Abstract
Rekombinante probiotische Bakterien, umfassendeine oder mehrere Nucleinsäuresequenzen, die für einen ersten heterologen Faktor kodieren, eine oder mehrere Nucleinsäuresequenzen, die für einen zweiten heterologen Faktor kodieren und vorzugsweise eine oder mehrere Nucleinsäuresequenzen, die für einen dritten heterologen Faktor kodieren, mit der Maßgabe, dass der erste Faktor, der zweite Faktor und der dritte Faktor jeweils funktionell unterschiedlich voneinander sind,wobei es sich beim ersten Faktor um einen Wachstumsfaktor handelt, wobei der zweite Faktor aus der aus M2-polarisierenden Faktoren bestehenden Gruppe ausgewählt ist und wobei der dritte Faktor vorzugsweise aus der aus M2-polarisierenden Faktoren und Wachstumsfaktoren bestehenden Gruppe ausgewählt ist.
Claims (23)
- Rekombinante probiotische Bakterien, umfassend eine oder mehrere Nucleinsäuresequenzen, die für einen ersten heterologen Faktor kodieren, eine oder mehrere Nucleinsäuresequenzen, die für einen zweiten heterologen Faktor kodieren und vorzugsweise eine oder mehrere Nucleinsäuresequenzen, die für einen dritten heterologen Faktor kodieren, mit der Maßgabe, dass der erste Faktor, der zweite Faktor und der dritte Faktor jeweils funktionell unterschiedlich voneinander sind, wobei es sich beim ersten Faktor um einen Wachstumsfaktor handelt, wobei der zweite Faktor aus der aus M2-polarisierenden Faktoren bestehenden Gruppe ausgewählt ist und wobei der dritte Faktor vorzugsweise aus der aus M2-polarisierenden Faktoren und Wachstumsfaktoren bestehenden Gruppe ausgewählt ist.
- Rekombinante probiotische Bakterien nach
Anspruch 1 , wobei die Nucleinsäuresequenz(en) an mindestens einem der Bestandteile Chromosom und Plasmid der rekombinanten probiotischen Bakterien lokalisiert sind. - Rekombinante probiotische Bakterien nach einem der
Ansprüche 1 oder2 , wobei eine Expression der Nucleinsäuresequenz(en) durch einen konstitutiven Promotor oder einen induzierbaren Promotor gesteuert wird. - Rekombinante probiotische Bakterien nach
Anspruch 3 , wobei die Expression der Nucleinsäuresequenz(en) durch einen induzierbaren Promotor gesteuert wird, wobei die Nucleinsäuresequenz(en) in Gegenwart mindestens eines Induktors exprimierbar sind. - Rekombinante probiotische Bakterien nach einem der
Ansprüche 3 oder4 , wobei es sich beim induzierbaren Promotor, der durch einen Induktor induzierbar ist, um einen Promotor für ein mikrobielles Gen handelt, das für ein lantibiotisches Peptid oder ein funktionelles Analoges davon kodiert, wobei es sich vorzugsweise beim induzierbaren Promotor um PnisA, PnisZ, PnisQ, PnisF, PnisU oder Kombinationen davon von Lactococcus lactis handelt. - Rekombinante probiotische Bakterien nach einem der
Ansprüche 4 oder5 , wobei es sich beim Induktor um mindestens ein lantibiotisches Peptid oder ein funktionelles Analoges davon handelt, die vorzugsweise aus der Gruppe, die aus Nisin A, Nisin Z, Nisin Q, Nisin F, Nisin U, funktionellen Analogen davon und Gemischen davon besteht, ausgewählt sind. - Rekombinante probiotische Bakterien nach einem der
Ansprüche 1 bis6 , wobei die probiotischen Bakterien ferner mindestens ein inaktiviertes Gen umfassen, das für ein essentielles Protein kodiert, das für die Lebensfähigkeit der probiotischen Bakterien notwendig ist. - Rekombinante probiotische Bakterien nach
Anspruch 7 , wobei das mindestens eine inaktivierte Gen, das für die Lebensfähigkeit der probiotischen Bakterien notwendig ist, aus der Gruppe ausgewählt ist, die aus Alanin-Racemase (alaR), Thymidylat-Synthase (thyA), Asparagin-Synthase (asnH), CTP-Synthase (pyrG), Tryptophan-Synthase (trpBA) und Kombinationen davon besteht. - Rekombinante probiotische Bakterien nach einem der
