Die Erfindung betrifft ein Verfahren zur Herstellung von organischen Alkinverbindungen der Formel (I), X-C=C-Y, durch Umsetzung von organischen Halogenverbindungen der Formel (Ia), X-Hal, mit organischen terminalen Alkinverbindungen der Formel (Ib), H-C=C-Y, wobei X und Y gleiche oder verschiedene organische Reste bedeuten und Hal für Chlor oder Brom steht, in inerten Lösungsmitteln unter Einwirkung von Mikrowellenstrahlung, in Gegenwart mindestens einer Metallverbindung und mindestens einer Base.
Description
Verfahren zur Herstellung von organischen Alkinverbindungen
Beschreibung
Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung von organischen Alkinverbindungen der Formel I
X-C≡C-Y (I)
durch Umsetzung von organischen Halogenverbindungen der Formel Ia
X-Hal (Ia),
mit organischen terminalen Alkinverbindungen der Formel Ib
H-C≡C-Y (Ib) ,
wobei X und Y gleiche oder verschiedene organische Reste bedeuten und Hai für Chlor oder Brom steht, in inerten Lösungsmitteln unter Einwirkung von Mikrowellenstrahlung, in Gegenwart mindestens einer MetallVerbindung und mindestens einer Base.
Unter den üblichen Bedingungen der Sonogashira-Reaktion werden Aryl- oder Alkenylhalogenide unter Katalyse mit Palladium- und Kupfersalzen bei erhöhter Temperatur mit terminalen Alkinverbindungen zu entsprechend substituierten Alkinverbindungen umgesetzt .
Eine deutliche Herabsetzung der Reaktionsdauer lässt sich erzielen, wenn man die Umsetzung unter Einwirkung von Mikrowellenstrahlung durchführt .
So beschreiben J.-X. Wang et al . (J. Chem. Research (S) , 2000, S. 536 - 537) Umsetzungen von verschiedenen terminalen Alkinen mit organischen Iodverbindungen in Gegenwart von Kupfer (I) iodid/Tri- phenylphosphin und Kaliumcarbonat in Dimethylformamid (DMF) . Die in dieser Publikation in Tabelle 2 aufgeführte Gegenüberstellung der Umsetzungen, einerseits unter Rückfluss des DMF, andererseits unter Einwirkung einer Mikrowellenstrahlungsquelle mit einer Leistung von 375 , zeigt in eindrucksvoller Weise, dass, bei vergleichbaren Ausbeuten, in letzterem Fall die Reaktionen um Faktoren von 48 bis 144 schneller verlaufen als in ersterem Fall.
Untersuchungen zur lösungsmittelfreien Umsetzung von Aryl-, Hete- roaryl- und Vinyliodiden mit terminalen Alkinen in Gegenwart von Palladium/Kupfer (I) iodid/Triphenylphosphin und auf Aluminiumoxid
geträgertem Kaliumfluorid unter Einwirkung von Mikrowellenstrahlung sind von G.W. Kabalka et al . (Tetrahedron Lett. 41, 2000, S. 5151 - 5154) durchgeführt worden. Die Autoren erwähnen hierbei (S. 5152), dass Arylchloride und -bromide nicht reagierten und die Ausgangsmaterialien unverändert zurückerhalten worden seien.
Es wurde nun überraschenderweise gefunden, dass sich auch organische Chlor- und Bromverbindungen in guten bis sehr guten Ausbeuten mit terminalen organischen Alkinverbindungen zu Alkinderiva- ten umsetzen lassen.
Dementsprechend wurde ein Verfahren zur Herstellung von organischen Alkinverbindungen der Formel I
X-C≡C-Y (I)
durch Umsetzung von organischen Halogenverbindungen der Formel Ia
X-Hal (Ia) ,
mit organischen terminalen Alkinverbindungen der Formel Ib
H-C≡C-Y (Ib) ,
wobei X und Y gleiche oder verschiedene organische Reste bedeuten,
in inerten Lösungsmitteln unter Einwirkung von Mikrowellenstrahlung,
in Gegenwart mindestens einer Metallverbindung und mindestens einer Base, gefunden, welches dadurch gekennzeichnet ist, dass Hai für Chlor oder Brom steht .
