Die vorliegende Erfindung betrifft eine Reihe von Carbonsäureamid-
und Carbonsäurehydrazidderivaten des 4,5-Diphenyloxazols
der allgemeinen Formel

In der obigen Formel ist η O, 1 oder 2 und R bedeutet einen
Monoalkylaminorest mit 1-4 C-Atomen, einen Dialkylaminorest nit 1-4 C-Atomen in jedem Alkylrest, einen N-heterocyclischen
Rest mit 5- oder 6-gliedrigen Ringen, die auch ein anderes
Heteroatom enthalten können, oder einen Hydrazin- oder N',N1-Dialkylhydrazinrest
mit 1-4 C-Atomen in jeder Alky!gruppe.
Spezielle Beispiele für Reste, die R bedeuten kann, sind NHCIU, ■ » N

Die Verbindungen der allgemeinen Formel (I) zeichnen sich durch eine bemerkenswerte analgetische Aktivität, oft zusammen mit
einer antiphlogistischen Aktivität,aus und durch eine hemmende
Wirkung auf das Zentralnervensystem.
4,5-diphenylsubstituierte Oxazolderivate, die eine sekundäre
oder tertiäre Aroinogruppe (italienische Patentanmeldung 18390/66
(37/499) der Anmelderin) oder eine Carboxylgruppe in 2-Stellung des Ringes tragen, welche eine oder mehrere Methylengruppen vom
heterocyclischen Ring weg liegen, sind dafür bekannt, daß sie antiphlogistische, analgetische und antihelkogenische Eigensshaft©R

Gemäß dieser Erfindung werden die Verbindungen der allgemeinen Formel (I) durch Aminolyse und Hydrazinolyse eines Esters der
allgemeinen Formel

bedeutet, unter Verwendung eines Amins oder Hydrazins der

in der R die obige Bedeutung hat, hergestellt. Der obige Ester (V) kann durch Acylierung von of.-Phenyl- oL-aminoacetophenon (II)
mit einem Dicarbonsäuremonoester-monochlorid der allgemeinen Formel (III) und anschließende Cyclisierung des erhaltenen Acylaminoderivates
(IV) mit Phosphoroxychlorid odef ähnlichen Kondensationsmitteln
nach folgendem Reaktionsschema hergestellt werden:

Im obigen Schema haben η, R und R1 die obige Bedeutung. Die Acylierung
des oC-Amino- oC-phenylacetophenons (II) mit dem Monoester-monochlorid
(III) wird in geeigneter Weise mit dem Hydrochlorid von (II) in einem inerten Lösungsmittel, wie Benzol
oder Toluol,bei der Siedetemperatur des Gemisches ausgeführt.
Das Mol-VerhSltnis der Reaktionspartner betragt vorzugsweise
etwa 1:1.
Die Acylaminoverbindung (IV) wird anschließend zu dem Oxazolderivat
(V) mit POCl3, PCl3 oder dergl. in einem inerten Lösungsmittel,
wie Benzol oder Toluol, bei Temperaturen im Pereich von Zimmertemperatur bis zur Siedetemperatur des Gemisches eyeIisiert.
Die Umsetzung des Esters (V) mit der passenden Base (VI)

wird in Anwesenheit oder Abwesenheit eines geeigneten organischen
Lösungsmittels (Eenzol, Toluol, Xylol usw.) in Abhängigkeit
von den spezifischen Reaktivitäten der Reaktionspartner
1-36 Stunden bei Temperaturen von 20 - 12O°C durchgeführt.
Eine Modifizierung des obigen Verfahrens, die nur auf Verbindungen
der allgemeinen Formel (I), in der η 1 oder 2 ist, angewendet
werden kann, besteht in der Hydrolyse eines entsprechenden Fsters (V), in der η 1 oder 2 ist, mit einem Alkali, um
eine entsprechende Carbonsäure zu erhalten, die anschließend in das Säurechlorid unter Verwendung von Thionylchlorid in Anwesenheit
oder Abwesenheit eines geeigneten Lösungsmittels, wie trockenes Benzol, Toluol oder Xylol, bei Zimmertemperatur umgewandelt
wird, und Umsetzung des erhaltenen Säurechlorides mit einem passenden Amin oder Hydrazin*von denen ein großer molarer
Überschuß erforderlich ist, in Inerten Lösungsmitteln , um das gewünschte Amid oder Hydrazid zu erhalten.
Die obige Modifikation bietet jedoch keine Vorteile gegenüber dem vorher angeführten Verfahren«
Die folgenden Beispiele erläutern die Erfindung.
4 f 5-Diphenyl-*2-oxagolylcagbons8ure-Nf N-diäthylamid
Eine Lösung von 8?8 g 2-Cas°foäthoxy»4,5-äiphenyloscszol (hergettellt
nach Chemical Abstracts |@? Seite 3408) in 31 ml gH8thyl&mln

