Die vorliegende Erfindung bezieht sich auf ein neues und verbessertes Verfahren zur Herstellung von 2,4-Bis-(dimethylamino)
-o-äthylenimino-l.S.S-triazin, welches
wertvolle pharmazeutische Eigenschaften besitzt.
2,4-Bis-(dimethylamino) -6-äthylenimino-l,3,5-triazin ist von Schäfer, Geoghegan und Kaiser im »Journal
of the American Chemical Society«, 1955, Bd. 77, S. 5921, beschrieben worden. Diese Autoren isolierten es als feste
Masse vom Schmelzpunkt 55 bis 57° C mit einer Ausbeute von 12%, indem sie Cyanurchlorid erst mit 1 Mol
Äthylenimin und dann mit 2 Mol Dimethylamin umsetzten, ohne ein Zwischenprodukt zu isolieren.
Gemäß der vorliegenden Erfindung wird ein zweistufiges Verfahren zur Herstellung des 2,4-Bis-(dimethylamino)-6-äthylenimino-l,3,5-triazins
vorgeschlagen, bei dem zunächst 2-Dimethylamino-4,6-dichlor-l ,3,5-triazin mit Äthylenimin und anschließend das erhaltene 2-Dimethylamino^-chlor-o-äthylenimino-l,3,5-triazin
mit Dimethylamin umgesetzt wird.
In der ersten Stufe dieses Verfahrens wird das 2-Dimethylamino^o-dichlor-l,3,5-triazin
mit Äthylenimin unter solchen Bedingungen umgesetzt, daß im wesentlichen nur ein Chlor atom in dem Triazin derivat ersetzt
wird. Diese Stufe wird praktisch so ausgeführt, daß nur etwa ein Äquivalent Äthylenimin verwendet und ferner
bei niedriger Temperatur, z. B. zwischen 5 und 250C, gearbeitet wird. Außerdem wird das Äthylenimin vorzugsweise
in Anteilen dem Dichlortriazinderivat zugesetzt und eine inerte organische Flüssigkeit, z. B.
Benzol, als Lösungsmittel und eine organische Base, z. B. Triäthylamin, als säurebindender Stoff verwendet. Das
Verfahren kann auch in einem wäßrigen Medium durchgeführt werden; da aber 2-Dimethylamino-4,6-dichlor-1,3,5-triazin
in Wasser im wesentlichen unlöslich ist, ist es unter diesen Umständen vorteilhaft, wenn das Reaktionsgemisch
ein inertes, mit Wasser mischbares organisches Lösungsmittel, z. B. Dioxan oder Aceton,
oder ein inertes, nicht mit Wasser mischbares organisches Lösungsmittel, z. B. Benzol, enthält. Darüber hinaus ist
es, wenn die Reaktion in einem wäßrigen Mittel ausgeführt wird, zweckmäßig, ein anorganisches, alkalisches,
säurebindendes Mittel zu verwenden, z. B. Kaliumcarbonat.
In der britischen Patentschrift 680 652 wird die Umsetzung von 2-Dimethylamino-4,6-dichlor-l,3,5-triazin
mit Äthylenimin beschrieben, wobei sich das Cytostaticum 2-Dimethylamino-4,6-bis-äthylenimino-l ,3,5-triazin ergibt.
Die neue Verbindung 2-Dimethylamino-4-chlor-6-äthylenimino-l
,3,5-triazin wird in der erwähnten Patentschrift nicht beschrieben und wird auch nicht unter
den in den dort angegebenen Umsetzungsbedingungen erhalten.

sind. Insbesondere ist die erstere Verbindung in Wasser schwer, die letztere dagegen in Wasser leicht
löslich. 2-Dimethylamino-4-chlor-6-äthylenimino-l,3,5-triazin
kann also leicht von etwaigem verunreinigendem, cytotoxischem 2 - Dimethylamino-4,6-bis - äthylenimino-1,3,5-triazin
befreit werden. Am Ende dieser Stufe des Verfahrens bekommt man reines 2-Dimethylamino-4-chlor-6-äthylenimino-l,3,5-triazin
in einer Ausbeute von 85 °/0.
Die zweite Stufe des vorliegenden Verfahrens besteht darin, daß man 2-Dimethylamino-4-chIor-6-äthylenimino-1,3,5-triazin
mit Dimethylamin umsetzt. Die Umsetzung wird zweckmäßigerweise so ausgeführt, daß das erwähnte
2-Dimethylamino-4-chlor-6-äthylenimino-l,3,5-triazin mit Dimethylamin in einem flüssigen Medium, z. B. Wasser,
zusammengebracht wird, und zwar in Gegenwart eines säurebindenden Stoffes, der z. B. Dimethylamin selbst
oder eine tertiäre organische Base, z. B. Triäthylamin, oder eine anorganische alkalische Substanz, z. B. Kaliumcarbonat,
sein kann. Falls gewünscht, kann auch in Gegenwart inerter organischer Lösungsmittel, z. B.
