Description
Die vorliegende Erfindung bezieht sich auf mit Hyaluronsäure ("HA") modifizierte polymere Artikel mit
verbesserter Bioverträglichkeit, wodurch es möglich ist, die modifizierten Artikel für zahlreiche In-vivo-Anwendungen
vorzusehen, zum Beispiel für verschiedene prothetische Vorrichtungen, einschließlich künstliche Herzklappen,
vaskuläre Transplantate etc.
Hyaluronsäure ist eine bekannte, natürlich vorkommende
Substanz, welche viele Anwendungen in Medizin und Biologie aufweist. Es sei zum Beispiel auf die
US-Patentschrift k 272 522 und die darin angegebenen
Veröffentlichungen hingewiesen.
Die vorliegende Erfindung ist auf polymere Artikel gerichtet, mit Hyaluronsäure (eine Substanz, die normalerweise
in tierischen Geweben vorhanden ist) modifi-
ziert, um solchen Artikeln einen beträchtlich höheren Grad an Bioverträglichkeit als den nichtmodifizierten
Artikeln zu verleihen. Die Erfindung schließt polymere Artikel, beispielsweise Gußartikel, Filme, Folien, Fasern,
Gewebe und Gewirke, ein, die Hyaluronsäure oder Salze davon enthalten, durch den Artikel hindurch dispergiert,
oder als Überzug bzw. Auflage an der Oberfläche davon, oder beides. Die Hyaluronsäure kann auch vernetzt
werden^nach der Herstellung des Artikels. Die auf diese Weise modifizierten polymeren Artikel sind daher gut
geeignet zur Verwendung bei verschiedenen prothetischen Vorrichtungen, beispielsweise künstlichen Herzklappen,
intraokularen Linsen, vaskulären Transplantaten, Schrittmacher-Leitungen
und dergleichen, sowie bei verschiedenen Arten von Arzneimittel-Zuführungssystemen.
Die Polymeren, die mit Hyaluronsäure gemäß der Erfindung modifiziert werden können, sind recht zahlreich
und schließen ein:
1.) Polyurethane, einschließlich aliphatische, aromatische
und araliphatische Polyätherurethane und Polyesterurethane,
2.) Polyolefine, beispielsweise Polyäthylen,
2.) Polyolefine, beispielsweise Polyäthylen,
3.) Vinylpolymere, beispielsweise Polyvinylchlorid, Polyvinylalkohol und Polyvinylacetat
und Copolymere davon,
k.) Akrylpolymere, beispielsweise Poly-(Hydroxyäthyl)-Methakrylat
und Copolymere von Methyl-
akrylat und Methjfmethakrylat,
5. ) Polyamide,
6.) die Polyester und
7·) Polysiloxane.
Es gibt zahlreiche Methoden, die Hyaluronsäure in ein Polymer einzubringen oder sie auf die Oberfläche
eines Polymers aufzubringen. Eine Methode besteht darin, Hyaluronsäure in einem geeigneten Lösemittel zu
lösen und die erhaltene Losung mit einer Polymerlösung oder einer Emulsion zu mischen und danach einen Artikel,
zum Beispiel einen Film, aus der Mischung zu bilden oder diese Mischung als einen Überzug bzw. Auftrag aufzubringen.
Die geeigneten Lösungsmittel für das Lösen von Hyaluronsäure sind Wasser, Dimethylsulfoxid und Dimethylformamid.
Die Oberfläche des polymeren Substrates kann aktiviert werden, dies gilt auch für die Hyaluronsäure
.
Ein weiteres Verfahren besteht darin, Hyaluronsäure in fester Form, vorzugsweise als Pulver, einer
Polymerlösung beizumischen und danach einen Artikel, zum Beispiel einen Film, aus der erhaltenen Mischung
zu formen oder diese Mischung als Überzug bzw. Auftrag aufzubringen.
Ein weiteres Verfahren besteht darin, partikuläres Material mit Hyaluronsäure von einer Lösung her zu
überziehen und die überzogenen Partikel in eine PoIymerlösung einzubringen und dann einen Artikel, zum Beispiel
einen Film, aus der erhaltenen Mischung herzustellen oder diese Mischung als Auftrag aufzubringen.
Beispiele geeigneter Feststoffpartikel sind Ionenaustauschharze,
Siliziumdioxid, Aluminiumhydroxid etc.
Schließlich kann bei jedem der oben beschriebenen
Verfahren die Hyaluronsäure vor oder nach dem Mischen mit dem Polymer unter Verwendung verschiedener Vernetzungsmittel
vernetzt werden.
