Die vorliegende Erfindung betrifft neue Phloroglucinderivate, die eine j5-Hydroxy-butyr ami din gruppe aufweisen, die durch
eine Ä'therfunktion in 4-Stelltmg mit der Phenylgruppe verbunden
ist. Die Erfindung betrifft auch ein Verfahren zur Herstellung der genannten Derivate und ihx'e therapeutische Anwendung.
Die neuen erfindungsgemässen Verbindungen werden aus einer Gruppe die

1. durch Verbindungen der allgemeinen Formel

in der R ein Wasserstoffatom und eine niedrige Alkylgruppe
mit 1 bis 5 C-Atomen und X ein Wasserstoffatom oder eine
Hydroxylgruppe darstellen,und
2. ihren Additionssalzen mit Säuren gebildet ist, ausgewählt.
Unter Additionssalzen mit Säuren versteht man Substanzen, die durch Reaktion der Verbindungen der Formel I mit Mineralsäuren
oder organischen Säuren erhalten werden. Unter den geeigneten Mineralsäuren kann man jene erwähnen, die nicht-giftige Additionssalze
bilden, insbesondere die Chlorwasserstoff--, Bromwasserstoff-·, Schwefel- und Phosphorsäure. An organischen
Säuren, die nichttoxische Additionssalze bilden, können die Oxalsäure, Fumarsäure, Maleinsäure, Glutaminsäure, Cyclohexylsulfaminsäure,
Asparaginsäure, Äpfelsäure, Benzolsäure,und
Ascorbinsäure erwähnt werden.
Zur Herstellung einer Verbindung geinäss Formel I, in der
X=H bedeutetj geht man nach einem Verfahren vor, das dadurch
gekennzeichnet ist, dass man
a) eine stochiometrische Menge Glyzerin-epichlorhydrin mit
einem j5.5-Dialkoxy-phenol (il)

in der R wie vorstehend definiert ist, umsetzt,
b) das derart erhaltene Epoxydderivät in ätherischem Medium mit einem HCl Gasstrom behandelt,
c) das somit erhaltene 1-Chlor 2-Hydroxy-propylderivat, das
durch nachstehende Formel wiedergegeben ist,
O - CH - CH - CH2Cl OH
mit einem Alkalimetallcyanid, insbesondere Kaliumcyanid, *
behandelt,
d) das derart erhaltene Nitril in ätherischem Medium mit
Äthanol behände! t und
e) den so gebildeten Imino-äthyl-äther einem gasförmigen
Ammoniakstrom aussetzt.
Um ein Derivat der Formel I herzustellen, in der X = OK ist,
lässt man nach einem an sich bekannten Verfahren Hydroxylamin auf ein 4-(3.5-DIaIkOXy-phenoxy)-3~IIydroxy~butyramidin, das
in Stufe e) erhalten ist, einwirken.
Vorzugsweise wird die Reaktion zur Gewinnung eines Amidoximdei'ivates
(X = OH) bei einer Temperatur zwischen 10 und 30 C,
vorzugsweise zwischen 15 und 25°C dadurch durchgeführt, dass man während ungefähr 24 Stunden 4.5 Mol 4-(3·5-Dialkoxy-phenoxy)
-3-Hydroxybutyramidin-hydrochlorid mit dem Reaktionsprodukt von
6 Mol eines Alkalimetallalkoliolates mit 6 Mol Hydroxylamin-hy-

drochlorid in Kontakt bringt.
Die Alkalimetallalkoholate die mit Vorteil verwendet werden,
sind insbesondere Natriummethylat und Natriumäthylat.
Die therapeutischen erfindungsgemässen Kompositionen werden
durch einen physiologisch geeigneten Träger zusammen mit zumindest einer Verbindung der Formel I oder einer ihrer Additionssalze
mit nichttoxischcn Säuren gebildet.

Zu einer Lösung von 12.3 g (0.08 Mol) 3.5-Dimethoxy-phenol
und 5.2 g (O.o8 Mol) Soda in 25 ml Wasser fügt man 7.4 g
(0.08 Mol) Glyzerin-epichlorhydrin und 25 ml Methanol hinzu und erhitzt 5 Minuten zum Sieden. Nach Vertreibung des Methanols
und Abkühlung extrahiert man das gebildete 3-(3·5-Dimethoxy-phenoxy)-1.2-epoxypropan
mit Kthyläther. Die ätherische Lösung, die mit Wasser gewaschen und anschliessend

