Claims (11)
1. Fremgangsmåde til fremstilling af et glycomodificeret protein, der omfatter: binding af et azido-modificeret sukker til en GIcNAc-rest på et første protein ved anvendelse af en Y289L-mutant-galactosyltransferase; og blanding af det første protein, med det azido-modificerede sukker bundet dertil, med et reporter-molekyle, bæremolekyle eller en fast bærer, der kan reagere med det azido-modificerede sukker; hvor det azido-modificerede sukker er UDP-GalNAz og reporter-molekylet, bæremolekylet eller den faste bærer mærkes med en alkyn eller en aktiveret alkyngruppe; og hvor reporter-molekylet, bæremolekylet eller den faste bærer binder til proteinet ved azidgruppen af det azido-modificerede sukker via alkynen eller den aktiverede alkyngruppe, hvorved der dannes et glycomodificeret protein.
2. Fremgangsmåde ifølge krav 1, hvor det første protein er et antistof.
3. Fremgangsmåde ifølge krav 1 eller krav 2, hvor før bindingstrinnet, fremgangsmåden omfatter spaltning af et oligosaccharid til stede på et første protein ved en GIcNAc-GIcNAc -binding for at opnå et protein, der omfatter en GIcNAc-rest, hvor eventuelt oligosaccharidet spaltes ved anvendelse af endoglycosidase H-spaltning ved GIcNAc-GIcNAc-bindingen eller endoglycosidase M-spaltning ved GlcNAc-GlcNAc-bindingen, efterfulgt af overførsel af oligosaccharidet til det første protein.
4. Fremgangsmåde ifølge et hvilket som helst foregående krav, der endvidere omfatter: spaltning af et oligosaccharid til stede på et andet protein ved en GIcNAc-GIcNAc - binding for at opnå et oligosaccharid med en GIcNAc-rest; binding af oligosaccharidet til reporter-molekylet, den faste bærer eller bæremolekylet, der kan reagere med den kemiske håndtering.
5. Fremgangsmåde ifølge krav 4, hvor oligosaccharidet med en GlcNAc-rest behandles med ammoniumbicarbonat og oligosaccharidet bindes til en alkyn via en succinimidylester.
6. Fremgangsmåde ifølge krav 4 eller krav 5, hvor det andet protein syntetiseres i en anden cellelinje eller celletype end det første protein, hvor eventuelt det andet protein syntetiseres i en human celle.
7. Fremgangsmåde ifølge et hvilket som helst foregående krav, hvor det modificerede oligosaccharid omfatter et reporter-molekyle valgt fra gruppen bestående af et fluorescerende farvestof, et enzym, et radioaktivt mærket stof, en metalchelator eller et detekterbart substrat, og/eller det modificerede oligosaccharid omfatter et bæremolekyle valgt fra gruppen bestående af terapeutiske midler, dna, protein, peptider og sukkere.
8. Fremgangsmåde ifølge et hvilket som helst foregående krav, hvor det glycomodificerede protein bliver mere eller mindre antigent sammenlignet med det første protein, inden det modificeres.
9. Fremgangsmåde ifølge et hvilket som helst foregående krav, hvor det første protein er afledt af en ikke-human kilde, hvor eventuelt det glycomodificerede protein er humaniseret.
10. Fremgangsmåde ifølge et hvilket som helst foregående krav, hvor bindingstrinnet er i en opløsning, der i alt væsentligt er fri for proteaser.
11. Antistof, der omfatter et mærket oligosaccharid, hvor antistoffet kan opnås ved fremgangsmåden ifølge et hvilket som helst foregående krav.
