塩基のMetから821塩基のLysのpDFH2T−vecOK
由来TfRに関するscFvを増幅した。pLP1ベクターは、Laukkenen ML, e
t al (Laukkanen ML, et al., Biochemistry (1994)) およびde Kruif et al(Kr
uif et al., FEBS Lett.(1996)399:232-236) によって述べられているように、
E.coliリポプロテインシグナルペプチド(ssLPP)とE.coliリポプリテイン
N末端の9個のアミノ酸(LPP)に関する配列をも含む。これらの配列の挿入
は、E.coli宿主において発現シグナルの脂肪酸アシル化を引き起こし、細菌膜
内にその導入がおこる。該ベクターは、不安定でない10個のアミノ酸リンカー
配列を有し、リピドタグ部位とscFvとの間の空間を広げる。細菌膜由来のリ
ピド修飾scFCv配列の精製によって、リポソーム中に結合するか挿入され得
る活性分子となる。

が好ましい。多くのクローンを選別し、最良の収率でscFvを生成する一つの
クローンが選択された。リピド修飾scFv(lpp-scFv)を、Kruif ら(
de Kruif et al., FEBS Lett.(1996)399:232-236)によって記載されたようにTr
iton X-100を使用する細菌膜から単離した。精製するために、単一コロニー
を5%グルコースと適当な抗菌剤を含むLB(200μl)中で再懸濁した。該混
合物を、5%グルコースと適当な抗菌剤を含む2枚のLB寒天プレート(99mm)
上で塗布し、一晩増殖した。翌日、該細胞を、プレートから洗浄し、0.1%グ
ルコースと適当な抗菌剤を含むLB(全量5リットル)の播種に使用した。該培養
物を、OD600で0.5〜0.7に達するまで、6時間、25℃、200rpmで
増殖した。IPTGを、1mMの終濃度まで添加し、該培養物をさらに一晩イン
キュベートした。翌日、該細菌培養物を、遠心分離で回収し、200mlの分解
用緩衝液で、室温で30分間分解した。該試料を、氷上で冷やしながら28ワッ
トで5分間音波処理を行った。該分解緩衝液は、20mM HEPES(pH7.
4〜7.9)、0.5mM NaCl、10%グリセロール、0.1mM PMSF
を含む。引用したプロトコールから唯一逸脱したことは、各々20および200
mM イミダゾールを含む、20mM HEPES(pH7.4〜7.9)、0.5
M NaCl、10%グリセロール、0.1mM PMSF、1%n−オクチルβ
−D−グルコシド(OG)、そして10%グリセロールを含む緩衝液でメタル・ア
フィニティーカラムの洗浄および溶出を包含する。lpp−scFvの溶出試料
は、抗c−myc抗体9E10を用いてSDS−PAGEおよびウエスタン・ブ
ロットで分析し、精製scFvが約30kDaのバンドを示すことを確認した。
溶化法に関する詳細な方法を開示する。クロロホルム中のリピド(5μl、DO
TAP/DOPE、1:1モル比)を、減圧下で蒸発させ、丸底ガラスフラスコ中
で乾燥リピド膜を得た。リピド膜に、0.5mlの、リピド修飾scFvを含む
1%OG、20mM HEPES、150mM NaCl(pH7.4)を添加した
。これを、室温で、10〜20分間インキュベーションし、次いでリピド膜を撹
拌で混合し、溶解した。次いで、滅菌水(2ml)を添加し、scFvリピド混
合物を希釈した。該溶液を、20℃で、バス型ソニケーターにより簡単に音波処
理し、透明にした。該scFv−リポソームは限定した量の界面活性剤OG残り
を有する透明な溶液である。該OGおよび非複合体scFvを、SepharoseCL-4B
もしくはSepharoseS500を使用するクロマトグラフィーによって除去し得るが、
それらは後の使用において干渉しない。
クト・アンカー法を提示する。丸底ガラスフラスコ内の乾燥リピド膜として調製
したリピド(20μmol、リピドA−H、組成および比は以下を参照)を、純水
(10ml)に添加し、バス型ソニケーターで、室温(リピドA、B、C)もし
くは65℃(リピドD、E、G、Hもしくは全てのコレステロール(Chol)との
組成物)で、10〜30分間音波処理した。調製した該カチオンリポソームは、
透明な溶液であり、その組成および比は以下である。
:3〜1:10の体積比で、1%OGを含む20mM HEPES、150mM N
aCl(0.5ml、pH7.4)中の該リピド修飾scFv(IPP−scFv)を添加し