mit wasserfreiem Natriumsulfat getrocknet wurde, wird mit·
einem trockenen gasförmigen Chlorwasserstoffstrom gesättigt.
Nach 4-tägigem Stehen wäscht man sie mit Wasser und anschliessend
mit einer verdünnten Natriumcarbonatlösung in Wasser.
Nach Trocknung mit Natriumsulfat und Vertreibung des Äthyläthers erhält man 17·5 g rohes 3-(3.5-Dimethoxy-phenoxy)
l-Chlor-2-propanol.
Diesem in 80 ml Äthanol gelösten Produkt fügt man eine Lösung
von 5»25 g Kaliumcyanid in 20 ml Wasser hinzu. Das Gemisch
wird bis zum Sieden erhitzt und sodann bei Rückflusstemperatur
des Lösungsmittels während einer Stunde gehalten. Nach Vertreibung des Lösungsmittels im Vakuum und erfolgter
Abkühlung fügt man Äthyläther hinzu. Die ätherische Lösung ergibt nach erfolgter Nachwaschung mit Wasser, Trocknung
mit wasserfreiem Natriumsulfat, anschliessendem Verdampfen 16.4 g rohes 4-(3.5"Dlmethoxy--phenoxy)—3-hydroxy-butyronitril.
Die Lösung dieses Produktes (0.07 Mol) in .16 ml Äthanol und
100 ml Äthyläther wird mit einem trockenen gasförmigen Chlorwasserstoffstrom kalt gesättigt. Anschiiessend wird das Gemisch
über Nacht im. Eisschrank gehalten. Man fügt Äthyläther hinzu und gewinnt durch Filtration das gebildete 4-(3.5-Dimethoxy-phenoxy)-3-Hydroxy-butyroiminoäthyläther-hydrochlorid.
Die Lösung dieses Produktes wird in 30 ml Äthanol mit einem
trockenen Ammoniakgasstrom gesättigt. Nach Stehen über Nacht
im Kühlschrank vertreibt man das Lösungsmittel im Vakuum. Der mit Äthyläther behandelte Rückstand wird abfiltriert.
Das derart erhaltene rohe 4~(3.5-Dimethoxy~phenoxy)«3-Hydroxy-

butyramidinhydrochlorld wird durch Umkristallisieren aus einem Äthanol/Äthyläthergemisch umkristallisiert. Das in einer Cesamtausbeute
von 30$ gewonnene Produkt liegt als weisses kristallines
Pulver mit einem scharfen Schmelzpunkt von l48°C vor und ist in Wasser, Äthanol und Methanol löslich und unlöslich
in Äther und Äthylacetat.

Zu einer kalten Lösung von 4.2 g (0.06 Mol) Hydroxylamin~hydro~
chlorid in 100 ml Methanol fügt man eine Lösung von 3·25 g
(0.06 Mol) Natriummethylat in 100 ml Methanol, rührt während einer halben Stunde, filtriert und fügt dem FiItrat 13 g (0.04?
Mol) 4-(3.5-Dimethoxy-phenoxy)-^-hydroxy-butyramidin-hydrochlorid
hinzu.
Man lässt 24 Stunden bei Raumtemperatur stehen, dampft im Vakuum zur Trockene ein, nimmt mit Wasser auf, extrahiert
mit Äthylacetat, wäscht dieses mit V/asser, trocknet und vertreibt das Lösungsmittel im Vakuum. Man erhält 10.4 g

der bei 90-920C schmelzenden Base. Die aoetonische Lösung,
der Base säuert man mit ätherischer Salzsäure an, filtriert und kristallisiert aus einem Gemisch Äthanol/Äther um.
Man erhält 4-(3.5-Dimethoxy-phenoxy)-3-Hydroxy-butyramidoxim
in einer Ausbeute von 6~[ tfo, wobei die Verbindung in Form
vjeisser bei 1380C schmelzenden Kristalle erhalten wird.
Sie löst sich in Wasser, Methanol, Äthanol und ist in Aceton, Äthylacetat und Äther unlöslich.
Die gemäss den Beispielen 1 und 2 erhaltenen Produkte wurden
Prüf versuchen unterzogen. Die Ergebnisse der phartnakologischen
und klinischen Versuche sind nachstehend zusammengefasst.
Die Vex*suehe mit dem Produkt des Beispiels 2 wurden durch
Einwirkung auf isolierte Herzkammern von Meerschweinchen durchgeführt. Hierbei wurden zwei Sätze Organe verwendet,
wobei das Produkt in Kontakt mit dem Organ vor dem Waschen während 5 Minuten gelassen wurde. Ab 10 /Ug/ml kann man
einen leichten negativ chronotropen Effekt beobachten. Bei einer Dosis von 100/Ug/ml übt das Produkt des Beispiels 2
eine stärkere. Wirkung aus. Die Ergebnisse sind nachstehend in der Tabelle I wiedergegeben*

Selbst bei einer Dosis von 100/Ug/ml blockiert das Produkt
des Beispiels 2 nicht die stimtnulierende Wirkung von Isoprenalin.
Dei den betäubten Hunden wurde das Produkt des Beispiels 2 mi ei Hunden auf intravenösem Meg j η anwachsenden
Dosen injiziert. Eine inotrope und negativ chronotrope Wirkung zeigte sich bei 100 mg/kg.

