Opfindelsen angår en analogifremgangsmåde til fremstilling af hidtil ukendte carbacyklinderivater.
I de tyske offentliggørelsesskrifter DE-OS 28 45 770, 29 00 352, 29 02 442, 29 04 655, 29 09 088 og 29 12 409 beskrives (5E) - og 5 (5Z) - 6a-carbaprostaglandin-12 - analoge. Nomenklaturen for forbin delserne fremstillet ifølge opfindelsen er baseret på et forslag af Morton og Brokow (J. Org. Chem. 44, 2880 [1979]) . Ved syntesen af disse forbindelser dannes der altid to dobbeltbindingsisomere, som bliver karakteriseret ved tilføjelsen (5E) eller (5Z). De 10 to isomere af denne prototype tydeliggøres ved hjælp af følgende strukturformel:

De fra DE offentliggørelsesskrift nr. 28 45 770 kendte 6a-carbaprostaglandin-I2-derivater virker såvel p.o., som i.v. stærkt blodtrykssænkende. De ved fremgangsmåden ifølge opfindel-15 sen fremstillede forbindelser har overlegen blodtrykssænkende virkning i forhold til disse kendte 6a-carbaprostaglandin-I2-derivater.
Fra den meget omfangsrige kendte teknik vedrørende prostacykliner og deres analoge ved man, at denne stofklasse på grund af deres 20 biologiske og farmakologiske egenskaber er egnet til behandlingen af pattedyr, specielt mennesker. Deres anvendelse som lægemiddel støder dog hyppigt på vanskeligheder, da de besidder en til terapeutiske formål forkort virkningsvarighed. Alle strukturændringer har det formål at forøge virkningsvarigheden samt selektivi- 2 DK 169815 B1 teten af virkningen.
Det har nu vist sig, at der ved substitution af methylengruppen i carbacyklinets 3-stilling med oxygen kan opnås en længere virknings varighed, større selektivitet og bedre virkning. For-5 bindeiserne fremstillet ved fremgangsmåden ifølge opfindelsen virker bronchodilatorisk og egner sig til inhiberingen af thrombocytaggregat ionen, til bl odtryks sænkning via en vasodilation og til hæmning af mavesyresekretionen.

Å hvori R-j^ betegner resten OR2, hvori R2 betyder hydrogen eller alkyl med 1-4 C-atomer, D betegner en ligekædet eller forgrenet, mættet alkylengruppe med 1-5 C-atomer, 15 R4 betegner en alkylgruppe med 1-2 C-atomer, 3 DK 169815 B1 eller, såfremt R2 har betydningen et hydrogenatom, deres salte med fysiologisk acceptable baser.
Forbindelserne af formlen I findes som såvel (5E) - som (5Z)-isomere.
5 Som alkylgruppe R2 egner sig ligekædede eller forgrenede alkyl-grupper med 1-4 carbonatomer, som f.eks. methyl, ethyl, propyl, isopropyl, butyl, isobutyl og tert.-butyl.
Som foretrukne alkylgrupper R2 skal nævnes methyl, ethyl, propyl, isobutyl og butyl.
10 Alkylgruppen R4 er methyl eller ethyl.
Som alkylengruppe D kommer ligekædede eller forgrenede, mættede alkylengrupper med 1-5 carbonatomer på tale. Eksempelvis skal nævnes: methylen, ethylen, 1,2-propylen, ethylethylen, trimethy-len , tetramethylen, pentarnethylen, 1-methyltetramethylen, 1-15 methyltrimethylen.
Til saltdannelsen med de frie syrer (R2=H) egner sig uorganiske og organiske baser, således som fagfolk kender det til dannelsen af fysiologisk acceptable salte. Eksempelvis skal nævnes: alkalihydroxider, såsom natrium- og kaliumhydroxid, jordalkalihydroxi-20 der, såsom calciumhydroxid, ammoniak og aminer, såsom ethanol-amin, diethanolamin, triethanolamin, N-methylglucamin, morpholin, tris-(hydroxymethyl)-methylamin, osv.