、リポソームを予め形成する。該混合物を、さらに1〜5分間撹拌にかけ、sc
Fvイムノリポソームの透明溶液を得た。残存OGと非複合体scFvは、クロ
マトグラフィーによって除去し得るが、それらは後の使用において干渉しない。
分離実験、すなわちCentricon−100(Amicon)を用いる限外濾過、Ficoll−4
00フローテション(Shen DF, et al.,Biochem Biophys Acta (1982)689:31-37
)またはSepharose CL-4B(Pharmacia)クロマログラフィーは、実際に、添加した
全てのリピド標的化scFvが、カチオンリポソームに結合またはアンカリングして
いることを示した。これは、対照的に、アニオンもしくは中性リポソームとlpp-
scFvと結合速度は非常に低い。そのため、scFvを除去するために、さら
なる精製工程は必要でない。
ソームの特徴を提供するものである。該ヒト前立腺癌細胞系DU145および頭
部および頚部のヒト扁平上皮細胞癌細胞系JSQ−3は、これらの研究に対する
TfR+標的細胞として機能する。
ムに結合する前後のlpp−scFvの免疫活性を測定した。96ウェルプレー
トにおける融合性JSQ−3細胞を、PBS中の0.5%のグルタルアルデヒド
を用いて、10分間、室温で固定した。該プレートを、PBS中の5%メス子牛
血清(FBS)を用いて、30℃で30分間ブロッキングした。lpp−scF
v、scFv−イムノリポソームおよびリポソームを、2つのウェルに添加し、
4℃で一晩インキュベーションした。3回のPBS洗浄の後、抗−c−mycモ
ノクローナル抗体を、PBS中3%のFBSの各ウェルに添加し、37℃、60
分間インキュベーションした。3回のPBS洗浄の後、3%のFBSで希釈した
HRP標識したヤギ抗マウスIgG(Sigma)を各プレートに添加し、30分間3
7℃でインキュベーションした。該プレートを、3回PBSで洗浄し、クエン酸
緩衝液(Sigma)中の基質(100μl)と0.4mg/mlOPDを各ウェルに添
加した。発色を2M スルホン酸(100μl)の添加により停止した。該プレー
トを、490nmで、ELISAプレートリーダー(Molecular Devices Corp)で
読取った。間接的細胞ELISAにより、抗TfR-scFvがリポソーム複合体中に
組込まれた後にその免疫活性を保持することを示した(表1)。 表1:JSQ−3への抗TfRscFvリポソームの結合
3とDU145細胞をインキュベーションし、次いでFITC標識ヒツジ抗マウ
スIgGとも、4℃でインキュベーションした。JSQ−3細胞とscFv−L
ipAのインキュベーションは、結合しない遊離の抗TfR lpp-scFv抗体に
よって観察されたものと同一の蛍光変化を示し、標的細胞に結合する量が非常に
多いことを示した。対照的に、非標的リポソームは、細胞への結合が非常に少な
いことを示した。類似の結果が、前立腺癌細胞系DU145で観察された。また
、明細書中、該scFv−Lip(A)複合体は、非標的化Lip(A)と比較
すると、腫瘍細胞への充分な結合を明確に示した。FACSデータは表2にまと
めた。表2では、蛍光変化を、蛍光を示す細胞の百分率として表示した。これら
の研究において、陽性細胞の百分率によって示した細胞への結合レベルは、非結
合の遊離scFvのものと類似し、さらにリポソーム複合体中への結合が、抗T
fR lpp−scFvの免疫学的活性を不活性にしないことを示唆した。DU
145を用いるこれらの初期実験に対して使用したリポソーム調製物が、JSQ
細胞に至適化されることを示すべきである。そのため、前立腺腫瘍細胞へのsc
Fv標的化リポソーム複合体の結合は、さらにこの細胞型に対して至適化された
リポソーム複合体の使用によって増幅される。 表2:JSQ−3およびDU145へのTfRscFvリポソームのFACS

)とFITC標識化抗マウスIgGの後に抗c−myc抗体を用いる間接的免疫
蛍光染色によって、JSQ細胞へのscFv標識化リポソーム複合体の結合を確
認した。形質移入細胞において赤色および緑色の蛍光濃度は、抗TfRscFv
(緑色蛍光としてFITC標識化抗c−myc抗体によって示される)が、細胞
への直接的なローダミン標識Lip(A)を示す。さらに、scFv Lip(
A)システムに関する細胞性結合の高いレベルは、高い%の赤/緑二重陽性蛍光
細胞によって示される。
