3. Verfahren zur Herstellung von Guanidinocapronsäurephenylestern
nach Anspruch i, dadurch gekennzeichnet, daß man in an sich bekannter Weise
e-Guanidinocapronsäurechlorid mit einer Phenolverbindung der allgemeinen Formel:

worin X die im Anspruch I angegebene Bedeutung besitzt, in Anwesenheit eines Chlorwasserstoff
abspaltenden Mittels umsetzt.

Das Säurechlorid der Guanidinocapronsäure kann in üblicher, an sich für die Herstellung von Säurechloriden
aus ihren Carbonsäuren bekannter Weise hergestellt werden. So kann Guanidinocapronsäure mit einem
Chlorierungsmittel wie SOCI2, POCI3, PCl3 oder PCI5 zur
Herstellung des Säurechlorids umgesetzt werden.
Als chlorwasserstoffabspaltendes Mittel kann ein beliebiges, bekanntes, organisches, basisches, Chlorwasserstoffsäure
abspaltendes Mittel, wie Pyridin oder Collidin verwendet werden.
Die Veresterungsreaktion zwischen dem Guanidinocapronsäurechlorid
und der Phenolverbindung der allgemeinen Formel (II) wird in Anwesenheit eines inerten, nichtpolaren, organischen Lösungsmittels für
beide Reaktionsteilnehmer, z. B. Tetrahydrofuran, Benzol oder Toluol, durchgeführt.
Da die Guanidinocapronsäurephenylester -J.er allgemeinen
Formel (I) basisch sind, werden sie vorteilhafterweise nach der Veresterungsreaktion, jedoch vor der
Isolierung und Reinigung in Säuresalze überführt. Beispiele von Säuren, die mit den Guanidinocapronsäurephenylestern
Säuresalze bilden können, sind Chlorwasserstoffsäure, Schwefelsäure, Oxalsäure, Weinsäure,
Toluolsulfonsäure. Ebenfalls ist es möglich, die Guanidinocapronsäure in das Säuresalz umzuwandeln, bevor
diese in das Säurechlorid umgewandelt wird, oder bevor dieses mit der Phenolverbindung umgesetzt wird.
Die folgenden Beispiele erläutern die Herstellung der erfindungsgemäßen Verbindungen:
Die Erfindung betrifft Guanidinocapronsäurephenylester und ein Verfahren zu ihrer Herstellung.
Die erfindungsgemäßen Guanidinocapronsäurephenylester werden durch die folgende allgemeine Formel
wiedergegeben:
worin X ein Wasserstoffatom, ein Chloratom, eine Nitnjgruppe, eine Methylgruppe, eine Methoxygruppe,
eine Carboxylgruppe, eine Hydroxylgruppe, ein Carbäthoxyrest oder ein ε-Guanidinocaproyloxyrest ist.
Niedere Alkyl- bzw. Alkoxyreste sind solche mit I bis 4 C-Atomen. Gemäß der Erfindung können die Verbindungen
der obengenannten Formel (I) hergestellt werden, indem das Säurechlorid von ε-Guanidinocapronsäure
mit einer Phenolverbindung der allgemeinen Formel:
7 g ε-Guanidinocapronsäure-p-toxylsalz und 21g
Thionylchlorid wurden zusammen vermischt, um bei Raumtemperatur miteinander zu reegieren. Es trat eine
endotherme Reaktion auf und die Säure löste sich allmählich auf. Dann wurde das Reaktionsgemisch 1 bis
2 h stehengelassen und mit 100 ml Äther extrahiert. Die
untere, ölige Schicht und die Ätherschicht wurden voneinander getrennt, und die unlere Schicht wurde
wiederholt mit Äther gewaschen. Dann wurde die ölige Substanz (Säurechlorid) mit 2 g Phenol in 50 ml
Tetrahydrofuran versetzt, und das Gemisch wurde gerührt. Nachdem die Mischung zu einer einheitlichen
Lösung geworden war, wurden 2 g Pyridin allmählich zugefügt. Es trat eine exotherme Reaktion auf, und in
der unteren Schicht trennte sich eine ölige Substanz ab. Nach Abschluß der Reaktion wurde dii ölige Substanz
mit Wasser gewaschen und dann aus heißem Wasser ein- oder zweimal umkristallisiert. Auf diese Weise
wurde das ε-Gurnidinocapronsäurephenylester-p-tosylsalz
als weiße Kristalle mit einem Schmelzpunkt von 101-1030C erhalten. Die Elementar-Analyse auf
C20H2JN3O5S ergab folgende Werte:

Die ε-Guanidinocapronsäurephenyiester der Erfindung sind neue Verbindungen. Sie besitzen eine niedrige
Toxizität (siehe Tabelle 1), und s:e besitzen eine inhibitorische Wirkung auf die Hydrolyse von N-Toxyl-L-argininmethylester durch Kallikrein, auf die Hydroly- Tabelle I
se von Casein durch Trypsin und die fibrinogenolytische
Aktivtät v<Jn Plasmin (siehe Tabellen II, III und IV). 30
