Opfindelsen angâr en fremgangsmâde til fremstilling af pyruvatoxidase, og denne fremgangsmâde er ejendommelig ved det i krav 1 anf0rte.
Et pyruvatoxidaseenzym kan indgâ som katalysator ved 5 folgende tre reaktioner: (1) Pyruvat + P + 02—^Ac/vP + C02 + H2 02 (2) Pyruvat + DPN+ + CoA —>Ac.f*/SCoA + DPNH + H+ + CC>2 (3) Pyruvat + l/202"^Ac + CC>2 hvor P = phosphat, Ac^vP = acetylphosphat, DPN = disphospho-10 pyridinnucleotid, CoA = coenzym A og Acr\/SCoA = acetyl- coenzym A.
Der findes med andre ord tre typer pyruvatoxidase, som hver katalyserer en af reaktionerne (1)-(3).
Pyruvatoxidasen ifolge opfindelsen er katalysator for 15 reaktionen (1).
Pyruvatoxidase er et kendt enzym. I "Methods in En-zymology", bind 1, side 482 (1955) og i "Enzymes", 2. ud-gave, side 684 (1964) er omtalt pyruvatoxidase, som katalysator for reaktionen (1Jhidrorende fra Lactobacillus del-20 brückii. Ved fremgangsmâden ifolge opfindelsen er de produ- cerende organismer Pediococcus,Streptococcus og Aerococcus, som aile er forskellige derfra.
I "Chemical Abstracts", bind 43, nr. 7l8i nævnes pyruvatoxidase hidrcrende fra Streptococcus faecalis, men denne 25 pyruvatoxidase er katalysator for reaktionen (2) eller (3), da der ikke dannes H2C>2 ved processen.
I "Chemical Abstracts", bind 57, nr. 17176f omtales pyruvatoxidase hidrorende fra Pediococcus casei, men denne pyruvatoxidase er afhængig af DPN, hvorfor den katalyserede 30 reaktion er reaktionen (2).
I det folgende menes ved "pyruvatoxidase" et enzym, som katalyserer reaktionen (1).

Det har vist sig, at enzymet pyruvatoxidase dannes i en bakteriestamme B-0667 tilh0rende slægten Pediococcus og B-0668 tilh0rende slægten Streptococcus isoleret fra en jordprove taget i en radisemark i Ohito-cho, Tagata-gun, 5 Shizuoka-ken, Japan, og at pyruvatoxidasen fremstillet der- fra kan benyttes til pyrodruesyreanalyse i en prove indehold-ende pyrodruesyre eller forskellige systemer, som frigor pyrodruesyre. Det har ogsà vist sig, at dette enzym kan benyttes til kvantitativ analyse af pyrodruesyre, mâling af en-10 zymaktiviteten af enzymreaktionssystemer, som danner pyruvat, kvantitativ bestemmelse af enzymet og substratet deraf. Pyrodruesyre kan analyseres ved omsætning af en prove indehold-ende pyrodruesyre med et reaktionsSystem omfattende i det mindste pyruvatoxidase, flavinadenindinucleotid (i det folg-15 ende kaldet FAD), thiaminpyrophosphat, oxygen og phosphat -

5 Enzymet pyruvatoxidase katalyserer en oxidativ reaktion fra pyrodruesyre, uorganisk phosphat og oxygen til dannelse af acetylphosphat, carbondioxid og hydrogen-peroxid. Enzymet fremstilles ved dyrkning af de pyruvat-oxidaseproôucerende mikrobestammer 10 Pediococcus sp. B-0667, Streptococcus sp, B-0668, Aero-coccus viridans IPO 12219 eller IPO 12317.

B-0668.

I en udforelsesform for den foreliggende opfindel-se dyrkes ovennævnte Pediocoocus sp, B-0667, Streptococcus 20 sp, B-0668, Aerococcus viridans IFO-12219 eller Aerococcus viridans IPO-12317 i et konventionelt medium til enzympro-duktion. Dyrkning kan foretages ved konventionel væskedyrk-ning, og submers luftningskultur foretrækkes til industriel produktion. Ber kan fortrinsvis benyttes et konventionelt 25 medium til mikroorganismer. Som kulstofkiîder kan benyttes assimilerbare carbonkilder sâsom glucose, saccharose, lactose, maltose, fructose, mêlasse og pyrodruesyre, Assimilerbare nitrogenkilder sâsom pepton, kcdekstrakt, gærekstrakt og caseinhydrolysat kan benyttes, Forskellige uorganiske 30 salte sâsom phosphat, carbonat og sulfat af magnésium, calcium, kalium, jern, mangan eller zink kan benyttes.

