Description
- Spiro[(4-cyclohexanon)-[3H]indol]-2'[ 1'H]-on- und Dispiro[(1,3-dioxolan)-2,4'-cyclohexan.-[3H]indol]-2''[1''H]-on-Derivate sind wichtige Zwischenstufen für die Vasopressin-V2-antagonistische Verbindung, SR 121 463. Beispilsweise kann Dispiro[(1,3-dioxo-lan)-2,4'-cyclohexan-1,3''(5''ethoxy)-[3H]indol]-2''[1''H]-on, wie in der Patentanmeldung WO 9715556 beschrieben (Verbindung der Formel VII), durch Umsetzung von 4-Ethoxyphenylhydrazin mit 4-(1,3-Dioxolan)cyclohexancarboxylatnatriumsalz und anschließende Cyclisierung des resultierenden 1-(4'-Ethoxyphenyl)-2-(4''-/ 1,3-dioxolan/cyclohexancarbonyl)-hydrazins hergestellt werden.
- Nach einem weiteren Syntheseweg (
EP 636608 - Die Nachteile von beiden obigen Synthesen sind die toxischen Ausgangsmaterialien, die vielstufigen Synthesen, die geringen Ausbeuten einiger Syntheseschritte, die teuren Reagentien und die extremen Reaktionsbedingungen bei bestimmten Umsetzungen.
- Zu unserer Überraschung; haben wir gefunden, dass im Gegensatz zu der analogen Reaktion, die in der Literatur beschrieben ist (Annalen 1941, 548, 117–146; J. Am. Chem. Soc. 1953, 75, 53015305; J. Chem. Soc. C, 1970, 796–800, J. Med. Chem. 1993, 36, 2459–2469), die Addition von Methyl- oder Ethylacrylat an 5-Ethoxyindolinon und die anschließende Dieckmann-Kondensation, Hydrolyse und Decarboxylierung nicht zu einem homogenen Produkt führen, darum sind die in der obigen Literatur beschriebenen Verfahrensweisen nicht zur industriellen Synthese geeignet. Wir haben gefunden, dass der Wasserstoff in Position 1 des 5-Ethoxyindolinons mit einer geeigneten Schutzgruppe substituiert werden muss, wenn wir wollen, dass die anschließenden Reaktionen in die gewünschte Richtung ablaufen.
- Falls der Substituent in Position 1 eine Vinylgruppe ist, wie in der USP 3395156 beschrieben, können Nebenreaktionen nicht vermieden werden. Vor der Dieckmann-Kondensation muss der erhaltene 1-Phenyloxindol-3,3-dipropionsäureester zur Darstellung eines reinen Ausgangsmaterials hydrolysiert und umgeestert werden.
- Gegenstand unserer Erfindung ist ein Verfahren zur Herstellung von Spiro[(4-cyclohexanon)-[3H]indol]-2'[1'H]-on-Derivaten der allgemeinen Formel I in der R1 und R2 unabhängig voneinander für Wasserstoffatome, C1-4-Alkyl, C1-4-Alkoxy, C1-4-Alkylthio, C1-4-Polyfluoralkyl, C1-4-Polyfluoralkoxy, C3-7-Cycloalkyloxy, C3-7Cycloalkylthio-, Phenoxy-, Benzyloxy- oder Nitrogruppen stehen, dadurch gekennzeichnet, dass man ein Indolin-2-on-Derivat der allgemeinen Formel II in der R1 und R2 die oben angegebenen Bedeutungen besitzen, mit einer Verbindung umsetzt, die dazu in der Lage ist, eine Schutzgruppe, ausgewählt aus 2-Tetrahydropyranyl-, 1-Diethoxymethylenoder C1-4-Alkoxycarbonylethylgruppen, einzuführen, die so erhaltene Verbindung der allgemeinen Formel III in der R1 und R2 die oben angegebenen Bedeutungen besitzen und A für eine Schutzgruppe, ausgewählt aus 2-Tetrahydropyranyl-, 1-Diethoxymethylen- oder C1-4-Alkoxycarbonylethylgruppen, steht, mit einem Acrylsäure-C1-4-ester kuppelt, die resultierenden Verbindung der allgemeinen Formel IV in der R1, R2 und A die oben angegebenen Bedeutungen besitzen und R3 für eine C1-4-Alkylgruppe steht, cyclisiert, die -COOOR3-Gruppe und die Schutzgruppe A des Ketoesters der allgemeinen Formel V in der R1, R2, R3 und A die oben angegebenen Bedeutungen besitzen, eliminiert, gegebenenfalls ohne Isolierung der Verbindungen der all-gemeinen Formel N
- Bezüglich Verbindungen, die in der Lage sind, die Schutzgruppe A einzuführen, können 2,3-Dihydropyran, Triethylorthoformiat oder ein Acrylsäure-C1-4-ester angewandt werden.
