Cyanoguanidinderivate und Verfahren zu deren Herstellung
Die Erfindung betrifft neue Cyanoguanidinderivate und deren Säureadditionssalze mit sehr guten pharmakologisehen
Eigenschaften. Die Erfindung betrifft auch ein Verfahren zur Herstellung der neuen Verbindungen.
Die neuen Cyanoguanidinderivate haben die allgemeine Formel (I)

Typische Substituenten für die substituierten Phenylgruppen

gemäss der Definition von A in der allgemeinen Formel (I)
schliessen ein eine Niedrigalky1-, Niedrigalkeny1-, Halogen,
Niedrigalkoxy-, Niedrigalkylthio-, Niedrigalkylsulfonyl-, halogensubstituierte Niedrigalkylgruppen und dergleichen.
Der Ausdruck "Niedrig", wie er hier verwendet wird, schliesst geradkettige oder verzweigte Kohlenstoffketten
mit 1 bis 6 Kohlenstoffatomen ein. Typische Beispiele für Niedrigalkylgruppen sind z.B. Methyl, Äthyl, n-Propyl, Isopropyl,
Isobutyl, 1-Methylpropyl, t-Butyl. n-Pentyl, 1-Äthylpropyl,
Isoamyl, n-Hexyl.,und dergleichen und Niedrig-alkeny!gruppen
können z.B. Isopropenyl, Allyl, Crotyl, Methallyl und dergleichen einschliessen.
Die für Y in der allgemeinen Formel (V) erwähnten Niedrigalkoxy- oder Niedrigalkylthiogruppen können auch 1 bis 6
Kohlenstoffatome haben.
Der Ausdruck "Halogen" bedeutet in der Praxis Fluor, Chlor, Jod oder Brom.
Die Cycloalky!gruppen schliessen Cycloalkylgruppen mit
5 bis 6 Kohlenstoffatomen ein. Die Niedrigalkylgruppen
und Niedrigalkeny!gruppen für R, R. und R_ haben die gleiche
Bedeutung wie die vorerwähnten.
Typische stickstoffhaltige heterozyklische Verbindungen, welche die Gruppen R- und R„ mit dem Stickstoffatom, an das
.sie gebunden sind, bilden, schliessen z.B. ein, Pyrolidin,
Piperidin, Piperazin oder Morpholin.
Alkylen, das für X steht, bedeutet eine Niedrigalkylengruppe.

wie beispielsweise Methylen, Äthylen und Propylen und diese Gruppe kann auch, ein endständiges Schwefelatom enthalten.
Die für B stehende Pyridylgruppe kann über jede der 2-, 3- oder 4-Stellungen des Pyridinringes gebunden
sein.
Die Verbindung (I) kann auch in tautomerer Form vorliegen
und diese Tautomeren sind.in die Erfindung eingeschlossen.
Die erfindungsgemässen Verbindungen (I) können leicht in
die pharmakologisch annehmbaren Säureadditionssalze überführt v/erden durch Umsetzen mit einer anorganischen oder
organischen Säure. Typische anorganische Säuren sind Chlorwasserstoffsäure, Bromwasserstoffsäure, Jodwasserstoff säure,
Schwefelsäure und dergleichen, und typische organische Säuren sind Maleinsäure, Fumarsäure, Bersteinsäure,
Essigsäure, Malonsäure, Zitronensäure, Benzoesäure und dergleichen.
Die erfindungsgemässen Verbindungen sind alle neu und bisher
nicht in der Literatur beschrieben worden. Sie haben eine sehr breite und sehr gute pharmakologische Wirksamkeit.
Sie sind z.B, pharmakologisch wirksam durch ihre die Magensaftsekretion inhibierende Wirkung sowie durch antisekretorische,
antidiarrhoische, antiödemische, diuretische, hypertonische, hypotonische, vasokonstriktorische, antiserotonische,
muskelentspannende, coronavasodilatorische und bradikardische (Herzschlag verlangsamende) Wirksamkeit.
Die Verbindungen (I) sind somit aufgrund ihrer pharmakologischen Wirkung sehr wertvolle Arzneimittel. Beispiele

für solche Arzneimittel sind antipeptische Geschwulstmittel, antidepressive Mittel, Lokalanästhetika, entzündungshemmende
Mittel, Diuretika, antiallergische Mittel, Antidiarrhoemittel, hypertonische, hypotonische, antiarrhytmische,
muskelrelaxierende oder coronavasodilatorische Mittel.
Da die inhibierende Wirkung der erfindungsgemässen Verbindung
auf die Magensaftsekretion nicht auf einer anticholinergischen Wirkung beruht, weisen die erfindungsgemässen
Verbindungen nicht die aufgrund von anticholinergischer
Wirkung bekannten Nebenwirkungen auf. Dies ist sehr charakteristisch für die erfindungsgemässen Verbindungen
und deshalb sind diese besonders hoffnungsvoll als antipeptische Geschwulstmittel.
Die erfindungsgemässen Verbindungen (I) kann man nach verschiedenen
Verfahren herstellen. Nachfolgend wird ein Verfahren, das man im allgemeinen anwendet, gezeigt:

