Es wurden nun verbesserte kardiovaskuläre Mittel gefunden, die für die Behandlung und Linderung
von Herzarrhythmien und anderen kardiovaskulären Störungen brauchbar sind.

dadurch gekennzeichnet, dass man 1- (2,3-Epoxy-l-propoxy )-5,8-dihydro-5*
8-methanonaphthalin oder ein am Kohlenstoffatom 4 substituiertes Derivat davon mit einem entsprechenden
Amin behandelt, um das entsprechende 1-(3-Amino-2-hydroxy-1-propoxy)-5,8-dihydro-5*8-methanonaphthalin
mit substituierter Aminogruppe herzustellen. Eine weitere
Substitution am Kohlenstoffatom 4 des Naphthalinringes kann auch nach der Amiribehandlung mit geeigneten Verfahren
ausgeführt werden.

Die Verbindungen der Formel I sind sowohl in dem bicyclischen Kern als auch in der Propanseitenkette
asymmetrisch und liegen daher in Form mehrerer Isomeren vor. Dementsprechend soll Formel I sowohl die betreffen-

Die Zwischenprodukte der Formel B werden im typischen Falle nach Stufe (1) hergestellt, indem man
die entsprechende Verbindung der Formel A, die den gewünschten Substituenten Y' aufweist, in einem geeigneten inerten
flüssigen Reaktionsmedium mit einem Epihalogenhydrin
umsetzt. Im typischen Falle erfolgt diese Behandlung bei Temperaturen im Bereich von ca. 50 bis 70 0C ca. 3
bis 16 Stunden lang, wobei man ca. 5 bis 10 Mol Epihalogenhydrin pro Mol Verbindung der Formel A verwendet.
Geeignete Verwendbare Epihalogenhydrine sind z.B. Epibromhydrin und Epichlorhydrin. Die besten Ergebnisse
werden im typischen Falle bei Verwendung von Epibromhydrin erhalten. Geeignete verwendbare inerte Reaktionsmedien sind z.B. Dimethylformamid, DimethyIsulfoxyd und

dergleichen. Im typischen Falle wird der Substituent am Kohlenstoffatom 4 bei dieser Behandlung nicht verändert;
wenn jedoch die Verbindung der Formel A, worin Yf Hydroxyl
darstellt, als Ausgangsmaterial verwendet wird, ergibt sieh ein Produkt der Formel B, das ein Gemisch aus

herkömmlicher Weise isoliert werden.
Die Verbindungen der Formel B, worin Y" Hydroxyl darstellt, können bequemer hergestellt werden, wenn man
eine labile Acyloxyschutzgruppe verwendet, d.h. Yf Acyloxy
darstellt, und dann das resultierende Produkt der Formel B durch herkömmliche Basenhydrolyse spaltet. Es
wurde ferner gefunden, dass das Zwischenprodukt der Formel B, worin Y" Allyloxy darstellt, bequemer aus
5,8-Dihydro-l- (2,3-epoxy-l-propoxy)~4-hydroxy«5,8-methanonaphthalin,
d.h. der Verbindung der Formel B, worin Y" Hydroxyl darstellt, als direkt aus einer Verbindung der

Formel B, worin Y! die entsprechende Bedeutung hat, hergestellt
werden kann. Dies kann zweckmässig ausgeführt werden, indem man die Verbindung der Formel B, worin Y"
-OH darstellt, in einem geeigneten inerten organischen Reaktionsmedium, z.B. Dimethylformamid, Dirnethylsulfoxyd
und dergleichen, mit Allylbromid behandelt. Im typischen Falle erfolgt diese Behandlung bei Temperaturen im Bereich
von ca. 18 bis 35 0G ca. 30 Minuten bis 20 Stunden
lang unter Verwendung von ca. 5 bis 10 Mol Allylbromid pro Mol der Verbindung der Formel B.
Die Verbindungen gemäss der Erfindung können über Stufe (2) hergestellt werden, indem man das entsprechende
Zwischenprodukt der Formel B mit einem primären Amih mit dem gewünschten Substituenten R behandelt.
Diese Behandlung erfolgt im typischen Falle von Temperaturen im Bereich von ca. 25 bis 100 0C ca. 1 bis 16 Stunden
lang unter Verwendung von ca. 5 bis 10 Mol primärem Amin pro Mol der Verbindung der Formel B. Jedoch können
auch Temperaturen, Molverhältnisse und Behandlungsdauern sowohl über als auch unter diesen Werten angewandt werden.
Vorzugsweise erfolgt die Behandlung in einem inerten organischen Lösungsmittel, wie beispielsweise Methanol,
■Aethanol, Benzol, Toluol und dergleichen. Wenn ein inertes
Lösungsmittel verwendet wird, erfolgt die Behandlung zweckmässig bei etwa der Rückflusstemperatur des Lösungs-