ョンの至適化 我々は、レポーター遺伝子としてβガラクトシダーゼを用いるJSQ細胞にに
おける抗TfRscFvLip(A)複合体のトランスフェクション効率を測定
した。これらの研究において、使用したレポーター遺伝子構築物は、CMVプロ
モータ(pCMVb)制御下でβガラクトシダーゼ遺伝子を含み、その同じプロ
モーターをpCMVp53に使用した(図3)。トランスフェクションした細胞
におけるβGla発現レベル(トランスフェクション効率と相関する)は、βGa
l酵素アッセイで評価した(XuL, et al.,Hum. Gene Ther.(1997)8:467-475)。
表3に示したように、抗TfRscFvのLipAへの結合は、非標的化リポソ
ーム複合体と比較すると、scFv-Lip(A)-pCMVbトランスフェクシ
ョンした細胞における酵素活性が2倍であった。この発現レベルは、トランスフ
ェリン自身を標的化リガンド(Tf-Lip(A)-pCMVb)として使用した場
合に観察される発現レベルと視覚的に実際に同一であった。さらに、遺伝子発現
における増加は、レポーター遺伝子用量依存性であることが示された。表4は、
JSQ-3細胞におけるscFvリポソーム介在トランスフェクションの至適化

を促進した抗−TfRscFv 抗TfR−scFv標的化Lip(A)−p53−3’Adによって遺伝子移入
したJSQ−3腫瘍細胞における外来性wtp53の発現を、p53応答性プロ
モーターの制御の下でルシフェラーゼレポーター遺伝子を含む(Chen L.et al.,
Proc. Natl. Acad. Sci. USA(1998)95:195-200)発現プラスミド(pBP10
0)のトランスフェクションによって評価した。結果として、外来性wtp53
の発現レベルが高くなると、ルシフェラーゼ活性のレベルも高かった。このルシ
フェラーゼ酵素活性は、相対的光単位(RUL)として表現した。上記に示したよ
うに、β−ガラクトシダーゼレポーター遺伝子と、Lip(A)−p53−3’
Ad複合体への標的化剤としての抗TfRscFvの添加は、非標的化Lip(
A)−p53−3’Ad複合体を超えるトランスフェクション効率とwtp53
タンパク質の発現において大きく増加した(ルシフェラーゼ活性のRLUによっ
て表現される)。再び、scFv−Lip(A)−p53−3’Adトランスフ
ェクション細胞におけるp53発現レベルは、トランスフェリンそれ自身を、標
的化リガンド(Lip(A)−p53−3’Ad)として使用した場合に観察さ
れたレベルと非常に類似する。そのため、これらの知見は、抗TfR単鎖抗体方
法が、腫瘍細胞への標的化カチオンリポソーム複合体を標的化し、生物学的に活
性なwtp53遺伝子を送達する有用な方法であることを示す。
53発現
ン(CDDP)の細胞毒性への腫瘍細胞を感受性にした p53誘導アポトーシスの研究に対して、マウス黒色腫細胞系B16を、2セ
ットの6ウェルプレートに、5μgのDNA/2×105細胞の用量で、p53
−3’Ad(図4)と、またはpCMVpRoプラスミド(図3)との抗TfR
scFvイムノリポソーム複合体でトランスフェクションを行った。比較のため
に、トランスフェリン−リポソーム−DNA(LipT−p53もしくはLIp
T−pRo)も、5μgDNA/2×105細胞の用量で遺伝子移入した。24
時間後、CDDPを、一組のプレートに、終濃度10μlまで添加した。薬物添
加後の24および48時間、結合および浮遊細胞の両方を、アポトーシスの染色
のために回収した。細胞を、使用プロトコールに従って、アネキシン(Annexin
)V−FITCキットで染色した(Trevigen, Inc., Gaithersburg, MD)。アネ
キシンVは、リポコルチンであり、天然に存在する血液タンパク質と抗凝固薬あ
る。染色細胞は、FACSサイトメーター上で測定した(Becton and Dickinson
)。表6は、アポトーシス分析の結果の要約である。 表6:リポソームp53遺伝子修復によって誘導されたB16細胞およびCD
DP細胞のアポトーシス
すると、scFvリガンドの添加によって24時間でアポトーシス細胞%におい
て増加しなかった。しかし、48時間までには、リポプレックスへの標的化sc
Fvの添加により、アポトーシス細胞%では、2倍以上に増加した。CDDPを
有する場合、非標的化リポソーム複合体と比較して、24時間であってもアポト