- Die Umsetzung der Verbindung der allgemeinen Formel II in der R1 und R2 die oben angegebenen Bedeutungen besitzen, mit einer Verbindung, die dazu in der Lage ist, die Schutzgruppe A einzuführen, wird vorzugsweise in Gegenwart eines Katalysators, vorzugsweise in Gegenwart von p-Toluolsulfonsäure, durchgeführt. Be züglich des Lösungsmittels können halogenierte Kohlenwasserstoffe, vorzugsweise Dichlormethan, verwendet werden.

- Unter Verwendung von Acrylsäure-C1-4-ester für die Schutzgruppe kann das Verfahren zweckmäßigerweise als "Eintopf″-Verfahren in einem polaren Lösungsmittel in Gegenwart einer Base durchgeführt werden.
-
- Weitere genaue Angaben bezüglich der Erfindung sind anhand der folgenden Beispiele gezeigt.
- Beispiele
- 1./Zu 38,33 g 5-Ethoxyindan-2-on werden 2,12 g p-Toluol-sulfonsäure und 880 ml Dichlormethan, anschließend unter Rühren 59 ml Dihydropyran zugesetzt. Das Reaktionsgemisch wird bis zur vollständigen Auflösung (ca. 2 h) gerührt, anschließend wird es 36 h stehen gelassen. Die resultierende braune Lösung wird mit 8-%iger wässriger Natriumhydrogencarbonatlösung gewaschen, über Natriumsulfat getrocknet und eingedampft. Der Rückstand wird langsam in 500 ml Petrolether gegossen. Der resultierende Niederschlag wird abfiltriert und mit einer kleinen Menge Petrolether gewaschen. Somit werden 42,4 g 1-(2-Tetrahydropyranyl)-5-ethoxyindolin-2-on erhalten, Fp.: 108–110°C, Ausbeute: 75%.
- Zu 41,8 g 1-(2-Tetrahydropyranyl)-5-ethoxyindolin-2-on werden 1,2 g Natriumethylat und 440 ml Toluol zugefügt. Der resultierenden Lösung werden langsam unter Rühren bei 25°C für die Dauer von 4 h 34 ml Ethylacrylat zugesetzt. Das Reaktionsgemisch wird für weitere 2 h gerührt, anschließend wird es mit 8-%iger wässriger Natriumhydrogencarbonatlösung gewaschen, über Natriumsulfat getrocknet, mit Aktivkohle geklärt, filtriert und in vacuo konzentriert. Der Rückstand wird langsam in 250 ml Petrolether gegossen. Das resultierende feste Material wird abfiltriert und mit Petrolether gewaschen. Es werden 58 g 1-(2-Tetrahydropyranyl)-3-(di-/ethoxycarbonylethyl/)-5-ethoxyindolin-2-on erhalten, Fp.: 84–86°C, Ausbeute: 78%.
- 178 g 1-(2-Tetrahydropyranyl)-3-(di-/ethoxycarbonylethyl/)-5-ethoxyindolin-2-on in 1300 ml Toluol werden mit 60,4 g Natriumethylat und 6 g Tetrabutylammoniumbromid bei 55°C 3,5 h gerührt. Nach dem Abkühlen wird das Reaktionsgemisch nacheinander mit 300 ml eiskaltem Wasser, 300 ml 1 N Chlorwasserstoffsäure und 150 ml Wasser extrahiert, mit Aktivkohle und Fuller's Erde geklärt und filtriert. Das Filtrat wird in vacuo eingedampft, der Rückstand wird unter Rühren bei Rückflusstemperatur 3, 5 h in dem Gemisch von 790 ml, 50% Ethanol und 315 ml konz. Chlorwasserstoffsäure erhitzt. Anschließend wird das Gemisch in 3000 ml Wasser gegossen, die wässrige Phase wird mit 2 × 600 ml und 3 × 300 ml Toluol extrahiert, über Natriumsulfat getrocknet und in vacuo ein gedampft. Der Rückstand wird aus Diisopropylether umkristallisiert. Das resultierende Material wird abfiltriert und mit Diisopropylether gewaschen. Somit werden 60,1 g Spiro[(4-cyclohexanon)-1,3'(5'-ethoxy)-[3H]indol]-2'[1'H]-on erhalten, Fp.: 171–172°C, Ausbeute: 60 % .