Die Ergebnisse sind in der Tabelle II nachstehend wiedergegeben .

Die Isoprenalinwirkung ist nicht wirksam selbst bei einer Menge
von 100 mg/kg vermindert. Zusammenfassend übt das Produkt des Beispiels 2 nur eine schwache negativ chronotrope Wirkung auf
die isolierte Herzkammer des Meerschweinchens und auf den be-

täubten Hund aus* Es besitzt keine ^-blockierenden Eigen-,
schäften.
Bei der menschlichen Therapie hat sich dieses Produkt als wirksam in der anti-arythmlschen Behandlung erwiesen.
Als Beispiele werden zwei Beobachtungen wiedergegeben.
M.R. ... - 90 Jahre - myocarditisehe Insufficient - Herzinsufficienz
- veiitriculäre Extrasystolen (5 auf jeweils 50 Zyklen). Die Substanz wird in einer
Dosis von 0.j50 g per os in einheitlicher Dosis
verabreicht. Verringerung der Extrasystolen. Erneute
Verabreichung nach 8 Stunden einer Tablette die O.io g-an Wirkstoff enthielt per os. Vollkommenes
Verschwinden der Extissystolen. Leichte bradyeardische
Wirkung.
M.G. ... polymorplie ventriculäre Extrasystolen. Die Substanz
wurde in Form einer Kapsel verabreicht, die O.^Og
des Wirkstoffs enthielt. Wirkung nach 1 Stunde J>0 Minuten.
Während J> Tagen eine Dosis von 4 Kapseln mit O,
täglich. Günstige Wirkung. Das zuvor intravenös in einer Dosis vor» 1 g täglich verabreichte Xylocain unterdrückte
die sporadischen Extrasystolen nicht.
Mit dem Produkt gemäss Beipsiol 1 hat man bei der isolierten
Herzkammer des Meerschweinchens beobachtet, dass, im Gegensatz
zu Propano.Lol, das die Wirkung von Isoprenalin in einer Dosis von 0.1, 't'/ml blockiert, das selbst bei einer Dosis von kO T/ml
keine Wirkung auftritt. Wax bor kann man feststellen, dass die
Frequenz eier Coronarleistimi, rton isolierten Herzens leicht erhöht
1st.

Bein Menschen übt die Substanz des Beispiels 1 eine anti-' arythmisohe Wirkung in einer Dosis von 0.10 g bis 0.30 g
an aktivem V/irkstoff in der Tablette oder Kapsel per os verabreicht, aus.

Claims (6)
1.) Neue Butyramidinphloroglucinderivate, die aus der Gruppe,
die durch
a) Verbindungen der allgemeinen Formel
O - CH2 - CH - CH2 - C^ (I)
ok
'MHX
in der R ein Wasserstoffatom oder eine Niedrigalkylgruppe
mit 1-5 C-atomen und X ein Wasserstoffatom oder eine Hydroxyl· gruppe bedeutet, und
b) deren Additionssalze mit Säuren gebildet ist, ausge-VJählt
sind.
2. Neue Derivate nach Anspruch 1, dadurch gekennzeichnet, dass die Additionssalze mit Säuren aus
der Gruppe, die durch das Hydrochloride Hydrobromid,
Sulfat, Phosphat, Oxalat, Fumarat, Maleat, Glutamat, Cyolo-

hexylsu]famat, Asparaginat, Malat, Benzoat, und Accorbat
von 4-(3.5-DJ alkoxy-phonoxy)-3-Hydroxybutyra.midin und
Butyraniidoxim gebildet ist, ausgevnihlt sind.
3· 4-(j5.5-Dimethoxy-phenoxy)-hydroxyV)utyramidin und seine
Additionssalzo mit Säuron.
4. 4- (3·5~Dimethoxy-phenoxy)-J-hydroxy-butyraniidoxitt und
seine Additionssalze mit Säuren.
5. Therapeutisohe Komposition, die in Verbindung mit einem
physiologisch geeigneten Träger, zumindest ejne Verbindung
der Formel I oder eines ihrer Additicnssalze mit nichttoxisohen Säuren enthält.
6. Therapeutische Komposition nach Anspruch 5* dadurch g e
kennzeichnet, dass sie in Form von Pillen, Tabletten oder Kapseln, die jeweils 0.10 bis 0.40 g aktiven
Wirkstoff enthalten, gebildet ist.