wurde 24 Ftunden unter Rückfluß erhitzt. Das überschüssige
Diethylamin wurde bei 20 mm Druck entfernt, der Rückstand in
100 ml siedendem 70 %igen Äthanol aufgenommen und nach dem Abkühlen
der Lösung 7 g kristallines 4,5-Diphenyl-2-oxazolylcarbonsSure-N,N-diäthylamid
mit einem Schmelzpunkt von 130 bis 132°C erhalten. Ausbeute 73 %.
4 , S-Diphenyl^-oxazolyl-carbonsSure-N' ,N'-dimethylhydrazid
Ein Gemisch von 11,6 g 2-Carbäthoxy-4,5-diphenyloxazol (hergestellt
wie in Beispiel 1 angegeben) und 18 g Ν,Ν-Dimethylhydrazin

mit einera Schmelzpunkt von 190 - 192 C erhalten. Ausbeute 73 %.
a) oL-Phenyl- 0C7 (N-carbSthoxyacetylamino) -acetophenon
Ein GeKiSGh von 6,2 g C^-Phenyl-oL-amlno-acetophenonhydrochlorid
und 5f2 g Carbäthoxy-acetylchlorid In 40 ml wasserfreiem Benzol
wurde 4 Sfeisnden tsnter Rückfluß erhitzt. Das siedende Gemisch
wurde filtriert, das Filtrat auf 15°C abgekühlt und mit 20 ml
Lig^eisi CSl@cl@bereieh 30 - 6O0C) verdünnt. Nach dem Abkühlen
auf Q0G Wtirdgsi if5 g kristallines cfe-Phenyl-o^-(N-carbäthoxyace-

tylamino)-acetophenon mit einem Schmelzpunkt von 90 - 92°C erhalten.
Ausbeute 80 %.
b) ^'thyl--4^5-diphenyl-2-oxazolylacetat
Eine Lösung von 6,5 g c^-Phenyl-cC-(N-carbSthoxyacetylamino)-acetophenon
und 8 g Phosphoroxychlorid in 15 ml wasserfreiem Benzol wurde 1 Stunde unter Rückfluß erhitzt. Das Lösungsmittel
vmrde bei 20 mm Druck entfernt und der ölige Rückstand mit 60 ml
50 %igen Äthanol aufgenommen, wobei 4,9 g kristallines iithyl-4,5-diphenyl-2-oxazolylacetat
mit einem Schmelzpunkt von 65 bis 67°C allmählich ausgeschieden wurden. Ausbeute 79 %.
c) 4,5-Diphenyl-2~oxazolylessigsgure-N-inethylamid
Ein Gemisch von 9 g iithyl~4,5-diphenyl-2-oxazolylacetat und
100 ml einer 20 %igen benzolischen Lösung von Methylamin wurde in einem verschlossenen Kolben 36 Stunden bei 22°C gehalten.
Nach dem Verdünnen der benzolischen Lösung mit 1OO ml Ligroin wurden 7,2g kristallines 4,5-Diphenyl-2-oxazolylessigsaure-N-methylamid
mit einem Schmelzpunkt von 139 - 142°C erhalten. Ausbeute 81 %.

6 g Kthyl-4,5-diphenyl-2-oxazolylacetat (hergestellt wie in Bei- .'
spiel 3 a-b beschrieben) und 17 g Morpholin wurden 6 Stunden unter Rückfluß erhitzt. Das überschüssige Morpholin wurde bei

20 nun Druck entfernt und der ölige Rückstand mit. 100 nl 50 %igem
Ethanol aufgenommen, wobei das kristalline 4,5-Diphenyl-2-oxazolylessigsSuremorpholid allmählich ausgeschieden wurde. Die
ümkristallisation dieser Verbindung aus einem Gemisch von absolutem Äthanol/wasserfreiem Äther/Ligroin ergab 4,2 g eines
Produktes mit einem Schmelzpunkt von 127 - 129°C. Ausbeute 60 %.
4,5-Diphenyl-2-oxazolylessigsäurehydrazid
Eine Lösung von 6 g Kthyl-4,5-diphenyl-2-oxazolylacetat (hergestellt wie in Beispiel 3 a-b beschrieben) in 50 ml absolutem
Äthanol wurde mit 10 ml 98 %igem Hydrazinhydrat behandelt. Das
Reaktionsgemisch wurde 1 Stunde auf 22 C gehalten, anschließend 50 ml destilliertes Wasser zugesetzt, um das 4,5-Diphenyl-2-oxazolylessigsäurehydrazid vollständig auszufällen, das anschließend durch Umkristallisieren aus 70 tigern Äthanol gereinigt
wurde. Es wurden 4,7 g eines Produktes mit einem Schmelzpunkt von 164 - 165°C erhalten. Ausbeute 79 t.
ä) ot-Phenyl-oL-fN- (B-carbmethoxypropionyl^ -aminoj-acetophenon
Ein Gemisch von 7,4 g ct-Phenyl-cCraminoacetophenonhydrochlorid
und 6 g B-Carbmethoxypropionylchlorid in 60 ml wasserfreiem
Benzol wurde 5 Stunden unter Rückfluß erhitzt. Nach dem Filtrieren des siedenden Gemisches und Abkühlen des Filtrates auf 15°C
wurden 20 ml Ligroin (Siedeberelch 30 - 60°C) zugesetzt. Nach dem