ーシス細胞において顕著に増加した(約1.5倍)。さらに重要なことは、CD
DPと組合せたアポトーシス細胞における増加は、Tf分子それ自身を用いる以
上に標的リガンドとしてTf受容体にscFvを使用すると、さらに顕著であっ
た。この増加は、トランスフェクション効率と相関する。
ンビボにおいて腫瘍細胞へ特異的に送達するために、scFv−Lip(A)p
53−3’Ad(図4)を、JSQ−3皮下異種移植片の腫瘍細胞を有するヌー
ド・マウスに静脈注射をおこなった。注射2日後、腫瘍細胞を摘出し、ウェスタ
ン・ブロッティングのために肝臓および皮膚に加えて該腫瘍細胞からタンパク質
を単離した(Xu L, et al., Hum. Gene Ther.(1997)8:467-475)。当量のタンパ
ク質(100μg、濃度によって決定する場合)を、各々のレーンに流した。図
2に示したように、マウス由来の腫瘍を、scFv−Lip(A)p53−3’
Ad複合体で全身的に処置し、図2でscFv−Lip(A)−p53を標識し
、非常に強いp53シグナルを示し、外来性のwtp53の高い発現レベルに関
するさらに追加の低いバンド表示を示、一方で、外来性マウスwtp53の非常
に低い発現が、皮膚おび肝臓の双方において明らかとなった。対照的に、我々の
初期段階の結果を基に期待していたように、非常低いレベルの外来性p53発現
が非標的化Lいp(A)p53−3’Ad注入マウス、図2中の標識化Lip(
A)−p53から単離した腫瘍細胞において明らかである。すなわち、我々の新
規でかつ独創的な抗TfRlpp−scFvリガンドによって標的化したリポソ
ーム複合体は、外来性遺伝子を、インビボで腫瘍細胞に選択的に送達することが
できることは明らかである。これらの結果は、効率よく標的化する新規の方法の
潜在性は、全身に、カチオンリポソーム複合体を特異的に送達したことを示すも
のである。
Fvタンパク質を結合した。このアプローチは、還元基、例えばメルカプト基に
よるカチオンリポソームへの単鎖タンパク質の接合を目的とする。好ましい態様
において、システイン残基を、TfRscFvタンパク質の3’末端で添加する
。このシステインの還元は、カチオンリポソームに接合し得る遊離のメルカプト
基において生じ、すなわちトランスフェリン受容体を発現する細胞にリポプレッ
クスを標的化するものである。一方、以下の実施例は、還元基としてシステイン
を用い、他の類似の還元基はこの方法で機能することもまた明らかである。
ヒスチジンタグを有する3’システインを含む発現ベクターの構築 実施例1のように、TfRのためのVH−リンカー−VκscFvを、プラス
ミド発現ベクター、pDFH2T−vecOKから得た(実施例1に記載)。PC
R増幅のための5’プライマー

gen)のNcoIおよびNotl部位にクローンした。このベクターは、Nco
I部位の5’、pelBリーダーシグナル配列も含む。発現ベクター中のこの配
列の存在は、ペリプラスミック(peliplasmic)スペースにこのタンパク質を輸
送する。該タンパク質の精製を助けるために、pET26b(+)ベクターもN
otl部位のヒスチジンタグ配列の3’を含む(図5)。
ジンタグを持たない3’システインを含む発現ベクターの構築 ヒトが治療用送達ビヒクルとして使用するためには、ヒスチジンタグなしにT
fRscFvが製造されることが好ましい。さらに、該構築は、実施例8、セク
ション1において記載した。 Aを、最終タンパク質生成物においてこのタグを排除するために修飾した。こ
れを達成するために、上記に記載した(実施例8、セクション1.A)同じ5’
プライマーを使用した。しかし、異なる3’プライマーを用いた。システイン残
基およびNotl制限部位に対するヌクレオチド配列に加えて、このプライマー
た(図6)。即ち、この構築物のタンパク質生成物は、ヒスチジンタグを含まな
い。
5’CBDTMタグを有する3’システインを含む発現ベクターの構築 接合法を用いるカチオンリポプレックスと結合するためにシステイン残基を含
む3つの選択的な構築物もおこなった。この構築物のために、実施例8、セクシ
ョン1.Bにおいて上記記載の2つの同じプライマーを使用した(図7)。即ち
、ヒスチジンタグは、タンパク質生成物中には存在しなかった。しかし、これら
の反応のPCR産物を、異なるベクター、pET37b(+)でクローンした。
このベクターは、セルロース結合ドメインタグ(CBDTMタグ)とSタグ、ベ
クター中のNcol部位の5’両方を含む。該CBDTMタグ配列は、ミクロバ