mittels. Geeignete verwendbare primäre Amine sind z.B. Methylamin, Aethylamin, Isopropylamin, tert.-Butylamin,
Hexylamin, 3i^-Dimethoxyphenyläthylamin, ß-(4-Chlorphenyl)-a-methyläthylamin
und dergleichen. Wenn ein flüchtiges Amin, wie Methylamin, verwendet wird, wird
die Behandlung vorzugsweise auch in einem geschlossenen System ausgeführt.
Im typischen Falle wird der Substituent am Kohlenstoffatom 4 bei dieser Behandlung nicht verändert;
wenn aber 1,4-Di- (2,3-epoxy-l-propoxy)-5,8-dihydro-5*8-methanonaphthaiin
als Ausgangsmaterial verwendet wird, besteht das erhaltene Produkt aus dem entsprechenden
1,4-Di- (3-amino-2-hydroxy-l-propoxy)-5,8-dihydro-5,8-methanonaphthaiin
mit substituierter Aminogruppe. Ferner werden wegen der alkalischen Bedingungen, die naturgemäss
durch das primäre Amin erzeugt werden, alle basenlabilen Gruppen gleichzeitig durch diese Behandlung abgespalten.
Wenn somit eine Verbindung der Formel B, worin Y" Acyloxy darstellt, für diese Behandlung verwendet wird, wird
ein Produkt der Formel I erhalten, worin Y Hydroxyl darstellt. Die Verbindungen der Formel I, worin Y Acyloxy
bedeutet, können ihrerseits zweckmässig nach dem folgenden schematisch wiedergegebenen Verfahren hergestellt

Die Stufe 1 dieses Verfahrens kann zweckmässig ausgeführt werden, indem man die Verbindung der Formel Ia
in Gegenwart eines niederen Alkanols, z.B. Aethanol, als Lösungsmittel mit einer wässrigen Formaldehydlösung behandelt.
Im typischen Falle erfolgt diese Behandlung bei ungefähr der Rückflusstemperatur ca. 2 bis 3 Stunden lang
unter Anwendung eines stöchiometrisehen Ueberschusses an
Formaldehyd.

• Stufe 2 dieses Verfahrens wird im typischen Falle in zwei Phasen ausgeführt. In der ersten Phase wird die
Verbindung der Formel C mit einer Suspension von Natriumhydrid in Aethyläther (oder dergleichen) behandelt. Im typischen
Falle erfolgt diese Phase bei Temperaturen im Bereich von 15 bis 25 0C, zweckmässig bei Raumtemperatur, Vorzugsweise
wird diese Phase unter Rühren ausgeführt. Die zweite Phase dieser Stufe wird im typischen Falle ausgeführt, indem
man das gewünschte Acylanhydrid zu dem erhaltenen Reaktionsgemisch zugibt und die Behandlung weitere 10 bis
20 Stunden lang fortsetzt. Vorzugsweise wird die gesamte Behandlung unter einer inerten Atmosphäre,· z.B. Stickstoff,
ausgeführt. Geeignete verwendbare Acylanhydride sind .z.B.
Essigsäureanhydrid, Propionsäureanhydrid, BenzoeSäureanhydrid
und dergleichen.
Der Oxazolidinring in der Verbindung der Formel D ist sehr labil und kann im typischen Falle durch Behandlung
mit Wasser und auf jeden Fall durch Behandlung mit einer milden Säure gespalten werden (Stufe 3).
Im typischen Falle werden die Produkte aller oben und in folgenden beschriebenen Verfahrensstufen vor der an-
schliessenden Weiterverwendung abgetrennt und gereinigt.
Derartige Abtrennungen und Reinigungen können mittels beliebiger geeigneter Methoden, beispielsweise durch Filtration,
Extraktion, Eindampfen, Kristallisation, Dünnschicht-

Chromatographie, Säulenchromatographie usw., ausgeführt werden, Beispiele von typischen Trennungs- und Reinigungsverfahren
finden sich in den entsprechenden Beispielen. Wenn man die reinen optischen Isomeren aus racemischen
Gemischen abtrennen will, können auch herkömmliche Aufspaltungsverfahren angewandt und durch Routineversuche
optimiert werden.
Die pharmazeutisch unbedenklichen Säureadditionssalze der Verbindungen gemäss der Erfindung können durch
Neutralisieren der eine Aminogruppe aufweisenden Stammverbindung mit der gewünschten Säure hergestellt werden.
Andere pharmazeutisch unbedenkliche Säureadditionssalze können dann zweckmässig durch Anionenaustausch mit einem
geeigneten Ionenaustauscherharz in der gewünschten Anionenform aus den durch Neutralisation erhaltenen Additionssalzen hergestellt werden. Entsprechend können die Salze
der 4-Carboxy!verbindungen durch Neutralisation mit der
gewünschten Base hergestellt werden.
Die Ausgangsmaterialien der Formel A, worin Y' •Acetoxy bedeutet, können zweckmässig durch selektive
Hydrolyse eines der Acetoxysubstituenten von 1,4-Dlacetoxy-5,8-dihydro-5,8-methanonaphthalin
hergestellt werden [siehe Meinwald et al., J. Am. Chem. Soc, Bd. 80, S. 3667 (1958)].

Dies kann zweckmässig durch ca. 8- bis 12-stündige Behandlung
mit Natriummethylat in einem inerten organischen Losungsmittel,
z.B. Aethylenglycoldimethylather oder Dimethylformamid,

Entsprechend kann das Ausgangsmaterial der Formel A, worin Y* Hydroxyl bedeutet,' nach dem gleichen Verfahren
erhalten werden, wobei man aber 2 bis 2,4 Mol Natriummethylat pro Mol !,^-Diacetoxy-SiS-dihydro-SiS-methanonaphthalin
verwendet, oder es kann auch nach den in der obigen Veröffentlichung in J. Am. Chem. Soc. beschriebenen
Verfahren erhalten werden.
Die Ausgangsmaterialien der Formel A, worin Y Alkoxy bedeutet, können hergestellt werden, indem man
5,8-Dihydro-l,4-dioxo-5,8-methanonaphthalin [siehe Albrecht,
"Ann., Bd. 348, S. 34 (1906) und Diels et al., Ber., Bd. 62,
S. 2337 (1929)] mit einem Tr!alkoxyphosphin mit der gewünschten
Alkoxygruppe in einem inerten organischen Reaktionsmedium,
z.B. Benzol, Toluol oder Xylol, umsetzt, wobei ein entsprechendes l-Alkoxy-4-(di-O-methylphosphono)-5,8-dihydro-5,8-metnanonaphthalin
erhalten wird. Diese Reaktion kann bei Temperaturen im Bereich von ca. 18 bis
25 0C im Verlauf von 30 Minuten bis 3 Stunden ausgeführt
werden. Die Phosphonogruppe kann dann durch herkömmliche

