Ejemplos 1 a 8: preparación de composiciones farmacéuticas que comprenden lubricantes de sal no metálica
Las composiciones farmacéuticas de los Ejemplos 1 a 3 se prepararon mediante el empleo de un compuesto de la fórmula (I) (en adelante, referido como "HM781-36B," Dongwoo Syntech Co., Ltd., Corea); manitol (Roquette);
5 Povidona® (BASF); Crospovidona® (BASF); y éster de ácido graso de sacarosa (Daiichi Kogyo Seiyaku, Japón), aceite vegetal hidrogenado (Lubritab®, JRS Pharma), o ácido esteárico (Emery Oleochemicals.), como un lubricante de sal no metálica, según la composición y la cantidad (unidad: mg) descritas en la Tabla 2.
Específicamente, HM781-36B y manitol se mezclaron y la mezcla se sometió a un proceso de granulación húmeda mediante un método convencional con el empleo de una solución aglutinante de povidona disuelta en agua 10 purificada. Los gránulos húmedos así obtenidos se secaron, se mezclaron con manitol y crospovidona, y posteriormente se añadió un lubricante, que se tamizó previamente a través de un tamiz de malla 30, para preparar una mezcla final. La mezcla final así preparada se transformó en un comprimido que tenía una dureza de aproximadamente 5 a 10 kp mediante una máquina de comprimidos (Sejong, Corea) según un método convencional.

Con el fin de observar los cambios en la estabilidad de las composiciones farmacéuticas preparadas según los Ejemplos 1 y 2 y Ejemplos Comparativos 1 y 3 frente a la temperatura y la humedad, las composiciones farmacéuticas se expusieron a 40º C y 75% de HR. Después de 1 y 2 semanas, respectivamente, el compuesto IV relacionado, un producto de degradación principal de HM781-36B, se extrajo con 60% de acetonitrilo como disolvente, y a continuación, se realizaron análisis de HPLC. 10 Con el fin de observar los cambios en la estabilidad de las composiciones farmacéuticas preparadas según los Ejemplos 1 y 2 y Ejemplos Comparativos 1 y 3 frente a la temperatura y la humedad en condiciones aceleradas, las composiciones fueron expuestos a 40º C y 75% HR en recipientes de HDPE sellados durante 1, 3 y 6 meses. El compuesto IV relacionado de cada composición se extrajo con 60% de acetonitrilo como disolvente, y a continuación, se realizaron análisis de HPLC. Los resultados se muestran en la Tabla 9 y en la Fig. 4.

Como se muestra en las Tablas 6 a 9 y Figs. 1 a 4, la formación del compuesto relacionado IV se redujo en aproximadamente de 4 a 10 veces o más en las composiciones farmacéuticas que comprendían cualquiera de los lubricantes de sal no metálica en comparación con las composiciones farmacéuticas que comprendían los
20 lubricantes de sal metálica. Por lo tanto, la estabilidad de almacenamiento de las composiciones farmacéuticas que contienen HM781-36B como un ingrediente activo puede ser mejorada de manera significativa mediante la adición de cualquiera de los lubricantes de sal no metálica a las composiciones farmacéuticas.
Según las directrices de la Conferencia Internacional sobre Harmonisation of Technical Requirements for Registration of Pharmaceuticals for Human Use (ICH), los límites de impurezas desconocidas y conocidas se
25 prescriben como 0,2% y 0,5%, respectivamente. Las composiciones farmacéuticas de los Ejemplos 1 y 2 según la presente invención mostraron resultados satisfactorios de menos de 0,5% a 40º C en un ensayo de estabilidad acelerada como se describe en la directriz ICH. En contraste, las composiciones farmacéuticas de los Ejemplos Comparativos 1 y 3 que comprenden lubricantes de sales metálicas convencionales superaron los límites predeterminados de la directriz ICH.
