Verfahren zur Herstellung von 4-Cyano-3-hvdroxybuttersäureestern
Die Erfindung betrifft ein Verfahren zur Herstellung von 4-Cyano-3 -hydroxybuttersäureestem durch Umsetzung von 4-substituierten 3-Hydroxy-buttersäureestem mit einem Cyanidsalz in Gegenwart eines weiteren Salzes.
4-Cyano-buttersäureester werden als wichtige Zwischenprodukte für die Herstellung von pharmazeutischen und agrochemischen Wirkstoffen verwendet. 4-Cyano-3-hydroxybuttersäureester werden als Zwischenprodukt für die Herstellung von Cholesterinsenkern wie beispielsweise Atorvastatin eingesetzt, vorzugsweise in einer enantiomerenangereicherten Form.
Bekannte Herstellungsmethoden für Cyanohydroxybuttersäureester gehen meist von Halogen- hydroxybuttersäureestem aus und setzen diese mit Cyanidsalzen um. In EP-A-0 970 947 wird beispielsweise die Umsetzung von Ethyl-4-chlor-3-hydroxybutyrat in Formamid und Wasser mit wässriger Natriumcyanidlösung bei 80 °C zu Ethyl-4-cyano-3-hydroxybutyrat in Reaktionsrohausbeuten bis 77.5 % für eine Batch-Umsetzung beschrieben. Nachteilig ist hierbei die Gegenwart von Wasser sowie der stark basische pH Wert einer solchen Reaktionslösung, der in beträchtlichem Maße zur Verseifung des Carbonsäureesters führt und dadurch die Ausbeute mindert. Femer wird in der EP-A-0 970 947 beschrieben, dass als unerwünschte Nebenprodukte ungesättigte Verunreinigungen in Form von Crotonsäureestem gebildet werden, die sich nur schwer von dem gewünschten Endprodukt des 4-Cyano-3-hydroxybuttersäureesters abtrennen lassen. Bereitgestellt wird gemäß EP-A-0 970 947 eine neue Methode zur Abtrennung dieser Verunreinigungen, indem ein Gemisch, welches vor allem den 4-Cyano-3-hydroxybuttersäureester der Formel
sowie in gewissen Mengen die unerwünschte ungesättigte Verbindung der Formel HOCH2-CH=CH-COOR enthält, mit Salzen der schwefeligen Säure, bevorzugt deren Alkali- und Erdalkalimetallsalzen, behandelt wird. Diese Salze der schwefeligen Säure wirken als sogenannte addition reagents und addieren an die Doppelbindung des Crotonsäureesters unter Bildung des Additionsprodukts ,

welches sich bezüglich Siedepunkt, Löslichkeit und anderen Eigenschaften so deutlich vom gewünschten Produkt unterscheidet, dass eine Abtrennung leicht möglich ist.
Es ist gemäß EP-A- 0 907 947 auch möglich, die Umsetzung der 4-substituierten 3-Hydroxy- buttersäureester mit einem Cyanidsalz direkt in Gegenwart der Salze der schwefligen Säure durchzuführen, so dass der Crotonsäureester sofort in-situ weiter umgesetzt wird. Nachteilig ist aber auch bei dieser Variante die Gegenwart von Wasser sowie der stark basische pH Wert der Reaktionslösung und die dadurch auftretende Verseifung des Carbonsäureesters.
Aufgrund der hohen Bedeutung der Cyanohydroxybuttersäureester als Agro- und Pharmazwischen- produkte bestand Bedarf nach einem neuen Verfahren zur Herstellung von Cyanohydroxybutter- säureestem in hohen Ausbeuten.
