Description
Die vorliegende Erfindung betrifft ein verbessertes
Verfahren zur Herstellung von 3-Amino-1-hydroxypro
pan-1,1-diphosphonsäure der Strukturformel
3-Amino-1-hydroxypropan-1,1-diphosphonsäure ist ein
nützlicher Wirkstoff eines Arzneimittels gegen
Osteoporose. Unter Osteoporose wird bekanntlich die
quantitative Verminderung des Knochengewebes bei
erhaltener Knochenstruktur durch gesteigerten Knochen
abbau und/oder verminderten -anbau verstanden, bis
hin zu einem abnormen Grad. Sie tritt überwiegend bei
älteren Menschen auf, besonders bei Frauen in den
Wechseljahren. Symptome dieser Krankheit können
starke Schmerzen, ähnlich denen bei Hexenschuß, die
Verkleinerung der Gestalt und Knochenbrüche, bezogen
auf die jeweilige Ernsthaftigkeit des Schwundes des
Mineralgehaltes der Knochen, sein.
Es ist bekannt, daß mit dem Ziel, die Behandlung von
Osteoporose positiv zu beeinflussen, bereits viele
Studien über die effiziente Herstellung von 3-Amino-1-hy
droxypropan-1,1-diphosphonsäure, bekannt als her
kömmliches, wirksames Arzneimittel, durchgeführt
wurden. So offenbart das DE 21 30 794 ein Verfahren
der Herstellung aus einer Lösung von β-Alanin, Phos
phortrichlorid und Phosphorsäure in Chlorbenzol.
Jedoch sind Ertrag und Reinheit des Endproduktes sehr
gering, und ein unbekanntes gelblich-rotes amorphes
Phosphoroxid tritt als Nebenprodukt auf. Die Entnahme
des Nebenproduktes gestaltet sich nicht nur schwie
rig, sie führt außerdem zu hohen Produktionskosten.
Einem anderen Verfahren zufolge wird 3-Amino-1-hy
droxypropan-1,1-diphosphonsäure hergestellt, indem
β-Alanin mit Phosphoroxychlorid und Phosphorsäure in
Chlorbenzol vermischt werden, mit nachfolgender
Hydrolyse. Dieses Verfahren ist ebenfalls unbefriedi
gend in Bezug auf Ausbeute und Reinheit des Endproduk
tes.
Aufgabe der vorliegenden Erfindung ist es daher, ein
Verfahren zur Herstellung von 3-Amino-1-hydroxypro
pan-1,1-diphosphonsäure bereitzustellen, das substan
tiell die Nachteile und Beschränkungen der beschrie
benen Verfahren unterbindet.
Erfindungsgemäß wird die Aufgabe durch ein verbesser
tes Verfahren zur Herstellung von 3-Amino-1-hydroxy
propan-1,1-diphosphonsäure gemäß den Merkmalen des
Anspruches 1 gelöst.
Das erfindungsgemäße Verfahren hat den Vorzug, daß
die Ausbeute an Endprodukt bei hoher Reinheit gestei
gert wird.
Weitere besondere Merkmale und Vorteile der Erfin
dung werden im folgenden beschrieben. Sie werden
teilweise unmittelbar aus der Beschreibung ersicht
lich oder können aus den praktischen Beispielen der
Anwendung der Erfindung in Erfahrung gebracht werden.
Bevorzugte Ausgestaltungen des Verfahrens ergeben
sich aus den Merkmalen der Unteransprüche 2 bis 5.
Um die genannten und anderen Vorteile der vorliegen
den Erfindung, wie dargestellt und ausführlich be
schrieben, zu erreichen, umfaßt die Herstellung von
3-Amino-1-hydroxypropan-1,1-diphosphonsäure folgende
Schritte: Mischung von N-Alkyloxycarbonyl-β-Alanin,
Phosphortrichlorid und Phosphorsäure im Verhältnis
1 : 1 : 1 bis 1 : 3 : 3 im Lösungsmittel Xylol bei
70°C bis 140°C, nachfolgend durch Hydrolyse Erhalt
des Endproduktes 3-Amino-1-hydroxypropan-1,1-diphosphon
säure.
Sowohl die vorangegangene überblickgebende, als auch
die folgende detaillierte Beschreibung des erfindungs
gemäßen Verfahrens sind beispielgebend und erklärend
und lassen weitere Erläuterungen der Erfindung gemäß
den Ansprüchen überflüssig werden.