Fremgangsmåden ifølge opfindelsen er ejendommelig ved det i kravets kendetegnende del angivne.
25 Omsætningen af forbindelserne med den almene formel II med et halogeneddikesyrederivat med den almene formel III foretages ved temperaturer fra OoC til XOOoC, fortrinsvis 10oC til 80°C, i et aprotisk opløsningsmiddel eller opløsningsmiddelblanding, f.eks. dimethylsulfoxid, dimethylformamid, tetrahydrofuran, osv. Som · 4 DK 169815 B1 baser kommer de for fagfolk til forethring kendte baser på tale, f.eks. natriumhydrid, kalium-tert.-butylat, butyllithium, osv.
Forsæbningen af prostaglandinesteren gennemføres ved hjælp af de for fagfolk kendte metoder, f. eks. med basiske katalysatorer.
5 Indføringen af esterguppen -0R2 for R-j_, hvor R2 betegner en alkyl gruppe med 1-4 carbonatomer, sker ved hjælp af de for fagfolk kendte metoder. Carboxyforbindelserne omsættes f.eks. meddiazo-hydrocarboner på i og for sig kendt måde. Forestringen med diazo-hydrocarboner sker f.eks. ved, at man blander en opløsning af 10 diazohydrocarbonet i et inaktivt opløsningsmiddel, fortrinsvis i diethylether med carboxyforbindelsen i det samme eller i et andet inaktivt opløsningsmidel, såsom f.eks. methylenchlorid. Efter endt omsætning i 1-30 min. fjernes opløsningsmidlet, og esteren renses på sædvanlig måde. Diazoalkaner er enten kendte 15 forbindelser eller kan fremstilles ved hjælp af kendte metoder [Org. Reactions, bind 8, side 389-394 (1954)].
Prostaglandinderivaterne af den almene formel I, hvor R-|_ betegner en hydroxygruppe, kan med ringe mængder af de tilsvarende uorganiske baser under neutralisering overføres i salte. F.eks. får 20 man ved opløsning af de tilsvarende PG-syrer i vand, som indeholder den støkiometriske mængde base, efter afdampning af vand eller efter tilsætning af et med vand blandbart opløsningsmiddel, f.eks. alkohol eller acetone, det faste uorganiske salt.
Fremstillingen af aminsaltene sker på sædvanlig måde. Dertil 25 sættes PG-syren, f.eks. opløst i et egnet opløsningsmiddel, såsom ethanol, acetone, diethylether eller benzen, og mindst den støkiometriske mængde amin, til denne opløsning. Derved udfældes saltet sædvanligvis i fast form eller isoleres efter fordampning af opløsningsmidlet på sædvanlig måde.
30 Den funktionelle afledning af de frie OH-grupper sker ved hjælp af de for fagfolk kendte metoder. Til indføringen af etherbeskyt-telsesgruppeme omsættes f. eks. med dihydropyran i methylenchlo- 5 DK 169815 B1 rid eller chloroform under anvendelse af et surt kondensations-middel, såsom f .eks. p-toluensulfonsyre. Dihydropyranet anvendes i overskud, fortrinsvis i en mængde, der er 4-10 gange så stor som den teoretisk nødvendige mængde. Omsætningen er normalt f or-5 løbet til ende ved 0-30°C efter 15-30 min. forløb.
Indføringen af acylbeskyttelsesgrupperne sker ved, at man omsætter en forbindelse af den almene formel I på i og for sig kendt måde med et carboxyl syrederivat, såsom f. eks. syrechlorid, syreanhydrid og andre.