- 2./In 318 ml Dimethylsulfoxid werden 112,7 g 5-Ethoxyindan-2-on vorgelegt, anschließend werden unter Rühren 3,82 g Kalium-tert.-butylat zugesetzt. Dieser Suspension werden nach 10 min Rühren
172 ,1 g Methylacrylat bei einer Temperatur von 40–45°C für die Dauer von 70 min zugetropft. Das Gemisch wird für weitere 65 min bei dieser Temperatur gerührt, anschließend werden hierzu während 30 min 161 g Kalium-tert.-butylat zugefügt, während die Temperatur unter 60°C gehalten wird. Das tert.-Butanol wird abdestilliert, der dicke Rückstand wird in 1780 ml Wasser gegossen, die Lösung wird mit Aktivkohle geklärt und filtriert. Das Filtrat wird in einem 85°C-Bad gerührt. Hat es die Temperatur von 68 °C (ca. 25 min) erreicht, wird es angeimpft, anschließend wird das Rühren noch 3 h bei einer Temperatur von maximal 81°C fortgesetzt. Anschließend wird das Gemisch auf Raumtemperatur abgekühlt, der resultierende Niederschlag wird abfiltriert und sorgfältig mit Wasser gewaschen. Somit werden 110,7 g Spiro[(4-cyclohexanon)-1,3'(5'ethoxy)-[3H]indol]-2'[1'H]-on erhalten, Fp.: 184–186°C, es ist für den nächsten Schritt geeignet. Ausbeute: 67%. - 3./11,2 g 5-Ethoxyindan-2-on und 220 ml Triethylorthoformiat werden bei 135–140°C 20 h gerührt, anschließend wird das Reaktionsgemisch in vacuo eingedampft. 17,2 g 1-(Diethoxymethy len)-5-ethoxyindolin-2-on werden als Öl erhalten. Seine Struktur wurde durch NMR-Spektroskopie bewiesen. Ausbeute: 92%.
- Dem Gemisch von 17,2 g 1-(Diethoxymethylen)-5-ethoxyindolin-2-on, 1,5 g Kalium-tert.-butylat und 170 ml Toluol werden bei 20–30°C während 1 h 12,6 ml Ethylacrylat zugetropft. Nach weiteren 90 min Rühren werden dem Reaktionsgemisch 50 ml Wasser zugesetzt, die Phasen werden getrennt, die organische Phase wird mit Wasser gewaschen, über Natriumsulfat getrocknet und in vacuo eingedampft. 22,2 g 1-(Diethoxymethylen)-3-(di-/ethoxycarbonylethyl/)-5-ethoxyindolin-2-on werden in Form eines gelbbraunen kristallisierenden Öls erhalten, das aus 110 ml n-Hexan umkristallisiert wird. Es werden 15,1 g kristallines Produkt erhalten, Fp.: 82-83°C, Ausbeute: 62%.
- 15 g 1-(Diethoxymethylen)-3-(di-/ethoxycarbonylethyl/)-5-ethoxyindolin-2-on werden in 150 ml Toluol gelöst, und der Lösung werden unter Rühren 7,2 g Kalium-tert.-butylat in 10 min zugesetzt. Das Reaktionsgemisch wird 2 h bei Raumtemperatur gerührt, anschließend wird hierzu Wasser zugefügt, die Phasen werden getrennt, die organische Phase wird mit Wasser gewaschen, über Natriumsulfat getrocknet und in vacuo eingedampft. Somit werden 10,2 g Spiro[(3-ethoxycarbonyl-4-cyclohexanon)-1,3'(1'-diethoxymethylen-5'-ethoxy-[3H]indol]-2'-on in Form eines braunen Öls erhalten, seine Struktur wurde durch NMR-Spektroskopie bewiesen. Ausbeute: 76%.
