Claims (26)
1. Genmodificeret plantecelle, der er kendetegnet ved, at den fremviser forøget aktivitet af et P-glucan-vand-dikinaseprotein sammenlignet med tilsvarende vildtypeplanteceller, som ikke er blevet genmodificeret, hvor genmodificeringen består af indførelse af mindst ét fremmed nukleinsyremolekyle i plantens genom, og hvor det fremmede nukleinsyremolekyle er udvalgt fra gruppen, der består af: a) nukleinsyremolekyler, der koder for et protein med aminosyresekvensen ifølge SEQ ID NO: 2 eller SEQ ID NO: 4; b) nukleinsyremolekyler, der koder for et protein, der indbefatter aminosyresekvensen, der kodes af insertionen i plasmid DSM16264 eller insertionen i plasmid DSM16302; c) nukleinsyremolekyler, der koder for et protein, hvis sekvens har en identitet på mindst 60 % med aminosyresekvensen ifølge SEQ ID NO: 2 eller SEQ ID NO: 4; d) nukleinsyremolekyler, der koder for et protein, hvis sekvens har en identitet på mindst 60 % med aminosyresekvensen, der kodes af det kodende område af insertionen i plasmid DSM16264 eller af det kodende område af insertionen i plasmid DSM16302; e) nukleinsyremolekyler, der indbefatter nukleotidsekvensen ifølge SEQ ID NO: 1 eller SEQ ID NO: 3 eller en komplementær sekvens; f) nukleinsyremolekyler, der indbefatter nukleotidsekvensen af insertionen, der er indeholdt i plasmid DSM16264 eller plasmid DSM16302; g) nukleinsyremolekyler, der har en identitet på mindst 60 % med nukleinsyresekvenserne beskrevet under a) , b), e) eller f) ; h) nukleinsyremolekyler, der under stringente betingelser hybridiserer med mindst den ene streng af nukleinsyremolekylerne beskrevet under a) , b), d) , e) eller f) ; i) nukleinsyremolekyler, hvis nukleotidsekvens afviger fra sekvensen af nukleinsyremolekylerne identificeret under a) , b), e) eller f) som følge af den degenererede kode; og j) nukleinsyremolekyler, som er fragmenter, allelvarianter og/eller derivater af nukleinsyremolekylerne identificeret under a), b), c), d), e), f), g), h) eller i).
2. Plantecelle ifølge krav 1, der syntetiserer en modificeret stivelse sammenlignet med de tilsvarende vildtypeplanteceller, som ikke er blevet genmodificeret.
3. Genmodificeret plantecelle ifølge krav 2, hvor den modificerede stivelse er kendetegnet ved, at den har et forøget stivelsesphosphatindhold og/eller en modificeret phosphatfordeling.
4. Genmodificeret plantecelle ifølge krav 3, hvor den modificerede stivelse er kendetegnet ved, at den har et modificeret forhold mellem C-3-phosphat og C-6-phosphat.
5. Plante, der indeholder genmodificerede planteceller ifølge et af kravene 1 til 4.
6. Plante ifølge krav 5, som er en stivelseslagrende plante.
7. Plante ifølge krav 6, som er en majsplante eller en hvedeplante.
8. Formeringsmateriale fra planter ifølge et af kravene 5, 6 eller 7, der indeholder planteceller ifølge et af kravene 1 til 4.
9. Høstbare plantedele af planter ifølge et af kravene 5, 6 eller 7, der indeholder planteceller ifølge et af kravene 1 til 4.
10. Fremgangsmåde til fremstilling af en genmodificeret plante, hvor a) et fremmed nukleinsyremolekyle indføres i plantens genom, hvor det fremmede nukleinsyremolekyle koder for et P-glucan-vand-dikinaseprotein, hvilket betyder et protein, der kræver allerede phosphoryleret stivelse som substrat, og hvor det fremmede nukleinsyremolekyle er udvalgt fra gruppen, der består af: i) nukleinsyremolekyler, der koder for et protein med aminosyresekvensen ifølge SEQ ID NO: 2 eller SEQ ID NO: 4; ii) nukleinsyremolekyler, der koder for et protein, der indbefatter aminosyresekvensen, der kodes af insertionen i plasmid DSM16264 eller insertionen i plasmid DSM16302; iii) nukleinsyremolekyler, der koder for et protein, hvis sekvens har en identitet på mindst 60 % med aminosyresekvensen ifølge SEQ ID NO: 2 eller SEQ ID NO: 4; iv) nukleinsyremolekyler, der koder for et protein, hvis sekvens har en identitet på mindst 60 % med aminosyresekvensen, der kodes af det kodende område af insertionen i plasmid DSM16264 eller af det kodende område af insertionen i plasmid DSM16302; v) nukleinsyremolekyler, der indbefatter nukleotidsekvensen ifølge SEQ ID NO: 1 eller SEQ ID NO: 3 eller en komplementær sekvens; vi) nukleinsyremolekyler, der indbefatter nukleotidsekvensen af insertionen, der er indeholdt i plasmid DSM16264 eller plasmid DSM16302; vii) nukleinsyremolekyler, der har en identitet på mindst 60 % med nukleinsyresekvenserne beskrevet under i), il), v) eller vi) ; viii) nukleinsyremolekyler, der under stringente betingelser hybridiserer med mindst den ene streng af nukleinsyremolekylerne beskrevet under i) , ii) , iv) , v) eller vi) ; ix) nukleinsyremolekyler, hvis nukleotidsekvens afviger fra sekvensen af nukleinsyremolekylerne identificeret under i), ii), v) eller vi) som følge af den degenererede kode; og x) nukleinsyremolekyler, som er fragmenter, allelvarianter og/eller derivater af nukleinsyremolekylerne identificeret under i), ii), iii), iv), v), vi), vii), viii) eller ix). b) en plante frembringes fra planteceller fra trin a).
