Die Erfindung betrifft das a-IsopropyI-a-[(N-methyl-N
- homoveratryl) - γ■■ aminopropyl] -3,4-äthylendioxyphenylacetonitril
der Formel

Es wurde gefunden, daß diese Verbindungen coronarerweiternde
Eigenschaften besitzen.
Es sind zwar basisch substituierte Phenylacetonitrile, wie die der deutschen Patentschrift 960462
bekannt, jedoch zeichnen sich diese durch eine gute analgetische Wirkung aus.
Ferner werden basisch substituierte Phenylacetonitrile in den deutschen Patentschriften 964499 und
964 500 beschrieben, wobei diejenigen der deutschen Patentschrift 964499 analgetiscb und spasmolytisch
wirksam sind und die Verbindungen der deutschen Patentschrift 964 500 gleichfalls analgetische Wirksamkeit
aufweisen.
Schließlich sind aus den deutschen Patentschriften 1 154810 und 1 158 083 weitere basisch substituierte
Phenylacetonitrile bekannt, die eine coronarerweiternde Wirkung zeigen, aber diese unterscheidet sich
nur in quantitativer Hinsicht von der des fiir den gleichen Zweck verwendeten /8-Oxyäthyl-theophyllins.
Die neuen Verbindungen der oben angegebenen Formel zeichnen sich durch eine die Kontraktilität
des Herzmuskels verbessernde Wirkung aus, was durch eine Zunahme der Druckanstiegsbeschleunigung
in der linken Herzkammer am narkotisierten Hund zum Ausdruck kommt.
Die dabei verwendete Versuchsanordnung am Hund iesteht darin, daß nach Morphin-Prämedikation
iblicherweise mit Chloralose intravenös narkotisiert vird. Die Dniickanstiegsbeschleunigung im linken
/entrikel wird! entweder mit einem elektrischen Katheterspitzenmanometer oder mit einem elektrischen
Manometer am Ende eines Stahlkatheters gemessen und über einen Differential-Rechen verstärker
in die mathematische Größe dp/dt umgewandelt
Die coronarcrweiternde Wirkung wurde in derselben Versuchsanordnung entweder mit einemelektromagnetischen
auf eine Coronararterie aufgesetzten Flußmeßkopf oder an Hand der Zunahme der Saueistoffsättigung
des Hämoglobins des Blutes im Coronarsinus gemessen.
Bei der ersteren Versuchsanordnung an narkotisierten
Bastard-Hunden ergaben sich mit dem a-Isopropyl-α
- [(N - methyl - N - homoveratryl) - γ - aminopropylj-3,4-äthyjendioxyphenylacetonitril
- hydrochloric! die in Tabelle I unter Substanz 1 ange/uhrten Zunahmen
der Coronardurchblutung. Als Vergleichspräparat wurde das aus der deutschen Auslegeschrift 1 210 883
bekannte 3 - {fl - Diäthylaminoäthy!) - 4 - methyl - 7 - äthoxy-carbonyl-methoxy-cumarin-hydrochlorid
(Substanz 2) mitgeprüft. Die Überlegenheit der neuen Verbindung ergibt sich aus dem therapeutischen Index,
der sich aus der Coronarwirksamkeit und der Toxizität (Tabelle I) errechnen läßt.
Die Toxizität des a-Isopropyl-a-[(N-m«thyl-N-homoveratryl)-y-aminopropyI]-3,4-äthylendioxy-phenylacetonitri!
hydrochloride wurde wie folgt ermittelt. Hierzu wurden fünf Bastard-Hunden (Gewicht 8 bis
14 kg) 20 mg/kg Körpergewicht dieser Verbindung i. v. appliziert. Von den Hunden überlebten vier Tiere;
diese zeigten während der ersten 4 bis 5 Minuten Seiteniage und zeitweilig Streckkrämpfe, die kurzfristig
auttragen, sowie Speichelfluß und Hecheln oder Schnappatmung. Die Erholung der einzelnen Tiere
erfolgte unterschiedlich schnell. Zwei Hunde richteten sich nach 4 bzw. 7 Minuten wieder auf, während 'ein
Hund noch nach 40 Minuten Krämpfe zeigte und sich erst nach 50 Minuten aufrichtete. Nach etwa