Unter inerten Lösungsmitteln sind hierbei solche Flüssigkeiten oder Flüssigkeitsgemische zu verstehen, welche unter den Umsetzungsbedingungen weder mit den Edukten noch mit den Produkten reagieren.
Insbesondere handelt es sich bei solchen inerten Lösungsmitteln um polare, aprotische Flüssigkeiten, da bei Verwendung von proti- schen Flüssigkeiten mit unerwünschten, durch Protonierung ausgelösten Nebenreaktionen gerechnet werden muss.
Vereinfachend wird im Folgenden auch dann von "Lösungsmittel" bzw. von "lösen" usw. gesprochen, selbst wenn im Einzelfall beispielsweise die verwendete (n) Base(n) oder MetallVerbindung (en)
nicht vollständig gelöst sind, sondern in Suspension (oder Emulsion) vorliegen.
Als MetallVerbindungen kommen vorzugsweise solche in Betracht, welche ein Metall ausgewählt aus der Gruppe bestehend aus Magnesium, Calcium, Strontium, Barium, Titan, Zirkonium, Hafnium, Eisen, Ruthenium, Osmium, Kobalt, Rhodium, Iridium, Nickel, Palladium, Platin, Kupfer, Silber, Gold, Zink, Cadmium und Quecksilber enthalten. Besonders bevorzugt werden Kupferverbindungen verwen- det.
Hervorzuheben sind als MetallVerbindungen die Halogenide, insbesondere die Chloride und Bromide, aber auch die Iodide, der genannten Metalle. Sofern diese Halogenide Addukte mit Triaryl- phosphinen, wie beispielsweise Triphenylphosphin, bilden, werden sie in vorteilhafter Weise in Form dieser Addukte verwendet.
Als MetallVerbindungen sollen darüberhinaus auch Metalle selbst, insbesondere die bereits genannten Metalle in elementarer Form verstanden werden. Desweiteren können auch Kombinationen mehrerer Metallverbindungen, mehrerer Metalle, aber auch Kombinationen von Metallen und MetallVerbindungen zur Verwendung kommen. Die in der Umsetzung katalytisch aktive metallhaltige Spezies uss nicht notwendigerweise mit den eingesetzten Metallverbindungen iden- tisch sein, sondern kann auch erst durch Reaktion mit den Edukten und/oder der Base/den Basen in situ entstehen.
Unter organischen Resten X und Y sind gesättigte oder ungesättigte Kohlenwasserstoffreste, sowie Kohlenwasserstoffreste zu verstehen, welche sowohl gesättigte als auch ungesättigte Molekülteile enthalten. Darüberhinaus können in den Kohlenwasserstoffresten auch übliche Heteroatome, wie etwa Stickstoff, Sauerstoff, Phosphor, Schwefel, Fluor, Chlor, Brom oder auch Iod enthalten sein. Die organischen Reste X und Y besitzen üblicherweise Molmassen bis etwa 600 g/mol. Im Einzelfall können die Molmassen der Reste X und Y jedoch auch höher sein.
Bevorzugte organische Reste X und Y enthalten gesättigte oder ungesättigte carbo- oder heterocyclische Reste, in welchen sowohl -Hai, d.h. Chlor oder Brom, als auch H-C≡C- direkt an die gesättigten oder ungesättigten carbo- oder heterocyclischen Reste gebunden sind.
Insbesondere entspricht X einem Rest der Formel Ha pl_γl_(Al_γ3)m,_(Tl_Bl_)m_T3_ (IIa)
und Y einem Rest der Formel Ilb

Bevorzugtes Alkyl für P1 und P2 sind die verzweigten oder unverzweigten Ci-Cε-Alkylketten, wie etwa Methyl, Ethyl, n-Propyl, 1- Methylethyl, n-Butyl, 1-Methylpropyl, 2-Methylpropyl, 1, 1-Dimethylethyl, n-Pentyl, 1-Methylbutyl, 2-Methylbutyl , 3-Methylbutyl , 2 , 2-Dimethylpropyl, 1-Ethylpropyl und n-Hexyl.
