Ved parenteral indgivelse af ikke-steroide inflammationshæmmende midler observeres der ofte lokale irritationer og hæmolytiske virkninger. Til grund for den foreliggende opfindelse ligger den opgave at tilvejebringe en mere acceptabel parenteral applikationsform for ikke-steroide inflammationshæmmende midler.

Fra tysk offentliggørelsesskrift 2730570 er det kendt at anvende blandingsmiceller af galdesyrer og lipider til solubilisering af i vand tungt- eller uopløselige farmaceutiske aktivstoffer i et vandigt medium. Det har nu overraskende vist sig, at vandige blandingsmicelleopløsninger af ikke-steroide inflammationshæmmende midler ved 10 parenteral administration er væsentligt bedre fordragelige end vandigt/organiske eller selv rent vandige opløsninger af sådanne inflammationshæmmende midler, som ikke er fremstillet under anvendelse af blandingsmiceller.
Den foreliggende opfindelse angår således vandige blandingsmicelle-opløsninger, 15 der indeholder et salt af en galdesyre, et lipid og en ikke-steroid inflammationshæmmer.
I et andet aspekt angår opfindelsen anvendelse af blandingsmiceller af galdesyresalte og lipider til solubilisering af ikke-steroide inflammationshæmmere i et van-20 digt miljø.
I de foreliggende blandingsmicelleopløsninger kommer de gaIdesyre-salte eller gal- i desyrederivater, der er nævnt i tysk offentliggørelsesskrift 2730570, i betragtning, især cholater, glycocholater og taurocholater, især alkalisaltene såsom natriumsal-25 tene. Na-glycocholat er særligt foretrukket.
Som lipider kommer især phosphatidylcholiner på tale, fx naturlige ledtiner eller syntetiske lecitiner med afledte sidekæder (fx sådanne, som er beskrevet i europæisk patentansøgning A2-0154977). Naturlige lecitiner såsom ægge- eller 30 soyalecitin foretrækkes.
Ikke-steroide inflammationshæmmere (non-steroidal anti-inflammatory drugs, NSAID’s) i forbindelse med den foreliggende opfindelse er forbindelser, som strukturelt adskiller sig fra steroider og udviser en inflammationshæmmende 35 virkning. Sådanne forbindelser er ofte karakteriseret ved tilstedeværelsen af en

H carboxylsyregruppe og/eller er derivater af eddikesyre eller propionsyre. Eksempler I
på sådanne ikke-steroide inflammationshæmmere er carprofen, ibuprofen, benoxa- I
profen, naproxen, sulindac, zomepirac, fenclofenac, alclofenac, ibufenac, flunixin, I
indomethacin eller salte deraf. En inden for rammerne af den foreliggende opfindel- I
5 se foretrukken ikke-steroid inflammationshæmmer er carprofen (6-chlor-a-methyl- I
carbozol-2-eddikesyre) og fysiologisk acceptable salte deraf med baser, fx alkali- I
H metalhydroxider, aminer eller basiske aminosyrer såsom arginin eller lysin. I
Molforholdet mellem lipid og galdesyresaltet ligger hensigtsmæssigt i størrelsesor- I
10 denen fra 0,1:1 til 2:1. Der foretrækkes blandingsforhold fra 0,8:1 til 1,5:1. I
Andelen af lipid plus galdesyre i injektionsopløsningen kan variere inden for brede I
intervaller og udgør fx 50-300 mg/ml injektionsopløsning. I

opløsningsmidler kommer sådanne på tale, i hvilke de komponenter, der skal op- I

I I en yderligere udførelsesform kan der først fremstilles en vandig blandingsmicelle- I
I opløsning af et lipid og et galdesyresalt efterfulgt af tilsætning af aktivstoffet. I

