Verfahren zur Herstellung von neuen Ergolderivaten der Formel I

Die Erfindung betrifft ein Verfahren zur Herstellung der Verbindungen der Formel I, worin R für Phenyl oder einen durch eine Alkoxygruppe mit 1 bis 4 Kohlenstoffatomen substituierten Phenylrest steht und ihrer Säureadditionssalze.
Aus E. Schreier, Helv. Chim. Acta 41, 1984 (1958) ist das 6-Methyl-8-piperidinomethyl-8-ergolen bekannt. Es wird aber für diese Verbindung keine therapeutische Anwendung angegeben. Zweck der vorliegenden Erfindung ist es, neue Verbindungen herzustellen, welche zur Behandlung von Depressionen und zerebraler Insuffizienz verwendet werden können. Die neuen Verbindungen unterscheiden sich von der bekannten strukturell naheliegendsten Verbindung dadurch, dass sie an Stelle der Piperidinogruppe eine 4-Phenylpiperazinogruppe aufweisen und sind aufgrund ihrer zentral serotoninerg stimulierenden Eigenschaften zur Behandlung von Depressionen und zerebraler Insuffizien indiziert.
Erfindungsgemäss gelangt man zu den Verbindungen der Formel I, indem man die Verbindungen der Formel II, worin R obige Bedeutung hat, mit selektiv die Carbonylgruppe angreifendem Reduktionsmittel reduziert.
Die Reduktion kann analog zu bekannten Methoden durchgeführt werden.
Als Reduktionsmittel wählt man z. B. ein komplexes Metallhydrid, vorzugsweise ein Aluminiumhydrid wie Lithiumaluminiumhydrid. Die Reduktion wird dann in Lösung, beispielsweise unter Verwendung von Dioxan oder Tetrahydrofuran, durchgeführt. Die Reaktionstemperatur wird zweckmässig zwischen etwa 70 bis 1100 gewählt. Man arbeitet vorzugsweise bei Siedetemperatur des Reaktionsgemisches.
Die Verbindungen der Formel I können in freier Form, als Base oder in Form von Additionssalzen mit Säuren, vorliegen.
Aus den freien Basen lassen sich in bekannter Weise Säureadditionssalze herstellen und umgekehrt.
Zur Salzbildung können z. B. folgende Säuren verwendet werden: Chlorwasserstoff-, Schwefel-, Phosphor-, Wein-, Malein- und Methansulfonsäure.
Die als Ausgangsstoffe verwendeten Verbindungen der Formel II kann man analog zu bekannten Methoden, beispielsweise wie im experimentellen Teil beschrieben, erhalten.
Die Verbindungen der Formel I in freier Form oder in Form von Additionssalzen mit physiologisch verträglichen Säuren zeichnen sich durch interessante pharmakologische Eigenschaften aus und können daher als Heilmittel Verwendung finden.
So reduzieren sie, bei einer Dosis von etwa 0,03 bis 0,1 mg/kg i.v., die Zahl der durch 0,5 mg/kg i.p. Reserpin an der Katze hervorgerufene Spitzenpotentiale am Corpus geniculatum laterale [PGO-Spikes; Froment und Eskagan, C.R.
Soc. Biol. (Paris) 165, 2153-2157 (1971)J. Die erfindungsge mässen Verbindungen besitzen daher zentral serotoninerg stimulierende Eigenschaften; aufgrund dieser Eigenschaften ist ihre Anwendung bei der Behandlung von Depressionen und zerebraler Insuffizienz indiziert.
Die zu verwendenden Dosen variieren naturgemäss je nach eingesetzter Substanz, Art der Administration und des zu behandelnden Zustandes. Im allgemeinen werden befriedigende Resultate mit täglichen Dosen von ungefähr 0,03 bis 0,3 mg/kg Tierkörpergewicht verabreicht, nötigenfalls in Teildosen, z. B. 2 bis 4 Anteilen täglich oder in Retardform, erreicht.
Die Tagesdosis für grössere Säugetiere beträgt ungefähr 2 bis ungefähr 20 mg. Geeignete Dosierungsformen für orale Verabreichung enthalten z. B. ungefähr 0,5 bis ungefähr 10 mg neben pharmazeutisch akzeptablen Träger- oder Hilfsstoffen.
Die Heilmittel, die eine Verbindung der Formel I in freier Form oder in Form ihrer physiologisch verträglichen Additionssalze mit Säuren enthalten, beispielsweise eine Lösung oder eine Tablette, können nach bekannten Methoden, unter Verwendung der üblichen Hilfs- und Trägerstoffe, hergestellt werden.