Als polymerisierbare oder zur Polymerisation geeignete Gruppe oder als Rest, welcher eine polymerisierbare oder zur Polymerisation geeignete Gruppe trägt (nachfolgend werden solche Gruppen oder Reste auch einfach als "reaktive Reste" bezeichnet) , kommen für P1 und P2 insbesondere in Frage:
-N=C=0, -N=C=S, -O-C≡N, -COOH, -OH oder -NH2, wobei die Reste R1 bis R3 gleich oder verschieden sein können und Wasserstoff oder Cι-C4-Alkyl, wie Methyl, Ethyl, n-Propyl, iso- Propyl, n-Butyl, iso-Butyl, sec.-Butyl oder tert.-Butyl bedeuten.
Als polymerisierbare Gruppe für P1 und P2 ist insbesondere der Acrylat-, Methacrylat- sowie der Vinylrest hervorzuheben.
Als Cι~C4-Alkyl für R in den unter den verknüpfenden Einheiten Y1 bis Y4 und B1, B2 aufgeführten Gruppen -CO-N(R)-, -(R)N-CO-, -O-CO-N(R)-, -(R)N-CO-O- und - (R)N-CO-N(R) - kommen Methyl, Ethyl, n-Propyl, iso-Propyl, n-Butyl, iso-Butyl, sec.-Butyl und tert.- Butyl in Betracht. Sofern in einer der Einheiten Y1 bis Y4 und B1, B2 ein oder zwei Reste R vorliegen, können gegebenenfalls in den übrigen Einheiten vorhandene Reste R gleich oder verschieden sein. Ebensolches gilt für den Fall, dass in einer Einheit zwei Reste R vorliegen.
Als Spacer A1 und A2 kommen alle dem Fachmann für diesen Zweck bekannten Gruppen in Betracht. Die Spacer enthalten in der Regel ein bis 30, vorzugsweise ein bis 12, besonders bevorzugt ein bis sechs Kohlenstoffatome und bestehen aus vorwiegend linearen ali- phatischen Gruppen. Sie können in der Kette, z.B. durch nicht benachbarte Sauerstoff- oder Schwefelatome oder Imino- oder Alkyl- iminogruppen, wie beispielsweise Methyliminogruppen, unterbrochen sein. Als Substituenten für die Spacerkette kommen Fluor, Chlor, Brom, Cyan, Methyl und Ethyl in Betracht.
Repräsentative Spacer sind beispielsweise:

Bevorzugte Spacer sind Ethylen, Propylen, n-Butylen, n-Pentylen und n-Hexylen.
Bei den Resten T1 bis T4 handelt es sich um Ringsysteme, welche durch Fluor, Chlor, Brom, Cyan, Hydroxy, Formyl, Nitro, Cι-C2o-Alkyl, Cχ-C2o-Alkoxy, Cι-C20-Alkoxycarbonyl, C!-C20-Mono- alkylaminocarbonyl , Cι-C o-Alkylcarbonyl , Cι-C2o-Alkylcarbonyloxy oder Cι-C2Q-Alkylcarbonylamino substituiert sein können.

Weiter kann anstelle der Hydroxy- beispielsweise auch eine Carbo- xylgruppe (P1 = Wasserstoff und Y1 = -0C0- und/oder P2 = Wasserstoff und Y2 = -C00-) vorliegen. Bei den difunktionellen Verbindungen können desweiteren sowohl Hydroxy- als auch Carboxylgrup- pen zugegen sein.
Diese beispielhaft aufgeführten Hydroxy- bzw. Carbonsäure- bzw. Hydroxy/Carbonsäure-Verbindungen sind wiederum als direkte Produkte des erfindungsgemäßen Herstellverfahrens anzusehen.
Die Edukte der Formeln Ia und Ib werden üblicherweise im Molverhältnis von 2:1 bis 1:2 zusammen mit der mindestens einen Metallverbindung und der mindestens einen Base im inerten Lösungsmittel gelöst. Die Herstellung der Lösung erfolgt im Normalfall bei Raumtemperatur, kann jedoch im Einzelfall auch bei darüber oder darunter liegender Temperatur stattfinden.