Den tid, som er nødvendig, indtil den således vundne blanding efter omrøring synes homogen, afhænger af arten af galdesyren, lipidet, aktivstoffet og koncentrationerne deraf, og kan i reglen forkortes ved kortvarig opvarmning.
5 Blandingsmicelleopløsningerne ifølge opfindelsen justeres hensigtsmæssigt til en pH-værdi på ca. 5,5-7,5.
Blandingsmicelleopløsningerne ifølge opfindelsen kan indeholde yderligere hjælpestoffer, fx buffere, isotoniserende tilsætninger, stabilisatorer og/eller konserve-10 ringsmidler, fx benzylalkohol. Som isotoniserende tilsætninger kommer især følgende i betragtning: Natriumchlorid, mannit eller glucose. Som buffer kan der anvendes tris-buffer, phospat-buffer, citrat-buffer, citrat-phosphat-blandingsbuffer osv. Det osmotiske tryk i injektionsopløsningerne ifølge opfindelsen bør i det ideelle tilfælde svare til blodets, dvs. ca. 300 mOsm men kan dog variere inden for visse 15 grænser.
Det kan endvidere være hensigtsmæssigt at foretage fremstillingen af opløsningerne ifølge opfindelsen under en inert gas og til opløsningen sætte en antioxidant såsom natriumascorbat, natriumhydrogensulfit eller natriumpyrosulfit.

En foretrukken blandingsmicelleopløsning ifølge opfindelsen indeholder natriumgly-cocholat, en naturlig lecitin og carprofen eller et salt deraf, især arginin- eller lysin-saltet.
25 En sådan opløsning egner sig især til anvendelse inden for veterinærmedicinen, fx til behandling af akut og kronisk laminitis; skeletsygdomme såsom navicular-syg-dom; myositis; smerter ved kofiker, især flatus og spastisk kolik; smerter ved sygdomme i åndedrætssystemet, akut mastitis, traumatiske smerter og til behandling af sygdomme i puerperalfasen såsom mastitis, manglende eller utilstrækkelig 30 uterusinvolution (postpartial involution).
Opfindelsen illustreres nærmere ved nedenstående eksempler.

Derefter gås der frem som i eksempel lb) og c), idet der dog anvendes 17,77 g lecitin. 2,5 g ibuprofen og 2,11 g L-arginin opløses i 20 ml methanol, hvorpå methanolen fjernes på rotationsfordamper. Det vundne I pulver sættes til blandingsmicelleopløsningen og opløses under omrøring. Derefter l justeres der til pH 7,1+0,1 med 2N HCI, fyldes op til 100 ml med N2-beluftet vand 15 til injektionsformål og gås frem som i Eksempel 1 g).
Eksempel 5 20 9,86 g glycocholsyre (med 5,6% H20) og 3,48 g L-arginin opløses i 50 ml N2-beluf-tet vand til injektionsformål. 2,5 g naproxen og 1,89 g L-arginin sættes til den vundne opløsning og opløses under omrøring. 17,77 g fintsnittet lecitin tilsættes og opløses under omrøring ved ca. 40-50°C. Derefter justeres pH som i Eksempel 3, og opløsningen fyldes op til 100 ml, filtreres og fyldes i ampuller.

Claims (10)
1. Vandige blandingsmicelleopløsninger, 5 kendetegnet ved, at de indeholder et salt af en galdesyre, et lipid og en ikke-steroid inflammationshæmmer.
2. Blandingsmicelleopløsning ifølge krav 1, kendetegnet ved, at galdesyren er cholsyre, glycocholsyre eller 10 taurocholsyre.
3. Blandingsmicelleopløsninger ifølge krav 1 eller 2, kendetegnet ved, at lipidet er en phosphatidylcholin, især en naturlig lecitin.
4. Blandingsmicelleopløsning ifølge et hvilket som helst af kravene 1-3, kendetegnet ved, at den ikke-steroide inflammationshæmmer er et carboxylsyrederivat.
5. Blandingsmicelleopløsning ifølge krav 4, 20 kendetegnet ved, at den ikke-steroide inflammationshæmmer er et eddikesyre- eller propionsyrederivat.
6. Blandingmicelleopløsning ifølge krav 4, kendetegnet ved, at den ikke-steroide inflammationshæmmer er carprofen, 25 ibuprofen, benoxaprofen, naproxen, sulindac, zomepirac, fenclofenac, alclofenac, ibufenac, flunixin, indomethacin eller et salt deraf.