Hierzu wird nun detailliert auf die gewünschte Dar
stellung der vorliegenden Erfindung hingewiesen.
Der Ausgangsstoff ist N-Alkyloxycarbonyl-β-Alanin,
welches aus der Reaktion von β-Alanin mit Chlorkohlen
säureester hervorgeht. 1 bis 3 mol N-Alkyloxycar
bonyl-β-Alanin, 1 bis 3 mol Phosphorpentachlorid oder
Phosphortrichlorid und 1 bis 3 mol Phosphorsäure
werden im Verhältnis 1 : 1 : 3 bis 1 : 3 : 3, bevor
zugt aber im Verhältnis 1 : 1 : 1 bei Temperaturen
zwischen 70°C bis 140°C miteinander vermischt.
Der Chlorkohlensäureester ist Chlorkohlensäureme
thylester, Chlorkohlensäureäthylester, Chlorkohlensäu
reisopropylester, Chlorkohlensäurepropylester, Chlor
kohlensäureallylester, Chlorkohlensäure-t-butylester
oder Chlorkohlensäurebenzylester.
Das Reaktionsgemisch wird bei einer Wassertemperatur
von 25°C bis 110°C hydrolysiert. Nach Abtrennen der
wäßrigen Phase erhält man aus dieser 3-Ami
no-1-hydroxypropan-1,1-diphosphonsäure.
Methanol kann hinzugegeben werden, um eine Erhöhung
der Ausbeute an 3-Amino-1-hydroxypropan-1,1-diphosphon
säure zu erreichen. Die zugegebene Menge an
Methanol beträgt ca. 150 ml bis 250 ml, vorzugsweise
aber 200 ml. Das Verhältnis von Xylol zu Methanol
beträgt 2,0 bis 3,2 : 1, vorzugsweise 2,5 : 1.
N-Alkyloxycarbonyl-β-Alanin kann beispielsweise nach
folgender Reaktion hergestellt werden:
wobei R für CH₃, CH₃CH₂, (CH₃)₂CH, CH₃CH₂CH₂,
CH₂=CHCH₂, (CH₃)₃C oder PhCH₂ steht.
1N NaOH wird zu der Lösung von 17.8 g (0,2 mol)
β-Alanin und 1N NaOH (200 ml) getröpfelt, damit sich
der pH-Wert auf einen Wert zwischen 9.0 und 9.5
einstellt. 105 mol Chlorkohlensäure werden für 1
Stunde 2-3 mol zugegeben. Die Temperatur des erhalte
nen Gemisches wird auf 0°C herabgesetzt. Daraufhin
wird das Gemisch dreimal mit 100 ml Äther extrahiert
und der pH-Wert mit Hilfe von Phosphorsäure auf
einen Wert von 1 eingestellt. Die wäßrige Phase wird
mit Natriumchlorid gesättigt und dreimal mit 100 ml
Methylenchlorid extrahiert. Nach der Verdunstung der
organischen Bestandteile erhält man 85% Ausbeute an
N-Alkyloxycarbonyl-β-Alanin.
Im folgenden soll die Erfindung an Ausführungsbeispie
len näher erläutert werden:
123.00 g (1.5 mol) Phosphorsäure werden zu der Suspen
sion von Phosphorpentachlorid (312.36 g, 1.5 mol) in
Xylol (500 ml) gegeben, um eine exotherme Reaktion
hervorzurufen. Die entstandene transparente Lösung
wird bis auf Raumtemperatur abgekühlt und 147.13 g
(1.5 mol) bereitgestelltes N-Alkyloxycarbonyl-β-Ala
nin werden der gekühlten Lösung zugegeben. Nachdem
sie 15 min. gerührt und langsam auf 100°C erhitzt
wurde, wird die Lösung für 5 Stunden unter Rückfluß
gehalten. Daraufhin wird das Gemisch auf Raumtempera
tur abgekühlt und 1000 ml Wasser werden ihm zugege
ben. Abermals wird das Gemisch langsam auf eine
Temperatur von 100°C erhitzt und für 5 Stunden einem
Rückfluß ausgesetzt. Der wäßrige Teil wird dann vom
Gemisch getrennt und gekühlt, um eine 65%ige Ausbeu
te an Endprodukt zu erhalten.