10 Frigørelsen af en funktionelt afledt OH-gruppe til forbindelserne af den almene formel I sker ved hjælp af kendte metoder. F.eks. gennemføres af spaltningen af etherbeskyttelsesgrupper i en vandig opløsning af en organisk syre, såsom f.eks. eddikesyre, propion-syre og andre, eller i en vandig opløsning af en uorganisk syre, 15 såsom f.eks. saltsyre. Til forbedring af opløseligheden tilsættes hensigtsmæssigt et med vand blandbart inaktivt organisk opløsningsmiddel. Egnede organiske opløsningsmidler er f.eks. alkoholer, såsom methanol og ethanol, og ethere, såsom dimethoxyethan, dioxan og tetrahydrofuran. Der anvendes fortrinsvis tetrahydrofu-20 ran. Af spaltningen gennemføres fortrinsvis ved temperaturer mellem 20°c og 80oC.
Af spaltningen af silyletherbeskyttelsesgryppeme sker f.eks. med tetrabutylammoniumchlorid. Som opløsningsmiddel egner sig f.eks. tetrahydrofuran, diethylether, dioxan, methylenchlorid, osv. Af-25 spaltningen gennemføres fortrinsvis ved temperaturer mellem 0°C og 80°C.
Forsæbningen af acylgruppeme sker f.eks. med alkali- eller jordalkalicarbonater eller -hydroxider i en alkohol eller den vandige opløsning af en alkohol. Som alkoholer kommer alifatiske 30 alkoholer i betragtning, såsom f.eks. methanol, ethanol, butanol, osv., fortrinsvis methanol. Som alkalicarbonater og/eller -hydroxider skal nævnes kalium- og natriumsalte. Kaliumsaltene foretrækkes dog. Som jordalkalicarbonater og -hydoxider egner sig DK 169815 B1 ξ f.eks. calciumcarbonat, calciumhydroxid og bariumcarbonat. Omsætningen sker ved -10oC til 70oC, fortrinsvis ved 25oC.
Såfremt udgangsproduktet indeholder OH-grupper i prostanresten, bringes disse OH-grupper også til reaktion. Man går derfor hen-5 sigtsmæssigt ud fra udgangsprodukter, hvori disse er beskyttede midlertidigt ved hjælp af fortrinsvis let afspaltelige ether-eller acylrester.

Forbindelserne fremstillet ifølge denne opfindelse virker bl odtryks sænkende og bronchodilatorisk. De egner sig desuden til 15 at hæmme thrombocytaggregationen. Følgelig udgør de hidtil ukendte carbacyklinderivater af formlen I værdifulde farmaceutisk virksomme stoffer. Desuden udviser de ved lignende virknings spektrum i sammenligning med de tilsvarende prostaglandiner en højere specificitet og frem for alt en væsentlig længere virkningstid.

Den høje vævsspecificitet af de nye carbacykliner viser sig ved undersøgelsen på glatmuskulære organer, som f.eks. på mar-svineileum eller på isoleret kanintrachea, hvor der konstateres en væsentlig ringere stimulering end ved applikationen af na-25 turlige prostaglandiner af E-, A- eller F-typen.
De hidtil ukendte carbacyklinanaloge besidder for prostacykliner typiske egenskaber, såsom f.eks. sænkning af den perifere arterielle og koronare vaskulære modstand, inhibering af throm- 9 DK 169815 B1 bocyt aggregat ionen og opløsningen af blodpladethromber, myocardial cytoprotektion og dermed sænkning af det systemiske blodtryk uden samtidigt at sænke slagvolumen og koronar gennemblødning; behandling af slagtilfælde, profylakse og terapi af koronare 5 hjertesygdomme, koronar thrombose, hjerteinfarkt, perifere arteriesygdomme, arteriesclerose og thrombose, chokterapi, inhibering af bronchokonstriktion, inhibering af mavesyresekretion og cytoprotektion af mave- og tarmslimhinden og i leveren og pankreas, antiallergiske egenskaber, sænkning af den pulmonære vaskulære 10 modstand og det pulmonære blodtryk, fremme af nyregennemblødning, behandling af svangerskabstoksikose, forhøjelse af den cerebrale gennemblødning, etc. Desuden besidder de hidtil ukendte carba-cyklinanaloge antiproliferative egenskaber. De hidtil ukendte carbacykliner kan endvidere finde anvendelse i kombination med 15 f.eks. SS-blokeringsmidler eller diuretika.
