Abstract
Description
リガンドおよび治療用のもしくは診断用の薬剤またはレポーター遺伝子をカプセル化する陽イオン性リポソームを含む複合体を安定化する量のショ糖を含む溶液と混合すること;および
複合体およびショ糖の生じた溶液を凍結乾燥して、凍結乾燥製剤を得ることを含み;
そこでは、再構成時に、製剤はその凍結乾燥前の活性の少なくとも約80%を保持する。
図1は、5%デキストロースもしくは5%ショ糖を含有する新鮮に作成されたおよび凍結乾燥された複合体(Tf−LipA−LucおよびTfRscFv−LipA−Luc)のサイズ(nm)を示す。
本発明にしたがって、リガンドおよび治療用のもしくは診断用の薬剤をカプセル化する陽イオン性リポソームの凍結乾燥された複合体の安定性を、凍結乾燥前に複合体を安定化する量のショ糖を含む水溶液と混合することにより増加させることができる。ショ糖の溶液は水中の単なるショ糖であり得るか、またはPBS、HEPES、TRISもしくはTRIS/EDTAのような、緩衝剤を含むことができる。典型的には、ショ糖の溶液は、約1%〜約80%のショ糖、典型的には、1%〜約10%、20%、30%もしくは40%のショ糖のような、1%〜約50%のショ糖、好ましくは約5%〜10%のショ糖、そして最も好ましくは約10%のショ糖、の最終濃度に複合体と混合される。凍結乾燥製剤は、約2〜8℃から約−80℃の範囲内で少なくとも6ヶ月の期間の間に著しい活性を損失すること無しに、安定である。好ましくは、製剤は、少なくとも6〜12ヶ月の期間の間に安定である。再構成時に、複合体は、それらの凍結乾燥前の活性の少なくとも約80%、好ましくはそれらの凍結乾燥前の活性の少なくとも約85%、そして最も好ましくはそれらの凍結乾燥前の活性の少なくとも約90〜95%、を保持する。
新鮮に作成されたおよび凍結乾燥された、炭水化物での複合体の作製ならびに活性およびサイズのインビトロでの評価
初期の実験は、凍結乾燥の前後で炭水化物で作成されたリガンド−リポソーム−核酸複合体のサイズならびにインビトロでのトランスフェクション効率の両方を試験するために実施された。二つの別々なリガンド、TfおよびTfRscFv、が試験された。複合体は、米国特許出願09/601,444ならびに公開米国特許出願09/914,046および10/113,927[Xu, L., et al. Human Gene Therapy 10:2941-2952 (1999); Xu, L., et al., Human Gene Therapy 13:469-481 (2002); and Xu, L., et al., Molecular Cancer Therapeutics 1:337-346 (2002)をまた参照]の中に記述されている方法論を用いて作成された。それぞれの複合体では、リポソームは、本明細書中でリポソームA(LipA)として確認される、1:1比のDOTAP:DOPEであった。使用されるDNAは、ホタルのルシフェラーゼ遺伝子をコード化する遺伝子を保持するプラスミドであった。全ての場合で、炭水化物溶液を、複合体の作製で最後の工程として加えた。
2〜8℃で一週間保存後の凍結乾燥された複合体によるインビボでのヒト前立腺腫瘍の標的化
10%ショ糖でのリポソーム複合体(新鮮に作成されたかもしくは凍結乾燥されて、2〜8℃で1週間保存された)のインビボでの腫瘍の標的化を、複合体の中でレポーター遺伝子として増強緑蛍光タンパク質(EGFP)を用いて試験した。複合体は、リポソームAがDOTAP:DOPE(1:1)である、TfRscFv−リポソーム A−pEGFPであった。三つの成分の比は0.3μg:14nmols:1μg(TfRscFv:リポソーム:DNA)であった。複合体を、実施例1で上述したように、作製して凍結乾燥した。凍結乾燥後に、複合体を2〜8℃で一週間冷蔵して保存した。試料を、実施例1で記述されたように、凍結乾燥前の容積に等しい容積にエンドトキシンの無い水の添加により再構成した。