Der Nachweis des Endproduktes erfolgt durch Messung
des Infrarotspektrums unter Verwendung von KBr-Granu
lat mit Hilfe des Infrarotspektralphotometers Modell
621, hergestellt von Perking-Elmer. Nach einer weite
ren Messung des Kernresonanzspektrums (NMR) mit Hilfe
von D₂O und einem EM-390-90 MHz Spektralphotometer
wurde das Endprodukt 3-Amino-1-hydroxypropan-1,1-di
phosphonsäure identifiziert. Die Ergebnisse der Analy
se waren folgende:
IR: starkes Maximum bei 2900 bis 3200 cm-1, Maximum
bei 1600 bis 540 cm-1,
NMR: Multiplett bei 2.2 ppm, Triplett bei 3.3 ppm.
NMR: Multiplett bei 2.2 ppm, Triplett bei 3.3 ppm.
Die Radionuklidreinheitsprobe des Produktes wurde mit
Hilfe von β-Alanin (1-14C) von ICN Chemical & Radioi
sotope Division als radioaktiv markiertes Startmate
rial durchgeführt. Die spezifische Aktivität von
β-Alanin (1-14C) betrug 13.9 mCi/mmol. Um die Rein
heit des Endproduktes zu messen, wurde ein Detektor,
Modell 7220/21 von Packard, zur radioaktiven Messung
genutzt, ein Flüssigkeits-Szintillationszähler LS
9000 zur Feststellung der spezifischen Aktivität und
eine polygram cell 300 Celluloseplatte für die
Thin-Layer-Chromatographie (TLC). Als Ergebnis konnte
das Endprodukt als alleinige reine Substanz identifi
ziert werden. Es wurde nur ein einziges Maximum für
das Produkt festgestellt.
Wasser : Ethanol : Aluminiumhydroxid = 80 : 10 : 15,
r.f. 0.8;
Wasser : Aceton = 70 : 30, r.f. 0.2;
Ameisensäure : Wasser = 1 : 1, r.f. 1.0.
Wasser : Aceton = 70 : 30, r.f. 0.2;
Ameisensäure : Wasser = 1 : 1, r.f. 1.0.
147,13 g (1 mol) N-Alkyloxycarbonyl-β-Alanin werden
500 ml Xylol zugegeben, gefolgt von der Zugabe von
137,33 g (1 mol) Phosphortrichlorid und 32,00 g (1
mol) Phosphorsäure. Das Gemisch wird anschließend
langsam auf 100°C erhitzt und für 5 Stunden unter
Rückfluß gehalten. Nach Abkühlung aus Raumtemperatur
werden dem Gemisch 1000 ml Wasser zugegeben. Das
Gemisch wird daraufhin wiederum auf 100°C erhitzt und
für 5 Stunden unter Rückfluß gehalten. Die wäßrige
Phase wird dann vom Gemisch abgetrennt und der Restbe
stand unter vermindertem Druck konzentriert, um das
Endprodukt in der wäßrigen Phase vorliegend, in
63%iger Ausbeute zu erhalten.
Durch IR- und NMR-Messungen wurde das Endprodukt als
3-Amino-1-hydroxypropan-1,1-diphosphonsäure identifi
ziert.
Um die Kristallisation zu unterstützen, werden 200 ml
Methanol zu der nach der Hydrolyse abgetrennten
wäßrigen Phase der Lösung zugegeben. Das weitere
Verfahren wird, wie im Ausführungsbeispiel 1 be
schrieben, durchgeführt, um das Endprodukt zu erhal
ten. Nach IR- und NMR-Messungen wurde als Endprodukt
3-Amino-1-hydroxypropan-1,1-diphosphonsäure festge
stellt; die Ausbeute betrug 72.0%.
Um die Kristallisation zu unterstützen, werden wieder
um 200 ml Methanol der nach der Hydrolyse abgetrenn
ten wäßrigen Phase der Lösung zugegeben. Anschlie
ßend wird das Verfahren wie in Ausführungsbeispiel 2
beschrieben weitergeführt, um das Endprodukt zu
erhalten. Durch IR- und NMR-Messungen wurde als
Endprodukt 3-Amino-1-hydroxypropan-1,1-diphosphonsäu
re identifiziert; die Ausbeute betrug 69.1%.
