Den foreliggende opfindelse angår en fremgangsmåde til fremstilling af et antibiotisk kompleks, kaldet Bu-2231 eller dets bestanddele Bu-2231 A og Bu-2231 B i kobberkomplekse former eller kobberfrie former og/eller syreadditionssalte deraf.
Fremgangsmåden ifølge opfindelsen er ejendommelig ved, at man dyrker en stamme af Streptoalloteichus hindustanus i et vandigt næringsmedium, som indeholder assimilerbare nitrogen- og carbon-kilder under submerse aerobe betingelser og udvinder det antibiotiske kompleks fra dyrkningsmediet, og om ønsket opdeler det i sine bestanddele og/eller omdanner den kobberkomplekse form til den kobberfrie form og/eller til syreadditionssalte.
Ifølge en foretrukket udførelsesform af fremgangsmåden ifølge opfindelsen er stammen Streptoalloteichus hindustanus ATTC 31158. Denne stamme er en ny stamme af Streptoalloteichus benævnt stamme Ξ465-94 i Bristol-Banyu kultursamlingen. Organismen er en Actino-mycetes-bakterie, som isoleredes fra en indisk jordprøve. En kultur af organismen er blevet deponeret i The American Type Culture Collection, Washington, D.C., og indføjet i dens permanente samling af microorganismer som ATCC 31158.
Det hidtil ukendte glycopeptidkompleks fremstillet ifølge den foreliggende opfindelse omfatter to hoved-glycopeptidbestanddele, benævnt Bu-2231 A og B, idet begge sådanne bestanddele er bio- 2 141372 aktive. Komplekset og enhver af de ovennævnte bestanddele har en hæmmende virkning på væksten af microbielle organismer, både bakterier og fungi, som er patogene i dyre- og planteliv, og disse antibiotika er derfor nyttige til terapi mod bakteriel infektion i mennesker eller dyr og til hindring eller undertrykkelse af sygdomme hos ris, ærter, hvede eller andre planter, hvilke sygdomme er forårsaget af plantepatogene organismer. Komplekset og de enkelte bestanddele er også nyttige i behandlingen af forskellige tumorsystemer i gnavere, indbefattet Walker 256 carcinosarcoma (ascitisk form) og Lewis Lung carcinoma.

De morfologiske, dyrkningsmæssige og fysiologiske egenskaber af stamme E465-94 ATCC 31158 er sammenfattet herunder:
Stamme E465-94 danner klase og sclerotium i luftmyceliet på de fleste agarmedier. Klasen er en fremherskende sporedannende struktur, og dannelsen af sclerotier er noget ustadig. Klasen består af buede eller L-formede korte sporekæder med mange forgreninger og udvikler sig ofte i en tyk masse. Nogle sporekæder stikker ud fra klasestrukturen og danner ofte åbne spiraler.

Sclerotiumsformen er oval og lejlighedsvis uregelmæssig. Opdeling af vegetativt mycelium fremkommer ikke. Der dannes ikke krans.
Stamme E465-94 danner kraftige luftmycelier på de fleste af de afprøvede agarmedier. Det modne luftmycelium er lysegulligt beige eller bleglyserødligt gult. Diffusibel pigment dannes ikke. Tyrosinasereaktion er negativ. Stammen kan modstå varme og vokser kraftigt ved 50 °C.