Basenhydrolyse gespalten werden. Beispielsweise kann die Hydrolyse durch Behandlung in einem niederen Alkanol,
z.B. Methanol, als Lösungsmittel mit wässrigem Natriumhydroxyd im Verlauf von ca. 2 bis 4 Stunden bei etwa der
Rückflusstemperatur erfolgen.
Die Verbindungen der Formel A, worin Yf Wasserstoff
bedeutet, können hergestellt werden, indem man ein l-Alkoxy-4- (di-0-alkylphosphono)-5,8-dihydro-5i8-methanonaphthalin
in flüssigem Ammoniak mit Lithium behandelt, wobei man ein entsprechendes l-Alkoxy-5,8-dihydro-5,8-methanonaphthalin
erhält. Im typischen Falle wird das Ammoniak unter Rühren ca. 3 bis 15 Minuten lang in das
l-Alkoxy-4»dialkylphosphato-5, 8-dihydr o-5,8- methanonaphthalinmaterial
eingeleitet, während es auf einer Temperatur von ca. -78 bis -30 0C gehalten wird. Dann
werden zu dem Reaktionsgemisch 2 Mol festes Lithium pro Mol 5»8-Methanonaphthalinderivat gegeben, worauf man die
Temperatur des Reaktionsgemisches ohne Regelung von aussen steigen lässt. Das l-Alkoxy-5,8-dihydro-5i8-methanonaphthalin,
das gebildet wird, kann durch Behandlung mit Natriumäthylmercaptid in einem inerten organischen Lösungsmittel,
wie beispielsweise Dimethylformamid, Dimethylsulfoxyd und Hexamethylphosphorsäureamid, bei einer Temperatur
im Bereich von ca. 8o bis 120 0C im Verlauf von ca. 12 bis 20 Stunden unter Verwendung von ca. 10 Mol

des Mercaptides pro Mol des 5#8-Methanonaphthalinderivates
zum entsprechenden 1-Hydroxyderivat entalkyliert werden.
Die Verbindungen der Formel A, worin Y Wasserstoff darstellt, können auch hergestellt werden, indem
man !,^-Diacetoxy-S^S-dihydro-SjS-methanonaphthalin in
einem inerten organischen Lösungsmittel, wie Aethylenglycoldimethyläther,
bei Temperaturen im Bereich von ca. 18 bis ca. 25 0C ca. 8 bis ca. 16 Stunden lang unter
Verwendung von ca. 1 Mol Natriummethylat pro Mol der
1,4-Diacetoxyverbindung mit Natriummethylat behandelt und dann das Produkt dieser Reaktion bei Temperaturen
im Bereich von ca. l8 bis ca. 25 0G ca. 30 Minuten bis
ca. 3 Stunden lang unter Verwendung von ca. 1,2 Mol der Aetherverbindung pro zu Anfang vorhandenes Mol der Diacetoxyverbindung
mit Chlormethylmethyläther behandelt, wobei 4-Acetoxy-5,8-dihydro-l-methoxymethylenoxy-5,8-methanonaphthalin
gebildet wird. Dieses Produkt kann dann unter den oben angegebenen Bedingungen mit Natriummethylat
umgesetzt werden, wobei aber anstelle von Aethylenglycoldimethyläther Aethyläther als Lösungsmittel
verwendet wird, worauf das Produkt dieser Reaktion seinerseits bei Temperaturen im Bereich von ca.
bis 25 0C ca. 4 bis 8 Stunden lang mit Chlorphosphonsäurediäthylester
und einem Ueberschuss an Triäthylamin umgesetzt wird, wobei man ein 4-Dialkylphosphato-5,8-dihydro-

A-methoxymethoxy-SiB-methanonaphthalin erhält. Dieses
Produkt kann dann nacheinander mit Lithium in flüssigem Ammoniak (unter den oben angegebenen Bedingungen) und
mit Salzsäure in Methanol bei etwa der Rückflusstemperatur des Methanols ca. 30 Minuten bis ca. 1 Stunde lang
behandelt werden, wobei man 5*8-Dihydro-I-hydroxy-5*8-methanonaphthalin
erhält.
Die Verbindungen der Formel A, worin Y eine Acylaminogruppe darstellt, können hergestellt werden,
indem man 5>8-Dihydro-l,4-dioxo-5i8-methanonaphthalin
in einem inerten organischen Lösungsmittel, wie beispielsweise Pyridin, Triäthylamin und dergleichen, mit einem
Alkoxylaminsalz, z.B. Methoxylaminhydrochlorid, umsetzt.
Im typischen Falle erfolgt diese Umsetzung bei Temperaturen im Bereich von ca. 18 bis 25 0C ca. 1 bis 3 Stunden
lang unter Verwendung von ca. 1 Mol Alkoxylamin . pro Mol Verbindung der Formel A. Bei dieser Reaktion
erhält man das entsprechende ^Alkoximino-SiS-dihydrol-oxo-5*8-methanonaphthalinderivat,
das dann durch Behandlung mit dem gewünschten Acylanhydrid in einem inerten organischen Medium, z.B. Diäthyläther, in Gegenwart
von metallischem Zink bequem zu dem entsprechenden 4-Acylamino-l-acyloxy-5,8-dihydro-5*8-methanonaphthalin
reduziert werden kann. Diese Reaktion erfolgt im typischen Falle bei Temperaturen im Bereich von ca. -20 bis 0 0C ca. 1 bis

