Kyseesså oleva keksinto koskee menetelmåå ionin-5 vaihtohartsien valmistamiseksi, joihin on sidottu kinoion ikarboksyy1ihappoj ohdanna is ia.
Farmaseuttisten vaikuttavien aineiden sitominen ioninvaihtohartseihin on jo kauan ollut tunnettua esim. selvan ominaishajun omaavien vaikuttavien aineiden kaytto-10 kelpoisuuden parantamiseksi (CH-PS 383 552). On myoskin tunnettua farmaseuttisten vaikuttavien aineiden sitominen ioninvaihtohartsiin vaikuttavan aineen tasaisen vapautumi-sen aikaansaamiseksi pidemmalla aikavålillå (EP-OS 42 818).
15 Lisaksi on tunnettua matolaåkkeinå kåytettåvien vaikuttavien aineiden sitominen ioninvaihtohartsiin, jol-loin vaikutetaan vaikuttavien aineiden makuun (DE-OS 3 028 082).
Ioninvaihtohartseihin sidotut kinolonikarboksyyli-20 hapot ja niiden johdannaiset eivat viela toistaiseksi ole olleet tunnettuja.
Kyseesså olevan keksinnon kohteena on menetelmå ioninvaihtohartsien valmistamiseksi, jotka pohjautuvat heikosti happamiin kationisiin ioninvaihtohartseihin, joi-25 hin on sidottu kinolonikarboksyylihappojohdannaisia, joil-la on kaava (I) 0 FVTVv^oor2 ITT (I) 30 R1 jossa R1 on syklopropyyli, R2 on vety, 35 R3 on 2 90494 R*_ R4-N N-.
Jr' 5 jossa R4 on vety, metyyli tai etyyli, R5 on vety ja R6 on vety.
10 Keksinnon mukaiselle menetelmalle on tunnusoroaista, ettå heikosti happamia kationisia ioninvaihtohartseja kå-sitellaan kaavan (I) mukaisten kinolonikarboksyylihappo-johdannaisten liuoksilla tai suspensioilla vedesså tai polaarisissa liuottimissa.
15 Ioninvaihtohartseina tulevat kysymykseen sekå ka- tioniset etta anioniset tyypit, jolloin niiden matriisi voi olla geelimainen tai makrohuokoinen. Perusmonomeereina ioninvaihtimia vårten tulevat kysymykseen polymeroitavissa olevat monomeerit, jotka vastaavaa funktionalisointia 20 kayttåen voidaan muuttaa kationintai anioninvaihtohart-seiksi. Monomeereina mainittakoon esimerkiksi metakryyli-happoesteri, metakryylinitriili sekå styreenijohdannaiset. Muina komonomeereina peruspolymeerien valmistukseen kayte-taan polyvinyyliyhdisteitå, kuten esimerkiksi divinyyli-25 bentseenia, etyleeniglykolidimetyyliakrylaattia tai mety- leenibisakryyliamidia. Myos kondensaatiohartsit, jotka johtavat kationintai anioninvaihtajiin, kuten esimerkiksi fenolin, formaldehydin reaktiossa polyamiinien kanssa muo-dostuvat hartsit, ovat sopivia kaavan (I) mukaisten kino-30 lonikarbksyylihappojohdannaisten kantajiksi.

Edullisilla makrohuokoisilla hartseilla voi olla 35 eri huokoskokoja. Soveltuvien ioninvaihtohartsien ris- 3 90494 tisidosasteen tulisi olla edullisesti 20 % ja erityisen edullisesti 12 %. Keinohartsien raekoot ovat 50-1300 μτη, edullisesti 100-300 μιη.
Erityisesti mainittakoon hienonnettujen ioninvaih-5 tohartsien kåytto. Hienontaminen voi talloin tapahtua en-nen tai jålkeen kaavan (I) mukaisten kinolonikarboksyyli-happojen sitomisen.
Kaupallisia ioninvaihtohartseja, joita voidaan é) /6) (g) kåyttåå, ovat Lewatit™ Amberiite^ ja Dowex™ 10 Kaavan (I) mukaiset kinolonikarboksyylihapot ja niiden valmistus ovat tunnettuja (DE-OS 3 03 3 157) .
Erityisesti mainittakoon vaikuttavina aineina seu-raavat kinolonikarboksyylihapot: l-syklopropyyli-6-f luori-1,4-dihydro-4-okso-7- (1-15 piperatsinyyli tai 4-metyyli- tai 4-etyyli-l-piperatsinyy- li)kinoloni-3-karboksyylihappo.
Aivan erityisen edullisina mainittakoon siproflok-sasiini ja enrofloksasiini.