2 Stunden liefen die vier Hunde wieder umher und zeigten am darauffolgenden Tage ein normales Verhalten.
Die in Anlehnung an die Methode von Litchfield

22,7 (19,9 bis 26,1) mg/kg Körpergewicht. Das geprüfte Hydrochlorid zeigt an Bastard-Hunden bereits
mit einer Dosis von 0,08 mg/kg J· v. eine deutliche
Wirkung auf die Herzkrankgefäße, wie nachfolgend aufgezeigt wird. Hieraus ergibt sich eine große
therapeutische Breite, da die intravenöse Toxizität der Substanz mehr als das 200fache der intravenös
therapeutisch wirksamen Dosis beträgt.

Die Messung der Sauerstoffsättigung im Coronarsinus
an narkotisierten Bastard-Hunden erfolgte mit Hilfe eines Katheters, durch den das venöse Blut
abgesaugt wurde. Die Messung des Oxybämoglobins im venösen Blut erfolgt in Durchflußküvetten eines
Atlasoxymeters. Die Versuche wurden ebenfalls mit dem Hydrochlorid der neuen Verbindung durchgeführt.
Die dabei erhaltenen Werte sind in der nachfolgenden Tabelle II unter Substanz 1 zusammengefaßt.
Als Vergleichspräparat wurde wiederum das 3 - (ß - Diäthylaminoäthyl) - 4 - methyl - 7 - äthoxycarbonyl-methoxy-cumarin-hydrochlorid
(Substanz 2) eingesetzt. Auch in dieser Versuchsanordnung ergibt sich die 'Überlegenheit der neuen Substanz 1, die
durch den günstigeren therapeutischen Index zum Ausdruck kommt. Eine gute Steuerbarkeit der neuen
Substanz ergibt sich aus der zeitlich dosisabhängigen und damit gut abgrenzbaren Wirkungsdauer (s.
Tabelle 11).

Es ist außerdem überraschend und neu, daß das a-Isopropyl-a-rjN-methyl-N-homoveratryO-y-aminopropyl]
- 3,4 - athylendioxy - phenylacetonitril - hydrochlorid im Bereich der therapeutischen coronarerweiternden
Wirkung bei der Messung der Sauerstoffsättigung des Coronarsinus keine direkt die Herzfrequenz
beeinflussende Wirkung aufweist. Eine ausgeprägte Blutdrucksenkung erfolgt ebenfalls nicht.
Die Herzfrequenz wurde über ein EK-Pulsgerät gemessen. Die Messung des Blutdrucks erfolgte mit
Statham-Druckmeßwandlern.
, Das a-Isopropyl-a-[(N-methyl-N-homoveratryl)- y - aminopropyl] - 3,4 - athylendioxy - phenylacetonitril
eignet sich auf Grund seiner pharmakologischen Eigenschaften für die Behandlung von herzkranken
Patienten mit Coronarinsuffizienz.
Das α- Isopropyl - α - [(N - methyl - N - homoveratryl)- y - aminopropyl] - 3,4 - athylendioxy - phenylacetonitril
wird dadurch hergestellt, daß man in Gegenwart eines basischen Kondensationsmittels in einem inerten
Lösungsmittel in an sich bekannter Weise entweder
a) α - Isopropyl - 3,4 - athylendioxy - phenylacetonitril
mit einem Homoveratrylamin der allgemeinen Formel

in der X einen reaktionsfähigen Säurerest, insbesondere ein Chlor- oder Bromatom, darstellt,
umsetzt oder
CH
in der X einen reaktionsfähigen Säurerest bedeutet, entweder mit N-Methylhomoveratrylamin
oder mit Homoveratrylamin umsetzt und das im letzteren Fall entstehende a-Isopropyl-a-[(N-homoveratryl)
-γ- aminopropyl] - 3,4 - äthylendioxy-
phenylacetonitril anschließend in üblicher Weise mit Formaldehyd und Ameisensäure methyliert.