Cellevægpræparatfremstilling udførtes efter den af T. Yama-guchi i J. Bacteriol. 8£: 444-453 (1965) beskrevne metode. Amino-syreanalysefremgangsmåden var som følger: Renset cellevæg (10 mg) hydrolyseredes i 1 ml 6N HC1 i et forseglet rør ved 120 °C i 18 timer. Hydrolysatet fortyndedes med et lige så stort volumen destilleret vand, filtreredes og inddampedes derpå i vakuum til tørhed. Halvdelen af slutproduktet genopløstes i 0,1 ml destilleret vand og undersøgtes ved to-dimensional TLC. Den anden halvdel opløstes i 2 ml citratpuffer (pH 2,2) og analyseredes ved væske-chromatografi. En 5 μΐ portion af hydrolysatet anbragtes på en silicagel-TLC plade (6OF254, E. Merck AG, Tyskland) og oparbejdedes med phenol-vand (4:1) i en retning og dernæst med n-butanol-eddikesyre-vand (3:1:1) vinkelret på den første udfoldelsesretning. Pletterne synliggjordes ved en påsprøjtning af 0,2% ethanolsk ninhydrinreagens, efterfulgt af opvarmning af pladen i 5 minutter ved 110 °C. Til adskillelse af meso og/ eller DD-DAP fra LL-DAP anbragtes 5 μΐ af hydrolysatet på en cellulosepulver-TLC plade. Pladen oparbejdedes med opløsnings-middelsysternet methanol-vand-10N HCl-pyridin (80:17,5:2,5:10) i 24 timer og oversprøjtedes derpå med 0,2% ninhydrinreagens.
I dette TLC-system bevægede LL-DAP sig hurtigere end den som referencestandard anvendte meso-DAP. Aminosyrer i cellevægpræparatet bestemtes også ved aminosyreanalyser. Aminosyre-analysen af cellevægpræparatet af stamme E465-94 viste tilstedeværelsen af meso-a, ε-diaminopimelinsyre (meso-DAP), alanin og glutaminsyre (tabel IV). Carbonhydrater i cellevæggen bestemtes som følger: En 50 mg prøve af den rå cellevæg opløstes i 3 ml 2N 0<2 hydro lyserede s i et forseglet rør ved 120 °C i 2 timer. Hydrolysatet neutraliseredes med mættet BaiOH^ opløsning, det udfældede BaSO^ fjernedes ved centrifugering og det ovenstående fluidum lyofiliseredes. Det således opnåede materiale 7 141372 trimethylsilyleredes, og produktet underkastedes gas-chromato-grafi og sammenlignedes med forskellige referencesukkerarter. Carbonhydratanalysen af cellevæggen af E465-94 viste nærværelsen af rhamnose, mannose, galactose og glucosamin (tabel V).

Stamme E465-94 producerer et hidtil ukendt antibiotisk kompleks Bu-2231, hvilket antages at være sammensat af antibio-tica af bleomycintype. Sammenligning udførtes derfor med adskillige Actinomycetes stammer, hvilke har vist sig at producere bleomycin eller bleomycinbeslægtede antibiotica. Resultaterne er sammenfattede i tabel VI. Stamme E465-94 skelnes klart fra enhver af disse microorganismer.

Actinomycetesstammen E465-94 dannede på agarmedier en klase og et sclerotium i luftmyceliet, hvilken morforlogi typisk findes hos nogle arter af Streptomyces eller Chainia.
Imidlertid var aminosyre- og sukker-sammensætningen i cellevægpræparatet af stamme E465-94 helt forskellig fra sammensætningen i enhver art i familien Streptomycetaceae. Cellevægpræparatet indeholdt meso-a,e-diaminopimelinsyre (meso-DAP) i stedet for LL-DAP, som har vist sig at være en specifik cellevægbestanddel i slægterne Streptomyces og Chainia. Ydermere var hovedsukker-bestanddelene i stammen galactose, mannose og rhamnose, hvilke er helt forskellige fra de for Streptomyces eller Chainia beskrevne. Under hensyn til de ovennævnte fakta antages det, at en hidtil ukendt slægt Streptoalloteichus (strepto- en streptomyceslignende organisme, allo-ændret og teichos-væg, dvs. en Streptomyceslignende organisme med usædvanlig cellevægsammensætning) må ud-nævnes under familien Streptomycetaceae til udskillelse af Actinomycetesstammer, som ligner Streptomyces i morfologi, men har cellevægsammensætningen af stamme E465-94-type; meso-DAP, alanin og glutaminsyre som hovedaminosyrer og galactose, mannose og rhamnose som diagnostisk neutrale sukkerarter. Således er 10 141372
Streptoalloteichus en blandt slægterne af familien Strepto-mycetaceae enestående slægt, som indeholder meso-DAP i stedet for LL-DAP i cellevægsairanensætningen. Det foreslås også, at stamme E465-94 kan betegnes Streptoalloteichus hindusta-nus gen. nov. og sp. nov., fordi organismen isoleredes fra Nordindien. Stamme E465-94 ser klart forskelllig ud fra microorganismerne, som har vist sig at danne bleomyciner og bleomycinbeslægtede antibiotika.
Ved fremgangsmåden ifølge opfindelsen kan anvendes stammer, som kan henføres til arten Streptoalloteichus hindu-stanus, d.v.s. hvis egenskaber ikke afviger væsentligt fra de her anførte egenskaber.