Die folgenden Beispiele, (bei welchen alle angegebenen Teile gewichtsmäßig angegeben sind, wenn nicht
anders spezifiziert), veranschaulichen die verschiedenen
Ausführungsformen der Erfindung, ohne jedoch eine Einschränkung
der Erfindung darzustellen, wobei die Erfindung allein durch die Patentansprüche umrissen ist.
Bei den Beispielen 1 bis k wurden verbesserte
Polyurethanartikel hergestellt, die aus einer Schicht Polyurethan und aus einer oder zwei Schichten einer elastischen,
filmbildenden Mischung von Hyaluronsäure mit
einem anderen Polymer oder Polymeren bestehen.
Ein Stück Polyätherurethangummifilm (Upjohn) wurde
dadurch aktiviert, daß er 10 Sekunden lang in ein Dimethylformamidbad getaucht wurde und anschließend mit
einem Wasser/Aceton-Gemisch von 1 : 30, volumenmäßig,
eine Minute lang gewaschen wurde und dann mit Wasser eine Minute lang.
Ein Gemisch, das Hyaluronsäure enthält, wurde wie folgt zubereitet: 31»0 g einer l-%igen Wasserlösung des
Natriumsalzes der Hyaluronsäure, (aus Hahnenkämmen bereitet, Proteingehalt 1,15 %, Grundviskosität fcij} 2,700) wurde
mit 6o,O g Wasser verdünnt und dann mit 6,7 g einer aliphatischen Polyurethanwasseremulsion (Neo Rez R9-62, Polyvinyl
Chemical Industries) gemischt. Ein durchscheinendes, moderat viskoses Gemisch wurde erzielt, Feststoffgehalt
2t96 %; das Hyaluronsäure/Polyurethan-Verhältnis war
etwa 1:4. Der aktivierte Polyurethanfilm wurde in das
zubereitete Gemisch getaucht, dann 30 Minuten lang an
der Luft getrocknet und schließlich in einem Vakuumofen bei 55 C und 4 3 mm Hg 30 Minuten lang getrocknet.
Der erhaltene Artikel bestand aus einer Schicht des PoIyätherurethangummifilms,
von beiden Seiten mit einem kontinuierlichen Film belegt, der aus dem aliphatischen
Polyurethan und Natriumhyaluronat bestand.
Das in Beispiel 1 beschriebene Verfahren wurde wiederholt, mit der Ausnahme, daß 0,3 g einer Mehrfunktions-Azirdinverbindung
(Polyvinyl Chemical Industries, cross-linker CX-IOO) beigemischt wurde. Der erhaltene
Artikel bestand aus einer Schicht des Polyätherurethan-Gummifilms,
der von beiden Seiten her mit einem kontinuierlichen Film bedeckt war, der aus einem aliphatischen
Polyätherurethan und vernetzten^ Natriumhyaluronat bestand.
Ein Glasstab wurde dadurch überzogen, daß er in eine 10%ige Lösung eines Polyätherurethans (The Upjohn
Company, Pellethane 80 AE) in Dimethylformamid getaucht
wurde. Der Polymerfilm wurde in einer Wasser/Aceton-Mischung von 3 : 1, volumenmäßig, koaguliert. Der Film
an dem Stab wurde in einem Ofen bei 80 C 30 Minuten lang getrocknet. Die zweite Schicht wurde dadurch/aufgebracht,
daß in das Polyurethan-Natriumhyaluronat-Gemisch gemäß Beispiel 1 eingetaucht wurde.
Der erhaltene rohrförmige Artikel bestand aus zwei Schichten, wobei eine davon ein Polyätherurethanfilm und
die zweite Schicht ein gemischter Film aus einem aliphatischen Polyurethan und vernetztem Natriumhyaluronat war.
l»09 g luftgetrocknetes Natriumhyaluronat wurde
in einem Gemisch von k,k0 g Wasser und 67 g Dimethylformamid
gelöst. 10 g der sich ergebenden Lösung wurden
mit 10 g einer 25%igen Lösung eines Polyätherurethans
(Upjohn Company, Pellethane 80 AE) in Dimethylacetamid gemischt. Der Feststoffgehalt bei dieser Lösung war
13,66 % mit einem Natriumhyaluronat/Polyurethan-Verhältnis von 1 : ^tO. Ein Glasstab wurde in diese viskose
Lösung eingetaucht. Die Polymere wurden in einem Wasser/ Aceton-Gemisch von 3 ; 1, volumenmäßig, koaguliert. Der
röhrenförmige Film wurde in Wasser 10 Minuten lang gewaschen
und dann in einem Vakuumofen bei k'y mm Hg und
50 C k.5 Minuten lang getrocknet.
Die folgenden Beispiele veranschaulichen die Anwendung eines Hyaluronsäureüberzuges auf einem Polyesterstoff
(Beispiel 5) und auf einem PVC-FiIm (Beispiel 6).