−80℃で一ヶ月間保存後の凍結乾燥された複合体によるインビボでのヒト膵臓腫瘍の標的化
凍結乾燥された複合体の安定性をまた、膵臓がん異種移植腫瘍への標的化により−80℃で一ヶ月保存後に試験した。複合体(実施例2で上述したのと同一の複合体および比)を10%ショ糖で作成し、そして、実施例1で記述したように、凍結乾燥した。凍結乾燥の後に、試料を−80℃で一ヶ月間保存し、次いで、実施例1で記述したように、エンドトキシンの無い水で再構成した。
サイズおよびインビトロでのトランスフェクション効率により評価されたように、2〜8℃で保存された凍結乾燥複合体の長期安定性
実行可能な臨床治療剤として我々のTfRscFv−リポソーム−DNA複合体の潜在能力を増加させるために、その安定性を増加させる手段を開発して、かくしてその腫瘍の標的化能、および使用期限を維持した。我々の試験の幾らかは、添加物として10%ショ糖で凍結乾燥された複合体は、−20℃、−80℃もしくは2〜8℃のいずれかで、成功裏に保存されることができたことを指示してきている。診療所での使用の便利性のために、好ましい保存方法は2〜8℃である。時間の長さを決定するために、凍結乾燥された複合体を2〜8℃で生物活性の損失無しに保存することができて、凍結乾燥されて、1,4および6ヶ月間2〜8℃で保存された複合体のインビトロでのトランスフェクション効率を評価した。複合体は、リポソームAがDOTAP:DOPE(1:1)である、TfRscFv−リポソーム A−p53であった。三つの成分の比は、0.34μg:10μg:1μgと同等である0.3μg:14nmols:1μg(TfRscFv:リポソーム A:DNA)であった。10%ショ糖を添加物として使用した。複合体の中のDNAは、ヒト野生型p53をコード化する約1.7KbのcDNA配列を含有するプラスミドベクターであった。複合体を作製し、凍結乾燥して、実施例1で記述したように、2〜8℃で保存後の適切な時間で再構成した。サイズ(数のパラメーター)およびゼータ電位をMalvern 3000H Zetasizerを用いて測定した。機能性活性をルシフェラーゼ共トランスフェクションアッセイを用いて評価した。ヒト前立腺がんDU145細胞を、BP100プラスミドDNAでおよび複合体で共トランスフェクトした。BP100プラスミドは、wtp53誘導性プロモーターの制御下にルシフェラーゼ遺伝子を保持する。かくして、トランスフェクトされた細胞の中の機能性p53のレベルは、ルシフェラーゼ活性のレベルにより反映される。トランスフェクションの24時間後に、細胞を溶菌して、ルシフェラーゼ活性を、Promegaルシフェラーゼ試薬を用い製造プロトコルにしたがって、アッセイした。表1に示すように、複合体のルシフェラーゼ活性、サイズおよびゼータ電位は、新鮮に作成された複合体および凍結乾燥されて、2〜8℃で6ヶ月までの間に保存された複合体との間で一貫している。
2〜8℃で保存後に全身的に投与された凍結乾燥複合体のインビボでの腫瘍特異的標的化
