Abstract
Whole blood or separated blood cells are diluted with water and the pH brought to 5.5-4.0 in the presence of an alum to precipitate a catalase complex. The latter may be separated and dried, or mixed with glycerol to give a stable product, or further purified by dispersing in water, bringing the pH to 7-8 (e.g. with sodium citrate), acidifying to pH 5.5-4, precipitating with ethanol and drying the precipitate. Preferably the temperature is kept around 5 DEG C. throughout. The initial fall in pH may be effected with the alum solution alone, or an acid such as sulphuric first added to bring the pH down to 6.0. Preferred pH values for the respective steps are 4.8 and 7.2. In a modification the initial water is treated with carbon dioxide (preferably in solid form) to bring its pH to 5.5-6.0. Examples are given of the process, using sodium aluminium sulphate as precipitant; chrome and ferric alums are also stated to be suitable. Specification 713,720 is referred to.