Eine Losung, die 1,5 Gewichtsprozent Hyaluronsäure
und lt0 Gewichtsprozent Vernetzungsmittel CX-IOO
enthielt, wurde zubereitet. Ein Stück Dacron-Strickstoff wurde in die Lösung 2 Minuten lang eingetaucht.
Der Überschuß der Lösung wurde ausgepreßt, und die Probe wurde über Nacht getrocknet. Der Hyaluronsäuregehalt in
bzw. bei dem überzogenen Gewirke war 3»7 Gewichtsprozent.
Eine Wasserlösung von Natriumhyaluronat wurde einer vinylakrylischen Wasseremulsion (Amsco-Res 9205» Union
Chemicals Division) beigemischt, um ein Gemisch zu erhalten, das 0,25 Gewichtsprozent Hyaluronsäure enthält.
Das Gemisch wurde dadurch aufgebracht, daß an einen Polyvinylchlorid- (PVC) -Film getaucht wurde und 2 Stunden lang
an der Luft getrocknet wurde. Der überzogene Film wurde in l^iger, gewichtsmäßig,Lösung des Vernetzungsmittels
CX-IOO in einem Wasser-Aceton-Geraisch, das 20 Volumenprozent
Aceton enthielt, behandelt und 3 Stunden lang an der Luft getrocknet. Ein Film mit einem gut haftenden
Überzug, der Hyaluronsäure enthält, wurde erzielt.
Die folgenden Beispiele veranschaulichen eine andere Herangehensweise, einen Polyurethanfilm, der Natriumhyaluronat
aufweist, zu erhalten, nämlich: Einbringen von Natriumhyaluronat in Pulverform, feinverteilt,
in eine Polyurethanlösung, (durch das Gießen des Gemisches
gefolgt), und Präzipitieren des Films durch das geeignete koagulierende Mittel.
Natriumhyaluronat (0,33 g» Grundviskosität 3·ΐ6θ)
in Pulverform, feinverteilt, wurde 10,69 g einer Polyurethanlösung
(Pellethane 8θΑΕ, Upjohn Company) in Dimethylacetamid
(Konzentration ΐ6,Ί Gewichtsprozent) zugegeben.
Das Gemisch wurde mit Hilfe eines Glasstabes auf einer Glasplatte ausgebreitet und in 100 ml 95prozentigen
Äthylalkohol gelegt. Der präzipitierte milchigweiße Film wurde 10 Minuten lang in dem Alkohol gelassen,
dann aus dem Bad entfernt, zweimal mit Äthanol gewaschen und an der Luft getrocknet. Der erhaltene elastische
Film enthielt 9»35 Gewichtsprozent Natriumhyaluronat.
Das folgende Beispiel veranschaulicht die Modifikation des erhaltenen Films, wie in Beispiel 7 oben beschrieben,
die zu der Bildung einer vernetzten Hyaluronsäureschicht an der Oberfläche des Films führt. Die Behandlung
besteht darin, daß der Film in eine Wasserlösung des Vernetzungsmittels gelegt wird, welche die Hyaluronsäure
beim Trocknen vernetzt. Bei dieser Behandlung quellen Hyaluronsäureteilchen, lösen sich teilweise im
Wasser und wandern zur Oberfläche des Films hin. Als Folge davon gibt es eine Schicht einer konzentrierten
Hyaluronsäurelösung an der Oberfläche. Beim Trocknen wird Hyaluronsäure durch das Vernetzungsmittel vernetzt,
und auf diese Weise entsteht eine Schicht vernetzter Hyaluronsäure an der Filmoberfläche.
Der in Beispiel 7 erhaltene Film wurde in eine Lösung einer Mehrfunktions-Arizidin-Verbindung in Wasser
(0,5 Gewichtsprozent; cross-linker, CX-IOO, Polyvinyl Chemical Industries) 1 Minute lang eingetaucht, dann aus
der Lösung entfernt und 2 Stunden lang an der Luft getrocknet.
Dann wurde der Film wiederholt in Wasser gewaschen und getrocknet. Der Film, der erzielt wurde,
hatte eine kontinuierliche Schicht aus vernetzter Hyaluronsäure auf seiner Oberfläche, welche dadurch bewiesen
wurde, daß der Film in einer Wasserlösung von Toluidine 0 Blue gefärbt wurde.