Die Temperatur bei der eigentlichen Umsetzung unter Einwirkung von Mikrowellenstrahlung ist unkritisch. Üblicherweise findet die Umsetzung bei Temperaturen zwischen Raumtemperatur und der Siede- temperatur des verwendeten Lösungsmittels statt.
Als Lösungsmittel wird vorzugsweise Dimethylformamid ("DMF"), N-Methylpyrrolidon ("NMP") oder auch eine Mischung aus beiden verwendet. Besonders bevorzugt verwendet man im erfindungsgemäßen Verfahren DMF als Lösungsmittel (bzw. als Suspendiermittel) .
Als mindestens eine Base kommt vorzugsweise eine Verbindung ausgewählt aus der Gruppe bestehend aus Alkalicarbonaten, Alkaliphosphaten und Tri (Cι-C4~alkyl) aminen in Frage, wobei die Alkali- carbonate hervorzuheben sind.
Die Gruppe der geeigneten Basen umfasst insbesondere Natriumcar- bonat, Kaliumcarbonat, Natriumphosphat, Kaliumphosphat, Trime- thyl-, Triethyl- und Triisopropylamin.
Besonders bevorzugt verwendet man Kaliumcarbonat.
Im Einzelfall kann auch die Zugabe von Kaliumiodid vorteilhaft für die Umsetzung sein. Ob ein solcher positiver Effekt zu ver- zeichnen ist und in welchen Mengen das Kaliumiodid gegebenenfalls
zugegeben werden muss, lässt sich dabei leicht durch Vorversuche ermitteln.
Die Leistung der verwendeten Mikrowellenstrahlungsquelle liegt üblicherweise bei einigen zehn bis einigen hundert Watt und ist dem Volumen des Reaktionsansatzes anzupassen. Die geeignete Dimensionierung der Strahlungsquelle ist dem Fachmann üblicherweise bekannt und/oder lässt sich durch Vorversuche leicht ermitteln.
Die Aufarbeitung und Reinigung der erhaltenen Alkinverbindungen erfolgt nach üblichen Methoden der organischen Synthese.

Für die nachfolgend beschriebenen Versuche wurden verwendet:
Versuchsdurchführung :
Allgemeine Reaktionsgleichung:
5 mmol 4-Halogenbenzoesäure (Halogen: Chlor, Brom oder Iod) , 7,5 mmol Phenylacetylen, 0,5 mmol Kupfer (I) iodid, 1,0 mmol Triphenylphosphin, 7,5 mmol Kaliumcarbonat und 10 ml DMF wurden unter Argonatmosphäre in einem mit Magnetrührer versehenen 100-ml-Vierhalskolben vorgelegt, innerhalb von 5 min auf eine Temperatur von 155°C erhitzt und für die Dauer von 20 min bei Rückfluss der maximalen Strahlungsleistung eines Mikrowellengeräts (MLS-Ethos 1600;
ungepulst; Magnetronfrequenz 2450 MHz; maximale Leistung 375 W) ausgesetzt .
Zur Aufarbeitung wurde der Feststoff (im Wesentlichen Kaliumcar- bonat) abfiltriert, mit 100 ml Dichlormethan nachgewaschen und die erhaltene Lösung dreimal mit je 50 ml einer gesättigten, wässrigen Natriumchlorid-Lösung extrahiert. Es erfolgte Trocknung der Dichlormethan-Lösung über Natriumsulfat und abschließende Entfernung des Lösungsmittels am Rotationsverdampfer.
Zu Vergleichszwecken wurden auch Versuche unter Zugabe von 0,5 mmol Kaliumiodid durchgeführt. Die Mengen der übrigen eingesetzten Substanzen blieben unverändert; ebenso erfolgte die Versuchsführung und Aufarbeitung völlig analog zum vorher Beschriebenen.
Ergebnisse:
Die Versuchsergebnisse sind in der nachfolgenden Tabelle aufgeführt .