Dosen af forbindelserne er 1-1500 /iig/kg/dag, når de gives til humane patienter. Enhedsdosen for den farmaceutisk acceptable bærer udgør 0,01-100 mg.
Ved intravenøs injektion i voksne hypertone rotter i doser på 20 5, 20 og 100 /Jg/kg legemsvægt udviser forbindelserne fremstillet ifølge opfindelsen en kraftigere blodtrykssænkende og i længere tid holdbar virkning end PGE2 og PGA2 uden som PGE2 at fremkalde diarre eller som PGA2 kardiale arrytmier.
Ved intravenøs injektion på narkotiserede kaniner udviser 25 forbindelserne fremstillet ifølge opfindelsen i sammenligning med PGE2 og PGA2 en kraftigere og væsentlig længere varende blodtrykssænkning, uden at andre glatmuskulære organer eller organfunktioner bliver påvirket.
Til den parenterale indgivelse anvendes sterile injicerbare, 30 vandige eller olieagtige opløsninger. Til den orale applikation egner sig f.eks. tabletter, drageer eller kapsler.

De virksomme stoffer, der fremstilles ifølge opfindelsen, skal i forbindelse med de i den galeniske farmaci kendte og sædvanlige hjælpestoffer tjene f .eks. til fremstilling af blodtrykssænkende midler. Enhedsdosisområdet for en ampul er 0,1-0,5 mg og for en 5 tablet 0,1-1 mg.
Eksempel 1 (5E) - (16RS) -16-methyl-3-oxa-18,18,19,19-tetradehydro-6a-carbapro-staglandin-I2.
Til en opløsning af 530 mg 2-{ (E) - (1S,5S, 6R, 7R) -7- (tetrahydropy-10 ran-2-yloxy) -6- [ (E) - (3S,4RS) -4-methyl-3- (tetrahydropyran-2-yl-oxy) -oct-l-en-6-yn-yl] -bicylo [3.3.0] octan-3-yliden}-ethan-l-ol i 3 ml tetrahydrofuran sættes ved 10°c 0,77 ml af en 1,52 molær butyllithiumopløsning i hexan, omrøres i 5 min., behandles med 3 ml dimethylformamid og 4 ml dimethylsulfoxid og tilsættes derpå 15 225 mg chloreddikesyre-lithiumsalt. Der omrøres i 24 timer ved stuetemperatur, hældes i isvand, symes med 10%' ig citronsyreopløsning, ekstraheres med ether, vaskes den organiske ekstrakt én gang med saltvand, tørres over natriumsulfat og inddampes i vakuum. Efter kromatografi af resten på kiselgel fås med eddike-20 ester/isopropanol (8+2) 290 mg (5E)-(16RS)-l6-methyl-3-oxa-18,18,19,19-tetradehydro-6a-carbaprostaglandin-I2-ll, 15-bis- (te-trahydropyranylether). Til afspaltning af beskyttelsesgrupperne omrøres 290 mg af bis-tetrahydropyranyletheren med 28 ml af en blanding af eddikesyre/vand/tetrahydrofuran (65+35+10) i 16 timer 25 ved stuetemperatur og inddampes derpå i vakuum. Resten kromato-graferes med eddikeester/eddikesyre (99+1) på kiselgel. Derved fås 105 mg af titelforbindelsen som en farveløs olie. IR: 3350 (bred), 2920, 1720, 1600, 1420, 965 cm"1.
Udgangsmaterialet for den ovennævnte titelforbindelse fremstilles 30 på følgende måde: 11 DK 169815 B1 la 2-{ (E) - (1S,5S,6R,7R) -7- (tetrahydropyran-2-yioxy) -6- [ (E) - (3S,4RS) - 4-methyl-3- (tetrahydropyran-2-yloxy) -oct-l-en-6-ynyl] -bicyk-lo[3.3.0] octan-3-yliden}-ethan-l-ol.