新鮮なならびに作製され、1,4もしくは6ヶ月間2〜8℃で保存されて、次いで実施例4に記載された試験のために再構成された凍結乾燥複合体を、全身的な(静脈)投与後にヒト前立腺異種移植腫瘍に到達してトランスフェクトするそれらの能力についてインビボでまた試験した。少なくとも100mm3の皮下ヒト前立腺腫瘍細胞株DU145異種移植腫瘍を持つ無胸腺ヌードマウスを、0.8mLの最終容積で注射当り40μgのDNAと同等な量で、複合体(新鮮なもしくは凍結乾燥されて、再構成された)で24時間にわたり3回静脈注射した。最後の注射の48時間後に、動物を人道的に安楽死させ、器官を除去し、タンパク質を単離して、発現をXu, L. et al., Tumor Targeting 4:92-104 (1999)により記述されたようにウエスタン分析により測定した。当分野で普通に知られている他の方法を、これに変えて使用しすることができた。全タンパク質溶菌液80μgを12%SDS−ポリアクリルアミドゲルのレーン上に装填した。ゲルを操作した後に、タンパク質をニトロセルロース膜へ転移しそして抗−p53マウスモノクローナル抗体(Oncogene Research製品)のプローブと反応させた。
バッチ間の一貫性および凍結乾燥後の安定性
同一の日の異なる時間に作製されて、凍結乾燥された、ならびに異なる日に作製されて、凍結乾燥された、複数のバッチの複合体が同様なサイズおよびトランスフェクション効率のレベルを有することを確立することは重要である。リポソームAがDOTAP:DOPE(1:1)である、複合体TfRscFv−リポソーム A−p53を、作製し、凍結乾燥し、保存して、実施例1に記載されたように、再構成した。成分およびp53DNAの比は実施例4に記載されたようであった。

ヒトの患者を処置するのに有用である凍結乾燥された複合体については、10%ショ糖の存在下における複合体の作製の加工法が、移行されて、商業的製造実体により大規模で成功裏に実施されることができたことを示すことが必要である。複合体を、委託および秘密保持の合意の下に、Cardinal Health, Albuquerque, NMによって攪拌により作製した。DNA溶液を、渦巻きが溶液の中に30秒〜1分の間丁度形成しつつあった、速度で攪拌する一方で、TfRscFv:リポソーム溶液に加えた。この溶液を室温で10〜20分間保持した後で、50%ショ糖の水溶液を30秒〜1分の間上のように攪拌しながら10%の最終濃度に加えて、室温で10〜20分間保持した。商業的に作製されたバッチは、50〜1000mlのサイズに分布した。この商業的施設でHull凍結乾燥機を用いる凍結乾燥プロトコルは以下のようであった:
・10%ショ糖での複合体5mLをそれぞれ含有する、10mLのバイアルを環境温度で装填した。
・棚の温度を1時間にわたり−45℃に下げていった。一旦棚の温度が−45±3℃に達すると、生成物を3時間保持した。
・この点で、冷却器を−55℃もしくはさらに低温に冷却して、真空を50ミクロンHgにセットした。
・次いで、棚の温度を1時間にわたり−35℃に上げていった。
・一旦棚の温度が−35℃に達すると、生成物を48時間保持した。
・棚の温度を4時間にわたり20℃に上げていって、生成物を12時間保持した。
・このサイクルの最後に、チャンバーの圧力を窒素で大気圧に復元し、適切な滅菌微生物拘束フィルターを通してNFろ過した。
・生成物の栓をし、ラベルを貼って、2〜8℃で保存した。
凍結乾燥された複合体中の未完のTfRscFvレベルのパーセントの一貫性