Organismen dyrkes i et næringsmedium, som indeholder en assimilerbar carbonkilde, f.eks. et assimilerbart car-bonhydrat. Eksempler på passende carbonkilder er glucose, ribose, galactose, fructose, mannose, saccharose, lactose, opløselig stivelse og glycerol. Næringsmediet bør også indeholde en assimilerbar nitrogenkilde, såsom f.eks. fiskemel, soyabønnemel, majsstøbevand, peptoner, kødekstrakt, jordnøddemel, gærekstrakt eller ammoniumsalte. Uorganiske salte, såsom natriumchlorid, kaliumchlorid, magnesiumsulfat, kalciumcarbonat, phosphater etc. tilsættes om nødvendigt. Sporelementer, såsom kobber, mangan, jern zink etc. tilsættes om ønsket mediet eller kan tilføres som urenheder i andre af mediernes bestanddele. Inkuberingstemperatu-ren kan være enhver temperatur, hvorved en Bu-2231 -dannende stamme er i stand til at vokse, f.eks. 20 - 54°C, men det foretrækkes, at fermenteringen udføres ved 25-35°C, især ved 27 - 32°C. En neutral eller nær neutral begyndelses pH- værdi, f.eks. pH svarende til 6-7, anvendes fortrinsvis i mediet, og fremstillingen af antibioticum 141372 11 udføres sædvanligvis i en tidsperiode på fra omkring 2 til 7 dage. Sædvanligvis opnås optimal fremstilling på 3 til 5 dage. Til fremstilling af forholdsvis små mængder kan rystekolber og overfladedyrkning anvendes, men til fremstillingen af store mængder foretrækkes submers aerobisk dyrkning i sterile tanke. Når tankfermentering skal udføres, er det ønskeligt, at man danner et vegetativt inoculum i et næringssubstrat ved inoculering af substratkulturen med en spore fra organismen og overføring, når et ungt aktivt vegetativt inoculum er blevet opnået, af dette inoculum aseptisk til fermenteringstanksubstratet. Beluftning i tanke og kolber kan tilvejebringes ved presning af steril luft gennem eller på det gærende mediums overflade. Yderligere bevægelse i tankene tilvejebringes ved hjælp af en mekanisk omrører. Et antiskummiddel, såsom olie fra svinefedt, kan om nødvendigt tilsættes.
Dannelsen af Bu-2231 i fermenteringsmediet kan let følges under fermenteringen ved hjælp af papirplade-agardiffusions-metoden under anvendelse af Bacillus subtilis PCI-219 og Mycobacterium 607 stamme M6-3 som testorganismer. Bu-2231 A og B er begge, såvel som de antageligt samtidigtdannede nebramycin-faktorer aktive mod B. subtilis PCI-219, men kun Bu-2231 A og B bestanddelene viste aktivitet mod M. 607 M6-3. I rystekolbe-fermenteringen dannede stamme E465-94 50-100 yg/ml Bu-2231 kompleks efter 3-5 dages forløb.
Efter opnåelse af optimal substratstyrke (f.eks. bestemt ved den ovennævnte prøvefremgangsmåde) skilles myceliet og uop-løste rester fra fermenteringssubstratet efter konventionelle metoder, såsom filtrering eller centrifugering. Den antibiotiske aktivitet indeholdes i filtratet og kan udvindes derfra under anvendelse af konventionelle adsorptionsteknikker. Adsorptions- 12 141372 midlerne, som mest formålstjenligt anvendes ved udvindingen, er cation-bytter-harpikserne, f.eks. de harpikser af IRC-5O-typen, som er kommercielt tilgængelige fra Rohm and Haas Company under varemærket "Amberiite IRC-50". Filtratet, som om nødvendigt neutraliseres til pH-værdi 7, sendes gennem en søjle, der er pakket med en cation-bytter-harpiks, såsom "Amberlite IRC-50" i ammoniumform. Harpiksen udvaskes dernæst med vand. Tillige med Bu-2231 komplekset danner organismen E465-94 samtidigt også et af nebra-mycinfaktorer bestående antibiotisk aminoglycosidkompleks.