- 4,05 g Spiro[(3-ethoxycarbonyl-4-cyclohexanon)-1,3'(1'diethoxymethylen-5'-ethoxy-[3H]indol]-2'-on werden, in dem Gemisch von 20 ml, 96% Ethanol und 0,5 ml 2 N Chlorwasserstoffsäure 2 h bei Raumtemperatur gerührt, das Reaktionsgemisch wird anschließend mit Eiswasser auf 5°C abgekühlt. Das ausgefallene Spiro[(3-ethoxycarbonyl-4-cyclohexanon)-1,3'(1'-formyl-5'-ethoxy)[3H]indol]-2'-on wird abfiltriert, Fp.: 133–136°C. Ausbeute: 52 %.
- 5 g Spiro[(3-ethoxycarbonyl-4-cyclohexanon)-1,3'(1'-formyl-5'ethoxy)-[3H]indol]-2'-on werden in 100 ml Essigsäure gelöst, hierzu werden 25 ml 5 N Schwefelsäure zugefügt. Das Gemisch wird unter Rühren unter Rückfluss erhitzt, anschließend wird es in vacuo eingedampft. Dem Rückstand wird Wasser zugesetzt, der pH wird mit Natriumhydroxidlösung auf pH 7 eingestellt. Es werden 3,1 g eines sich verfestigenden öligen Niederschlags erhalten, der mit dem Spiro[(4-cyclohexanon)-1,3'(5'-ethoxy)-[3H]indol]-2'[1'H]-on, das auf einem anderen Weg erhalten wurde, identisch ist, Fp.: 139–140°C. Ausbeute: 86%.
Claims (10)
- Verfahren zur Herstellung von Spiro[(4-cyclohexanon)[3H]indol]-2'[1'H]-on-Derivaten der allgemeinen Formel I – in der R1 und R2 unabhängig voneinander für Wasserstoffatome, C1-4-Alkyl-, C1-4-Alkoy-, C1-4-Alkylthio-, C1-4-Polyfluoralkyl-, C1-4-Polyfluoralkoxy-, C3-7Cycloalkyloxy-, C3-7-Cycloalkylthio-, Phenoxy-, Benzyloxy- oder Nitrogruppen stehen -, dadurch gekennzeichnet, daß man ein Indolin-2-on-Derivat der allgemeinen Formel II – worin R1 und R2 die oben angegebenen Bedeutungen besitzen – mit einer Verbindung umsetzt, die dazu in der Lage ist, eine Schutzgruppe ausgewählt aus 2-Tetrahydropyranyl-, 1-Diethoxymethylen- und C1-4-Alkoxycarbonylethylgruppen einzuführen, die erhaltene Verbindung der allgemeinen Formel III in der R1 und R2 die oben angegebenen Bedeutungen besitzen und A für eine Schutzgruppe ausgewählt aus 2-Tetrahydropyranyl-, 1-Diethoxymethylen- und C1-4-Alkoxycarbonylethylgruppen steht – mit einem Acrylsäure-C1-4-ester kuppelt, die erhaltene Verbindung der allgemeinen Formel IV – in der R1, R2 und A die oben angegebenen Bedeutungen besitzen und R3 für eine C1-4-Alkylgruppe steht, kuppelt, die -COOR3-Gruppe und die Schutzgruppe A des Ketoesters der allgemeinen Formel V in der R1, R2, R3 und A die oben angegebenen Bedeutungen besitzen, eliminiert, gegebenenfalls ohne Isolierung der Verbindungen der allgemeinen Formeln IV
- Verfahren nach Anspruch 1, dadurch gekennzeichnet, daß man als Verbindung zur Einführung der Schutzgruppe A 2,3-Dihydropyran, Triethylorthoformiat oder einen Acrylsäure-C1-4-ester verwendet.
- Verfahren nach den Ansprüchen 1 bis 2, dadurch gekennzeichnet, daß man die Umsetzung der Verbindung der allgemeinen Formel II – in der R1 und R2 die in Anspruch 1 angegebenen Bedeutungen besitzen – mit der Verbindung, die dazu in der Lage ist, die Schutzgruppe A einzuführen – worin A die in Anspruch 1 angegebenen Bedeutungen besitzt – in Gegenwart eines Katalysators durchführt.
- Verfahren nach Anspruch 3, dadurch gekennzeichnet, daß man als Katalysator einen sauren Katalysator, vorzugsweise eine p-Toluol-sulfonsäure, oder einen alkalischen Katalysator, vorzugsweise ein Alkalialkoholat, verwendet.
- Verfahren nach Anspruch 4, dadurch gekennzeichnet. daß man als Katalysator einen alkalischen Katalysator, vorzugsweise ein Alkali alkoholat, einsetzt.
