Claims (1)
- 【特許請求の範囲】 1.(a)配列1の911位から1795位に示すコーディング鎖またはその相 補鎖のヌクレオチド配列、 (b)55〜65℃で0.5×SSCから2×SSC、0.1%SDSに等価の 洗浄緊縮性条件下で前記(a)のヌクレオチド配列にハイブリダイズでき且つポ リヒドロキシアルカノエート産生に好適な活性を有しているタンパク質をコード するヌクレオチド配列、 (c)前記(a)のヌクレオチド配列に少なくとも約40%の相同性を有してお り且つポリヒドロキシアルカノエート産生に好適な活性を有しているタンパク質 をコードするヌクレオチド配列、 (d)前記(a)のヌクレオチド配列と同じ遺伝情報をコードしているが遺伝コ ードの縮重性に従って縮重しているヌクレオチド配列、 (e)前記(b)のヌクレオチド配列と同じ遺伝情報をコードしているが遺伝コ ードの縮重性に従って縮重しているヌクレオチド配列、 から成るグループから選択されたヌクレオチド配列を含む単離されたDNA分子 。 2.配列1の911位から1795位までのDNA配列を有することを特徴とす る請求項1に記載のDNAセグメント。 3.宿主細胞中でRNA配列を産生させるべく機能するプロモーター領域と前記 プロモーターに作用可能に連結された請求項1に記載のDNA配列とを含んで成 る組換えDNAセグメント。 4.配列1の911位から1795位までのDNA配列を有することを特徴とす る請求項3に記載の組換えDNAセグメント。 5.請求項1から4のいずれか一項に記載のDNAセグメントを含む組換えベク ター。 6.請求項1から4のいずれか一項に記載のDNAセグメントを含んでおり、前 記DNAセグメントがプロモーター領域にヘテロロガスであることを特徴とする 組換え宿主細胞。 7.微生物または植物の細胞であることを特徴とする請求項6に記載の組換え細 胞。 8.Pseudomonas fluorescens rRNA相同グループ Iの細菌であることを特徴とする請求項6に記載の組換え細胞。 9.少なくとも20ヌクレオチドの長さを有しており少なくとも20個の連続す る請求項1に記載のヌクレオチドに対応する配列を有することを特徴とする単離 されたDNAセグメント。 10.ポリヒドロキシアルカノエートを産生させるために有用な細胞を形質転換 させる方法であって、 機能性PHAシンターゼ遺伝子を有する宿主細胞を選択する段階と、 宿主細胞中でRNA配列を産生するために機能するプロモーター領域と前記プ ロモーターに作用可能に連結されており前記プロモーター領域にヘテロロガスな 請求項1に記載のDNA配列とを含む組換えベクターを作製する段階と、 選択された宿主細胞を形質転換させる段階と、 形質転換細胞を再生する段階と、 から成る細胞の形質転換方法。 11.組換えベクターが配列1の911位から1795位までのDNA配列を含 むことを特徴とする請求項10に記載の方法。 12.宿主細胞が微生物または植物の細胞であることを特徴とする請求項10に 記載の方法。 13.宿主細胞がPseudomonas fluoresc ens rRNA相同グループIの細菌から選択されることを特徴とする請求項 10に記載の方法。 14.3〜14個の炭素原子を有する反復単位から成るポリヒドロキシアルカノ エート(PHA)の産生方法であって、 請求項10から13のいずれか一項に記載の方法で作製した形質転換細胞をフ ァージ遺伝子の発現が得られるように培養する段階と、 細胞からPHAを回収する段階とから成る方法。 15.形質転換した微生物細胞を単純糖質基質を含む培地で培養することを特徴 とする請求項14に記載の方法。 16.培地がグルコネート、グルコース、スクロース、フルクトースまたはラク トースから成ることを特徴とする請求項15に記載の方法。 17.形質転換細胞をコーンシロップまたは糖蜜から成る培地で培養することを 特徴とする請求項14に記載の方法。 18.配列2のアミノ酸配列を有する実質的に精製されたタンパク質。 19.PhaGを発現する遺伝的に形質転換された植物を産生する方法であって 、 (i)植物細胞にRNA配列を産生させる機能を有しているプロモーターと、 (ii)前記プロモーターに作用可能に連結された請求項1に記載のPhaG DNA配列と、 を含んで成る組換え二重鎖DNA分子を宿主植物細胞のゲノムに挿入する段階と 、 形質転換植物細胞を作製する段階と、 形質転換植物細胞を再生させる段階と、 から成る方法。 20.PhaG DNA配列が配列1の911位から1795位までの配列であ ることを特徴とする請求項19に記載の方法。 21.宿主植物細胞のゲノムが更に機能性PHAシンターゼ遺伝子を含むことを 特徴とする請求項19に記載の方法。 22.更に、形質転換された植物または植物の部分を採取する段階を含む請求項 19に記載の方法。 23.採取した植物の部分が葉、根、種子または塊茎であることを特徴とする請 求項21に記載の方法。 24.宿主植物が、トウモロコシ、ジャガイモ、テンサイ、タバコ、小麦、また は、大豆、キャノーラ、ナタネ、ヒマワリ、 亜麻、落花生及びArabidopsisから成るグループから選択された高脂 肪種子植物であることを特徴とする請求項19に記載の方法。 25.請求項18から23のいずれか一項に記載の方法によって産生される植物 。 26.グルコネートからPHAを合成する能力を有していないがオクタン酸から PHAを合成する能力を有している突然変異微生物。 27.微生物がPseudomonas putidaの突然変異体であること を特徴とする請求項26に記載の突然変異体。 28.微生物が細菌または真菌の突然変異体であることを特徴とする請求項26 に記載の突然変異体。 29.微生物が、Streptomyces、Nocardia、Alcali genes、Bacillus、Escherichia、Klebsiell a、Lactobacillus、Methylomonas,Pseudom onas、Rhodospirillum、Aspergillus、Ceph alosporium、Penicillium、Saccaromyces、 Rhodotorula、Candi da、HansenulaまたはPichiaの突然変異体であることを特徴と する請求項26に記載の突然変異体。 30.グルコネートからPHAを合成する表現型の能力が配列1のDNA配列と の相補性によって回復することを特徴とする請求項26に記載の突然変異体。 31.微生物が放線菌類Actinomycetesまたは酵母の突然変異体で あることを特徴とする請求項28に記載の突然変異体。