Denne aminoglycosidurenhed kan adskilles fra Bu-2231 komplekset ved eluering med en fortyndet base, f. eks.
0,25 N· ammoniumhydroxid. Det ønskede Bu-2231 kompleks, som forbliver adsorberet på harpiksen, kan derpå elueres med en mineralsyreopløsning, f.eks. saltsyre med pH-værdi 2, og eluatets Bu-2231 fraktioner kan opsamles og kombineres.
Partiel rensning af komplekset kan udføres ved chromatografi af de kombinerede Bu-2231 fraktioner over et passende adsorptionsmiddel, såsom aktiveret carbon, og eluering med f.eks. vandig butanol med sur pH-værdi. Butanollaget skilles fra, og det vandige lag lyofiliseres eller koncentreres i vakuum til ydelse af det faste Bu-2231 kompleks. Komplekset kan yderligere underkastes chromatografi og/eller gelfiltrering til dannelse af det rensede kobberholdige faste kompleks.
Glycopeptidbestanddelene Bu-2231 A og B kan særlig fordelagtigt adskilles fra komplekset ved gradient-eluerings-chranatografi, især over en modificeret dextranderivat-cationbytter, f.eks. en modificeret poly-sacchariddextran af den kommercielt under varemærket "CM-Sephadex C-25" solgte type. En vandig opløsning af Bu-2231 komplekset, fortrinsvis renset som ovenfor beskrevet, sættes til en søjle, som indeholder den modificerede dextran-ionbytter. Som i tilfældet med bleomycinerne (J. Antibiotics, 19^ 210-215 (1966)) er det lettere at adskille komponenterne efter denne fremgangsmåde, hvis komplekset er tilstrækkeligt chelateret med kobber. Således omfatter den foretrukne adskillelsesfremgangsmåde opløsning af komplekset i en cuprichloridopløsning til sikring af, at det er 13 141372 en tilstrækkeligt kobber-chelateret form, og dernæst tilføring af denne opløsning til søjlen. Bestanddelene elueres derpå på en trinvis måde med vandig ammoniumformiatopløsning af fra 1 til 7% varierende koncentrationer. Bu-2231 B fremkommer i eluatets tidlige fraktioner, medens senere fraktioner indeholder Bu-2231 A. De fraktioner, som indeholder de samme bestanddele, forenes, afsaltes ved gelfiltrering, koncentreres og lyofiliseres til dannelse af de rensede Bu-2231 A og B bestanddele i deres kobberkompleksformer.
Anvendelse af ammoniumformiat som elueringsmiddel i den ovenfor beskrevne chromatografiske adskillelsesfremgangsmåde resulterer i udvinding af de lyofiliserede bestanddele i form af formiatsalte. For fagmanden kendte standardfremgangsmåder kan anvendes til omdannelse af formiatsaltene til deres respektive frie baser og/eller til omdannelse af formiatsaltene eller de frie baser til andre ønskede syreadditionssalte, f.eks. hydrochloridsaltene.
Glycopeptidbestanddele Bu-2231 A og B har den egenskab, at de chelaterer med kobber ligesom bleomycineme gør det, og således kan bestanddelene og deres syreadditionssalte forefindes enten i den kobberkomplekse form eller i den kobberfrie form.
De kobberfrie former af Bu-2231 A og B kan fremstilles ud fra de kobberkomplekse former ved anvendelse af H2S i methanol som beskrevet i OSA Patentskrift nr. 3.646.197.
De to antibiotiske glycopeptidbestanddele kan skelnes fra hverandre og de beslægtede bleomyciner og phleomyciner'enten i deres kobberkomplekse eller kobberfrie form ved hjælp af TLC-systemerne S-102 og S-123 som nedenfor vist i Tabel VII.