Herstellungsverfahren (1)
Die erfindungsgemässe Verbindung (I) kann einstufig hergestellt
werden, indem man ein Aminderivat der allgemeinen Formel (II) mit einem Isothioharnstoffderivat oder einem
Isoharnstoffderivat der obigen Formel (III) umsetzt. Die
Umsetzung kann in Gegenwart oder Abwesenheit eines Lösungsmittels erfolgen. Dabei kann man einen Überschuss des
Aminderivates als Lösungsmittel verwenden. Es ist jedoch wünschenswert als Lösungsmittel z.B. Methanol, Äthanol,
Isopropanol, Acetonitril oder Chloroform zu verwenden. Die Umsetzung wird bei Raumtemperatur oder erhöhter Temperatur
vorgenommen. Man kann die Umsetzung unter Rückfluss durchführen, sofern ein Lösungsmittel verwendet wird.
Herstellungsverfahren (2)
Die erfindungsgemässen Verbindungen der allgemeinen Formel
(I) kann man auch nach einem Zweistufenverfahren herstellen, indem man ein Aminderivat der allgemeinen Formel
(II) mit einer Verbindung der allgemeinen Formel (IV) unter Bildung einer Verbindung der allgemeinen Formel (V) umsetzt,
worauf man die letztere dann mit einem Amin der

allgemeinen Formel (VI) umsetzt. Bei diesem Verfahren
können beide Umsetzungen in den beiden Stufen unter ähnlichen Bedingungen durchgeführt werden wie die Umsetzung des Aminderivats (II) mit dem Isothioharnstoffderivat
oder Isoharnstoffderivat (III) gemäss den vorhergehenden Ausführungen. Die Umsetzung kann auch vorgenommen werden, ohne dass man die Verbindung (V) aus dem Reaktionsgefäss nach der Umsetzung des Aminderivates (II) mit der Verbindung (IV) abtrennt.

Diese Ergebnisse zeigen, dass die erfindungsgemässen Guanidinderivate sehr starke pharmakologische Wirkungen jeweils im Vergleich zu bekannten Verbindungen aufweisen.
Die Erfindung wird in den folgenden Beispielen näher
beschrieben, ohne dass diese Beispiele limitierend auszulegen sind.

(1)(a) Dimethylcyanodithioimidocarbonat in einer Menge
von 11,4 g (0,1 Mol) und Ji,- (2-Pyridyl) -o-methylbenzylamin
in einer Menge von 18,4 g (0,1 Mol) wurden in 300 ml Äthanol gelöst. Das ganze wurde über Nacht umsetzen gelassen.
Die abgeschiedenen Kristalle wurde durch Filtration abgetrennt, das Filtrat wurde konzentriert, wobei eine
weitere Menge an Kristallen gewonnen wurde. Diese Kristalle wurden zusammengegeben und aus Äthylacetat umkristallisiert,
wobei man 17,7 g (Ausbeute 66,4 %) N-Cyano-S-methyl-N-/φ-(2-pyridyl)-orthomethylbenzy!/-isothioharnstoff
erhielt. Schmelzpunkt: 162 bis 164°C.

(b) Es wurde eine Mischung aus 5,9 g (0.02 Mol). N-Cyano-S-methy1-N-/cC -(2-pyridyl)-orthomethylbenzyl7-isothioharnstoff,
hergestellt gemäss (a), 30 ml einer 20 %-igen Lösung von Methylamin in Äthanol und 70 ml
Äthanol hergestellt und über Nacht bei Raumtemperatur umgesetzt. Die Reaktionslösung wurde unter vermindertem
Druck konzentriert. Der Rückstand wurde aus Äthylacetat umkristallisiert, wobei man 4,7 g N-Cyano-N1-methyl-
~(2-pyridyl)-orthomethylbenzylZ-guanidin (Ausbeute:
85,0 %) erhielt. Schmelzpunkt: 187 bis 189°C.

(2) 1,8 g (0,01 Mol) ώ -(2-Pyridyl)-orthomethylbenzylamin

Die Reaktionslösung wurde unter vermindertem Druck konzentriert. Der Rückstand wurde aus Äthylacetat umkristallisiert,
wobei man 1,6 g N-Cyano-N'-methyl-N-^öo -(2-pyridyl)-orthomethylbenzyl7-guanidin
(Ausbeute: 59,0 %) erhielt. Schmelzpunkt: 187 bis 189°C.

5 g (0,018 Mol) N-Cyano-N' -ßj - (2-pyridyl) -benzyl7-S-methylisothioharnstoff
und 5 g (0,057 MoI) N,N-Dimethyläthylendiamin wurden in 100 ml Äthanol gelöst und das
Ganze wurde unter Rückfluss 10 bis 15 Stunden umgesetzt.
Die Reaktionslösung wurde unter vermindertem Druck konzentriert und der Rückstand mit Wasser gewaschen und
dann aus Äthylacetat umkristallisiert, wobei man 3,4 g N-Cyano-N1-(2-dimethylamino)-äthyl-N-^^C -(2-pyridyl)-benzyl7~guanidin
in einer Ausbeute von 58,7 % erhielt. Schmelzpunkt: 142 bis 144°C.
