Description
Die Erfindung betrifft das S-Methoxy-o-mercaptopyridazin-1-oxyd
oder dessen Salze der allgemeinen Formel (I)

in der R ein Wasserstoffatom oder Alkalimetallatom bedeutet, und ein Verfahren zu seiner Herstellung,
indem man ein 3-Methoxy-6-halogenpyridazin-l-oxyd der allgemeinen Formel (II)

in der X ein Halogenatom bedeutet, in an sich bekannter Weise mit einem Alkalimetallhydrosulfid
kondensiert und gegebenenfalls das erhaltene Alkalimetallderivat durch Behandlung mit einer Säure in
die entsprechende freie Mercaptoverbindung überführt.
Beispiele für verwendbare Alkalimetallhydrosulfide sind Natrium- und Kaliumhydrosulfid. Die Umsetzung
erfolgt bereits bei Raumtemperatur glatt und in hoher Ausbeute.
Das erfindungsgemäße Verfahren erfolgt nach an
sich in der Chemie der Pyridazinverbindungen bekannten Methoden (vgl. J. Pharm. Soc. Japan, 75
[1955], S. 778 bis 781).
Das erfindungsgemäß herstellbare 3-Methoxy-6-mercaptopyridazin-l-oxyd
und dessen Alkalisalze haben eine hohe antibiotische Aktivität gegenüber verschiedenen Krankheitserregern. Dies geht aus der
nachstehenden Tabelle hervor, die auch die Ergebnisse von Vergleichsversuchen mit konstitutionell
ähnlichen Verbindungen enthält. Außerdem haben sie bei äußerlicher Anwendung fungizide Wirkung.
Trotz ihrer geringen Löslichkeit in Wasser haben die Alkalisalze gegenüber Trichophyton interdigitale die
gleiche Wirkung wie die freie Mercaptoverbindung. Pyridazinverbindungen mit antibiotischer Aktivität
sind bis jetzt nicht beschrieben. Aus der Tabelle ist ersichtlich, daß die Verbindungen der Erfindung
besonders gegen Trichophyton interdigitale wirken und Griseofulvin überlegen sind. Griseofulvin ist als
»Schmalband«-Antibiotikum bekannt und völlig unwirksam bei einer Reihe von Mikroorganismen,
insbesondere Bakterien und hefeähnlichen Pilzen.

Das Beispiel erläutert die Erfindung.
30 ml einer 2 η-Lösung von Kaliumhydrosulfid in Alkohol werden mit 3 g o-Chlor-S-methoxypyridazin-1-oxyd
versetzt, und das Gemisch wird 3 Stunden bei Raumtemperatur gerührt. Nach dem Stehenlassen
über Nacht werden die aus der Lösung ausgefallenen Kristalle abfiltriert, dann in Wasser gelöst und von
6o unlöslichen Stoffen durch Filtrieren der Lösung befreit.
Beim Ansäuern des Filtrats mit Salzsäure fallen gelblichbraune Kristalle aus, die aus Alkohol
umkristallisiert werden. Die plättchenförmigen Kristalle haben einen Schmp. von 140 bis 1410C.

Durch Auflösen der Kristalle in wenig Wasser und Eingießen der Lösung in eine alkoholische Lösung
von Kalilauge wird das entsprechende hellgelbe Kaliumsalz erhalten, das in Wasser löslich ist. Das
Salz schmilzt unter Zersetzung bei 3200C. Die Ausbeute beträgt 2,3 g (80% der Theorie).