5 Til en opløsning af 4,0 g phosphoneddikesyretriethylester i 80 ml tetrahydrofuran sættes ved 0°C 1,73 g kalium-tert.-butylat, omrøres i 10 min., behandles med en opløsning af 4,45 g (1R,5S, 6R,7R) - 7- (tetrahydropyran-2-yloxy) -6- [ (E) - (3S,4RS) -4-methyl-3-(tetrahydropyran-2-yloxy) -oct-l-en-6-ynyl-bicyklo [3.3.0] octan-3-10 on i 45 ml toluen og omrøres i 20 timer ved stuetemperatur. Der fortyndes med 600 ml ether, rystes én gang med vand, én gang med 20%' ig natriumhydroxid, vaskes neutral med vand, tørres over magnesiumsulfat og inddampes i vakuum. Resten filtreres med hexan/ ether (3+2) over kiselgel. Derved fås 3,7 g af den umættede ester 15 som en farveløs olie.
IR: 2945, 2870, 1700, 1655, 970/cm.
1,2 g lithiumaluminiumhydrid sættes portionsvis ved 0oC til en omrørt opløsning af 3,9 g af den ovenfor fremstillede ester i 130 ml ether, og der omrøres i 30 min. ved 0°C. Reagensoverskud-20 det dekomponeres ved dråbevis tilsætning af eddikeester, hvorpå der tilsættes 6 ml vand, omrøres i 2 timer ved 20°C, filtreres og inddampes i vakuum. Resten kromatograferes med ether/hexan (3+2) på kiselgel. Derved fås som mere upolær forbindelse 1,05 g 2-{(Z)-(IS,5S,6R,7R)-7-(tetrahydropyran-2-yloxy)-6-[(E)-25 (3S,4RS) -4-methyl-3- (tetrahydropyran-2-yloxy) -oct-l-en-6-yn-yl] - bicyklo[3.3.0]octan-3-yliden}-ethan-l-ol og 2,2 g af titelforbindelsen som en farveløs olie.

Eksempel 2 (5Z) - (16RS) -16-methyl-3-oxa-18,18,19,19-tetradehydro-6a-carbapro-staglandin-I2.
Analogt som beskrevet i eksempel 1 fås af 490 mg af den ifølge 5 eksempel la fremstillede Z-konfigurerede allylalkohol 85 mg af titelforbindelsen somen farveløs olie. IR (CHC13) : 3340 (bred) , 2920, 1722, 1600, 1420, 966/cm.
Eksempel 3 (5E)-(16RS)-16,20-dimethyl-3-oxa-18,18,19,19-tetradehydro-6a-10 carbaprostaglandin-I2.
Analogt som beskrevet i eksempel 1 fås af 0,5 g 2-{ (E) - (1S,5S, -6R, 7R) -7- (tetrahydropyran-2-yloxy) -6- [ (E) - (3S,4RS) -4-methyl-3-(tetraiiydropyran- 2 -yloxy) -non-1 - en- 6 -yn-yl ] -bicyklo [3.3.0] octan- 3-yliden}-ethan-l-ol og 220 mg chl or eddikesyr e -1 i thiumsal t 260 15 mg(5E)-(16RS)-16,20-dimethyl-3-oxa-18,18,19,19-tetradehydro-6a-carbaprostaglandin-I2-ll, 15-bis- (tetrahydropyranylether) . Efter afspaltning af beskyttelsesgrupperne fås 90 mg af titelforbindelsen som en farveløs olie.

20 Udgangsmateriale for den ovennævnte titelforbindelse fremstilles på følgende måde: 3a (1R,5S, 6R, 7R) -3,3-ethylendioxy-7-benzoyloxy-6- [ (E) - (3S,4RS) -3-hydroxy-4-methyl-non-l-en-6-ynyl] -bicyklo [3.3.0] octan.