イアルセルロースから誘導されたセルロース結合ドメインをコードする。そのた
め、このタグは、高い特異性、タンパク質生成物の低いコストアフィニティー精
製のためのセルロースを基本とする支持体の使用を可能にした。この構築物中に
存在するSタグの存在は、ウスタン・ブロットでのタンパク質生成物を簡単に検
出すること、タンパク質量の簡単に酵素的な定量を可能にする。
(DE3)を、実施例8に記載したような発現ベクター(3つ全てを個々に用い
た)で形質転換した。多くのクローンを選択し、最も収率のよくTfRscFv
産生するものを選抜した。ヒスチジンタグを有する実施例8、セクション1.A
において、上記の構築物由来のタンパク質の精製は、実施例に詳細に記載するが
、同じ方法を、実施例8、セクション1.Aで記載した3つ全ての構築物由来の
TfRscFvタンパク質を含有するシステインの精製に使用した。主要なTf
Rsタンパク質(約90%)は、可溶性で存在するのではなく、封入体内に存在
することが明らかとなった。そのため、システインリンカーを含有するTfRs
cFvを、以下のように封入体から精製した。単一クローンを、50μg/ml
のカナマイシン含有LB培地(5〜10ml)で培種し、37℃、250rpm
、0.5〜0.7のOD600のなるまで(4〜5時間)増殖させた。この少量の
培養物を沈殿させ、LB培地(ブロス)で懸濁し、50μg/mlのカナマイシ
ン含有LB培地(1L)に添加し、37℃、250rpm、OD600が0.5
〜0.7になるまで(4〜5時間)増殖させた。TfRscFvタンパク質の発
現を誘導するために、終濃度1mMのIPTGをこの時点で添加し、さらに4時
間インキュベーションを継続した。この時点がタンパク質発現の最高レベルをも
たらすことを測定した。次いで、バクテリア培養物を遠心分離で回収し、15分
間、30℃で、リゾチーム(100μg/ml)を含有するコールドの20mM Tris−HCl(pH7.5、100ml)で分解した。試料を、氷上、10
ワットで5分間(30秒で破裂)音波処理した。この封入体を、遠心分離(13
000g、15分間)で単離した。得られた沈殿物を、3回、コールドの20m
M Tris−HCl緩衝液(pH7.5)で3回洗浄した。封入体の精製度と量
を、可溶化する前にSDSポリアミドゲル電気泳動によって決定した。
uHCl緩衝溶液)を含有する100mMTris−HCl緩衝液(pH8.0)
に溶解し、遠心分離(12,300g、15分間)を行い、不溶性堆積物を除去
した。2−メルカプトエタノールを、タンパク質濃度の約50モル倍に等しい終
濃度まで上清に添加し、1時間、室温で回転させながらインキュベーションした
。このような高濃度のグアニジン−HClと還元剤の存在により、全く折りたた
まれていないタンパク質を生じる。TfRscFvタンパク質の再折りたたみは
、4℃で、透析を行って、2−メルカプトエタノールの非存在下、グアニジン−
HCl濃度を低下させて行った。100mM Tris−HCl(pH8.0)およ
び200mM NaCl中:次の6M、3M、2M、1Mおよび0.5Mのグアニ
ジン−HCl濃度に対して各々、24時間、透析を行った。最初の透析は、10
0mM Tris−HCl(pH8.0)および200mM NaClを3回取り替え
た。4回目の透析溶液(1M グアニジン−HCl)は、2mM グルタチオン(
酸化型)と500mM L-アルギニンも含有する。これらの試薬は、部分的に再
折りたたみされたタンパク質を適切なジスルフィド結合を形成させ、正しいタン
パク質構造を形成させる。該溶液を、凝集物を除去するために遠心分離(130
00g)によって分離した。該試料を、遠心分離用フィルター(Centrplus cent
rifugal filter)(Amicon)を用いて、約1.5倍に濃縮した(3000g、90
分間)。SDS-PAGEは、ほんの微々たる混在物を含む、約28kDaの分
子量を有するTfRscFvを含む可溶化システインのシングルバンドを示した
(図8)。

中のscFvに、1M DTTを、1〜50mM の終濃度になるまで添加した。
室温、5〜10分間の回転後、該タンパク質を、10−DGカラム(Bio-Read)
で脱塩した。遊離SH基を5,5’ジチオビス−(2−ニトロベンゼン酸)(D
TNB、Ellman's 試薬)によって測定し(G.L. Ellman (1959)Arch. Biochem.