Abkühlen auf O0C wurden 8,5 g kristallines otPhenyl-oL-[n-(ßcarbmethoxypropionyl)aminoj-acetophenon
mit einem Schmelzpunkt von 1O3 - 1O4°C erhalten. Ausbeute 87 %. ■
b) Methyl-3-(4,5-diphenyl-2-oxazolyl)-propionat
Eine Lösung von 6,5 g d-Phenyl-cL-[N-(ß-carbmethoxypropionyl)-aminoTJ
-acetophenon und 8 g Phosphoroxychlorid in 20 ml wasserfreiem
Benzol wurde 1 Stunde unter Rückfluß erhitzt. Das Lösungsmittel wurde bei 20 mm Druck entfernt und der ölige Rückstand
mit 60 ml 50 %igem Äthanol aufgenommen. Aus der erhaltenen
Lösung wurden bei Zimmertemperatur 5,2 g kristallines Methyl-3-(4,5-diphenyl-2-oxazolyl)-propionat
mit einem Schmelzpunkt von 58 - 59°C erhalten. Ausbeute 85 %.
c) 3-(4,5-Diphenyl-2-oxazolyl)-propionsaurepyrrolidid
9,2 g Methyi-3-(4,5-diphenyl-2-oxazolyl)-propionat und 25 ml
Pyrrolidin wurden 3 Stunden unter Rückfluß erhitzt. Das überschüssige
Pyrrolidin wurde bei 20 mm Druck entfernt und der ölige Rückstand mit 100 ml 70 %igem Äthanol aufgenommen« Nach
dem Abkühlen auf O0C wurden 8,S g kristallines 3-(4,5-Diphenyl-2-Qxazolyl)-pr©pionsäiirQjpyrr©lidid
aus der erhaltenen Lösung rait einem SchmslzpuRkt -won §2 - 66°C erhalten. Ausbeute 82 %.

v/ie in Beispiel 6 a-b beschrieben) wurden in einer Lösung von
4 g Natriumhydroxyd in 40 ml Wasser und 30 ml Äthanol gelöst. Die Lösung wurde 1 Stunde unter Rückfluß erhitzt, auf 5°C abgekühlt
und mit 10 %iger Salzsäure auf pH 2 angesäuert, um die 3-(4,5-Diphenyl-2-oxazolyl)-propionsäure auszufällen. Das Reaktionsprodukt
wurde durch Umkristallisieren aus 70 %igem Äthanol gereinigt und 5,3 g eines Produktes mit einem Schmelzpunkt von
160 - 162°C erhalten. Ausbeute 90 %.
b) 3~ (4 ,S-Diphenyl-^-oxazolyl) -propionsäure-N^-diäthylamid
12 g 3-C4,5-Diphenyl-2-oxazolyl)-propionsäure wurden in 100 ml
wasserfreiem Benzol suspendiert und 8,6 ml Thionylchlorid zugesetzt.
Das Reaktionsgemisch wurde 1 Stunde auf Zimmertemperatur
gehalten und anschließend das überschüssige Lösungsmittel und Thionylchlorid bei 5 mm Druck entfernt. Der ölige Rückstand
wurde in 50 ml wasserfreiem Benzol aufgenommen und die erhaltene Lösung tropfenweise unter Rühren zu einer Lösung von 6 g Diäthylamin
in 100 ml wasserfreiem Benzol zugesetzt, die vorher auf O0C gekühlt worden war. Nach der Zugabe wurde die Temperatur
a«f 22 - 25°C gesteigert, das Reaktionsprodukt mit Wasser
gewaschen und die abgetrennte Bensolphase bei 20 mm Druck destilliert« Der ölig© Rückstand wurde in 50 ml wasserfreiem