Diese Aufgabe wird gelöst durch ein Verfahren zur Herstellung von Verbindungen der allge- meinen Formel (I),
wobei
R geradkettiges oder verzweigtes Cι-C20-Alkyl, C3-Cg-Cycloalkyl, substituiertes oder un- substituiertes Aryl oder Benzyl bedeutet,
und die Verbindungen der allgemeinen Formel (I) racemisch oder enantiomerenangereichert vorliegen können,
indem man Verbindungen der allgemeinen Formel (II)
worin
R die für die allgemeine Formel (I) genannte Bedeutung besitzt,
X Chlor, Brom, Iod, Methansulfonyloxy oder Phenylsulfonyloxy bedeutet und
die Verbindungen der allgemeinen Formel (II) racemisch oder enantiomerenangereichert vorliegen können,
mit einem Cyanidsalz in einem organischen Lösungsmittel umsetzt, dadurch gekennzeichnet, dass man die Umsetzung in Gegenwart eines oder mehrerer Salze der Formel (HI), welche von dem Cyanidsalz verschieden sind, durchführt

In den allgemeinen Formeln (I) und (II) kann der Rest R für geradkettiges oder verzweigtes Cι-C2o-, bevorzugt Q-Ciz-Alkyl und besonders bevorzugt Ci- -Alkyl stehen. Dieser Alkylrest ist gegebenenfalls weiter substituiert, beispielsweise durch Ci-GrAlkoxy-Restβ. R steht geeigneterweise für Methyl, Ethyl, n-Propyl, Isopropyl, n-Butyl, tert.-Butyl, n-Pentyl, n-Hexyl, n-Heptyl, n- Octyl, iso-Octyl, n-Decyl, n-Dodecyl, n-Hexadecyl oder n-Octadecyl.
R kann auch für einen C3-C8-Cycloalkyl-Rest, bevorzugt für Cyclopentyl oder Cyclohexyl stehen.
R kann femer Aryl bedeuten. Aryl umfasst carbocyclische Reste und heteroaromatische Reste, in denen keines, ein, zwei oder drei Gerüstatome pro Cyclus, im gesamten Rest mindestens jedoch ein Gerüstatom, durch Heteroatome ausgewählt aus der Gruppe Stickstoff, Schwefel und Sauer- stoff ersetzt sind. Bevorzugt sind Cs-C^-Aryl-Reste, besonders bevorzugt sind Phenyl, Pyridyl, o-, m-, oder p-Tolyl, sowie Anthracenyl.
R kann femer einen Benzyl-Rest darstellen.
In der allgemeinen Formel (II) bedeutet X Chlor, Brom, Iod, Methansulfonyloxy oder Phenylsulfonyloxy. Der Phenylsulfonyloxy-Rest kann dabei auch substituiert sein, bewährt haben sich als weitere Substituenten beispielsweise AlkyLreste. Bevorzugt steht X für Chlor und Brom, besonders bevorzugt ist Chlor.

Die Verbindungen der allgemeinen Formel (II) werden bevorzugt enantiomerenangereichert eingesetzt, besonders bevorzugt ist (S)-Ethyl-4-chlor-3-hydroxybutyrat.
„Enantiomerenangereichert" im Sinne der Erfindung bedeutet die enantiomeren-reinen Verbindungen oder Mischungen der Enantiomere, in denen ein Enantiomer in einem Enantiomeren- überschuss, auch ee („enantiomeric excess") genannt, im Vergleich zum anderen Enantiomer vorliegt.
Verbindungen der Formel (H) können beispielsweise durch Carboalkoxylierung des korrespon- dierenden Epihalohydrins (Journal of Organic Chemistry 1967, 32, 3888), durch chemische oder enzymatische Reduktion des korrespondierenden ß-Ketoesters (Tetrahedron Letters 1994, 35, 8119) oder durch Sulfonierung des korrespondierenden ß,γ-Dihydroxyesters hergestellt werden.
Durch das erfindungsgemäße Verfahren können Verbindungen der allgemeinen Formel (I) hergestellt werden, die sich beispielsweise aus den zuvor genannten Verbindungen der allgemeinen Formel (IT) ergeben, indem der Rest X durch eine Cyano-Gruppe ausgetauscht wird. Beispielsweise seien genannt: Methyl-4-cyano-3-hydroxybutyrat, Ethyl-4-cyano-3 -hydroxybutyrat, n- Propyl-4-cyano-3-hydroxy-butyrat, i-Propyl-4-cyano-3-hydroxybutyrat, s-Butyl-4-cyano-3- hydroxybutyrat, t-Butyl-4-cyano-3-hydroxy-butyrat und n-Butyl-4-cyano-3-hydroxybutyrat. Besonders bevorzugt ist Ethyl-4-cyano-3 -hydroxybutyrat.
Die Verbindungen der allgemeinen Formel (I) werden bevorzugt als enantiomerenangereicherte
Verbindungen hergestellt, besonders bevorzugt ist (R)-Ethyl-4-cyano-3-hydroxybutyrat.
