Die Erfindung betrifft den in den Ansprüchen gekennzeichneten
Gegenstand.
Die neuen Cephalosporinderivate der allgemeinen Formel I sind Antibiotika mit breitem Wirkungsspektrum und eignen sich zur
Bekämpfung von durch gram-negative und gram-positive Keime hervorgerufenen Infektionen. Zu diesem Zweck können sie entweder
parenteral oder oral in Form der freien Säuren oder in Form von Salzen, z, B, als Natrium-, Kalium-, Calcium- oder Magnesiumsalze,
verabreicht werden.
Die Verbindungen der allgemeinen Formel I können in an sich bekannter Weise dadurch hergestellt werden, daß Cephalosporine

in der R die vorstehend angegebene Bedeutung hat, einschließlich Cephalosporin C, mit den entsprechenden Mercaptopyrazinen umgesetzt
werden. Die Verbindungen der allgemeinen Formel I können auch dadurch hergestellt werden, daß eine 7-Aminocephalosporansäure
der allgemeinen Formel VI

Acylierungsmittel, z. B. einem Säurechlorid, Säureanhydrid, Säureazid oder aktivierten Ester, umgesetzt wird. Der Ersatz
der Acetoxygruppe der Cephalosporinderivate erfolgt in Anwesenheit eines inerten Lösungsmittels, z. B. Aceton, Dioxan, Methanol,
Äthanol, Tetrahydrofuran oder in einem Gemisch dieser Lösungsmittel, in einer wäßrigen Lösung der Lösungsmittel, in

Die Thiole können entweder in freier Form oder als Metallsalze verwendet werden.

Die Verbindungen der allgemeinen Formel VI können entweder aus 7-Aminocephalosporansäure oder aus Cephalosporin C durch ümsetzen
mit dem entsprechenden Mercaptopyrazin hergestellt werden· Im Falle von Cephalosporin C als Ausgangsmaterial wird das erhaltene
3-Pyrazinylthiomethyl-cephalosporin C in an sich bekannter
Weise hydrolysiert; vgl. E. H. Flynn, a.a.O., S, 27. Es
wird die entsprechende 7-Aminocephalosporansäure-Verbindung erhalten.
Die Hydrolyse (Spaltung der Säureamidgruppe) wird mit Phosphorpentachlorid
an einem geschützten Derivat» wie dem silylierten 3-thiolierten Cephalosporin C-ester, durchgeführt. Das imidchlorid
wird mit einem Alkohol in den entsprechenden Iminoäther überführt. Nach Hydrolyse wird die 3-thiollerte 7-Aminocephalosporansäure
erhalten. Diese Umsetzungen werden durch folgendes Reaktionsschema erläutert.

Die Zwischenprodukte der allgemeinen Formel VI sind neue Verbindungen.
Ein weiterer Gegenstand der vorliegenden Erfindung sind daher die entsprechenden Acylamidomethylester, Acyloxymethylester,
5-Oxotetrahydro-2-furylester und Phthalidylester der Säuren
der Formel I.
In Tabelle I ist die MHK einiger Verbindungen der Erfindung und bekannter Verbindungen zusammengefaßt. Die Bestimmung der MHK

Ferner wurden in vivo-Versuche an Mäusen durchgeführt, die experimentell mit Staphylococcus aureus und Salmonella abortivo
equina infiziert worden waren (Infektionsdosis 2 LD_0/Maus i.p.);
es wurden Gruppen von sechs männlichen Swiss Cobs-Albinomäusen verwendet. Die infizierten Tiere wurden subkutan 4 Stunden nach
der Infektion behandelt. Die Sterblichkeitsrate wurde täglich während eines Zeitraums von sieben Tagen aufgezeichnet. Es wird
die Dosis curativa 50 (DCj-q) bestimmt; vgl. L. J. Reed, H.
Muench, Amer. J. Hyg., Bd. 27 (1938), S. 493 bis 497.
Die Ergebnisse sind in Tabelle II zusammengefaßt.