Biophys. 82:70-77. P.W.Riddles, R.L.Blakely, B.Zeruer(1993)Methods Enzy
mol. 91:49-60)、SH/タンパク質モル比もしくは遊離SH/scFv分子の
数として計算した(表7)。この結果は、1〜10mMDTTが、scFv還元
に供されたことを示す。 表7:TfRscFvの還元
(Avanti Polar Lipids)は、リピド総量の5〜8%モルまで、実施例3に記載
した7つのリポソーム化合物を包含する。MPBリポソームを、実施例3に記載
したと同じ方法で調製した。他のリポソーム調製方法を、カチオンリポソームを
調製するために使用することができる。例えば、Campbell MJ (Biotechniques 1
995 Jun; 18(6):1027-32)によって記述した方法から修飾したエタノール注射法
は、本発明において上手く使用した。簡単に言うと、全てのリピドを、エタノー
ル中で可溶化させ、混合し、ハミルトン(Hamilton)シリンジを用いて、50〜
60℃の純水を撹拌しながら注入した。該溶液を、さらに10〜15分間、撹拌
した。この溶液の終濃度は、1〜2mMの全リピドであった。エタノール注入方
法は、敏速かつ容易で確実である。終濃度10〜20mMになるまで、1M H
EPES、pH7.5(pH7.0〜8.0)を添加した。我々は、マレイミド基
が水溶液中(pH7.0以上)で不安定であるとわかったので、該リポソームを
、水(pH5〜6.5)中で調製した。pHは、後の被覆反応を利用するために
、scFv−SHに結合する前に、1M HEPES緩衝液(pH7.0〜7.0
〜8.0)を用いて7.0〜8.0に調整することができる。
ましくは1/10〜1/20の比で、MPB−リポソームに添加した。該溶液を
ゆっくりと、30分間、室温で回転して混合し、scFv−Lipを生成した。
scFv−Lipを精製せずに使用したが、Sepharose CL-4Bクロマトグラフィ
ーで精製することが可能である。プラスミドDNAを水で希釈し、1/6〜1/
20、好ましくは1/10〜1/14のDNA/リピド(μg/nmol)比で
scFv−Lipに添加した。該溶液を、数回反転させて、5−15分間充分に
混合し、scFv−DNA複合体を生成した。scFv−Lip−DNAは精製
せずに使用したが、Sepharose CL-4Bクロマトグラフィーで精製することが可能
である。80〜100%のscFvがリポソームに接合していたことが分かった
。
/14のDNA/リピド(μg/nmol)比で、MPBリポソームに添加した
。該溶液を、数回反転させて、5〜15分間充分に混合し、MPB−Lip−D
NA複合体を生成した。次いで、scFv−SHを該複合体に、1/5〜1/4
0、好ましくは1/10〜1/20のタンパク質/リピド(w/w)比で、複合
体に添加した。該溶液を、ゆっくりと、30分間、室温で回転させて混合し、最
終的なscFv−Lip−DNA複合体を生成した。該scFv−Lip−DN
Aは精製せずに使用したが、Sepharose CL-4Bクロマトグラフィーで精製するこ
とが可能である。80〜100%のscFvがリポソームに接合していたことが
分かった。 4.静脈注射のために、50%のデキストリン溶液を、終濃度5%までscFv
−Lip−DNAに添加した。
接合法によって生成した抗TfRscFv−イムノリポソームに関する特性付与
を示す。ヒト扁平上皮細胞の頭頚部癌細胞系JSQ−3は、これらの研究に関し
てTfR(+)標的細胞として機能する。
SA)を用いて、リポソームに結合する前後のTfRscFv免疫反応活性を測
定した。96ウェルプレートで融合性JSQ−3細胞を、PBS中の0.5%の
グルタルアルデヒドを用いて、10分間、室温で固定した。該プレートを、PB
S中の5%メス子牛血清(FBS)を用いて、30℃で30分間ブロッキングし
た。システイン含有TfRscFv単独、カチオンリポソームに接合したTfR
scFv(TfRscFvイムノリポソームおよび非標的化リポソームを、3つ
のウェルに添加した。抗トランスフェリン受容体モノクローナル抗体(Hb21
、David Fitzgerald, NIHから入手)を陽性対照として、一連のウェルに使用し
た。該プレートを、4℃で一晩インキュベーションした。該ウェルを、3回PB
Sで洗浄し、PBS中3%のFBSの抗Hisモノクローナル抗体(Qiagen)を
各ウェルに添加し、60分間37℃でインキュベーションした。3回PBS洗浄
の後、3%のFBSで希釈したHRP標識ヤギ抗マウスIgG(Sigma)を、各
ウェルに添加し、30分間37℃でインキュベーションした。該プレートを、P
BSで3回洗浄し、クエン酸リン酸緩衝液(Sigma)中の基質(100μl)と0.