Als Cyanidsalze kommen beispielsweise Alkali und Erdalkalicyanide, Ammonium-, Phosphonium- oder Trialkylsilylcyanide in Frage. Bevorzugt sind beispielsweise Natriumcyanid, Kaliumcyanid, Lithiumcyanid, Cäsiumcyanid, Magnesiumcyanid, Calciumcyanid, Tetrabutylammoniumcyanid, Tetraphenylammoniumcyanid, Tetrabutylphosphoniumcyanid, Tetraphenylphosphoniumcyanid oder Trimethylsilylcyanid. Besonders bevorzugt sind Natriumcyanid und Kaliumcyanid. Insbesondere bevorzugt ist Natriumcyanid.
Die Cyanidsalze können auch in situ erzeugt werden, bevorzugt aus anderen Cyanidquellen, wie z.B. Acetoncyanhydrin, in Gegenwart einer Base. Die Cyanidsalze können einzeln oder in Mischung eingesetzt werden, in Mengen von 0,1 - 100 Äquivalenten bezogen auf die Verbindung der allgemeinen Formel (II), bevorzugt 0,5 - 5 Äquivalenten, besonders bevorzugt 0,9 - 2 Äquivalenten. Das Cyanidsalz kann fest oder gelöst vorgelegt oder als Feststoff oder gelöst zudosiert werden. Es ist bevorzugt, das Cyanid vorzulegen.
Das Salz der allgemeinen Formel (HI) [Kj Am x n H20] besteht aus ein oder mehreren organischen oder anorganischen Kationen und aus einem oder mehreren Anionen organischer oder an- organischer Säuren mit einem pKa von 1-10, ausgenommen der schwefligen Säure, wobei die Indizes 1, m = 1,2 oder 3 und n = 0-20 sein können. Bevorzugt sind Salze der allgemeinen Formel (HI), bei denen n = 0-12 beträgt.
Als Kationen können beispielsweise Alkalimetall-, Erdalkalimetallkationen oder Ammonium- Ionen fungieren, bevorzugt sind Natrium, Kalium und Ammonium.
Als Anionen haben sich besonders Anionen organischer Säuren bewährt, bevorzugt sind C2-Cι2- Mono-, Di- oder Tri-Carboxylate, besonders bevorzugt Phthalate, Citrat oder Acetat.
Als Anionen haben sich femer Anionen anorganischer Säuren bewährt, bevorzugt sind Phosphate,
Hydrogenphosphate, DihydrogenphoΞphate oder Borale.
Bevorzugt werden im erfindungsgemäßen Verfahren als Salze Natriumacetat, Natriumacetat Tri- hydrat, Ammoniumacetat, Kaliumaeetat, Natriumcitrat, Kaliumeitrat, Kaliumhydrogenphthalat, Natriumdiethylbarbiturat, Natriumborat, Kaliumborat, Natriumglycinat, Kaliumglycinat, Ammo- niumdihydrogenphosphat, Kaliumdihydrogenphosphat, Dikaliumhydrogenphosphat, Dinatrium- hydrogen-phosphat, Natriumdihydrogenphosphat, Natriumdihydrogenphosphat-Monohydrat oder Natriumdihydrogenphosphat-Dihydrat eingesetzt. Besonders bevorzugt sind Natriumdihydrogen- phosphat, Natriumdihydrogenphosphat-Monohydrat und Natriumdihydrogenphosphat-Dihydrat.
Das Salz K] Am x n H20 oder Mischungen verschiedener dieser Salze werden üblicherweise in Mengen von 0,05 - 20 Äquivalenten pro Äquivalent des Cyanidsalzes zugesetzt. Bevorzugt sind
0,2 - 5 Äquivalente, besonders bevorzugt sind 0,7 bis 2 Äquivalente. Das Salz oder die Mischung verschiedener Salze kann vorgelegt oder zudosiert werden. Möglich ist es auch, das Salz aus der Säure und der entsprechenden Base in situ zu generieren.
Als organische Lösungsmittel können im erfindungsgemäßen Verfahren beispielsweise Alkohole, bevorzugt Methanol, Ethanol, i-Propanol, Ethylenglykol oder n-Butanol, Ketone, Stickstoff enthaltende organische Lösungsmittel, bevorzugt Dimethylformamid, Methylformarnid, Formamid, Acetonitril oder Acetamid, halogenierte Lösungsmittel, bevorzugt Methylenchlorid oder Chloroform, oder Schwefel-enthaltende Lösungsmittel, bevorzugt Dimethylsulfoxid, eingesetzt werden.
