Claims (8)

1. Spoj, koji je [image] pri čemu R je (CH2CH20)2H, (CH2)3C(O)NH2, (CH2)4CH(NH2)COOH (CH2)3COOH, (CH2)3NHC(O)NH2 ili (CH2CH2O)3H; q se kreće od 1 do 4; a Ab je trastuzumab.
2. Spoj prema zahtjevu 1, naznačen time, da R je (CH2CH2O)2H.
3. Spoj prema zahtjevu 1 ili 2, naznačen time, da je prosjek q približno 2.
4. Postupak za pripremu spoja prema bilo kojem od zahtjeva 1 do 3 sadrži sljedeće korake: a) Redukcija trastuzumaba uporabom 1.1 molarnih ekvivalenata TCEP inkubiranjem na 20°C tijekom 90 minuta; b) Dodavanje kap po kap konjugata veznika-sredstva otopljenog u DMSO u omjeru od 1.3 molarnih ekvivalenata po slobodnom tiolu do konačne koncentracije DMSO od 10%; c) Miješanje daljnjih 50 minuta na 20 ° C; d) Gašenje reakcije dodavanjem 1 molarnog ekvivalenta N-acetilcisteina po konjugatu veznika-sredstva; e) Desalinizacija konjugata u pufer PBS pH 7.4; f) Pročišćavanje uporabom r-proteina A stupca; i g) Filtriranje preko filtra od 0.2 µm.
5. Farmaceutski pripravak koji sadrži spoj prema bilo kojem od zahtjeva 1 do 3 i farmaceutski prihvatljiv nosač, poželjno za parenteralnu primjenu.
6. Spoj prema bilo kojem od zahtjeva 1 do 3 ili farmaceutski pripravak prema zahtjevu 5 za uporabu kao lijek, posebno za liječenje ili prevenciju tumora kod sisavca.