Das folgende Beispiel veranschaulicht eine Variante der oben umrissenen Vorgehensweise. Bei dieser Variaute
wird ein Material, das aus kleinen Partikeln besteht, zum Beispiel Ionenaustauschharz, Siliziumdioxid, Aluminiumhydroxid
etc., mit Hyaluronsäure bedeckt, und diese überzogenen Teilchen werden in ein Polyurethan eingebracht
·
Ein Ionenaustauschharz (5,0 g Dowex 5OW-2x, Maschenweite
200-400) wurde gemischt mit 10,6 g Hyaluronsäure in Wasser mit einer Konzentration von 7»2 mg/ml.
Das Gemisch wurde in einem Vakuumofen getrocknet. Das erhaltene Produkt enthielt 1,5 % Hyaluronsäure. Das belegte
Harz (0,5 g) wurde mit 5 g der im vorhergehenden Beispiel benutzten Polyurethanlösung vermischt. Der Film
wurde präzipitiert und behandelt, wie oben. Der Hyaluronsäuregehalt
in dem Film war 0,38 %.
Das folgende Beispiel veranschaulicht die Modifikation der Oberflächeneigenschaften von Silikonkautschuk
mit einem hyaluronatenthaltenden Auftrag.
Ein Stück Silikongummi ( Silastic, Dow Corning) wurde in eine lOprozentige Lösung eines aminenthaltenden
Silans (Silane A-1100, Union Carbide) in Toluol eingetaucht,
10 Minuten an der Luft getrocknet und dann in einem Ofen bei 60 C 10 Minuten lang getrocknet. Ein
auf diese Weise behandeltes Gummi wurde in eine 0,5 % Natriumhyaluronat und 0,12 % Cross-Linker CX-IOO enthaltende
Lösung eingetaucht. Die Probe wurde an der Luft und dann in einem Ofen bei 70 C 10 Minuten lang getrocknet.
Die modifizierte Probe besaß eine benetzbare Oberfläche. Diese Eigenschaft war permanent, wie durch wiederholtes
Waschen der Probe mit Wasser herausgefunden wurde.
Die Bioverträglichkeit der Zubereitungen gemäß der Erfindung wurde durch den nachstehend beschriebenen Versuch
demonstriert.
Die Freisetzung von Η-Serotonin durchlmenschliche
-Ik-
Blutplättchen wurde in vorausgehenden Studien angewendet, um die Blutreaktivität des Produktes des Beispiels 5 abzuschätzen.
Normales menschliches venöses Blut wurde in Kunststoffspritzen abgezogen und unmittelbar in Kunststoffröhrchen
übermittelt, die 3,8 % Natriumeitrat (ein Teil Citrat zu neun Teilen Vollblut) enthielten. Plättchenreiches
Plasma wurde durch Zentrifugierung bei 4 C
Minuten lang mit 125 x S zubereitet und durch eine serologische Pipette in eine Kunststoff- oder silizierte
Teströhre entfernt. H-Serotonin ( H-5-Hydroxytryptamin, 5H-5HT; New England Nuclear, 26,3 Ci/mmol, lmCi/ml
Äthanol-Wasser) wurde dem plättchenreichen Plasma (platelet rich plasma = PRP), 0,2-0,5 ul/ml PRP, beigemischt
und 15 Minuten lang bei 37 C inkubiert. Bei der Prüfung wurden silizierte Teströhren oder
Polypropylen-Teströhren verwendet; Thrombin wurde als Positivkontrolle benutzt, belegte und unbelegte Proben
wurden geprüft. 1,0-2,0 ml H-5HT-PRP wurde jeder von Duplikatröhren, die zu untersuchende Proben enthielten,
zugegeben; eine 50 ul Aliquote wurde zur Ermittlung der
Gesamtradioaktivität aus dem Kontrollgemisch entfernt. Im Anschluß an die entsprechende Inkubationszeit
(10-120 Minuten) wurden 0,2-0,5 ml Aliquoten der Suspension entfernt und zwei Minuten bei 12.000 χ g über
Silikonöl in einer Eppendorf-Microfuge zentrifugiert. 50 ul des Überstandes wurden aus jeder Röhre entfernt,
mit 5 ml eines flüssigen Szintxllationsfluids vermischt,
und die Radioaktivität wurde durch Betaspektrometrie gemessen. Der Betrag von H-5HT, durch Thrombin freigesetzt,
oder die Testproben, war die Zunahme an Radioaktivität des Überstandes (Radioaktivität der experimentellen
Proben minus Radioaktivitätskontrolle). Die überzogenen Proben des Produktes des Beispiels 5 waren
konsistent weniger reaktiv, vom Betrag des freigesetzten H-5HT aus betrachtet; die nichtüberzogenen Proben
bewirkten eine 52 % größere Freisetzung als die über-
zogenen Proben.