De antibiotiske stoffer Bu-2231 A og B er begge amorfe baser, som i den kobberkomplekse form fremtræder som blå faststoffer og i den kobberfrie form som hvide faststoffer. Begge stoffer er opløselige i vand og methanol, tungtopløselige i ethanol og praktisk taget uopløselige i andre organiske opløsningsmidler.
Bu-2231 A og B er i stand til dannelse af salte med syrer, f.eks. farmaceutisk acceptable syreadditionssalte.
Eksempler på velegnede farmaceutisk acceptable syreadditionssalte omfatter de ikke-toksiske salte med organiske og uorganiske syrer, såsom f.eks. saltsyre, svovlsyre, phosphorsyre, eddikesyre, myresyre, stearinsyre, propionsyre, vinsyre, ma-leinsyre, benzoesyre, ravsyre og lignende.

Forholdet mellem de specifikke UV ekstinktioner ved de to absorptionsmaxima for Bu-2231 (forholdsmæssig specifik ekstinktion ved 240 my og 290 my: 1,2 - 1,3) antyder det hidtil ukendte kompleks' lighed .ned bleomycingruppen af antibiotica snarere end phleomycingruppen.
De infrarøde og kernemagnetiske resonansspektre for det kobberfrie hydrochloridsalt af Bu-2231 A og det kobberfrie formiatsalt af Bu-2231 B er vist på tegningen i fig. 1 til 4.
Begge antibiotiske bestanddele giver positive reaktioner med ninhydrinreagens.
Bu-2231 A's og B's nære strukturelle lighed med bleomycin antydedes ved de ovenfor beskrevne fysisk-kemiske egenskaber.
De kobberfrie præparater af de to bestanddele sammen med bleomycin A2 hydrolyseredes i et forseglet rør med 6N HC1 i 20 timer ved 105 °C. Ethvert af hydrolysaterne undersøgtes sammenligningsvis ved højspændings-papirelektroforese. Lokaliteten af amino-syrerne påvistes ved hjælp af ninhydrinreagens og tillige ved UV lys.

Som vist i Tabel VIII identificeredes enhver af de 6 aminosyrer (I til VI) og 1 amindel (VII) af bleomycin A2, hvilken er beskrevet i litteraturen (J. Antibiotics 21: 79, 1968) ved hjælp af papirelektroforese. Hydrolyseprodukteme af Bu-2231 A og B udviste ninhy dr in-positive zoner svarende til aminosyreme I, II, IV og V fra bleomycin A2, og påvisningen af, at enhver af de 4 fra Bu-2231 A og B isolerede aminosyrer var identiske med aminosyreme I, II, IV og V fra bleomycin A2 udførtes ved kemiske og spektrale metoder. Aminosyre III (zone ved 13,5 cm) fra bleomycin A2 var ikke til stede i Bu-2231 A og B, hvilke i stedet udviste en yderligere ninhydrin-positiv zone ved 14,8 cm (betegnet som aminosyre VIII). Strukturen af aminosyre VIII er blevet bestemt som 4-amino-3-hydroxy-n-valerianesyre, som er en desmethylanalog af aminosyre III.
Aminosyre VI (zone ved 8,5 cm, UV absorptionsmaksimum ved 290 my) fra bleomycin A2 var ikke til stede i Bu-2231 A og B.
Imidlertid isoleredes en sur forbindelse (IX) med et UV absorp- 1% tionsmaksimum ved 283 my (E. =280) som et bundfald fra i cm syrehydrolysatet af Bu-2231 A og B.
Den endestillede amindel (VII i Tabel VIII) i bleomycin A2 fremkom ved zone 31,5 cm. Bu-2231 A og B gav den samme zone som amin VII i bleomycin A2, men adskillige amindele i bleomycin-kompleks viser lignende bevægelighed i denne papirelektroforese-metode. Yderligere analyse viser faktisk, at den endestillede amindel i Bu-2231 A og B er spermidin (X), amindelen i bleomycin A5: Bu-2231 A gav en yderligere ninhydrin-positiv zone ved 23,0 cm. (XI i Tabel VIII), hvilken ikke var til stede i bleomycin A2 og Bu-2231 B. Forbindelse XI identificeredes som β-lysin.