Beispiel
6-Methyl-8-(4'-p-methoxy-phenyl-1 piperazinomethyl)-8-ergolen
0,4 g Lithiumaluminiumhydrid werden in 15 ml Dioxan suspendiert und bei 95 innerhalb 2 Minuten eine Lösung von 0,44 g 6-Methyl-8-ergolen-8-carbonsäure- (4'-p-methoxyphenyl)-1'-piperazid in 15 ml Dioxan zugetropft. Nach weiteren 2 Minuten wird auf 00 abgekühlt und das überschüssige LiAIH4 mit 1 ml Wasser zersetzt. Die organische Phase wird mehrmals mit Diäthyläther ausgeschüttelt; die Äther-Extrakte über Kaliumcarbonat getrocknet und eingedampft. Die Titelverbindung kristallisiert aus Aceton (Smp. 208-210", [a] = - 1090 (c = 0,4 in Pyridin).
Das Maleinat der Titelverbindung kristallisiert aus Methanol (Smp. 174 bis 1760, [a120 = -630 (c = 0,4 in 50%igem wässrigem Äthylalkohol).
Das als Ausgangsprodukt verwendete 6-Methyl-8-ergolen 8-carbonsäure-(4'-p-methoxyphenyl)-1'-piperazid kann folgendermassen hergestellt werden: a) 6-Methyl-8-ergolen-8-carbonsäurechloriddime thylformamid-Addukt Hydrochlorid
Eine Lösung von 1,1 ml Oxalylchlorid in 10 ml Chloroform wird bei 200 zu 2,4 ml N,N-Dimethylformamid in 10 ml Chloroform zugetropft und nach ca. 15 Minuten in die Suspension von 2,68 g 6-Methyl-8-ergolen-8-carbonsäure und 7,7 ml Dimethylformamid in 50 ml Chloroform bei -200 gegeben und 15 Minuten gerührt. Das entstandene Addukt aus 6-Methyl-8ergolen-8-carbonsäurechlorid Hydrochlorid und Dimethylformamid wird direkt für die nachfolgende Kondensationsstufe verwendet.
b) 6-Methyl-8-ergolen-8-carbonsäure- (4'-p-methoxy phenyl)-1 '-piperazid
5,3 g 4-p-Methoxyphenylpiperazindihydrochlorid werden in 50 ml Chloroform und 15 ml tert. Butylalkohol suspendiert und mit 4 g Triäthylamin versetzt. Bei -200 lässt man das nach Stufe a) hergestellte Addukt zutropfen und rührt anschliessend 30 Minuten bei 00. Die Chloroformlösung wird danach mit 100 ml gesättigter, wässriger Natriumhydrogencarbonatlösung von 0 ausgeschüttelt, über Natriumcarbonat getrocknet und danach im Vakuum das Chloroform abgedampft, wobei die Titelverbindung als Rückstand verbleibt, der weiterverarbeitet werden kann.

From E. Schreier, Helv. Chim. Acta 41, 1984 (1958) 6-methyl-8-piperidinomethyl-8-ergolene is known. However, no therapeutic application is specified for this compound. The purpose of the present invention is to produce new compounds which can be used for the treatment of depression and cerebral insufficiency. The new compounds differ from the known structurally most obvious compound in that they have a 4-phenylpiperazino group instead of the piperidino group and, because of their central serotoninergic stimulating properties, are indicated for the treatment of depression and cerebral insufficiency.
According to the invention, the compounds of the formula I are obtained by selectively reducing the compounds of the formula II, in which R has the above meaning, using a reducing agent which attacks the carbonyl group.
The reduction can be carried out analogously to known methods.
As a reducing agent you choose z. B. a complex metal hydride, preferably an aluminum hydride such as lithium aluminum hydride. The reduction is then carried out in solution, for example using dioxane or tetrahydrofuran. The reaction temperature is expediently chosen between about 70 to 1100. It is preferably carried out at the boiling point of the reaction mixture.
The compounds of the formula I can be present in free form, as a base or in the form of addition salts with acids.
Acid addition salts can be prepared from the free bases in a known manner and vice versa.
For salt formation z. B. the following acids are used: hydrochloric acid, sulfuric acid, phosphoric acid, wine acid, maleic acid and methanesulfonic acid.
The compounds of formula II used as starting materials can be obtained analogously to known methods, for example as described in the experimental part.
The compounds of the formula I in free form or in the form of addition salts with physiologically tolerable acids are distinguished by interesting pharmacological properties and can therefore be used as medicines.