Stunden lang unter Verwendung eines grossen Ueberschusses an Zink. Geeignete verwendbare Acylanhydride sind z.B.
Essigsäureanhydrid, Propionsäureanhydrid, Benzoesäureanhydrid
und dergleichen. Die 1-Aoylgruppe kann dann durch
herkömmliche Basenhydrolyse, z.B. durch Behandlung mit wässrigem Natriumhydroxyd bei -20 bis 0 0C, im Verlauf
von ca. 15 Minuten bis 1 Stunde abgespalten werden, wobei man das entsprechende ^-Acylamino-5»8- dihydro-1-nydroxy-5,&-metnanonaphthalinderivat
erhält.
Die obigen Ausführungen beschreiben die Herstellung der wichtigsten anfänglichen Ausgangsm&terlalien.
Andere Ausgangsmaterialen können nach bekannten Verfahren sowie nach den in den folgenden Beispielen dargelegten
Verfahren oder naheliegenden Abwandlungen dieser Verfahren,
z.B. durch Verwendung von Homologen usw., hergestellt werden.
. Die Verbindungen gemäss der Erfindung sind verbesserte
kardiovaskuläre Mittel und eignen sich demzufolge für die Behandlung und Linderung von kardiovaskulären Abnormalitäten
bei Menschen und Säugetieren. Diese Verbindungen sind besonders brauchbar für die Behandlung und
Linderung von Herzarrhythmien, hypertropischer Subaortenstenose, Hypertonie, Schilddrüsenüberfunktion, hyperhormonalem
Nebennierenmark-Syndrom, hyperkinetischen Syndromen
und allgemeinen ischämischen Zuständen, z.B. Angina pectoris.

Die Verbindungen gemäss der Erfindung können
in den verschiedensten Darreichungsformen entweder allein oder in Kombination mit anderen pharmazeutisch verträglichen
Medikamenten in Form von für die orale oder parenterale Verabreichung geeigneten pharmazeutischen Präparaten
verabreicht werden. Die Verbindungen werden im typischen Falle verabreicht als pharmazeutische Präparate, die als
wesentliche Bestandteile die pharmazeutisch unbedenklichen Salze der oben beschriebenen Verbindungen sowie
einen pharmazeutischen Träger enthalten. Der pharmazeutische Träger kann entweder ein festes Material oder
eine Flüssigkeit, in der die Verbindung gelöst, dispergiert oder suspendiert vorliegt, sein und gegebenenfalls
kleine Mengen Konservierungsmittel und/oder pH-Puffermittel enthalten. Geeignete verwendbare Konservierungsmittel
sind z.B. Benzylalkohol und dergleichen. "Geeignete Pufferungsmittel sind z.B. Natriumacetat und
pharmazeutische Phosphorsäuresalze und dergleichen."
Die flüssigen Präparate können beispielsweise in Form von Lösungen, Emulsionen, Suspensionen, Sirupen
oder Exilieren vorliegen. Eine bevorzugte Verabreichungsart und ein bevorzugtes Mittel ist die intravenöse Verabreichung
einer einfachen Lösung der pharmazeutisch unbedenklichen Additionssalze der oben beschriebenen Verbindungen
in sterilem Wasser, die gegebenenfalls kleine

Mengen Konservierungsmittel und/oder Puffermittel enthält.
Die festen Präparate können in Form von Tabletten, Pulvern, Kapseln, Pillen oder dergleichen vorliegen,
vorzugsweise in Form von Dosierungseinheiten für die einfache Verabreichung oder genauen Dosierungen, vorliegen.
Geeignete feste Träger sind z.B. pharmazeutische Sorten von Stärke, Lactose, Natriumsaccharin, Natriumbisulfit
und dergleichen.
Die Verbindungen gemäss der Erfindung werden im
typischen Falle in Dosierungen von ca. 0,01 bis 5 rag pro
kg Körpergewicht verabreicht. Die genaue wirksame Dosierung hängt natürlich von der Art der Verabreichung, dem
behandelten Zustand und dem Patienten ab.
Die folgenden Beispiele erläutern die Erfindung. Alle Temperaturen und Temperaturbereiche sind in 0C angegeben,
wenn nicht ausdrücklich etwas anderes erwähnt ist, und die Begriffe "Umgebungstemperatur" oder "Raumtemperatur"
beziehen sich auf ca. 20 0C. Die Prozente sind Gewichtsprozente.
Wenn nicht ausdrücklich etwas anderes ausgesagt wird, werden raeemische oder diastereomere Gemische als
Ausgangsmaterialien verwendet und entsprechend raeemische oder diastereomere Gemische als Produkte erhalten; erforderlichenfalls
werden Beispiele wiederholt, um genügend grosse Mengen Ausgangsmaterialien für folgende Beispiele

