Claims (14)
1. Förfarande för framställning av ett terapeutiskt 5 användbart konjugat, som omfattar en toxindel och en del, som binder sig tili en mälcell, och är selektivt mot ga- strointestinala tumörer och varvid delen, som binder sig tili en mälcell, omfattar en C242-antikropp eller en del, som binder sig tili en mälcell, som har väsentligen samma 10 cellbindande egenskaper, kännetecknat av att förfarandet omfattar: (a) för de konjugaters del, i vilka delen som binder sig tili en mälcell kopplas direkt tili toxindelen, fär toxindelen reagera med den del som binder sig tili 15 mälcellen; eller (b) för de konjugaters del, i vilka delen som binder sig tili en mälcell kopplas tili toxindelen via en bindningsdel, fär den med bindningsdelen derivatiserade delen som binder sig tili en mälcell reagera med 20 toxindelen eller fär den med bindningsdelen derivatiserade toxindelen reagera med delen som binder sig tili en mälcell.
2. Förfarande enligt patentkrav 1, kännetecknat av att delen som binder sig tili en mälcell ·. 25 omfattar en C242-antikropp eller ett fragment därav.
3. Förfarande enligt patentkrav 1 eller 2, kännetecknat av att toxindelen omfattar rekombinant ricin-A.
4. Förfarande enligt patentkrav 1, 2 eller 3, 30 kännetecknat av att delen som binder sig tili . en mälcell och toxindelen kopplas tili varandra med hjälp av en bifunktionell bindningsdel.

30 CDR3: LeuGlnHisLeuGluTyrProPheThr Tung kedja CDR1: TyrThrGlyMetAsn CDR2: TrpIleAspThrThrThrGlyGluProThrTyrAlaGluAspPheLysGly 35 CDR3: ArgGlyProTyrAsnTrpTyrPheAspVal 106928 varvid CDR-omrädena är ordnade sä att delen som binder sig till en mälcell uppvisar väsentligen sarama bindningsegen-skaper som C242-antikroppen.

11. Förfarande enligt patentkrav 1, kanne-tecknat av att delarna för konjugatet, som omfattar en C242-antikropp och rekombinant ricin-A, kopplas till varandra frän en aminogrupp i C242 till en tiol i en 30 cysteingrupp i ricin-A via en 3-mercaptosmörsyreradikal (-CO.CH2.CH(CH3) -S-) .

13. Förfarande enligt patentkrav 1, k ä n n e - 15 tecknatav att konjugatet, som omfattar en toxindel och en del som binder sig tili en mälcell, är specifikt mot en epitop av en gastrointestinal tumör.