Sukkerbestanddelen i Bu-2231 A og B undersøgtes også i sammenligning med bleomycin. En blanding af bestanddelene A og B opvarmedes under tilbagesvaling i methanol med en stærkt sur harpiks (Amberlyst 15) i 20 timer. Basiske fragmenter fra hydrolysen adsorberedes på harpiksen, og i opløsningen frigjorte produkter koncentreredes i vakuum til en klæbrig sirup. Dette materiale trimethylsilyleredes og underkastedes derefter gaskromatografisk analyse. Bleomycin gav toppe, som kan henføres til gulose og mannose, og Bu-2231 bestanddelene viste omtrent det samme elueringsmønster som bleomycin, hvilket antyder den Η1372

20 samme struktur af carbonhydratdelen i antibiotikaene.

De minimale inhiberingskoncentrationer (MIC) af Bu-2231 A og B bestemtes mod mange forskellige bakterier og fungi ved dobbelt-agar fortyndingsmetoden. Mueller-Hinton agarmedium anvendtes til bestemmelsen af bakterielle MIC'er (gram-positive og gram-negative), medium nr. 1001 [glycerol (3%), Na-glutaminat (0,3%), pepton (0,2%), Na2HP04 (0,31%), KH2P04 (0,1%), ammoniumcitrat (0,005%), MgS04.7H20 (0,001%), agar (1,5%)] til syrefaste 21 141372 bakterier og Sabouraud's agar til fungi. Resultaterne*er vist i tabellerne IX og X i sammenligning med bleomycin og phleo-mycin. De antibakterielle og antifunglelle aktiviteter af Bu-2231 A og B er meget højere end de samme af reference-antibiotikaene. Bu-2231 A viste noget højere antibakterlel aktivitet end Bu-2231 B, men var mindre kraftig end den sidstnevnte i anti-fungiel aktivitet.

In vivo-aktiviteten af Bu-2231 kompleks evalueredes i eksperimentelle musinfektioner mod S. aureus Smith og E. coli NIHJ. Bleomycin afprøvedes til sammenligning som et referencean tibioticum. Resultaterne er vist i Tabel XI. Bu-2231 var omkring. 20 gange (S. aureus) til 80 gange (E. coli) mere aktivt end bleomycin udtrykt som middelbeskyttelsesdosisværdi (PD^q).

In vivo-aktiviteten af de enkelte bestanddele af Bu-2231 i hver af de kobberfrie og kobberkomplekse former bestemtes også i E. coli-infektion. Som vist i Tabel XII var Bu-2231 A noget 25 141372 mere aktivt end Bu-2231 B. Tilstedeværelsen af kobber i Bu-2231 molekylet påvirkede ikke kendeligt in viνο-aktiviteten.

Den akutte toksicitet af kobberfrit Bu-2231 A og B bestemtes i mus ved en enkelt subkutan injektion. Grupper på 3 mus anvendtes til hver dosisniveau, og legemsvægten registreredes i 16 dage. Resultaterne er vist i Tabel XIII. Bu-2231 A var noget mere toksisk end Bu-2231 B. Ingen død fremkom med Bu-2231 B op til en dosis på 50 mg/kg, men tab i legemsvægt bemærkedes i alle de testede dyr.

Foruden den ovenfor viste antimikrobielle aktivitet viste Bu-2231 kompleks og dets hovedbestanddele Bu-2231 A og B sig også at inhibere forskellige eksperimentelle dyriske tumorer. Testforsøg, som udførtes på to transplanterede gnaver-tumorsys terner, Walker 256 carcinosarcoma i ascitisk form og Lewis Lung carcinoma med intraperitonalt implanteret tumorvævssuspension, er beskrevet herunder. Den anvendte metodik fulgte i almindelighed protokollerne fra The National Cancer Institute som rapporteret i Cancer Chemotherapy Reports: 3(2):1-103 (1972). Eksperimenternes væsentlige detaljer er fremført i tabellerne Ia-IIIa herunder.