4mg/mlOPDを各ウェルに添加した。発色は、各ウェルに2M スルホン
酸(100μl)を添加して停止した。該プレートを、490nmで、ELISA
プレートリーダー(Molecular Devices Corp)によって測定した。
、該免疫活性を保持することを明確に示した。OD490値は、タンパク質(0
.6μg)で0.060±0.6から、TfRscFv(1.5μg)で0.100
±0.0038およびTfRscFv(3μg)で0.132±0.0031まで
TfRscFvタンパク質量の増加と共に増加した。さらに、このTfR-scF
vタンパク質は、陽性対照として使用した親のHb21抗トランスフェリン受容
体以上にさらに高い結合活性を示すことが分かった。Hb21(100μl)の
最高濃度に関するOD490は、約2〜4倍低い(0.033±0.0086)。
(A)およびLip(B))中に本発明(実施例9)の接合方法によって同じT
fRscFvを組み込んだ後に実施し、カチオンリポソームを用いるこの方法の
万能性を示す。実施例9において詳細に記載されたリポソーム調製物の前および
後接合法の両方法を用いた。表8に示したように、接合法によって調製したTf
RscFvの免疫反応活性は、2つのリポソーム組成物の両方と複合体化するこ
とによって失わない。これは、イムノリポソーム複合体を生成するために使用し
た前および後の結合法の両方に対して真実である。該TfRscFv標的化リポ
プレックスを、細胞に結合することを示した。この結合は、TfRscFvを含
まないリポソームの結合以上に非常に高く、この結合は、実際、細胞へのトラン

合法によって調製したTfRscFvイムノリポソーム複合体のインビトロでの
トランスフェクション効率を決定した。標的リガンドとしてTfRscFvの万
能性を示すために、ここでもまた実施例10のように、2つの異なるリポソーム
複合体(LIp(A)とLip(B))を、TfRscFvタンパク質に接合し
た。ヒト肺癌細胞系MDA−MB−435および頭および頚部のヒト扁平上皮瘍
細胞を、これらの研究で使用した。インビボでのトランスフェクションを、24
ウェルプレートで行った(Xu L, et al., Hum. Gene Yher.(1999)10:291-2952)
。トランスフェクション溶液を、10%の血清存在下、細胞に添加した。24時
間後、細胞を洗浄し、ルシフェラーゼ活性およびタンパク質濃度を測定した。該
結果は、表9Aおよび9Bに示したように、分解物中のμgタンパク質あたりの
相対的光単位(RLU)として表現した。
ポソームは、非標的化リポソームよりも3〜6倍高く、トランスフェリン標的化
リポソームよりも2〜3倍高い、インビトロで非常に高いトランスフェクション
活性を有することを示す。これは、両方のヒト腫瘍細胞系および両方のリポソー
ム組成物に関して真実である。即ち、それらは、その免疫反応活性を保持し、標
的受容体に結合することができる。表9Aを基にすると、scFvリポソームを
、インビトロにおける有効な遺伝子トランスフェクション薬物として使用するこ
とができ、市販入手し得るカチオンリポソーム(DOTAP/DOPEおよびD
DAB/DOPE)およびトランスフェリンよりも有効であった。本発明で開示
した該TfRscFvイムノリポソームは、トランスフェリン受容体と哺乳類細
胞のトランスフェクションに有用なインビトロ遺伝子トランスフェクションキッ
トのために使用することができる。
ため、得られた複合体はよりコンパクトで、高いトランスフェクション効率を示
す細胞によって簡単に執り込まれる。これらの結果は、ヒトに使用するための全
身送達のためにTfRscFvイムノリポソームに対する利点を示す。類似サイ
ズは、微小管からの腫瘍細胞へのアクセスを増加させた。最も重要なことは、T