Variationen und Modifikationen können natürlich
vorgenommen werden, ohne von dem Gedanken und Umfang der Erfindung abzuweichen.
Claims (23)
1. Stoffzusammensetzung, gekennzeichnet durch
ein polymeres Material, das durch den Einschluß einer Hyaluronsäure darin, oder eines Salzes davon, modifiziert
ist.
2. Stoffzusammensetzung nach Anspruch 1, dadurch
gekennzeichnet, daß das polymere Material ein Polyurethan, Polyolefin, ein vinylisches Polymer oder Copolymer,
ein Akrylpolymer oder -copolymer, ein Polyamid, ein Polyester oder ein Polysiloxan ist.
3· Stoffzusammensetzung gemäß Anspruch 2, dadurch
gekennzeichnet, daß das polymere Material ein Polyurethan ist.
k. Stoffzusammensetzung nach Anspruch 3, dadurch
gekennzeichnet, daß das Polyurethan ein aliphatisches, aromatisches oder araliphatisches Polyätherurethan oder
Polyesterurethan ist.
5. Stoffzusammensetzung nach Anspruch 3, dadurch
gekennzeichnet, daß die eingeschlossene Hyaluronsäure, oder das Salz davon, auf die Oberfläche des polymeren Materials
aufgebracht ist.
6. Stoffzusammensetzung nach Anspruch 3» dadurch
gekennzeichnet, daß die eingeschlossene Hyaluronsäure an die Oberfläche des polymeren Materials kovalent gebunden
ist.
7. StoffZusammensetzung nach Anspruch 3, dadurch
gekennzeichnet, daß die Hyaluronsäure durch den ganzen Körper und an der Oberfläche des polymeren Materials
verteilt ist.
8. Stoffzusammensetzung nach Anspruch 5» dadurch
gekennzeichnet, daß die Hyaluronsäure an der Oberfläche des polymeren Materials vernetzt ist.
9· StoffZusammensetzung nach Anspruch 6, dadurch
gekennzeichnet, daß die Hyaluronsäure an der Oberfläche des polymeren Materials vernetzt ist.
10. StoffZusammensetzung nach Anspruch 7» dadurch
gekennzeichnet, daß die Hyaluronsäure an der Oberfläche des polymeren Materials vernetzt ist.
11. Geformter Artikel, aus der Stoffzusammensetzung
nach Anspruch 3 hergestellt.
12. Artikel nach Anspruch 11, dadurch gekennzeichnet, daß er aus einem Gußartikel, einem Film, einer
Faser oder aus einem Gewebe oder einem Gewirke besteht.
13· Stoffzusammensetzung nach Anspruch 2, dadurch
gekennzeichnet, daß das polymere Material ein Polyester ist.
ΐΛ. Stoffzusammensetzung nach Anspruch 13» dadurch
gekennzeichnet, daß die eingeschlossene Hyaluronsäure, oder ein Salz davon, auf die Oberfläche des polymeren
Materials aufgebracht ist.

gekennzeichnet, daß die Hyaluronsäure an oder auf der Oberfläche des polymeren Materials vernetzt ist.

daß dieser ein Gußartikel, ein Film, eine Faser oder ein Gewebe oder Gewirke ist.
18. Stoffzusammensetzung nach Anspruch 2, dadurch
gekennzeichnet, daß das polymere Material ein Vinylpolymer
ist.
19· Stoffzusammensetzung nach Anspruch 18, dadurch
gekennzeichnet, daß das VinylpoLymer ein Polyvinylchlorid ist.
20. StoffZusammensetzung nach Anspruch 19» dadurch
gekennzeichnet, daß die eingeschlossene Hyaluronsäure, oder ein Salz davon, auf die Oberfläche des polymeren
Materials aufgebracht ist.
21. StoffZusammensetzung nach Anspruch 20, dadurch
gekennzeichnet, daß die Hyaluronsäure an der Oberfläche
des polymeren Materials vernetzt ist.
22. Geformter Artikel, aus der Stoffzusammensetzung
nach Anspruch 18 hergestellt.
23. Artikel nach Anspruch 22, dadurch gekennzeichnet, daß der Artikel ein Gußartikel, ein Film, eine Folie,
eine Faser, ein Gewebe oder ein Gewirke ist.