herzustellen. Wenn ferner von sauren Lösungen usw. die Rede ist, ohne dass ein Lösungsmittel erwähnt ist, handelt
es sich um wässrige Lösungen.
Eine Lösung von 615 mg 5,8-Dihydro-l,4-dihydroxy-5,8-methanonaphthalin
in 20 ml Dimethylsulfoxyd wird mit 396 mg Kalium-tert.-butylat versetzt. Das Gemisch wird
bei Raumtemperatur gerührt, bis es homogen ist, und-dann
im Verlauf von 2 Stunden tropfenweise zu einer Lösung von 7*0 g Epibromhydrin in 20 ml Dimethylsulfoxyd, die
auf 70 0G gehalten wird, zugesetzt. Nach 90 Minuten wird
das Reaktionsgemisch in 5OO ml eines Misehlösungsmittels
aus Hexan und Aether (2:1) gegossen und mit Wasser gewaschen. Die organische Lösung wird über Magnesiumsulfat
getrocknet und zur Trockene eingedampft, wobei ein öliges Material erhalten wird. Dieses ölige Material wird der
Säulenchromatographie (Kieselgel, Gemische aus Hexan und Aceton) unterworfen, wobei 5*8-Dihydro-1-(2,3-epoxy-lpropoxy)-4-hydroxy-5#8-methanonaphthalin
und 1,4-Di-(2,3-epoxy-1-propoxy)-5»8-dihyäro-5*8-methanonaphthalin
als getrennte Produkte erhalten werden.
Eine Lösung von 202 mg des 5*8-Dihydro-1-( 2,3-epoxy-1-propoxy)-4-hydroxy-5,8-methanonaphthalins
von

Beispiel 1 in 20 ml Methanol wird mit 20 ml tert.-Butylamin
versetzt. Diese Lösung wird bei Raumtemperatur 24 Stunden lang unter einer Stickstoffatmosphäre'gerührt.
Das Lösungsmittel sowie das überschüssige Amin werden im Vakuum entfernt. Das als Rückstand zurückbleibende
gebildete 1-(3-tert.-Butylamino-2-hydroxy-1-propoxy)-5,8-dihydro-4-hydroxy-5,8-methanonaphthalin
wird in einem Gemisch aus Diäthyläther und Methylenchlorid gelöst und mit Salzsäure behandelt, wobei man das Hydrochlorid
als Niederschlag erhält. Der Niederschlag wird abfiltriert, worauf Spuren Lösungsmittel verdampft werden.
Wenn man das gleiche Verfahren befolgt, aber Isopropylamin, Hexylamin, 3,4-Dimethoxyphenyläthylamin,
ß-(4-Chlorphenyl)-a-methyläthylamin bzw. Propargylamin
anstelle von tert.-Butylamin verwendet, erhält man in
ähnlicher Weise 1-(3-Isopropylamino-2-hydroxy-1-propoxy )-5,8-dihydro-4-hydroxy-5,8-methanonaphthalin,
1- (3-Hexylamino-2-hydroxy-1-propoxy)-5,8-dihydro-4-hydroxy-5,8-methanonaphthalin,
1- [3- (β—.3' ,^'-Dimethoxyphenyläthylamlno)-2-hydroxy-1-propoxy]-5,8-dihydro-4-hydroxy-5,8-methanonaphthalin,
1-[3-(ß-4l-chlorphenyl-a-methyläthylamino)-2-hydroxy-1-propoxy]-5,8-dihydro-5,8-methanonaphthalin

Hydrochloride. Wenn das Amin nicht flüchtig ist, wird das Produkt durch präparative DünnschichtChromatographie
über Kieselgel unter Verwendung eines Gemisches von Methanol, Chloroform und Formaldehyd als Eluierungsmittel
isoliert.
In diesem Beispiel werden 400 mg 5,8-Dihydro-1-
(2,3-epoxy-1-propöxy )-4-hydroxy-5j8-methanonaphthalin
zu 5 ml mit Methylamin gesättigtem Aethanol gegeben, worauf
man das Gemisch 20 Stunden lang bei Raumtemperatur stehen lässt. Das Lösungsmittel Aethanol wird dann durch
Verdampfen entfernt und der resultierende Rückstand durch Dünrischichtchromatographie über Kieselgel unter
Eluieren mit einem Gemisch aus Chloroform, Methanol und Formaldehyd gereinigt, wobei man l-(3-Methylamino-2-hydroxy-1-pr
opoxy)- 5 , 8- dihydro- 4- hydroxy- 5,8- me thanonaphthalin
erhält. Das Hydrochlorid wird dann in der gleichen Weise wie in Beispiel 2 beschrieben hergestellt.
Die Verfahrensweisen der Beispiele 2 und 2A werden wiederholt, wobei man aber das 1,4-Di-(2,3-epoxyl-propoxy)-5,8-dihydro~5,8-methanonaphthalin

von Beispiel 1 in 1 ml Dimethylformamid werden bei Raumtemperatur tropfenweise und unter einer Stickstoffatmosphäre zu einer Suspension
von 24 mg Natriumhydrid in 5 ml Dimethylformamid und 2 ml Allylbromid zugesetzt. Das Reaktionsgemisch wird 2 Stunden
lang kräftig gerührt und dann in Wasser gegossen. Das resultierende Produkt wird mit Aether extrahiert und durch