25 Til en suspension af 1,46 g natriumhydrid (55%,ig suspension i olie) i 130 ml dimethoxyethan (DME) dryppes ved 0oC en opløsning . af 9,02 g 3-methyl-2-oxo-oct-5-ynyl-phosphonsyredimethylester i 67 ml DME og omrøres i 1 time ved 0<>C. Derpå behandler man ved 13 DK 169815 B1 -20oc med en opløsning af 9,4 g (lR,5S,6R,7R)-3,3-ethylendioxy-7-benzoyloxy-6-formylbicyklo[3.3.0]-octan i 130 ml DME, omrøres i 1,5 time ved -20oC, hældes på 600 ml mættet ammoniumchl or idopløsning og ekstraheres tre gange med ether. Den organiske 5 ekstrakt vaskes neutral med vand, tørres over magnesiumsulfat og inddampes i vakuum. Efter kromatografi af resten på kiselgel fås med ether/hexan (1+1) 9,1 g af den a, /?-umættede keton som en olie.
Til en opløsning af 9,1 g af ketonen i 300 ml methanol sættes 10 ved -40©c portionsvis 5,2 g natriumborhydrid og omrøres i 1 time ved -40°C. Derpå fortyndes med ether, vaskes neutral med vand, tørres over magnesiumsulfat og inddampes i vakuum. Ved søjlekromatografi på kiselgel med ether/hexan (7+3) fås først 3,9 g af titelforbindelsen (PG-nomenklatur: lSa-hydroxy) samt som mere 15 polær komponent 3,2 g af den isomere af 15o;-hydroxyforbindelse.
IR: 3600, 3400 (bred), 2942, 1711, 1603, 1588, 1276, 968, 947/cm.
3b (IR, 5S, 6R, 7R) -7- (tetrahydropyran-2-yloxy) -6- [ (E) - (3S,4RS) -3-tetrahydropyran-2-yloxy)-4-methyl-non-l-en-6-ynyl]-bicyk- 20 lo[3.3.0]octan-3-on.
En blanding af 3,6 g af den ifølge eksempel 3a fremstillede a-alkohol og 1,4 g kaliumcarbonat i 120 ml methanol omrøres i 16 timer ved stuetemperatur under argon. Derpå inddampes i vakuum, fortyndes med ether og vaskes neutralt med saltvand. Der tørres 25 over magnesiumsulfat og inddampes i vakuum. Inddampningsresten omrøres i 16 timer ved stuetemperatur ved 75 ml af en blanding af eddikesyre/vand/tetrahydrofuran (65+35+10) og inddampes derpå i vakuum. Efter filtrering af resten over kiselgel fås med eddikeester/hexan (7+3) 2,2 g af ketonen som en olie.
30 En opløsning af 2,2 g af ketonen, 2,4 ml dihydropyran og 23 mg p-toluensulfonsyre i 75 ml methylenchlorid omrøres i 30 min. ved 0°C. Derpå fortyndes med ether, rystes med fortyndet natrium- 14 DK 169815 B1 bicarbonatopløsning, vaskes neutralt med vand, tørrres over magnesiumsulfat og inddampes i vakuum. Der fås 3,4 g af bis-tetra-hydropyranyletheren, der anvendes uden yderligere rensning.
IR: 2960, 2865, 1738, 970/cm.

Udgangsmaterialet for den ovennævnte forbindelse fremstilles på følgende måde: 10 5a 3-methyl-2-oxo-oct-6-ynylphosphonsyredimethylester.
Til en opløsning af 40 g 3-pentin-l-ol i 240 ml pyridin sættes 109 g p-toluensulfonsyrechlorid og omrøres i 48 timer ved 0oc. Derpå tilsættes 30 ml vand, omrøres i 2 timer og fortyndes med 15 ether. Der rystes efter hinaden med 5%'ig svovlsyre, med vand, med 5%'ig natriumbicarbonatopløsning, og vaskes neutralt med vand, tørres over magnesiumsulfat og inddampes i vakuum. Resten omkrystalliseres fra hexan/ether. Derved fås 80 g af tosylatet (smeltepunkt 43°C) .