De i tabellerne viste resultater fortolkes som følger:
Tabel la: Testforsøg udførtes på Walker 256 ascitisk tumor med Bu-2231 kompleks og med bleomycin kompleks som kontrol. Dosen 30 141372 på 2560 yg/kg er aktiv men toksisk, siden T/C var mindre end for den næste lavere dosis, og ingen langtids-overlevende (44. dag) observeredes. Den minimale effektive dosis (MED) af bleomycin var 640 yg/kg, hvorimod MED af Bu-2231 var 160 yg/kg, hvad der antyder, at Bu-2231 er omkring 4 gange så kraftigt som bleomycin.
Tabel Ila: Bu-2231 A og B i form af deres kobberfrie formiatsalte afprøvedes på Walker 256 ascitisk tumor. Den sandsynlige minimale effektive dosis (MED) er 160 yg/kg i dette eksperiment. Sammenlignelig overlevelsesforøgelse (T/C = 157) sås med begge bestanddele ved 40 yg/kg, hvad der således antyder 4 gange større styrke for Bu-2231 A og B. A-bestand-delen kan være lidt mere kraftig end B-bestanddelen (aktivitet ved 10 yg/kg), men forskellen kan ikke betragtes som signifikant.
Tabel Ilia; Bleomycin viser lejlighedsvis skarpt afgrænsede effekter i testforsøg mod Lewis Lung carcinoma. I dette eksperiment var Bu-2231 A aktivt lige under et niveau på 1 mg/kg (T/C = 123), medens Bu-2231 B var aktivt ved doser på 2 og 1 mg/kg.
De følgende eksempler tjener til yderligere belysning af den foreliggende opfindelse.
tM-Sephadex C-25" er et varemærke for et tørt opløseligt pulver (fremstillet af Pharmacia Fine Chemicals Inc.), hvilket ar sammensat af mikroskopiske kom, som er syntetiske organiske forbindelser, der indeholder carboxymethyl-funktionelle grupper, og som udvindes fra polysaccharid-dextran. "Amberlyst 15" er et varemærke for en ionbytter-harpiks, som fremstilles af Rohm and Haas Company. "Amberlite XAD-2" er et varemærke for en adsorberende harpiks, som består af styrendivinylbenzen-copolymer og fremstilles af Rohm and Haas Company.
Eksempel 1.
Fermentering af kompleks.
En veludviklet agar~skråglaskultur af den Bu-2231 produce- 31 141372 rende organisme E465-94 anvendtes til inokulering af flydende vegetativt medium indeholdende de følgende bestanddele: 1,5% glucose, 0,5% polypepton, 0,2% gærekstrakt, 0,05% i^HPO^ og 0,05% MgS04.7H20. Podekulturen inkuberedes ved 28 °C i 2 dage på en rotationsryster (250 omdr./minut), og 2 ml af væksten overførtes til 100 ml af fermenteringsmediet i en 500 ml Erlen-meyer kolbe, hvilket medium havde en sammensætning af 2% glycerol, 1% pharmamedier, 1% majsstøbevand, 0,3% 0,003% ZnS04.7H20 og 0,4% CaCO^. Antibioticumproduktion nåede et maksimum efter 3 til 5 dages omrystning ved 28 °C.
Eksempel 2.
Ekstraktion og oprensning.
Det i overensstemmelse med Eksempel 1 fremstillede udvindingssubstrat (ca. 10 1, 50 yg/ml) filtreredes, og bioaktiviteten i filtratet adsorberedes ved pH-værdi 7 af "Amberlite •j· IRC-50" (NH^ form, 900 ml). Harpiksen udvaskedes med vand, (5 1) og derefter med 0,25N NH^OH opløsning (4 1) til eluering af nebramycinfaktorer. Bu-2231 kompleks elueredes derefter fra harpiksen med HCl-opløsning (11x3) ved pH-værdi 2. Bu-2231 fraktionerne kombineredes, adsorberedes af carbon (30 g) og elueredes med vandig butanol (11x3) ved pH-værdi 2. Butanol-laget fraskiltes, og det vandige lag koncentreredes i vakuum til tørhed. Det således opnåede rå faststof (1,2 g) oprensedes ved "Amberlite XAD-2"-chromatografi og derefter ved "Sephadex LH-20"-chromatografi til dannelse af et svagt grønligt pulver af Bu-2231 kompleks (150 mg, kobberkompleksform).
Eksempel 3.
Adskillelse af bestanddele.
Til adskillelsen i hver komponent opløstes Bu-2231 kompleks (140 mg) i 3 ml 0,7% cuprichloridopløsning og anbragtes på en "CM-sephadex C-25usøjle (85 ml), som elueredes gradvist med vandig ammoniumformiatopløsning på 1 til 7%. Bu-2231 B elueredes med 3% ammoniumformiatopløsning og Bu-2231A ved 5% koncentration.