Ein Kolben wird mit einer Lösung von 8,14 g 5,8-Dihydro-4-(di-0-methylphosphono)-l-methoxy-5,8-methanonaphthalin
in 15 ml Aether beschickt, worauf 100 ml flüssiges Ammoniak zugegeben werden. Der Reaktionskolben
wird auf -78 0C abgekühlt und in kleinen Portionen mit 344 mg Lithiumdraht versetzt. Die Reaktion
ist praktisch augenblicklich zu Ende, worauf das flüssige Ammoniak verdampft wird. Wasser wird zugesetzt
und das Produkt mit Aether extrahiert. Die Aetherlösung wird mit Wasser gewaschen und mit Magnesiumsulfat getrocknet.
Das Aetherlösungsmittel wird im Vakuum entfernt und die zurückbleibende Flüssigkeit durch Destillation
(85 °C/0,5 mm) gereinigt, wobei man 5>8-Dihydro-lmethoxy-5,8-methanonaphthalin
erhält.
Eine Lösung von 172 mg 5,8-Dihydro-l-methoxy-5,8-methanonaphthalin
in 5 ml Dimethylformamid wird mit 500 mg Natriumäthylmercaptid versetzt. Die resultierende
Lösung wird in einer Stickstoffatmosphäre 24 Stunden lang auf 120 0C erhitzt. Das Gemisch wird in 25 ml Wasser gegossen,
mit 50$iger Natriumhydroxydlösung auf pH = 12 bis l4 gebracht und wiederholt mit Aether gewaschen.
Die basische Lösung wird mit Salzsäure angesäuert und dann mit Aether extrahiert. Die Aetherlösung wird mit

Eine gekühlte Lösung von 1,72 g 5,8-Dihydro-1-methoxy-5,8-methanonaphthalin
und 790 mg Acetylchlorid
in 10 ml Nitrobenzol wird unter Rühren in kleinen Portionen
mit 150 mg Aluminiumchlorid versetzt. Das Gemisch wird über Nacht bei Raumtemperatur gehalten, auf Eis gegossen
und mit Benzol extrahiert. Die Benzollösung wird mit Wasser gewaschen, mit Magnesiumsulfat getrocknet und im'
Vakuum eingedampft. Das Nitrobenzol wird unter vermindertem Druck entfernt und das Produkt durch Destillation

Nacht im Dunkeln bei 0 0C gehalten, mit Sulfaminsäure behandelt, mit Natriumbicarbonat neutralisiert und in
Eiswasser gegossen, wobei man 5,8-Dihydro-l-hydroxy-5,8-methanonaphthalin-4-diazoniumchlorid
als Niederschlag erhält, der dann abfiltriert wird. 2,0 g dieses -Niederschlages werden darauf in β ml 24$iger Bromwasserstoffsäure
gelöst und die resultierende Lösung zu einer siedenden Lösung von 790 mg Cuprobromid in 5 ml 48$iger

die auf 0 0C gehalten wird, zugegeben. Man lässt das Geraisch sich in 2 Stunden auf Raumtemperatur erwärmen
und isoliert das Produkt durch Extrahieren mit Diäthylather.
Die Diäthylatherlösung wird mit Wasser gewaschen,
mit Magnesiumsulfat getrocknet, zur Trockene eingedampft und das Produkt durch Säulenchromatographie
(Kieselgel, Gemische aus Hexan und Aceton) isoliert, wobei man 4-Cyano-5,8-dihydro-1-hydroxy-5,8-methanonaphthalin
erhält.

Diazomethan wird das Lösungsmittel entfernt, indem man einen Stickstoffstrom durch das Reaktionsgemisch perlen

wobei man 5,8-Dihydro-l-hydroxy-4-methoxycarbonyl-5,8-methanonaphthalin

Eine Lösung von 172 mg 5>8-Dihydro-l,4-dioxo-5/8-methanonaphthalin
in 5 ml Pyridin, die auf 0 0C gekühlt ist, wird mit einer Lösung von 85 mg Methoxylaminhydrochlorid
in 5 ml Pyridin versetzt. Die Reaktionslösung wird über Nacht auf 0 0C gehalten und dann in Wasser
gegossen. Das Produkt wird mit Aether extrahiert. Die AetherlÖsung wird mit Wasser, verdünnter (0,1-normaler)
Salzsäure und Kochsalzlösung gewaschen und dann über Magnesiumsulfat getrocknet. Das Lösungsmittel wird im
Vakuum entfernt, wobei man ein Produkt erhält, das durch Säulenchromatographie (Kieselgel, Gemische aus Hexan und
Aceton) weiter gereinigt wird und 5*8-Dihydro-4-methoxyimino-l-oxo-5,8-methanonaphthalin
ergibt.
Eine Lösung von 201 mg 5,8-Dihydro~4-methoxyimino-l-oxo-5,8-methanonaphthalin
in 5 ml Essigsäureanhydrid und 3 Tropfen Essigsäure, die auf Q 0C gekühlt
ist, wird in kleinen -Portionen mit 1 g Zinkstaub versetzt. Das Gemisch wird 2 Stunden lang bei 0 0C gerührt
und dann in Wasser gegossen. Das Produkt wird, mit Aether extrahiert und die AetherlÖsung mit Wasser
gewaschen, über Magnesiumsulfat getrocknet und zur

Trockene eingedampft. Der Rückstand wird durch Säulenchromatographie
(Kieselgel, Gemische aus Hexan und Aceton) gereinigt, wobei man 4-Acetamido-l-acetoxy-5,8-dihydro-5,8-methanonaphthalin
erhält.
Eine Lösung von 257 mg 4-Acetamido-l-acetoxy-5,8-dihydro-5,8-methanonaphthalin
in 10 ml Methanol wird mit 1,2 ml 1-normalem Natriumhydroxyd versetzt';
Nach einer Stunde wird die Lösung angesäuert und der grösste Teil des Methanols im Vakuum entfernt. Der Rückstand
wird zwischen Wasser und Aether verteilt und die wässrige Schicht mit zusätzlichem Aether extrahiert.
Die vereinigten Aetherextrakte werden mit Wasser gewaschen, mit Magnesiumsulfat getrocknet und zur Trockene
eingedampft. Der Rückstand wird durch Säulenchromatographie (Kieselgel, Gemische aus Hexan und Aceton)
gereinigt, wobei man ^Acetamido-l-hydroxy-5,8-dihydro-5,8-methanonaphthalin
erhält.