Die Umsetzungen der betreffenden 3,4-Äthylendioxy-phenylacetonitrile
mit den entsprechenden Homo-
veratrylaminen werden unter Rühren bei erhöhter
Temperatur durchgeführt. Dabei können die 3,4-Äthylendioxy-phenylacetonitrile
mit dem basischen Kondensationsmittel, vorzugsweise Natriumamid, in einem inerten organischen Lösungsmittel zunächst so lange
erhitzt werden, bis die Ammoniakentwicklung abklingt, worauf anschließend die betreffende Homoveratrylaminverbindung
zugegeben wird. Es ist aber auch möglich, die beiden Reaktionskomponenten zu vermischen und anschließend das Kondensationsmittel
in die siedende Lösung derselben einzuführen. Als basische Kondensationsmittel eignen sich außer
Natriumamid auch Natriumhydrid, Natriumhydroxid, Kaliumamid, Lithiumamid, Kaliumcarbonat oder
tertiäre Amine, wie Pyrimidin oder Triäthylamin.
· Als Lösungsmittel eignen sich insbesondere aro-. matische Kohlenwasserstoffe, wie Benzol, Toluol
oder Xylole, Äther, wie Diäthyläther oder Dioxan bzw. flüssiges Ammoniak.

additionssalzen eignen sich beispielsweise Essigsäure, Propionsäure, Diäthylessigsäure, Malonsäure, Bernsteinsäure,
Fumarsäure, Maleinsäure, Milchsäure, Weinsäure, Äpfelsäure, Zitronensäure, Schwefelsäure,
Bromwasserstoffsäure, Orthophosphorsäure.
Die neuen Verbindungen können in Form von üblichen pharmazeutischen Zubereitungen in der
Humanmedizin verwendet werden.
Eine Lösung, bestehend aus 86,5 g a-Isopropyl-3,4
- athylendioxy - phenylacetonitril (Kp.^ 156 bis
159° C) und 500 ml Toluol, wurde mit 99 g N-(y-Chtorpropyl)
- N - methyl - homoveratrylamin versetzt und unter Rühren zum Sieden erhitzt. Anschließend wurde
die Mischung mit einer Suspension von 15,6 g Natriumamid in 100 ml Toluol versetzt und 4 Stunden
am Rückfluß gekocht. Dann wurde das Reaktionsgemisch abgekühlt auf 700 ml Eiswasser gegossen,
die Toluollösung abgetrennt, mit Wasser gewaschen und mit verdünnter Salzsäure ausgeschüttelt Dann
wurde die saure Phase mit N-jtronlauge alkalisch
gemacht, die dabei abgeschiedene Base in Benzol aufgenommen, mit Wasser gewaschen, über Natriumsulfat
getrocknet und eingedampft. Der Ruckstand wurde · in Äthanol aufgenommen, in die Lösung
bis zur sjuren Reaktion Chlorwasserstoffgas eingeleitet
und hierauf Äther zugegeben. Dabei fiel das Hydrochlorid des α - Isopropyl - α - [(N - methyl-N-
homoverati yl)- y- aminopropyl]- 3,4- äthylendioxyphenylacetonitrils
aus, das nach der Umkristallisation aus Äthanol bei 172 bis 1740C schmolz. Die Ausbeute
betrug 122,9 g (=69% der Theorie).
23,9g a-Isopropyl^^-äthylendioxy-phenylacetonitril
wurden in 140 ml absolutem Benzol gelöst und zum Sieden erhitzt. Dann wurden unter Rühren
4.3 g Natriumainid, die in 20 ml absolutem Benzol
suspendiert waren, zugetropft. Nach 2 Stunden wurde dann eine Lösung von 27,2 g N-Methyl-N-(y-chlorpropyl)-homoveratrylamin
in 30 ml absolutem Benzol innerhalb von 40 Minuten in die Mischung eingetropft. Das Reaktionsgemisch wurde nach 2 Stunden auf
Eis gegossen und gemäß Beispiel 1 aufgearbeitet. Es wurden 41,2 g der Rohbase als Rückstand erhalten,
aus dem, wie im Beispiel 1 beschrieben, nach der Umkristallisation aus Äthanol 34,3 g a-Isopropyl-
a - [(N - methyl - N - homoveratryl) -γ- aminopropyl]-
3.4 - äthylendioxy - phenylacetonitril - hydrochlorid
erhalten wurden und dessen Mischschmelzpunkt mit dem nach Beispiel 1 erhaltenen Produkt keine Depression
zeigte.
