Claims (8)
- Mormolták ko-dltterencsálása aliogén donorokból Szabadalmi igénypontok 1. E|árés nem-kimerűlt éretlen dendrites séfek (DC-k) előállítására, amely éliras az alábbi lépéseket tartalmazza: - aliogén leukoeiták keverékét állítjuk elő, amely ailogén íeukociíákat legalább két különböző aliogén donorból szerezzük be, - az említett aifogén leukoclta keverékből Izoláljuk az aliogén roonocstákat és így monoeita-dúsított aliogén leukoeitákat állítunk ele, és * az említett monoGlta-dúsított aliogén ieuköciiákból nem-kimerült éretlen DOket állítunk elő, almi a nem kimerni éretlen dendrites sejtek {DC-k) előállítását az említett mrmoeiía-dúsioi aliogén leukocíták nem-bumán szérumtól mentes vizes sejttenyész-tö közegben 2-7 napon át történd egyidejű tenyésztésével végezzük, ahol az említett közeget ínterieukín4~eí (IL-4) és granulocíta-makrofág kolónia stimuláló faktorral (GM-CSF) egészítjük ki.
- 2. Az 1. Igénypont szerlbtiepris, ahol az említett sejttenyészíő közeg legalább egy a traszfemn, albumin és inzulin közül választott humán polipeptídef tartalmaz.
- 3. Az 1, vagy 2, igénypont szerinti eljárás* ahol az említett monocita-dúsitott aliogén leukoeiták aliogén nautröieket tartalmaznak,
- 4. Az 1-3. igénypontok bármelyike szerinti eljárás, ahol az említeti aliogén; ietiko-clta keveréket legalább két, leukoeitákat tartalmazó buífy-coat összegyűjtésével képezzük, amely büffy-ooaí-et legalább két különböző donortól szerezzük be.
- 5. Az 1-3, igénypontok bármelyike szerinti eljárási, aboi az említett ailogén leuko-cita keveréket az alábbiak szerint állítjuk elő: - leukoeitákat; eluáiunk iegalébb két leukoela ülepítő szűréről, amely szőröket korábban leukodláknak a teles vérből történő kíüiepítésére használtak, ahol az említet tek jós vért legalább két különböző aliogén donorból gyűjtöttük bej és - a kapót leukoeitákat egybegyűjtve előállítjuk az említett aliogén leukoclta keveréket;; vagy - leukocitákat efuâiunk egy ieokocka ülepítő szűrőről amely szűrőt korábban leukocl-táknak összegyűjtött feyfy-coaí-bol történő ktűtsptésére használtak, ah©!; az említett összegyűjtött butfy-coai-ot legalább két különböző allogén donorból gyűjtöttük be.
- 6. Az előző Igénypontok bármelyike szerinti eljárás, ahol az említett allogén monocitákat ülepítéssei vagy antitest/gyöngy Izolálással állítjuk elő
- 7. Az előző Igénypontok bármelyike szerinti eljárás, ahol az említett együttes tenyésztést kb. 5 napig végezzük.
- 8. Az előző Igénypontok bármelyike szerinti eljárás, amelynél további lépésként a nem-kimerült éretlen DC-ket egy antigénnel töltjük be.