凍結乾燥された複合体の安定性をさらに評価するために、複合体化されていないリガンドの量を2〜8℃で6ヶ月までの間に保存後に測定した。TfRscFv−LipA−p53複合体の中の複合体化されていないTfRscFvの量を評価するために、4%〜20%グラジエントの非変性のかつ非還元のポリアクリルアミドゲル電気泳動に引き続くウエスタン分析を当業者に普通に知られる方法を用いて使用した(図8)。TfRscFvタンパク質に対するポリクローナルウサギ抗体を第一の抗体(Animal Pharm, Healdsburg, CAにより製造された)として使用して、HRP−標識マウス抗−ウサギモノクローナル抗体(Sigma)を二次抗体として使用した。それぞれの中にTfRscFv134ngを含有する、新鮮に作成されたかもしくは凍結乾燥された複合体を、実施例1で記述されたように、作製して凍結乾燥した。凍結乾燥された試料を2〜8℃で1、4、もしくは6ヶ月の間に保存した後に、それらを、実施例1でのように、再構成した。一旦リポソームに複合体化されると、TfRscFvタンパク質はPAGEゲルには入ることができないであろう。それ故に、複合体化されていない遊離のTfRscFvのみが検出されるであろう。非変性のかつ非還元の条件下では、遊離のTfRscFvの単量体の量を正確に測定することは困難である。かくして、13.4ng(複合体のそれぞれの中のものの10%)、26.8ng(20%)もしくは40.2ng(30%)の単一薬剤のTfRscFvを含有する、複合体化されていない試料を、試験複合体のそれぞれの中の複合体化されていないTfRscFvの量の粗い評価のための濃度標準として同一のゲルの中でまた操作した。
アンチセンスHER−2オリゴヌクレオチドを含有するリガンド−リポソーム−核酸複合体の凍結乾燥
上の試験は、複合体の中でプラスミドDNAを用いた。プラスミドDNAおよびオリゴヌクレオチドは必ずしも相互交換可能ではなく、そして異なる化学を有し得るので、凍結乾燥手順がオリゴヌクレオチド(ODN)を含有するリガンド−リポソーム複合体に適用できたことを立証するために、実験をまた行った。使用されたODNは、5’-TCC ATG GTG CTC ACT-3’の配列を持つHER−2遺伝子(AS HER−2)の開始コドン領域に相補的な15マーのホスホロチオエート化配列に特異的なアンチセンスHER−2ODNであった。アッセイシステムとしてMDA−MB−453ヒト乳がん細胞株を用いて、TfRscFv−LipA−AS HER−2複合体による細胞致死を、増加するODN濃度での異なる砂糖との凍結乾燥後に評価した。複合体は、実施例1で記述されたように、作製され、そして1nmol対15nmol(ODN:リポソーム)および1μg対30μg(TfRscFv:リポソーム)の比で、TfRscFv、リポソームA(1:1でDOTAP:DOPE)ならびにODNから成った。
AS ODNを保持する複合体の凍結乾燥および6ヶ月の保存後のサイズ、ゼータ電位ならびに有効性の維持
AS HER−2ODNを含有して、10%ショ糖で作製されたリガンド−リポソーム−核酸複合体のサイズ、ゼータ電位およびトランスフェクション活性を、凍結乾燥の前後に検査した。複合体のサイズは、凍結乾燥および−20℃で6ヶ月までの間の保存の前後で本質的に同一であると見出された。例えば:凍結乾燥前で、新鮮なおよび、実施例9で記述されたように作製された、6ヶ月凍結乾燥複合体についての強度、容積ならびに数平均によるサイズ(nm)の値は、それぞれ、410(I)、454(V)および368(N)対339(I)、427(V)および397(N)であった。加えて、HER−2レベル(SC-ODN_ (5’-CTA GCC ATG CTT GTC-3’)に影響しないもう一つのオリゴヌクレオチドを、同一の比でまた複合体化し、凍結乾燥し、−20℃で6ヶ月まで間に保存して、実施例9におけるように再構成した。ここでまた、凍結乾燥および保存は、複合体のサイズもしくはゼータ電位に著しい効果を何も有しなかった。かくして、任意のODNを複合体化して、凍結乾燥することができる。
siRNAを保持する複合体の凍結乾燥後のサイズおよびゼータ電位の維持