Im Falle der Verwendung von 4-Iodbenzoesäure (Beispiel 1 (Vergleich) ) wurde die mit Abstand geringste Ausbeute, im Falle der Verwendung von 4-Brom- und insbesondere 4-Chlorbenzoesäure (Beispiele 4 und 5 bzw. 2 und 3) wurden dagegen deutlich höhere Ausbeuten an gewünschter ZielVerbindung erhalten. In den hier durchgeführten Versuchen wurde bei Zusatz von Kaliumiodid (Beispiele 3 und 5) eine Verschlechterung gegenüber der Kaliumiodid-freien Versuchsführung (Beispiele 2 und 4) festgestellt. Denkbar ist jedoch, dass im Einzelfall die Zugabe von Kaliumiodid eine vorteilhafte Wirkung mit sich bringen kann.
Claims
Patentansprüche
1. Verfahren zur Herstellung von organischen Alkinverbindungen der Formel I
X-C≡C-Y (I)
durch Umsetzung von organischen Halogenverbindungen der For- mel Ia
X-Hal (Ia) ,
mit organischen terminalen Alkinverbindungen der Formel Ib
H-C≡C-Y (Ib),
wobei X und Y gleiche oder verschiedene organische Reste bedeuten,
in inerten Lösungsmitteln unter Einwirkung von Mikrowellenstrahlung,
in Gegenwart mindestens einer Metallverbindung und mindestens einer Base,
dadurch gekennzeichnet, dass Hai für Chlor oder Brom steht.
2. Verfahren nach Anspruch 1, dadurch gekennzeichnet, dass es in Gegenwart mindestens einer MetallVerbindung durchgeführt wird, welche ein Metall ausgewählt aus der Gruppe bestehend aus Magnesium, Calcium, Strontium, Barium, Titan, Zirkonium, Hafnium, Eisen, Ruthenium, Osmium, Kobalt, Rhodium, Iridium, Nickel, Palladium, Platin, Kupfer, Silber, Gold, Zink, Cad- mium und Quecksilber enthält.
3. Verfahren nach Anspruch 1, dadurch gekennzeichnet, dass es in Gegenwart einer Kupferverbindung durchgeführt wird.
4. Verfahren nach einem oder mehreren der Ansprüche 1 bis 3, dadurch gekennzeichnet, dass X und Y gleiche oder verschiedene gesättigte oder ungesättigte carbo- oder heterocyclische Reste enthaltende organische Reste bedeuten, in welchen sowohl -Hai als auch H-C= C- direkt an die gesättigten oder ungesät- tigten carbo- oder heterocyclischen Reste gebunden sind.
Verfahren nach einem oder mehreren der Ansprüche 1 bis 3, dadurch gekennzeichnet, dass

P1 und/oder P2 entsprechen jeweils einem Rest P1' und/oder P2', welcher eine unter den Umsetzungsbedingungen stabile Vorläufergruppe bezeichnet, die zu den entsprechenden po- ly erisierbaren oder zur Polymerisation geeigneten Gruppen pi und/oder P2 oder den Resten P1 und/oder P2, welche eine polymerisierbare oder zur Polymerisation geeignete Gruppe tragen, umgesetzt oder durch diese substituiert werden können,

Verfahren nach Anspruch 5, dadurch gekennzeichnet, dass die Reste T1 bis T4 in den Formeln Ila und Ilb ausgewählt sind aus der Gruppe bestehend aus
Verfahren nach einem oder mehreren der Ansprüche 1 bis 6, dadurch gekennzeichnet, dass als inertes Lösungsmittel Di e- thylformamid oder N-Methylpyrrolidon oder eine Mischung aus beiden verwendet wird.
Verfahren nach einem oder mehreren der Ansprüche 1 bis 6, dadurch gekennzeichnet, dass als inertes Lösungsmittel Di e- thylformamid verwendet wird.
Verfahren nach einem oder mehreren der Ansprüche 1 bis 8, dadurch gekennzeichnet, dass als mindestens eine Base eine Verbindung ausgewählt aus der Gruppe bestehend aus Alkalicarbo- naten, Alkaliphosphaten und Tri (Cι-C4-alkyl) aminen verwendet wird.
10. Verfahren nach einem oder mehreren der Ansprüche 1 bis 8, dadurch gekennzeichnet, dass als Base mindestens ein Alkalicar- bonat verwendet wird.