Ansprüche 7 oder8 , wobei das Gen inaktiviert ist durch Deletion des Gens, Mutation des Gens, epigenetische Modifikation des Gens, RNA-Interferenz (RNAi)-vermittelte Genabschaltung des Gens, translationale Hemmung des Gens oder Kombinationen davon. - Rekombinante probiotische Bakterien nach einem der
Ansprüche 1 bis9 , wobei mindestens einer der Bestandteile erster Faktor, zweiter Faktor und dritter Faktor aus den rekombinanten probiotischen Bakterien freisetzbar ist. - Rekombinante probiotische Bakterien nach einem der
Ansprüche 1 bis10 , wobei der M2-polarisierende Faktor aus der aus Cytokinen, Chemokinen und Gemischen davon bestehenden Gruppe ausgewählt ist. - Rekombinante probiotische Bakterien nach einem der
Ansprüche 1 bis11 , wobei der M2-polarisierende Faktor aus der aus Interleukin 4 (IL-4), Interleukin 10 (IL-10), Interleukin 13 (IL-13), kolonienstimulierender Faktor-1 (CSF-1), Interleukin 34 (IL-34) und Gemischen davon bestehenden Gruppe ausgewählt ist. - Rekombinante probiotische Bakterien nach einem der
Ansprüche 1 bis12 , wobei der Wachstumsfaktor aus der Gruppe ausgewählt ist, die aus Fibroblasten-Wachstumsfaktoren (FGF), vaskulären, endothelialen Wachstumsfaktoren (VEGF), epidermalen Wachstumsfaktoren (EGF), heparinbindendem, EGF-artigem Wachstumsfaktor (HB-EGF), transformierendem Wachstumsfaktor α (TGF-α), insulinartigen Wachstumsfaktoren (EGF), von Blutplättchen abgeleiteten Wachstumsfaktoren (PDGF), transformierendem Wachstumsfaktor beta (TGF-β) und Gemischen davon besteht, vorzugsweise Fibroblasten-Wachstumsfaktor 2 (FGF-2). - Rekombinante probiotische Bakterien nach einem der
Ansprüche 1 bis13 , wobei es sich bei den rekombinanten probiotischen Bakterien um Milchsäurebakterien, vorzugsweise um eine Lactobacillus- oder eine Lactococcus-Spezies, handelt. - Rekombinante probiotische Bakterien nach
Anspruch 14 , wobei es sich bei der Lactococcus-Spezies um Lactococcuslactis , vorzugsweise Lactococcuslactis Subspezies cremoris, handelt. - Rekombinante probiotische Bakterien nach einem der
Ansprüche 1 bis15 , wobei die rekombinanten probiotischen Bakterien in Lösung, gefroren oder getrocknet vorliegen, vorzugsweise lyophilisiert oder sprühgetrocknet. - Rekombinante probiotische Bakterien nach einem der
Ansprüche 1 bis16 , wobei die rekombinanten probiotischen Bakterien topisch und/oder durch subkutane Injektion zu verabreichen sind. - Rekombinante probiotische Bakterien zur Verwendung bei der Behandlung einer entzündlichen Hautdysfunktion, wobei es sich bei den rekombinanten probiotischen Bakterien um rekombinante probiotische Bakterien nach einem der
Ansprüche 1 bis17 handelt. - Rekombinante probiotische Bakterien nach
Anspruch 18 , wobei es sich bei der entzündlichen Hautdysfunktion um eine entzündliche Hautkrankheit oder eine entzündliche Bindegewebskrankheit handelt. - Rekombinante probiotische Bakterien nach
Anspruch 19 , wobei es sich bei der entzündlichen Hautkrankheit um eine Erfrierung, ein Ekzem, Psoriasis, Dermatitis, ein Ulcus, eine Wunde, Lupus erythematodes, Neurodermitis oder Kombinationen davon handelt. - Rekombinante probiotische Bakterien nach
Anspruch 20 , wobei es sich bei der Wunde um eine Brandwunde, eine Chemiewunde, eine durch Strahlen herbeigeführte Wunde, eine ischämische Wunde oder um eine mechanische Wunde handelt. - Rekombinante probiotische Bakterien nach
Anspruch 20 , wobei es sich beim Ulcus um einen Druckulcus (decubitus ulcer) oder ein Ulcus cruris handelt. - Verfahren zur Behandlung einer entzündlichen Hautdysfunktion, wobei das Verfahren die Stufe der Verabreichung von rekombinanten probiotischen Bakterien nach einem der
Ansprüche 1 bis22 an ein Individuum, das an der entzündlichen Hautdysfunktion leidet, umfasst.
