Abstract

Bacterias probióticas recombinantes, que comprenden (a) secuencia o secuencias de ácido nucleico que codifican un primer factor heterólogo, (a) secuencia o secuencias de ácido nucleico que codifican un segundo factor heterólogo y preferiblemente (a) secuencia o secuencias de ácido nucleico que codifican un tercer factor heterólogo, con la condición de que dicho primer factor, dicho segundo factor y dicho tercer factor son funcionalmente diferentes entre sí, en las que dicho primer factor es un factor de crecimiento, en las que dicho segundo factor se selecciona del grupo que consiste en factores polarizantes de M2, y en las que preferiblemente dicho tercer factor se selecciona del grupo que consiste en factores polarizantes de M2 y factores de crecimiento.

Claims (23)

  1. REIVINDICACIONES
    1. Bacterias probióticas recombinantes, que comprenden
    (a) secuencia o secuencias de ácido nucleico que codifican un primer factor heterólogo, (a) secuencia o secuencias de ácido nucleico que codifican un segundo factor heterólogo y preferiblemente (a) secuencia o secuencias de ácido nucleico que codifican un tercer factor heterólogo, con la condición de que dicho primer factor, dicho segundo factor y dicho tercer factor son funcionalmente diferentes entre sí,
    en las que dicho primer factor es un factor de crecimiento, en las que dicho segundo factor se selecciona del grupo que consiste en factores polarizantes de M2, y en las que preferiblemente dicho tercer factor se selecciona del grupo que consiste en factores polarizantes de M2 y factores de crecimiento.
  2. 2. Bacterias probióticas recombinantes, según la reivindicación 1, en las que dicha secuencia o secuencias de ácido nucleico están situadas en al menos uno de un cromosoma y un plásmido de dichas bacterias probióticas recombinantes.
  3. 3. Bacterias probióticas recombinantes, según cualquiera de las reivindicaciones 1 ó 2, en las que la expresión de dicha secuencia o secuencias de ácido nucleico está controlada por un promotor constitutivo o un promotor inducible.
  4. 4. Bacterias probióticas recombinantes, según la reivindicación 3, en las que la expresión de dicha secuencia o secuencias de ácido nucleico está controlada por un promotor inducible, siendo dicha secuencia o secuencias de ácido nucleico expresables en presencia de al menos un inductor.
  5. 5. Bacterias probióticas recombinantes, según cualquiera de las reivindicaciones 3 ó 4, en las que dicho promotor inducible, que es inducible por un inductor, es un promotor para un gen microbiano que codifica un péptido lantibiótico o un análogo funcional del mismo, en las que preferiblemente dicho promotor inducible es PnisA, PnisZ, PnisQ, PnisF, PnisU o combinaciones de los mismos de Lactococcus lactis.
  6. 6. Bacterias probióticas recombinantes, según cualquiera de las reivindicaciones 4 o 5, en las que el inductor es al menos un péptido lantibiótico o un análogo funcional del mismo, preferiblemente seleccionado del grupo que consiste en nisina A, nisina Z, nisina Q, nisina F, nisina U, análogos funcionales de las mismas, y mezclas de las mismas.
  7. 7. Bacterias probióticas recombinantes, según cualquiera de las reivindicaciones 1 a 6, en las que dichas bacterias probióticas comprenden, además, al menos un gen inactivado que codifica una proteína esencial necesaria para la viabilidad de dichas bacterias probióticas.
  8. 8. Bacterias probióticas recombinantes, según la reivindicación 7, en las que dicho al menos un gen inactivado necesario para la viabilidad de dichas bacterias probióticas se selecciona del grupo que consiste en alanina racemasa (alaR), timidilato sintasa (thyA), asparagina sintasa (asnH), CTP sintasa (pyrG), triptófano sintasa (trpBA), y combinaciones de los mismos.
  9. 9. Bacterias probióticas recombinantes, según cualquiera de las reivindicaciones 7 u 8, en las que dicho gen se inactiva por deleción de dicho gen, mutación de dicho gen, modificación epigenética de dicho gen, silenciamiento génico mediado por ARN de interferencia (ARNi) de dicho gen, inhibición de la traducción de dicho gen, o combinaciones de los mismos.