5 g α - fsopropyl - α - [y - chlorpropyl) - 3,4 - äthylendioxy-phenylacetonitril
(im Gemisch mit 10,6 g nicht umgesetztem Ausgangsprodukt a-Isopropyl-3,4-äthylendioxy-phenylacetonitril)
wurden mit 6,65 g N-Methyl-homoveratrylamin 7 Stunden auf 130° C erhitzt.
Das 80° C heiße Reaktionsprodukt wurde dann mit 50 ml Benzol versetzt und nach dem Abkühlen
mit Wasser extrahiert. Dann wurde die Benzolphase mit verdünnter Salzsäure ausgeschüttelt und die
saure, wäßrige Lösung unter Eiskühlung mit konzentrierter Natronlauge alkalisch gemacht. Die alkalisch
wäßrige Phase wurde hierauf mit Chloroform extrahiert, der Extrakt getrocknet und das Lösungsmittel
verdampft. Es wurden 8,9 g Rohbase erhalten, die mit Benzol über 20 g Aluminiumoxid der Aktivitätsstofe
II filtriert wurden. In das Filtrit wurde Chlorwasserstoffgas
eingeleitet, wobei ein Hydrochlorid erhalten wurde, dessen Mischschmelzpunkt mit dem
nach Beispiel 1 erhaltenen Produkt keine Depression zeigte. Die Ausbeute an o-Isopropyl-a-[(N-methyl-N-homoveratryl)-y-aminopropyl]-3,4-äthylendioxyphenylacetonitril-hydrochlorid
betrug 2,1 g.
7,2 g rohes α - Isopropyl - a - [(N - homoveratryl)-
γ - aminopropyl] - 3,4 - äthylendioxy - phenylacetonitril
(der Schmelzpunkt des Hydrochloride betrug 160 bis 162° C) wurden in 7 ml Isopropanol gelöst und mii
4 ml Formalinlösung, 0,4 ml Wasser und 28 ml konzentrierter
Ameisensäure 8 Stunden auf 9O0C er-
wärmt. Dann wurde das Gemisch abkühlen gelassen, mit konzentrierter Natronlauge versetzt, die Schichten
wurden getrennt und die organische Phase über festem Natriumhydroxid getrocknet. Nach dem Abdampfen
des Lösungsmittels wurde der Rückstand
destilliert, wobei das fi-Isopropyl-«-[(N-methyl-N-homoveratryl)-y-aminopropyl]-3,4-äthylendioxy-phenylacetonitril
vom Κρ-ο,ι 285 bis 290'C erhalten wurde.
Die Ausbeute an dem anschließend hergestellten Hydrochlorid, dessen Mischschmelzpunkt mit dem
nach Beispiel 1 erhaltenen Produkt keine Depression zeigte, betrug 0,4 g.
6,4 g α - [(N - Methyl - N - homoveratryl) - γ - amino-
propyl] - 3,4 - äthylendioxy - phenylacetonitril vom Κρ.01 270 bis 28O0C wurden in 150 ml flüssigem Ammoniak,
in dem 0,5 g Natrium als Natriumamid enthalten sind, gelöst. Unter Rühren wurden dann bei
-350C 3,3 g Isopropylbromid zugetropft. Die ammo-
niakalische Lösung wurde 2 Stunden unter Rückfluß gekocht. Dann wurde etwas Ammoniumchlorid zugesetzt
und das Ammoniak verdampft. In dem Maße, wie das Ammoniak verdampfte, wurde Äther zugegeben.
Nachdem alles Ammoniak verdampft war,
wurde die Ätherlösung mit Wasser (100 bis 200 ml) gewaschen und anschließend mit verdünnter Salzsäure
ausgeschüttelt. Die saure, wäßrige Phase wurde mit konzentrierter Natronlauge alkalisch gemacht. Die
abgeschiedene Base wurde in Benzol aufgenommen,
mit Wasser neutral gewaschen und über Natriumsulfat getrocknet. Nach Verdampfen des Lösungsmittels
wurden 4,2 g Rohbase erhalten, die in 20 ml Benzol gelöst über 20 g Aluminiumoxid der Aktivitätsstufe II
filtriert wurden. In das Filtrat wurde Chlorwasserstoffgas eingeleitet und der Niederschlag abfiltriert. Das
erhaltene Hydrochlorid war mit dem des Beispiels 1 identisch. Der Mischschmelzpunkt zeigte keine Depression.
Die Ausbeute betrug 0,3 g.