Besonders bevorzugt sind Ethanol, Methylformarnid und Formamid, insbesondere bevorzugt ist Formamid.
Diese organischen Lösungsmittel können als alleinige Lösungsmittel eingesetzt werden. Möglich ist es aber auch, diese in Kombination mit Wasser einzusetzen. Bevorzugt ist der Einsatz von Mischungen aus einem großen Überschuss an organischem Lösungsmittel und einer geringeren Menge Wasser. Besonders bevorzugt ist der Einsatz von 0 - 10 ol Wasser auf 1 1 organisches Lösungsmittel.
Bevorzugt ist femer der Einsatz von einem organischen Lösungsmittel allein oder aber einer Mischung organischer Lösungsmittel. Besonders bevorzugt ist der Einsatz von Formamid, welches wasserfrei oder aber mit geringen Mengen Wasser versetzt ist.
Das organische Lösungsmittel wird üblicherweise in Mengen von 0,1-10 kg pro Mol Edukt der Formel (II) eingesetzt, bevorzugt sind 0,25-5 kg pro Mol Edukt und besonders bevorzugt sind 0,5 - 2 kg pro Mol Edukt.
Die Reaktion wird üblicherweise bei einer Temperatur im Bereich von 0 - 200°C durchgeführt, bevorzugt im Bereich von 25 - 125 °C und besonders bevorzugt im Bereich von 50 - 100°C.
Die Reaktion kann unter Druck oder unter Vakuum durchgeführt werden (z.B. für Vakuumsiede- kühlung), bevorzugt ist jedoch die Durchführung unter Normaldruck.
In einer typischen Ausführung des erfindungsgemäßen Verfahrens werden das Salz oder die Mischung der Salze der allgemeinen Formel (DT) und das Cyanidsalz in dem Lösungsmittel vorgelegt und auf die Reaktionstemperatur gebracht. Dann wird die Verbindung der allgemeinen Formel (JJ) unter Rühren zudosiert und anschließend für eine gewisse Zeit gerührt. Die Aufarbeitung kann nach dem Fachmann bekannten, gängigen Methoden erfolgen, z.B. durch Extraktion oder Destillation. Es hat sich beispielsweise bewährt, das Reaktionsgemisch auf 20°C zu bringen, an-
schließend Wasser sowie ein Extraktionsmittel zuzusetzen, das Gemisch zu filtrieren und die wässrige Phase wiederholt zu extrahieren. Die vereinigten organischen Extrakte werden eingeengt und das Konzentrat anschließend einer fraktionierten Feindestillation unterworfen.
Die Reaktion kann auch so durchgeführt werden, dass entweder alle im erfindungsgemäßen Ver- fahren einzusetzenden Substanzen und Lösungsmittel vorgelegt und auf Reaktionstemperatur gebracht werden oder ein oder mehrere Reaktionspartner zudosiert werden; beispielsweise können auch parallel oder in Folge das Cyanidsalz, das Salz der Formel (UI) und/oder die Verbindung der allgemeinen Formel (Tf) zudosiert werden.
Das erfindungsgemäße Verfahren lässt sich nicht nur als Batchverfahren, sondern auch kontinuierlich durchführen.
Das erfindungsgemäße Verfahren zeichnet sich dadurch aus, dass die gewünschten 4-Cyano-3- hydroxybuttersäureester in hoher Ausbeute erhalten werden können. Das Verfahren hat femer den Vorteil, dass unerwünschte Nebenkomponenten wie freie Säuren und Lactone nur in extrem geringen Mengen gebildet werden, wodurch sich die bevorzugte destillative Aufarbeitung deutlich vereinfacht.
Beispiele
Beispiel 1
Es werden in der angegebenen Reihenfolge 2265 g Formamid, 227,4 g Natriumcyanid und 507,9 g Natriumdihydrogenphosphat Monohydrat im Reaktions-gefäß vorgelegt und mit weiteren 964 g Formamid versetzt. Dieses Reaktionsgemisch wird unter Rühren auf 65 °C geheizt, anschließend in 15 Minuten 524 g (S)-Ethyl-4-chlor-3-hydroxybutyrat zudosiert und die Leitung mit 170 g Formamid nachgespült. Nach Dosierende wird das Reaktionsgemisch für 60 Minuten gerührt und das Reaktionsgemisch auf Raumtemperatur abgekühlt. Das Reaktionsgemisch wird gaschromato- graphisch untersucht und enthält 400 g (R)-Ethyl-4-cyano-3-hydroxybutyrat, entsprechend 85 % der Theorie.
