Abstract
Настоящее изобретение относится к способам получения библиотек эукариотических клеток, кодирующих набор связывающих молекул ("связывающие молекулы"), причем в указанных способах используется сайт-специфическая нуклеаза для направленного расщепления клеточной ДНК с целью улучшения сайт-специфического встраивания генов связывающих молекул посредством эндогенных клеточных механизмов репарации. Получают популяции эукариотических клеток, в которых набор генов, кодирующих связывающие молекулы, встроен в требуемый локус в клеточной ДНК (например, геномный локус), что обеспечивает экспрессию кодируемой связывающей молекулы, в результате чего получают популяцию клеток, экспрессирующих разные связывающие молекулы.