Es wird Chlorbenzol an Stelle von 500 ml Xylol als Re
aktionsmedium genutzt. Das Verfahren wird dann je
weils wie in den Ausführungsbeispielen 1 und 2 be
schrieben durchgeführt, um 3-Amino-1-hydroxypro
pan-1,1-diphosphonsäure zu erhalten; die Ausbeute belief
sich auf 35% bzw. 32%.
Aus den Ausführungsbeispielen 1 bis 4 und den verglei
chenden Beispielen wird ersichtlich, daß die Ausbeute
an Endprodukt geringer wird, sobald Xylol als Reak
tionsmedium durch Chlorbenzol ausgetauscht wird. Man
nimmt an, daß der Produktionsertrag an Nebenprodukt
bei Verwendung von Chlorbenzol an Stelle von Xylol
größer ist und die Trennung der wäßrigen Phase,
welche das Endprodukt enthält, von der organischen
Phase, enthaltend das Nebenprodukt, nur unzureichend
vor sich geht. Die spezifische Dichte der wäßrigen
Phase, das Produkt enthaltend, wird somit größer als
die des Wassers und ähnelt der von Chlorbenzol, in
der organischen Phase vorliegend. Letztendlich stellt
man zwischen der spezifischen Dichte der wäßrigen
Phase mit dem Endprodukt und der der organischen
Phase mit dem Nebenprodukt eine Ähnlichkeit fest,
welche die effiziente Trennung der beiden Phasen er
schwert.
Wie in den Ausführungsbeispielen 3 und 4 aufgezeigt,
wird die Ausbeute an 3-Amino-1-hydroxypropan-1,1-di
phosphonsäure erhöht, wenn Methanol zugegeben wird.
Entsprechend der vorliegenden Erfindung wird die
Menge an unidentifizierten Nebenprodukte vermindert
und eine effektive Phasentrennung der Nebenprodukte,
gelöst in einer Xylolschicht, von dem Endprodukt in
der wäßrigen Schicht, erreicht. Folglich kann das
Endprodukt, hochreines 3-Amino-1-hydroxypropan-1,1-di
phosphonsäure, mit hoher Ausbeute hergestellt werden.
Man kann davon ausgehen, daß die Produktionskosten
durch den Gebrauch von Xylol als Reaktionsmittel ver
ringert werden, weil es nicht so teuer ist wie Chlor
benzol.
Es ist offensichtlich, daß aufgrund des vorliegenden
erfinderischen Verfahrens verschiedene Abwandlungen
und Variationen der Herstellung von 3-Amino-1-hydroxy
propan-1,1-diphosphonsäure möglich sind, ohne den An
wendungsbereich der Erfindung zu mißachten. Die vor
liegende Erfindung umfaßt daher auch die Abwandlungen
und Variationen des Verfahrens, soweit sie mit den Pa
tentansprüchen und den Äquivalenten der Merkmale über
einstimmen.
Claims (5)
1. Verfahren zur Herstellung von 3-Amino-1-hydroxypro
pan-1,1-diphosphonsäure, umfassend folgende Schrit
te:
- - Mischen von N-Alkyloxycarbonyl-β-Alanin, Phos phorchlorid und Phosphorsäure im molaren Verhält nis von 1 : 1 : 1 bis 1 : 3 : 3 in dem Lösungs mittel Xylol bei Temperaturen von 70-140°C;
- - Hydrolyse und anschließende
- - Phasentrennung zum Erhalt von 3-Amino-1-hydro xypropan-1,1-diphosphonsäure als Endprodukt.
2. Verfahren nach Anspruch 1, dadurch gekennzeichnet,
daß 150 ml bis 250 ml Methanol hinzugefügt werden,
im Verhältnis von Xylol zu Methanol
2,0 bis 3,2 : 1.
3. Verfahren nach Anspruch 1, dadurch gekennzeich
net, daß N-Alkyloxycarbonyl-β-Alanin, Phosphorchlo
rid und Phosphorsäure in einem molaren Verhältnis
von 1 : 1 : 1 gemischt werden.
4. Verfahren nach Anspruch 1, dadurch gekennzeich
net, daß als Phosphorchlorid Phosphorpentachlorid
oder Phosphortrichlorid eingesetzt werden.
5. Verfahren nach Anspruch 3, dadurch gekennzeich
net, daß als Phosphorchlorid Phosphorpentachlo
rid oder Phosphortrichlorid eingesetzt werden.