10%ショ糖でのTfRscFv、リポソームAおよびsiRNAの複合体の凍結乾燥の後の安定性を、凍結乾燥の前後で複合体のサイズおよびゼータ電位を測定することにより評価した。複合体は、33.3μgにおけるTfRscFv、リポソームA(1:1のモル比ででDOTAP:DOPE)およびsiRNAから成った。複合体の全容積は500μLであった。成分の比は、1μg対7nmol(siRNA:リポソーム)および1μg対30μg(TfRscFv:リポソーム)であった。ショ糖を10%の最終濃度に複合体へ加えた。複合体を、実施例1で記述されたように、作製して、凍結乾燥した。凍結乾燥の後に、複合体を、実施例1で記述されたように、再構成して、サイズおよびゼータ電位をMalvern Zetasizer 3000Hを用いて測定した。結果を表4に示す。
ペプチド−リポソームで作成された複合体の凍結乾燥後のサイズの維持
本発明の一般的な性質をさらに立証するために、修飾リポソームを含んだ複合体をまた作製した。複合体を形成するために使用されたリポソームをペプチドに結合した。ペプチドは、ヒスチジンおよびリシンを含んで、長さで31個のアミノ酸の枝分かれしたペプチドであって、構造(5’-K[K(H)-K-K-K]5-K(H)-K-K-C-3’)を持つヒスチジンおよび非−ヒスチジンアミノ酸の組合せから成った。この試験におけるリポソームはDOTAP:DOPE(1:1)から成った。HKペプチドは、末端システインおよびリポソーム中のマレイミド基を通してリポソームに共有結合で結合された。複合体は、TfRscFv−HK−リポソーム−DNAから成って、そこでは成分の比は以下のようであった:1μg:30μgのTfRscFv対HK−リポソーム(μg:μg)および1μg:14nmolのDNA対HK−リポソーム(μg:nmol)であった。使用されたDNAは、複合体の全容積300μLについて18μgのDNAにおいてp53(実施例4を参照)であった。10%ショ糖は最終の複合体の中に含まれた。複合体を、実施例1で記述されたように、作製して、凍結乾燥した。凍結乾燥の後に、複合体を2〜8℃で3日間保存して、次いで、実施例1で記述されたように、再構成した。凍結乾燥の前のおよび2〜8℃で三日間保存後の複合体のサイズをMalvern Zetasizer 3000H上で測定した。凍結乾燥の前には、サイズ(数平均)は601nmであった。保存および再構成の後では、それは588であった。かくして、もう一度、10%ショ糖を用いる複合体の凍結乾燥は、HKペプチドの包含をもってしても、複合体のサイズに如何なる著しい変化をもたらさなかった。
異なる治療的遺伝子(RB54)を保持する複合体の凍結乾燥および2〜8℃で保存後でのインビトロおよびインビボでの評価
ルシフェラーゼ遺伝子、増強緑蛍光タンパク質をコード化する遺伝子およびp53遺伝子に加えて、他のプラスミドDNAを保持するリポソーム複合体を凍結乾燥して、サイズおよび生物活性を保持することができる。さらに立証するために、もう一つの治療的遺伝子、腫瘍サプレッサー遺伝子RB94を保持するこの複合体をまた作製した。複合体は、リポソームAがDOTAP:DOPE(1:1)である、TfRscFv−リポソーム A−RB94であった。三つの成分(TfRscFv:リポソーム:DNA)の比は0.34μg:10μg:1μgであった。複合体はまた、10%ショ糖と共に、0.5μLの全容積でRB94プラスミドDNAの30μgを含有した。実施例1で記述したように、複合体を作製して、サイズおよびゼータ電位の試験のために実施例1で記述した方法を用いて凍結乾燥したかもしくはインビトロおよびインビボでの標的化試験に使用のためにCardinal Healthにより実施例7におけるように作製した。