  10. 10. Bacterias probióticas recombinantes, según cualquiera de las reivindicaciones 1 a 9, en las que al menos uno de dicho primer factor, dicho segundo factor y dicho tercer factor es liberable de dichas bacterias probióticas recombinantes.
  11. 11. Bacterias probióticas recombinantes, según cualquiera de las reivindicaciones 1 a 10, en las que dicho factor polarizante de M2 se selecciona del grupo que consiste en citocinas, quimiocinas y mezclas de las mismas.
  12. 12. Bacterias probióticas recombinantes, según cualquiera de las reivindicaciones 1 a 11, en las que dicho factor polarizante de M2 se selecciona del grupo que consiste en interleucina 4 (IL-4), interleucina 10 (IL-10), interleucina 13 (IL-13), factor 1 estimulante de colonias (CSF-1), interleucina 34 (IL34) y mezclas de los mismos.
  13. 13. Bacterias probióticas recombinantes, según cualquiera de las reivindicaciones 1 a 12, en las que dicho factor de crecimiento se selecciona del grupo que consiste en factores de crecimiento de fibroblastos (FGF), factores de crecimiento endotelial vascular (VEGF), factores de crecimiento epidérmico (EGF), factor de crecimiento similar a EGF (HB-EGF) de unión a heparina, factor de crecimiento transformante a (TGF-a), factores de crecimiento similares a la insulina (IGF), factores de crecimiento derivados de plaquetas (PDGF), factor beta de crecimiento transformante (TGF-p) y mezclas de los mismos, preferiblemente el factor 2 de crecimiento de fibroblastos (FGF-2).
  14. 14. Bacterias probióticas recombinantes, según cualquiera de las reivindicaciones 1 a 13, en las que dichas bacterias probióticas recombinantes son bacterias de ácido láctico, preferiblemente una especie de Lactobacillus o Lactococcus.
  15. 15. Bacterias probióticas recombinantes, según la reivindicación 14, en las que dicha especie de Lactococcus es Lactococcus lactis, preferiblemente la subespecie cremoris de Lactococcus lactis.
  16. 16. Bacterias probióticas recombinantes, según cualquiera de las reivindicaciones 1 a 15, en las que dichas bacterias probióticas recombinantes están en una solución, congeladas o secas, preferiblemente están liofilizadas o secadas por pulverización.
  17. 17. Bacterias probióticas recombinantes, según cualquiera de las reivindicaciones 1 a 16, en las que dichas bacterias probióticas recombinantes se administran por vía tópica y/o mediante inyección subcutánea.
  18. 18. Bacterias probióticas recombinantes para usar en el tratamiento de una disfunción inflamatoria de la piel, en las que dichas bacterias probióticas recombinantes son bacterias probióticas recombinantes, según cualquiera de las reivindicaciones 1 a 17.
  19. 19. Bacterias probióticas recombinantes, según la reivindicación 18, en las que dicha disfunción inflamatoria de la piel es una enfermedad inflamatoria de la piel o una enfermedad inflamatoria del tejido conectivo.
  20. 20. Bacterias probióticas recombinantes, según la reivindicación 19, en las que dicha enfermedad inflamatoria de la piel es la congelación, eczema, psoriasis, dermatitis, úlcera, herida, lupus eritematoso, neurodermitis o combinaciones de las mismas.
  21. 21. Bacterias probióticas recombinantes, según la reivindicación 20, en las que dicha herida es una herida por quemadura, una herida química, una herida inducida por radiación, una herida isquémica o una herida mecánica.
  22. 22. Bacterias probióticas recombinantes, según la reivindicación 20, en las que dicha úlcera es una úlcera de decúbito o una úlcera venosa.
  23. 23. Método para el tratamiento de una disfunción inflamatoria de la piel, en el que dicho método comprende la etapa de administrar bacterias probióticas recombinantes, según cualquiera de las reivindicaciones 1 a 22, a un individuo que padece dicha disfunción inflamatoria de la piel.