Antokokeet osoittavat, ettå nesteissa, joiden pH-arvot ovat valillå 1-3, vapautuu vaikuttava aine erityisen hyvin.

Kaavan (I) mukaisia kinolonikarboksyylihappojohdan-naisia sisaltavien ioninvaihtohartsien valmistus tapahtuu vedessa tai polaarisissa orgaanisissa liuottimissa, kuten esim. alkoholeissa, kuten metanolissa tai etanolissa, ke-5 toneissa, kuten asetonissa tai niiden seoksissa. Erityisen edullinen on vesi. Ioninvaihtohartsia ja vaikuttavaa ai-netta sekoitetaan talloin esim niin kauan (esim. 5-24 tun-tia) vedessa huoneen lampotilassa tai kohotetussa låmpoti-lassa, kunnes vaikuttava aine on sitoutunut taydellisesti. 10 Kuten jo on mainittu, ioninvaihtohartseja, joihin on sidottu kaavan (I) mukaisia kinolonikarboksyylihappo-johdannaisia, on mahdollista kåyttåå lååkeaineiden valmis-tukseen. 15 Elaimille soveltuvia lååkeainevalmisteita ovat esim. sellaiset, joiden yhteydessa maun parantamisella on merkitysta annon yhteydessa tai joiden yhteydessa pyritaan hidastuneeseen vaikuttavan aineen vapautumiseen annon jal-keen.
20 Naita ovat esim. kiinteåt valmisteet, kuten pulve- rit, esiseokset tai tiivisteet, rakeet, pelletit, table-tit, kerta-annokset, kapselit; suspensiot, joita kåytetaan esim. oraalisesti tai iholle. Ne valmistetaan suspendoi-malla vaikuttavaa ainetta sisaltåva hartsi kantajanestee-25 seen mahdollisesti lisaten muita apuaineita, kuten kostut-teita, variaineita, imeytymista ediståviå aineita, sailon-tåaineita, antioksidantteja, valolta suojaavia aineita.
Lisaamalla aineita, jotka lisaavat viskositeettia, voidaan naita suspensioita antaa myos niin sanottuina 30 "puolikiinteina” valmisteina, kuten esim. salvoina. Eri-tyisesti voidaan taman kaltaisia valmisteita kayttaa uta-resairauksien (mastitis) hoitoon tai oraalisina pastoina kissoille, koirille ja hevosille.
Kiinteiden valmisteiden valmistamiseksi vaikuttavaa 35 ainetta sisaltåva hartsi sekoitetaan sopivien kantaja-ai- i, 5 90494 neiden kanssa lisåten apuaineita ja saatetaan haluttuun muotoon.
Kantaja-aineina mainittakoon kaikki fysiologisesti siedetyt kiinteåt inertit aineet. Sellaisina toimivat epå-5 orgaaniset ja orgaaniset aineet. Epaorgaanisia aineita ovat esim. keittosuola, karbonaatit, kuten kalsiumkar-bonaatti, vetykarbonaatti, alumiinioksidit, piihapot, sa-vimaat, saostettu tai kolloidinen piidioksidi, fosfaatit. Orgaanisia aineita ovat esim. sokeri, selluloosa, ravinto-10 ja rehuaineet, kuten maitopulveri, elåinjauhot, vilja- jauhot ja -rouheet, tSrkkelykset.
Apuaineita ovat såilontåaineet, antioksidantit, variaineet, jotka on jo esitetty aikaisemmin edella.
Muita soveltuvia apuaineita ovat voitelu- ja liukuaineet, 15 kuten esim. magnesiumstearaatti, steariinihappo, talkki, bentoniitti, hajoamista edistavMt aineet, kuten tårkkelys tai ristisidoksellinen polyvinyylipyrrolidoni, sidosai-neet, kuten esim. tarkkelys, gelatiini, suoraketjuinen polyvinyylipyrrolidoni seka kuivat sidosaineet, kuten mik-20 rokiteinen selluloosa.
Suspensioiden valmistamiseksi sekoitetaan vaikutta-vaa ainetta sisaltavat hartsit kantaja-aineeseen mahdolli-simman homogeenisesti, mahdollisesti kayttaen lisaapuna * muita apuaineita, kuten kostutteita, sailontaaineita tai • 25 viskositeettia lisSaviM aineita.