20 Til en suspension af 8,7 g natriumhydrid (55%'ig suspension i olie) sættes 34 g methylmalonester og koges i 4 timer under tilbagesvaling. Derpå tilsættes ved stuetemperatur 35 g 1-to-syloxy-3-pentyn og 130 ml DME, og der opvarmes i 8 timer under tilbagesvaling. Der neutraliseres med eddikesyre, behandles med 25 130 ml vand, ekstraheres med ether, udrystes tre gange med vand, tørres over magnesiumsulfat og inddampes i vakuum. Resten renses ved vakuumdestillation ved 12 mm Hg. Ved 150-156°C fås 24 g af den alkylerede methylmalonsyreester, som man opvarmer i 160 ml dime thyl sul f oxid og 1,5 ml vand i 6 timer med 7,5 g lithiumchlo-30 ridunder tilbagesvaling. Derpå hældes i 400 ml isvand, ekstraheres med ether, udrystes den organiske ekstrakt to gange med vand, tørres over magnesiumsulfat og inddampes i vakuum. Destillationen af resten giver ved 92©C og 12 mm Hg 14 g 2-methyl-hept-5-ynsy- 16 DK 169815 B1 reethylester, som en farveløs væske.
Til en opløsning af 18,6 g methanphosphonsyredimethylester i 280 ml tetrahydrofuran dryppes 73 ml af en 1, 7 molær butyllithiumop-løsning i hexan ved -70°C, omrøres i 15 min. og tilsættes lang-5 somt en opløsning af 10,5 g 2-methyl-hept-5-ynsyreethylester i 48 ml tetrahydrofuran. Der omrøres i 4 timer ved -70oC, neutraliseres med eddikesyre og inddampes i vakuum. Destillationen af resten i vakuum ved 0,6 mm Hg og 130°C giver 10,8 g af titelforbindelsen som en farveløs væske.
10 5b (IR,5S, 6R, 7R) -3,3-ethylendioxy-7-benzoyloxy-6- [ (E) - (3S,4RS) -3-hydroxy-4-methyl-non-l-en-7-ynyl] -bicyklo [3.3.0] octan.

Til denne opløsning drypper man derefter ved 0oC en opløsning af 29 g l-brom-2-pentyn i 44 ml absolut tetrahydrofuran, omrører 3 timer ved 0oC, neutralise-25 rer med 3N saltsyre og inddamper i vakuum. Man tilsætter 50 ml saltopløsning, ekstraherer tre gange med hver gang 200 ml methy-lenchlorid, udryster den organiske ekstrakt to gange med hver gang 50 ml saltopløsning, tørrer med magnesiumsulfat og inddamper i vakuum. Efter destillation af remanensen ved 0,6 Torr og 125oc 30 opnås 23 g af titelforbindelsen som farveløs væske.

Derefter inddampes i 23 DK 169815 B1 vakuum, fortyndes med ether og vaskes neutral med saltopløsning. Inddampningsresten omrøres i 16 timer ved stuetemperatur med 40 ml af en blanding af eddikesyre/vand/tetrahydrofuran (65+35+10) og inddampes derefter i vakuum. Efter filtrering af remanensen 5 over kiselgel opnås med eddikesyreester/hexan (7+3) 1,15 g af ketonen som olie.
En opløsning af 1,15 g af ketonen, 1,2 g ml dihydropyran og 10 mg p-toluensulfonsyre i 40 ml methylenchlorid omrøres i 30 minutter ved 0oC. Derefter fortyndes med ether, rystes med fortyn-10 det natriumbicarbonatopløsning, vaskes neutral med vand, tørres over magnesiumsulfat og inddampes i vakuum. Der opnås 1,65 g bistetrahydropyranylether, der anvendes uden yderligere rensning.