Dann werden 3500 g Wasser und 2700 g Ethylacetat zu der Reaktionsmischung hinzugegeben und 15 Minuten nachgerührt. Das Gemisch wird filtriert und die organischen Phasen des Filtrats getrennt. Die wässrige Phase wird noch fünf Mal mit Ethylacetat extrahiert und das Lösungsmittel aus den vereinigten organischen Phasen entfernt. Es werden 1050 g eines Rohproduktes erhalten, das 365 g (R)-Ethyl-4-cyano-3-hydroxybutyrat enthält, entsprechend 76,4 % der Theorie.
Beispiel 2
Es werden 74 g Natriumcyanid und 191 g Natriumdihydrogenphosphat Dihydrat in 1 L Formamid unter Rühren vorgelegt und auf 65°C erhitzt. Anschließend werden 167 g Ethyl-4-chlor-3-hydroxy- butyrat innerhalb von 15 Minuten zudosiert und das Reaktionsgemisch 1 h gerührt. Es wird auf Raumtemperatur abgekülilt und 500 ml Wasser und 800 ml Ethylacetat zugegeben. Nach Filtration des Reaktionsgemischs werden die Phasen getrennt und die wässrige Phase noch 5 mal mit Ethylacetat extrahiert. Die vereinigten organischen Phasen werden vom Lösungsmittel befreit, wobei ein Rohprodukt erhalten wird, das 113 g Ethyl-4-cyano-3-hydroxybutyrat enthält, entsprechend 72 % der Theorie.
Beispiel 3
Es werden 74 g Natriumcyanid und 167 g Natriumdihydrogenphosphat Monohydrat in einer Mischung aus 500 ml Formamid und 150 ml Ethanol unter Rühren vorgelegt und auf 65°C erhitzt. Anschließend werden 167 g Ethyl-4-chlor-3-hydroxybutyrat innerhalb von 30 Minuten zudosiert und das Reaktionsgemisch für 1 Stunde gerührt. Es wird auf Raumtemperatur abgekühlt und 500 ml Wasser und 800 ml Ethylacetat zugegeben. Nach Filtration des Reaktionsgemischs werden die Phasen getrennt und die wässrige Phase noch fünf Mal mit Ethylacetat extrahiert. Die ver-
einigten organischen Phasen werden vom Lösungsmittel befreit, wobei ein Rohprodukt erhalten wird, das 105 g Ethyl-4-cyano-3-hydroxybutyrat enthielt, entsprechend 70 % der Theorie.
Beispiel 4
Es werden 74 g Natriumcyanid und 191 g Natriumdihydrogenphosphat Dihydrat in 1 L Formamid und 167 g Ethyl-4-chlor-3 -hydroxybutyrat unter Rühren vorgelegt und auf 65°C erhitzt. Die Temperatur steigt auf 82°C und klingt dann wieder auf 70°C ab. Bei dieser Temperatur wird das Reaktionsgemisch 1 Stunde gerührt. Es wird auf Raumtemperatur abgekühlt und 500 ml Wasser und 800 ml Ethylacetat zugegeben. Nach Filtration des Reaktionsgemischs werden die Phasen getrennt und die wässrige Phase noch fünf Mal mit Ethylacetat extrahiert. Die vereinigten organischen Phasen werden eingeengt, wobei ein Rohprodukt erhalten wird, das 118 g Ethyl-4- cyano-3 -hydroxybutyrat enthält, entsprechend 75 % der Theorie.
Beispiel 5
Es werden 74 g Natriumcyanid und 174 g Natriumdihydrogenphosphat Monohydrat in 800 ml Formamid unter Rühren vorgelegt und auf 70°C erhitzt. Anschließend werden 167 g Ethyl-4-chlor- 3-hydroxybutyrat innerhalb von 1 Stunde zudosiert und das Reaktionsgemisch für 30 Minuten gerührt. Es wird auf Raumtemperatur abgekühlt und 400 ml Wasser und 700 ml Ethylacetat zugegeben.