Erityisen edullisia ovat ei-ioniset tensidit.
10 Muita apuaineita ovat esim.:
Variaineet, ts. kaikki elaimille kaytettaviksi sallitut variaineet, jotka voidaan liuottaa tai suspendoi-da.
Antioksidantit, kuten esim. sulfiitit tai metabi-15 sulfiitit, kuten kaliummetabisulfiitti, askorbiinihappo, butyylihydroksitolueeni, butyylihydroksianisoli, tokofero- li.
Sakeuttamisaineet esim. viskositeettia lisaavSt aineet, esim. epaorgaaniset sakeuttamisaineet, kuten ben-20 toniitti, kolloidinen piihappo, alumiinimonostearaatti, orgaaniset sakeuttamisaineet, kuten selluloosajohdannai-set, polyvinyylialkoholit ja niiden kopolymeerit, akrylaa-tit ja metakrylaatit, alginaatit, gelatiini, polyvinyyli-pyrrolidoni, polyetyleeniglykolit, vahat, arabikumi ja 25 ksantaanikumi tai mainittujenaineiden seokset.
Ioninvaihtohartsit, joihin on sidottu vaikuttavia aineita, voidaan lisata sellaisinaan tai esiseosten tai ravintotiivisteiden muodossa ravintoon.
Esiseokset tai ravintotiivisteet ovat vaikuttavan 30 aineen seoksia soveltuvan kantaja-aineen kanssa.
Kantaja-aineiksi katsotaan yksittaiset ravintoai-neet tai niiden seokset, sekå aikaisemmin edellå mainitut inertit kantaja-aineet.
Ne voivat sisåltåa edelleen muita apuaineita, kuten 35 esim. - aineita, jotka saatelevat juoksevuutta ja sekoit- I: 7 90494 tuvuutta, kuten esim. piihappoja, bentoniittia, ligniini-sulfonaatteja. Muita vaikuttavia aineita voivat olla esim. pe-nisilliinit, niiden suolat ja johdannaiset, kuten esim. penisilliini G:n prokaiinisuola, tai sen johdannaiset, 15 kuten oksasilliini tai kloksasilliini.
Vaikuttavaa ainetta sisaltSvien ioninvaihtohartsien anto tapahtuu edullisesti yhdessS ravinnon kanssa.
Ravinnoksi katsotaan kasviperaiset yksittaiset ra-vintoaineet, kuten heina, juurikkaat, vilja, viljan sivu-20 tuotteet, elainperaiset yksittaiset ravintoaineet, kuten liha, rasvat, maitotuotteet, luujauho, kalatuotteet, edelleen yksittaiset ravintoaineet, kuten vitamiinit, prote-.. iinit, aminohapot, esim. DL-metioniini, suolat, kuten kal- kki ja keittosuola. Ravinnoksi katsotaan myos lisa-, val-25 mis- ja seosrehuaineet. Lewatit^ S 100:aa H+ -muodossa, kunnes vesifaasi kirkas-tuu. Sen jalkeen toistetaan tåmå toimenpide, kunnes myos 24-tuntisen sekoituksen jålkeenmitåån kirkasta vesifaasia ei ole. Esimerkki 2 (g) 2714 ml Lewatit** SPC 108:aa H+-muodossa sekoitetaan 10 yhdessa 5000 ml:n kanssa demineralisoitua vettå ja 782 g:n kanssa enrofloksasiinia yli yon. Valmiste-esimerkkeia 4. Esisekoite ruokintalaåkeainetta vårten esimerkin 2 mukaista enrofloksasiini-25 ioninvaihtajaa 4,85 kg (4,85 g vastaa 2,5 g enrofloksasiinia) vehnajauhoa 95.15 ka 100.00 kg
Valmistus: 30 Aineet sekoitetaan sekoittajassa homogeeniseksi.

Valmistus:
Kaksiprosenttinen metyy1ihydroksipropyyliselluloo-15 sageeli valmistetaan tavalliseen tapaan. Siihen liuotetaan bentsyylialkoholi ja aromiaineet ja suspendoidaan keksin-ηδη mukainen vaikuttava aine.