Dyrkningstemperaturen kan vælges i omrâdet for dyrkning af mikrobeceller og produktion af pyruvatoxidase, fortrinsvis 25-37°0. Dyrkningstiden kan ændrea afhængende af betingelserne og afsluttes nâr pyruvatoxidaseproduktio-5 nen er praktisk taget fuldstændig, normalt 18-48 timer.
Pyruvatoxidase ekaisterer i cellerne af mikroorga- nismer.
Œil adskillelse af pyruvatoxidase fra de dyrkede celler filtreres eller centrifugeres den dyrkede masse 10 til samling af cellerne, og den oplukkes ved behandling med mekaniske midler eller en enzymatisk procès sâsom et lysozym. EAD ved 0, 40, 50, 60 og 70°C i 10 mi-nutter efter metoden til enzymbestemmelse. Som det ses af fig. 2 bliver enzymerne opnâet fra B-0667, IP0-12219 25 og IPO-12317 en smule aktiveret ved 40°C og denatureret over 60°C. Det enzym, som opnâs fra B-0668, aktiveres ikke ved 40°G og denatureres næsten over 60°C.

En analyse foretages ved inkubering med en prsve og reagensblanding. Reagensblandingen er fortrinsvis et udstyr af de fornsdne reagenser. En indikator for hydrogenperoxid er en kombination af en eller flere chromogen- eller fluoressensindi- katorer, som pâvirkes ved kobling med hydrogenperoxid.
Eksempler pâ sàdanne indikatorer er kombinationer af 25 tetravalent titaniumforbindelse og xylenolorange, som kobles med hydrogenperoxid til dannelse af en stabil red farve, eller kombinationer af phénol eller Ιί,Ιϊ'-dimethyl-anilin eller homovanilinsyre, 4-aminoantipyrin og peroxi-dase til mâling af farve eller fluoressens, 4-aminoanti-30 pyrin kan erstattes med 4-aminophenazon. En kombination af 2,6-diohlorphenolindophenol og peroxidase og af guaia-col og peroxidase kan ogsâ benyttes. Indikatoren kan frera-stilles i forvejen som oplesning. Hver 5 portion (1,0 ml) af oplzsningen anbringes separat i smâ reagensglas og forinkuberes ved 37°C i 5 minutter (for-inkuberet oplzsning af reagens (I))# Standardserumoplzs-ningen (20 μϋΐθΓ) (Calbioohem Co,, handelsnavn: Maxitol, indeholdende G-PT 700 K enheder/ml og GOT 1000 K/ml) for-10 tyndet med konstant forhold tilsættes og inkuberes ved 37°G i 10 minutter. Reaktionen standses ved tilssstning af stopreagens (III) (2,0 ml), og man mâler absorptionen ved 565 nm.
Som vist i fig. 10 har begge enzymaktiviteter 15 linearitet op til den optiske tæthed pâ ca, 1,0 (enzym-aktivitet: ca. 750 K enheder/ml).
Eksempel 12.
Til hver forinkuberet oplzsning af reagens (I) fremstillet i eksempel 11 sættes 20 μΐ^βτ humant sérum 20 (45 prover), og man inkuberer ved 37°C i 20 minutter.
Der tilsættes stopreagens (III) (2,0 ml), og man mâler absorptionen ved 565 nm.
Samme prover af humant sérum analyseres ligeledes ved EKB-metoden (ultraviolet absorptionsmetode, GOT-ana-25 lyseudstyr og GPT-analyseudstyr, fremstillet af LKB Oorp.) og korrelationen.afhlndes. grafisk.med aktiviteten af GPT og GOT.
Som det ses i fig, 11, iagttages korrelations-koefficienten r = 0,998 og regressionsligningen ; 30 y = 0,00295 x + 0,0032 for GPT-analysen,
Yed GOT-analysen fremgâr korrelationsmonstret af fig, 12, hvor man fâr korrelationskoefficienten r = 0,966 og regressionsligningen y = 0,00287 x + 0,0180.

- DK 156253B Fremgangsmâde til fremstilling af pyruvatoxidase, som katalyserer en reaktion fra pyrodruesyre, phosphat og oxygen til dannelse af acetylphosphat, carbondioxid og hydrogenperoxid, kendetegnet ved, at den omfat-5 ter dyrkning af en pyruvatoxidaseproducerende mikroorganis- me udvalgt blandt Pediococcus sp. B-0667 FERM-P nr. 4438, Streptococcus sp. B-0668 FERM-P nr. 4439, Aerococcus viri-dans IFO-12219 og Aerococcus viridans IFO-12317 i et nærings-dyrkningsmedium og isolation af den sâledes dannede pyru-10 vatoxidase fra de dyrkede celler.
