Claims (1)
- 【特許請求の範囲】 1. 吸着剤としてシリカゲルを用いるクロマトグラフィー方法の適用下にシクロ スポリン含有粗製抽出物からのシクロスポリンAおよび関連シクロスポリン類を 精製する方法において、 a)第1クロマトグラフィー工程において分取HPLCまたは“シミュレート 移動層”技術を用いて粗製抽出物を分離された濃度プロフィルでの画分カットに より無極性随伴物質を含有する有用画分1およびより大きい極性の随伴物質を含 有する有用画分2に分離し、 b)有用画分1(ラフィネート)および有用画分2(エキストラクト)を、次 の第2クロマトグラフィー工程においてSMB技術を用いさらに精製することを 特徴とする高度に精製されたシクロスポリンAおよび関連シクロスポリンを得る ためのクロマトグラフィー方法。 2. a)第1クロマトグラフィー工程ならびに第2クロマトグラフィー工程を順 相系(酢酸エチル)または逆相系(アセトニトリル/水)中で実施し、 b)有用画分1(ラフィネート)をSMB技術によって逆相系(アセトニトリ ル/水)中で第2クロマトグラフィー工程にかけ、有用画分2(エキストラクト )を順相系(酢酸エチル)中で第2クロマトグ ラフィー工程にかけ、 c)有用画分2(エキストラクト)をSMB技術によって逆相系(アセトニト リル/水)中で第2クロマトグラフィー工程にかけ、有用画分1(ラフィネート )を順相系(酢酸エチル)中で第2クロマトグラフィー工程にかけることを特徴 とする請求項1記載の方法。 3. 第5ゾーンの導入により洗浄工程を初めにアルコールを用い、引き続き溶離 剤を用いて実施し、第5ゾーン中に数個のカラムが存在する場合、これらのカラ ムを並列接続で流過させることを特徴とする請求項1記載の方法。 4. 第2クロマトグラフィー工程における分離を、逆相系(アセトニトリル/水 )中で、アセトニトリル/水の40〜80:20〜60、とくに60:40(v /v)の比を用いて実施することを特徴とする請求項2記載の方法。 5. SMBプラント中のカラムおよび溶離剤を40〜80℃の温度範囲内、とく に60℃に保つことを特徴とする請求項1または2記載の方法。 6. 逆相系(アセトニトリル、水)中で、溶離剤のpH値を2〜5の範囲内、と くに3に調節することを特徴とする請求項2記載の方法。