11. Fremgangsmåde til fremstilling af en modificeret stivelse, der indbefatter trinet ekstraktion af stivelsen fra en plantecelle ifølge et af kravene 1 til 4.
12. Fremgangsmåde til fremstilling af en modificeret stivelse, der indbefatter trinet ekstraktion af stivelsen fra en plantecelle ifølge et af kravene 5, 6 eller 7.
13. Fremgangsmåde til fremstilling af en modificeret stivelse, der indbefatter trinet ekstraktion af stivelsen fra høstbare plantedele ifølge krav 9.
14. Anvendelse af genmodificerede planter ifølge et af kravene 5, 6 eller 7 til fremstilling af en modificeret stivelse.
15. Fremgangsmåde til fremstilling af mel, der indbefatter trinet formaling af plantedele ifølge kravene 5, 6 eller 7 eller formeringsmateriale ifølge krav 8 eller høstbart materiale ifølge krav 9.
16. Anvendelse af genmodificerede planteceller ifølge et af kravene 1 til 4 eller af planter ifølge et af kravene 5, 6 eller 7 til fremstilling af mel.
17. Nukleinsyremolekyle, der koder for et protein med den enzymatiske aktivitet af et P-glucan-vand-dikinaseprotein, der er udvalgt fra gruppen, der består af: a) nukleinsyremolekyler, der koder for et protein med aminosyresekvensen ifølge SEQ ID NO: 2 eller SEQ ID NO: 4; b) nukleinsyremolekyler, der koder for et protein med en aminosyresekvens, der har en identitet på mindst 60 % med aminosyresekvensen ifølge SEQ ID NO: 2 eller SEQ ID NO: 4; c) nukleinsyremolekyler, der indeholder nukleotidsekvensen ifølge SEQ ID NO: 1 eller SEQ ID NO: 3 eller indeholder en sekvens, der er komplementær til disse sekvenser; d) nukleinsyremolekyler, der har en identitet på mindst 60 % med nukleinsyresekvenserne beskrevet under a) eller c); e) nukleinsyremolekyler, der under stringente betingelser hybridiserer med mindst den ene streng af nukleinsyremolekylerne beskrevet under a) eller c) ; f) nukleinsyremolekyler, der har en nukleotidsekvensen, der afviger fra sekvensen af nukleinsyremolekylerne nævnt under a) eller c) som følge af den degenererede kode; og g) nukleinsyremolekyler, som er fragmenter, allelvarianter og/eller derivater af nukleinsyremolekylerne anført under a) , b) , c) , d) , e) eller f) .
18. Rekombinant nukleinsyremolekyle, der indeholder et nukleinsyremolekyle ifølge krav 17.
19. Vektor, der indeholder et nukleinsyremolekyle ifølge krav 18 .
20. Vektor ifølge krav 19, hvor nukleinsyremolekylet er koblet med regulatorsekvenser, der initierer transkription i prokaryote eller eukaryote celler.
21. Værtscelle, der er genmodificeret med et nukleinsyremolekyle ifølge krav 17, med et rekombinant nukleinsyremolekyle ifølge krav 18 eller med en vektor ifølge kravene 19 eller 20.
22. Sammensætning, der indeholder nukleinsyremolekyler ifølge krav 17, et rekombinant nukleinsyremolekyle ifølge krav 18 eller en vektor ifølge kravene 19 eller 20.
23. Anvendelse af en sammensætning ifølge krav 22 til identificering af planteceller, der har forøget aktivitet af et P-glucan-vand-dikinaseprotein sammenlignet med vildtypeplanteceller, der ikke er blevet genmodificeret.
24. Protein, der fremviser stivelsesphosphoryleringsaktivitet og kræver phosphoryleret stivelse som substrat, hvilket protein fremviser et phosphohistidindomæne, der har en identitet på mindst 70 % med aminosyresekvensen ifølge SEQ ID NO: 5.
25. Protein ifølge krav 24, der behøver phosphoryleret stivelse som substrat og overfører det resterende phosphat af ATP til phosphoryleret stivelse.
26. Fremgangsmåde ifølge krav 10, der omfatter et ekstra trin c) , hvor yderligere planter frembringes ved hjælp af planterne ifølge trin b) , hvor trin c) udføres ved hjælp af vegetativ formering.