Hver fraktion af saltedes ved’Bephadex LH-20l,-chromatografi, koncentreredes i vakuum og lyofiliseredes til dannelse af de respektive formiatsalte af Bu-2231 A (84 mg) og B (21 mg) (kobber-komplekse fcrmer). De kobberfrie formiatsaltpræparater opnåedes ved behandling af de kobberkomplekse former med H^S i methanol i overensstemmelse med den almene fremgangsmåde i Eksempel 1 i USA Patentskrift nr. 3.646.197.
Eksempel 4.
Bu-2231 A, hydrochloridsalt.
En opløsning af kobberfrit Bu-2231 A formiat (750 mg) i 100 ml methanol indstilledes til pH-værdi 2,0 ved en tilsætning af 2N methanolsk saltsyreopløsning. Opløsningen sattes derpå dråbevis til 300 ml acetone under omrøring. Det dannede bundfald fraskiltes ved filtrering og tørredes i vakuum til dannelse af det rå Bu-2231 A hydrochloridsalt som et hvidt pulver (673 mg). En portion af dette pulver (140 mg) overførtes til en søjle indeholdende 70 ml 'Bephadex LH-20"(varemærke for en modificeret alkyleret dextran-gel, som markedsføres af Pharmacia Fine Chemicals Inc.), som er pakket i 98% methanol. Ejøjlen udvikledes med 98% methanol, og de bioaktive fraktionen samledes derpå, koncentreredes i vakuum og lyofiliseredes til dannelse af det kobberfrie hydrochlorid af Bu-2231 A (105 mg).

Fremgangsmåde til fremstilling af et antibiotisk kompleks, kaldet Bu-2231, eller dets bestanddele Bu-2231 A og Bu-2231 B, i kobberkomplekse former eller kobberfrie former og/eller syreadditionssalte deraf, kendetegnet ved, at man dyrker en stamme af Streptoalloteichus hindustanus i et vandigt næringsmedium, som indeholder assimilerbare nitrogen- og carbonkilder, under submerse aerobe betingelser, og udvinder det antibiotiske kompleks fra dyrkningsmediet, og om ønsket opdeler det i sine bestanddele og/eller omdanner den kobberkomplekse form til den kobberfrie form og/eller til syreadditionssalte.
2. Fremgangsmåde ifølge krav 1 kendetegne t ved, at stammen er Streptoalloteichus hindustanus ATCC 31158.
3. Fremgangsmåde ifølge krav 1 eller 2 kendete gnet ved, at det antibiotiske kompleks udvindes fra dyrkningsmediet ved fraskillelse" af myceliet, adsorbering af komplekset på en cationbytter-harpiks, fraskillelse af det samtidigt dannede aminoglycosidkompleks fra Bu-2231-glycopeptidkomplekset ved eluering af aminoglycosidkomplekset med en fortyndet base og eluering af Bu-2231 komplekset fra adsorptionsmidlet med en mineralsyreopløsning.
4. Fremgangsmåde ifølge krav 1-3 kendetegne t ved, at man opdeler Bu-2231 komplekset i de enkelte bestanddele Bu-2231A og B i de kobberkomplekse former ved gradient-eluerings-chromatografi og eventuelt behandler de kobberkomplekse former med hydrogensulfid i methanol til dannelse af de tilsvarende kobberfrie former.