2 Stunden lang gerührt und dann in Wasser gegossen. Das wässrige Gemisch wird darauf mit
1-normalem wässrigem Natriumhydroxyd neutralisiert. Das organische Produkt wird aus dem Gemisch mit Di-
äthyläther extrahiert und der resultierende Aetherextrakt mit Wasser gewaschen, über Magnesiumsulfat getrocknet
und dann zur Trockene eingedampft. Der resul-

Eine Lösung von 173 mg 4-Amino-5,8-dihydro-1-hydroxy-5,8-methanonaphthalin
und 2 ml Ameisensäure in 20 ml Toluol wird unter einer Stickstoffatmosphäre 3
Stunden lang zum Rückfluss erhitzt. Dann werden die überschüssige Ameisensäure und das Lösungsmittel Toluol
durch Verdampfen im Vakuum entfernt, worauf.das als Rückstand zurückbleibende Produkt durch Säulenchromatographie
über Kieselgel unter Eluieren mit Gemischen aus Hexan und Aethylacetat weiter gereinigt wird, wobei
man 5,8-Dihydro-4-formamido-1-hydroxy-5,8-methanonaphthalin
erhält.

Eine Suspension von 24θ mg Natriumhydrid in 20 ml wasserfreiem Aethylenglycoldimethylather wird bei
Raumtemperatur unter kräftigem Rühren mit 1,73 S 4-Amino-5,8-dihydro-l-hydroxy-5,8-methanonaphthalin
versetzt. Das Gemisch wird bei Raumtemperatur unter einer Stickst off atmosphäre gerührt, bis keine weitere Gasentwicklung
beobachtet wird (ca. 2 Stunden lang) und dann mit 0,8l g gereinigtem Chlormethylmethyläther versetzt. Das
resultierende Gemisch wird 15 Stunden lang bei Raumtemperatur gerührt und dann in Wasser gegossen. Das wässrige
Gemisch wird mit Diäthyläther extrahiert und der re-

sultierende Diäthylätherextrakt mit Wasser gewaschen,
über Magnesiumsulfat getrocknet und dann zur Trockene eingedampft. Der Rückstand wird durch Säulenchromatographie
über Kieselgel unter Eluieren-mit Gemischen aus Hexan und Aethylacetat gereinigt, wobei man reinen
4-Amino-5 * 8-dihydro-5,8-methanonaphthalin-1-methoxymethyläther
erhält.
4 ml 42$ige wässrige Fluoroborsäure werden mit 4 ml Wasser'verdünnt und dann mit 217 mg 4-Amino-5,8-dihydro-5»8-methanonaphthalin-1-methoxymethyläther
versetzt, worauf das resultierende Gemisch auf 0 0C gekühlt
wird. Eine Lösung von 69 mg Natriumnitrit in 1 ml Wasser
wird dann langsam zugesetzt, wobei als Niederschlag 5,8-Dihydro-5,8-methanonaphthalin-1-methoxymethyläther-"diazoniumtetrafluoroborat
erhalten wird. Der Niederschlag wird abfiltriert und nacheinander mit 4 ml 5$iger wässriger Pluoroborsäure, 5 ml Methanol von
0 0C und Diäthylather gewaschen. Das Produkt ist so rein,
dass es ohne weitere Reinigung für die folgende Reaktion verwendet werden kann..
151 mg SiS-Dihydro-Sie-methanonaphthalin-lmethoxymethyläther-4-diazoniumtetrafluoroborat

Eine Lösung von 70 mg Natriummercaptid in 5 ml Dimethylformamid wird mit 322 mg des nach Beispiel
27 hergestellten 5*8-Dihydro-5>8-methanonaphthalin-lmethoxymethyläther-4-diazoniumtetrafluoroborats
versetzt. Das resultierende Gemisch wird erhitzt, bis keine Gasentwicklung mehr beobachtet wird, und dann in Wasser
gegossen. Die organische Phase wird mit Diäthyläther extrahiert und der resultierende Diäthylätherextrakt mit
Wasser gewaschen, über Magnesiumsulfat getrocknet und zur Trockene eingedampft. Der resultierende Rückstand
wird durch Säulenchromatographie über Kieselgel unter

Stunden lang bei 0 0C gerührt und dann in Eiswasser gegossen
und mit Diäthyläther extrahiert. Der Diäthylatherextrakt
wird nacheinander mit wässriger Natriumbicarbonatlosung und Wasser gewaschen, dann über Magnesiumsulfat getrocknet
und zur Trockene eingedampft. Der resultierende Rückstand wird durch Säulenchromatographie über Kieselgel
unter Eluieren mit Gemischen aus Hexan und Aceton gereinigt, wobei man 5i8-Dihydro-4-methylsulfinyl-5,8-methanonaphthalin-1-methoxymethylather
erhält.