実施例1におけるように作製されて、凍結乾燥され、2〜8℃で4日間保存されて、実施例1におけるように、再構成された複合体のサイズおよびゼータ電位を、凍結乾燥および保存の前後でMalvern Zetasizer 3000Hを用いて比較した。凍結乾燥の前では、サイズ(nm)は、283(強度)および392(容積)であったが、一方では後でそれは303(強度)および347(容積)であると見出された。かくして、10%ショ糖が含まれるときには、凍結乾燥および4日間2〜8℃で保存の後では、サイズに著しい変化は何も無かった。同様に、ゼータ電位は何も主要な相違を示さないで、両方は+20〜+30の範囲[19(前)および30.7(後)]にあった。
腫瘍細胞を特異的に標的にして、凍結乾燥および2〜8℃で長期間の間の保存の後でそれらを効率よくトランスフェクトする複合体の能力を、ヒト前立腺細胞株DU145およびヒト膀胱がん腫細胞株HTB−9を用いて細胞培養液の中でまた試験した。両方の細胞株を、新鮮に作成された複合体かもしくは商業的受託者により作製されて、凍結乾燥されて(実施例7)、実施例1におけるような再構成の前におよそ4ヶ月間2〜8℃で保存されてきていた複合体のいずれかを用いて、インビトロでトランスフェクトした。細胞の中のRB94タンパク質発現のレベルを、当業者に既知の標準的プロトコルを用いるウエスタン分析により測定した。新鮮に作成されたかもしくは凍結乾燥された複合体でのトランスフェクションの後に検出されたタンパク質の量の間にいずれのヒト腫瘍細胞株において著しい相違は何も無かった。
Claims (42)
- リガンドおよび治療用の薬剤、レポーター遺伝子もしくは診断用の薬剤をカプセル化する陽イオン性リポソームを含む安定な細胞標的化複合体を作製するための方法であって、
リガンドおよび治療用の薬剤、レポーター遺伝子もしくは診断用の薬剤をカプセル化する陽イオン性リポソームを含む複合体を提供すること;
当該リポソーム複合体を安定化する量のショ糖を含む溶液と混合すること;および
リポソーム複合体およびショ糖の当該溶液を凍結乾燥して、凍結乾燥製剤を得ること:を含み、
そこでは、再構成時に、当該製剤はその凍結乾燥前の活性の少なくとも約80%を保持する、
方法。 - 再構成時に当該製剤がその凍結乾燥前の活性の少なくとも約85%を保持する、請求項1に記載の方法。
- 再構成時に当該製剤がその凍結乾燥前の活性の少なくとも約90%を保持する、請求項2に記載の方法。
- 再構成時に当該製剤がその凍結乾燥前の活性の少なくとも約95%を保持する、請求項3に記載の方法。
- 当該複合体がショ糖の溶液と約1%〜約80%のショ糖の最終濃度に混合される、請求項1に記載の方法。
- 当該複合体がショ糖の溶液と約1%〜約50%のショ糖の最終濃度に混合される、請求項1に記載の方法。
- 当該複合体がショ糖の溶液と約1%〜約20%のショ糖の最終濃度に混合される、請求項1に記載の方法。
- 当該複合体がショ糖の溶液と約5%〜約10%のショ糖の最終濃度に混合される、請求項1に記載の方法。
- 当該複合体がショ糖の溶液と約10%のショ糖の最終濃度に混合される、請求項8に記載の方法。
- 当該リガンドが、標的細胞上で差次的に発現される受容体を含む、請求項1に記載の方法。
- 当該リガンドが、トランスフェリン、フォレート、抗体もしくは抗体フラグメントを含む、請求項10に記載の方法。
- 当該リガンドが、トランスフェリンを含む、請求項11に記載の方法。
- 当該リガンドが、抗−TfRモノクローナル抗体を含む、請求項11に記載の方法。
- 当該リガンドが、抗体の単鎖Fvフラグメントを含む、請求項11に記載の方法。