neten Wirksamkeit gegen Staphylokokken von besonderem Interesse,
da diese ^offensichtlich wesentlich besser ist als Jene bekannter Cephalosporine, wie Cephalozin(GB-PS 1 206 305) und Cephalothin
(BE-PS 6l8 663).
Die Verbindungen der Erfindung sind gegenüber gram-negativen Keimen im allgemeinen weniger wirksam. Jedoch zeigten die Verbindungen
314, 317 und 321 eine therapeutische Wirksamkeit, die
jener von Cephalothin bei experimenteller Infektion von Mäusen mit Salmonella abortivo equina ähnlich ist. Insbesondere zeigte
sich, daß die therapeutische Wirksamkeit der Verbindung 321 bei Mäusen, die mit Salmonella abortivo equina experimentell infiziert
waren (0Cc0 mg/kg) gleich oder nur unwesentlich geringer
ist als Jene von Cephalothin bzw. Cephalozin (321=50; Cephalothin=50; Cephalozin=25)i dieses Verhalten beweist eine andere
Pharmakokinetik oder eine größere BioVerfügbarkeit der neuen
Verbindung.
Die folgenden Beispiele erläutern die Herstellung der Verbindungen
der Erfindung.
7-Phenylacetamido-3-(6-carboxamidopyrazin-2-yl-thiomethyl)-3-cephem-4-carbonsäure

Eine Lösung von 3 g 7-Phenylacetamidocephalosporansäure,
1,3 g 2-Mercaptopyrazin und 1,36 g Natriumbicarbonat in 45 ml
einee Gemisches aus Wasser-Aceton (2:1) wird 3 Stunden bei 65
l_ bis 700C gerührt. Danach wird das Aceton unter vermindertem j

Druck abdestilliert und die wäßrige Lösung mit 2N Salzsäure unter Kühlung auf O bis 5°C auf einen pH-Wert von 2,0 einge-
stellt. Der erhaltene rohe Niederschlag wird abfiltriert, mit Wasser gewaschen und aus wäßrigem Aceton umkristallisiert. Es
werden 2,6 g (70 % d. Th.) der Titelverbindung in gelblichen Kristallen vom P. Hl2oC erhalten.

Gemäß Beispiel 1 werden unter Verwendung von 7-Phenylacetamldocephalosporansäure
und der entsprechenden Mercaptopyrazine die folgenden Verbindungen erhalten:
a) 7-Phenylacetamido-3-pyrazinyIthiomethyl-3-cepham»4-carbonsäure
(356/308), 68 % Ausbeute; F. 2O8°C.

Ausgehend von 7-Phenoxyacetamidocephalosporansäure und den entsprechenden
Mercaptopyrazinen werden nach dem in Beispiel 1 beschriebenen Verfahren folgende Verbindungen erhalten:
a) 7-Phenoxyacetamido-3-pyrazinylthiomethy1-3-cephem-4-carbonsäure
(52 % Ausbeute). F, 190°C.

Beispiel 5 7-Pyrazinylthloacetamidocephalosporansäure (356/312)
Eine Lösung von 7 g Natriumsalz der 7-Chloracetamidocephalosporansäure
in 20 ml Wasser wird mit einer Lösung von 2,7 g 2-Mercaptopyrazin in wäßrigem Natriumbicarbonat bei 0,5°C versetzt,
wobei der pH-Wert mit gesättigter Natriumbicarbonatlösung auf 7 eingestellt wird. Nach 3stündigem Rühren bei Raumtemperatur wird
die gekühlte Lösung auf einen pH-Wert von 2 angesäuert. Der entstandene Niederschlag wird abfiltriert und aus Äthanol umkristallisiert.
Es werden 7,8 g (74 % d. Th.) der Titelverbindung
vom F. 145°C erhalten.

Das gleiche Produkt wird auch ausgehend sowohl von 7-Bromacetamidocephalosporansäure
als auch von 7-Jodacetamidocephalosporansäure erhalten.
Unter Verwendung von 7-Pyrazinylthioacetamidocephalosporansäure
und der entsprechenden Mercaptopyrazine werden gemäß Beispiel 1 folgende Verbindungen erhalten:
a) V-Pyrazinylthioacetamido^-pyrazinylthiomethyl-S-cephem-^-
carbonsäure (65 % Ausbeute); F; 175°C.