fR分子の場合、TfRscFvはヒト血液産物ではない。そのため、ヒトの治
療用のトランスフェリンそれ自身の使用に関連する懸念および技術的問題は回避
される。
ボで腫瘍細胞に優先的に野生型p53(wtp53)遺伝子を持つリポプレック
スを指向するTfRscFvの能力を示す。標的リガンドとしてTfRscFv
の万能性を示すために、明細書中の実施例10に示したように、2つの別のリポ
ソーム組成物(Lip(A)およびLip(B))を、接合法によってシステイ
ン含有TfRscFvタンパク質と複合体化した。実施例9において詳細に説明
したような前接合法だけを、この研究で用いた。2.5×106MDA−MB−
435ヒト肺癌細胞を、4−6週令のメス無胸腺ヌード・マウスに皮下注射した
。Martrigelコラーゲン基部膜[Collaborative Biomedical Produsts(登録商標)]
中で懸濁した1.1×107DU145ヒト前立腺癌細胞を、4−6週令のメス
無胸腺ヌード・マウスに皮下注射して腫瘍を発病させた。50−200mm3の
腫瘍を持つ動物を本実験で使用した(1個体/1試験試料)。wtp53遺伝子を
持つ接合TfRscFvイムノリポソーム、さらに非標的化Lip(B)および
wtp53本来のDNAを動物の尾血管中に静脈注射した。追加対照として、p
53含有ベクタの位置にある空ベクターを保持する接合TfRscFv−Lip
(A)を、マウスに注入した。実施例7に示したように、注射後の約60時間後
、該動物を屠殺し、腫瘍、肝臓を採取した。タンパク質を組織から単離し、各々
の試料(100μg)を、抗53モノクローナル抗体を用いるウエスタン・ブロッ
ト分析のために10%ポリアクリルアミドゲルに流した。これら両方の腫瘍細胞
のタイプでは、外来性マウスと外来性ヒトp53は、同じ位置で移動する。明細
書においてこの結果は、実施例7における記載を反映するものであった。図9に
示したように、接合法によって調製したTfRscFv−Lip(A)−pCM
Vp53リポプレクッスまたはTfRscFv−Lip(B)−pCMVp53
リポプレックスによって静脈注射した動物由来のDU145およびMDA−MB
−435腫瘍細胞の両方は、強いp53シグナルとさらに低いバンドによって示
されるような、DU145腫瘍細胞において最良の発現と共に、外来性wtp5
3の高い発現レベルを示した。一方、両方の腫瘍細胞型において、Lip(A)
組成物はいくらかLip(B)よりも良好であり、両方のリポソーム組成物はこ
の方法の万能性を示す。外来性マウスp53タンパク質のみ、これらの動物の肝
臓に顕性であった。対照的に、外因性マウスp53タンパク質のみは、空のベク
ターを有する接合TfRscFv−Lip(B)または本来wtp53DNAを
用いて注入したマウスから発生した腫瘍に顕性であった。また、p53の発現に
おける若干の増加を、非標的化Lip(B)−p53と共にDU145腫瘍にお
いて観察した。即ち、優先的に腫瘍細胞へwtp53遺伝子を送達した接合Tf
RscFvイムノリポソームは、2つの異なる腫瘍細胞において顕性であり、こ
の発明の方法の広い有用性を示すものである。そのため、実施例において記載し
た本発明の方法は、カチオンリポソームへの結合能を保持するだけでなく、イン
ビボおよびインビトロにおいてもトランスフェリン受容体への結合能を保持する
TfRscFvタンパク質を発生させる、即ち、遺伝子治療用に、腫瘍特異性の
、標的化イムノリポソームを生成する我々の目的を満たすものであった。

に結合し得る、請求項1記載のイムノリポソーム。 - 【請求項3】 該核酸がDNAである、請求項1記載のイムノリポソーム。
- 【請求項4】 該核酸が野生型p53をコードする、請求項1記載のイムノ