- 当該抗体フラグメントが、抗−TfRモノクローナル抗体に基づくscFvを含む、請求項14に記載の方法。
- 当該リポソームが、少なくとも一つの陽イオン性脂質および少なくとも一つの中性のもしくはヘルパーの脂質を含む、請求項1に記載の方法。
- 当該陽イオン性脂質がリン酸ジオレオイルトリメチルアンモニウム(DOTAP)もしくは臭化ジメチルジオクタデシルアンモニウム(DDAB)を含みそして当該中性のまたはヘルパーの脂質がジオレオイルホスファチジルエタノールアミン(DOPE)もしくはコレステロール(chol)を含む、請求項16に記載の方法。
- 当該リポソームがDOTAPおよびDOPEの混合物を含む、請求項16に記載の方法。
- リポソームが少なくとも約10個のアミノ酸のペプチドに結合し、そこでは当該ペプチドが約5〜100%のヒスチジンおよび0〜95%の非−ヒスチジン残基から成る、請求項1に記載の方法。
- 当該ペプチドの当該非−ヒスチジン残基の少なくとも10%がリシン残基である、請求項19に記載の方法。
- 当該ペプチドが5'-K[K(H)-K-K-K]5-K(H)-K-K-C-3'の構造を有する、請求項20に記載の方法。
- 当該治療用の薬剤が遺伝子、プラスミドDNA、オリゴヌクレオチド、オリゴデオキシヌクレオチド、アンチセンスオリゴヌクレオチドもしくはsiRNAを含む、請求項1に記載の方法。
- 約−80℃〜8℃の範囲内の温度で少なくとも6ヶ月の間に少なくとも約80%の活性を保持しながら安定である、リガンドの複合体および治療用の薬剤、レポーター遺伝子もしくは診断用の薬剤をカプセル化する陽イオン性リポソームを含む凍結乾燥製剤であって、当該製剤が当該複合体および当該複合体の安定性を増加させるための有効量のショ糖を含む、凍結乾燥製剤。
- 約1%〜約80%のショ糖を含む、請求項23に記載の凍結乾燥製剤。
- 約1%〜約50%のショ糖を含む、請求項23に記載の凍結乾燥製剤。
- 約1%〜約20%のショ糖を含む、請求項23に記載の凍結乾燥製剤。
- 約5%〜約10%のショ糖を含む、請求項23に記載の凍結乾燥製剤。
- 約10%のショ糖を含む、請求項27に記載の凍結乾燥製剤。
- 再構成時にその凍結乾燥前の活性の少なくとも約80%を保持する、請求項23に記載の凍結乾燥製剤。
- 再構成時にその凍結乾燥前の活性の少なくとも約85%を保持する、請求項23に記載の凍結乾燥製剤。
- 再構成時にその凍結乾燥前の活性の少なくとも約90%を保持する、請求項23に記載の凍結乾燥製剤。
- 再構成時にその凍結乾燥前の活性の少なくとも約95%を保持する、請求項23に記載の凍結乾燥製剤。
- 当該リガンドが、標的細胞上で差次的に発現される受容体を含む、請求項23に記載の凍結乾燥製剤。

- 当該リポソームが、少なくとも一つの陽イオン性脂質および少なくとも一つの中性のもしくはヘルパーの脂質を含む、請求項23に記載の凍結乾燥製剤。
- 当該陽イオン性脂質がDOTAPもしくはDDABを含みそして当該中性のまたはヘルパーの脂質がDOPEもしくはコレステロールを含む、請求項37に記載の凍結乾燥製剤。
- 当該リポソームがDOTAPおよびDOPEの混合物を含む、請求項38に記載の凍結乾燥製剤。
- 請求項23に記載の凍結乾燥製剤であって、リポソームが少なくとも約10個のアミノ酸のペプチドに結合し、そこでは当該ペプチドが約5〜100%のヒスチジンおよび0〜95%の非−ヒスチジン残基から成る、凍結乾燥製剤。
- 当該ペプチドの当該非−ヒスチジン残基の少なくとも10%がリシン残基である、請求項40に記載の凍結乾燥製剤。
- 当該ペプチドが5'-K[K(H)-K-K-K]5-K(H)-K-K-C-3'の構造を有する、請求項41に記載の凍結乾燥製剤。