1-(3-Hexylamino-2-hydroxy-1-propoxy )-5,8-dihydro-4-fluor-5i8-methanonaphthalin,

1- [3- Cß-3' ,4'-Dimethoxyphenyläthylamino)-2-hydroxy-1-propoxy]-5,8-dihydro-4-fluor-5,8-methanonaphthalin,

und die entsprechenden Hydrochloride erhält.
Eine Lösung von Il4 mg 1-(3-tert.-Butylamino-2-hydroxy-1-propoxy)-5,8-dihydro-4-hydroxy-5*8-methanonäphthalin
und 60 mg 4o$igem wässrigem Formaldehyd in 40 ml Aethanol wird 2,5 Stunden lang zum Rückfluss erhitzt.
Die flüchtigen Flüssigkeiten werden dann durch Eindampfen im Vakuum entfernt, worauf der Rückstand durch
präparative Dünnschichtchromatographie über Kieselgel unter Eluieren mit Gemischen aus Aceton, Hexan und
Formaldehyd gereinigt wird, wobei man 5*8-Dihydro-4-hydroxy-5*8-methanonaphthaiin-1-(N-tert.-butyl-5-oxazolidinyl)-methylather
erhält.
Eine Suspension von 26 mg Natriumhydrid in 20 ml Diäthyläther wird unter kräftigem Rühren bei
Raumtemperatur zu 301 mg 5,8-Dihydro-4-hydroxy-5,8-

methanonaphthalin-l-(N-tert .-butyl-5-oxazolidinyl )-methyläther
gegeben. Das resultierende Gemisch wird, unter einer Stickstoffatmosphäre gerührt, bis'keine
Gasentwicklung mehr beobachtet wird, worauf 102 mg Essigsäureanhydrid zugesetzt werden und das Gemisch
15 Stunden lang bei Raumtemperatur gerührt wird. Das Gemisch wird dann in V/asser gegossen und mit Chloroform
extrahiert. Der Chloroformextrakt wird mit Wasser gewaschen, über Magnesiumsulfat getrocknet und zur
Trockene eingedampft. Der resultierende Rückstand wird durch präparative Dünnschichtchromatographie über
Kieselgel unter Eluieren mit Gemischen aus Chloroform und Methanol gereinigt, wobei man 1-(3-tert.-Butylamino-2-hydroxy-1-propoxy)-4-acetoxy-5,8-dihydro-5,8-methanonaphthalin
erhält.
In ähnlicher Weise wird 1-(3-tert.-Butylamino-2-hydroxy-1-propoxy)-4-benzoyloxy-5*8-dihydro-5,8-methanonaphthalin
erhalten, wenn man die obige Verfahrensweise befolgt, aber Benzoesäureanhydrid anstelle
von Essigsäureanhydrid verwendet.
In ähnlicher Weise werden die entsprechenden 4-Acetoxy- und 4-Benzoyloxyderivate der Produkte der
Formel I aus den Beispielen 2 und 2A hergestellt.

- 3. Verbindung nach Anspruch 1, dadurch gekennzeichnet, dass Υ Wasserstoff, Hydroxyl, Methyl oder Halogen bedeutet.
- 4. Verbindung nach Anspruch 3, dadurch gekennzeichnet, dass Y Wasserstoff bedeutet.
- 5. Verbindung nach Anspruch 3, dadurch gekennzeichnet, dass R Isopropyl, tert.-Butyl, β-(3,4-Dimethoxyphenyl) <-äthyl oder Propargyl bedeutet.
- 6. Verbindung nach Anspruch 5> dadurch gekennzeichnet, dass es sich um 1- (3-Isopropylamino-2-hy droxy-1- pr opoxy)- 5,8- dihydro- 5*8- methanonaphthalin oder ein pharmazeutisch unbedenkliches Salz davon handelt.
- -7· Verbindung nach Anspruch 5, dadurch gekennzeichnet, dass es sich um 1-(3-tert.-Butylamino-2- hydroxy-1- pr opoxy)- 5,8- dihy dr 0-5,8- methanonaphtha lin oder ein pharmazeutisch unbedenkliches Salz davon handelt. - ;
- 8. Verbindung nach Anspruch 5* dadurch gekennzeichnet, dass es sich um 1-[3-(ß-3',4'-Dimethoxyphenyläthylamino)-2-hydroxy-1-propoxy]-5,8-dihydro-5,8-methanonaphthalin oder ein pharmazeutisch unbedenkliches Salz davon handelt.
- 9. Verbindung nach Anspruch 5, dadurch gekennzeichnet, dass es sich um 1-(3-Prοpargylamino-2»408825/1149hydroxy-1- propoxy)- 5,8- dihydro- 5 j 8-methanonaphthalin oder ein pharmazeutisch unbedenkliches Salz davon handelt.
- 10. Verfahren zur Herstellung der Verbindungen nach Anspruch 1, dadurch gekennzeichnet, dass man eine Verbindung der Formel:(B)worin Y" aus der für Y in Anspruch 1 definierten Gruppe von Substituenten sowie 2,3-Epoxy-l-propoxy gewählt ist, mit einem primären Amin der Formel HpNR, worin R wie-ϊη Anspruch 1 definiert ist, in einem inerten organischen Lösungsmittel unter zur Reaktion führenden Bedingungen behandelt, wodurch die entsprechende, durch R substituierte Verbindung nach Anspruch 1 gebildet wird.
- 11. Verfahren nach Anspruch 10, dadurch gekennzeichnet, dass man die Behandlung in einem inerten organischen Lösungsmittel bei Temperaturen im Bereich von ca. 25 bis 100 0C ca. 1 bis 16 Stunden lang unter409825/1U9Verwendung von ca. 5 Ms 10 Mol des primären Amins pro Mol der Verbindung der Formel B ausführt.
- 12. Heilmittel, enthaltend eine Verbindung gemäß Anspruch 1 - 9.INSPECTED409825/1 U9