Aceton, Dioxan oder Benzol, gearbeitet werden. Die Umsetzung kann auch in einem wasserfreien Medium ausgeführt
werden, z. B. in einem organischen Lösungsmittel, wie Benzol. Es wird dabei eine Ausbeute von 85%
erreicht.

2,4-Bis- (dimethylamino) -6-äthylenimino-l ,3,5-triazin,
das gemäß dem vorliegenden Verfahren gewonnen wird, hat einen Schmelzpunkt von 74° C und wird reiner erhalten,
als wenn es nach dem Verfahren von Schäfer und Mitarbeitern, wie bereits angegeben, hergestellt wird.
Außerdem wird beim vorliegenden Verfahren vermieden, daß das gewünschte Produkt durch 2-Dimethylamino-4,6-bis-äthylenimino-l
,3,5-triazin verunreinigt ist. Da bekanntlich 2-Dimethylamino-4,6-dichlor-l ,3,5-triazin durch
Umsetzung von Cyanurchlorid mit Dimethylamin in fast quantitativer Ausbeute hergestellt werden kann, stellt
das vorliegende ein dem Schäferschen weit überlegenes Verfahren dar.
Auf Cyanurchlorid ergibt sich eine Ausbeute von etwa 70%, während bei dem Verfahren von Schäfer nur
12°/0 Ausbeute erzielt werden.
Die vorliegende Erfindung wird durch die folgenden Beispiele, in denen sich die Teile auf das Gewicht beziehen,
erläutert, aber nicht beschränkt.
Eine Mischung von 4,7 Teilen Äthylenimin, 11,1 Teilen
Triäthylamin und 80 Teilen Benzol wird allmählich zu einer gerührten Mischung von 19,3 Teilen 2-Dimethylamino-4,6-dichlor-l
,3,5-triazin und 160 Teilen Benzol gegeben, die unterhalb von 100C gehalten wird. Die Mischung
wird dann weitere 30 Minuten lang bei 100C gerührt und dann ohne Kühlung 30 Minuten gerührt. Sie
wird anschließend filtriert und das Filtrat unter vermindertem Druck unterhalb von 400C bis zur Trockne
eingeengt. Der Rückstand wird pulverisiert und mit 400 Teilen Wasser vermischt. Die Mischung wird 20 Minuten
lang gerührt und dann filtriert. Der feste Rückstand wird getrocknet und aus Leichtbenzin (Kp. 60 bis 80° C)
umkristallisiert. Es werden 17,0 Teile 2-Dimethylamino-4-chlor-6-äthylenimino-l,3,5-triazin,
das eine feste kristalline Masse vom Schmelzpunkt 104 bis 1050C bildet,
erhalten.
Eine Mischung von 4,7 Teilen Äthylenimin, 15,2 Teilen Kaliumcarbonat und 100 Teilen Wasser wird langsam zu
einer gerührten Mischung von 19,3 Teilen 2-Dimethylammo-4,6-dichlor-l
,3,5-triazin und 160 Teilen Aceton, die bei 5 bis 100C gehalten wird, gegeben. Die Mischung
wird dann eine weitere Stunde ohne Kühlung gerührt. Anschließend werden 200 Teile Wasser zugegeben und die
Mischung filtriert. Der feste Rückstand wird mit Wasser gewaschen und getrocknet. Er wird dann aus Leichtbenzin
(Kp. 60 bis 800C) umkristallisiert; dabei werden ll,2Teile
2 - Chlor - 4 - dimethylamino - 6 - äthylenimino -1,3,5- triazin vom Schmelzpunkt 104 bis 1050C erhalten.
Eine Mischung von 4,7 Teilen Äthylenimin und 100 Teilen Wasser wird langsam zu einer gerührten
Mischung von 19,3 Teilen 2-Dimethylamino-4,6-dichlor-1,3,6-triazin,
120 Teilen Benzol, 15,2 Teilen Kaliumcarbonat und 100 Teilen Wasser, die bei 1O0C gehalten
wird, gegeben. Die Mischung wird dann 90 Minuten lang ohne Kühlung gerührt. Die Benzolschicht wird anschließend
abgetrennt und mit Wasser gewaschen. Die Benzollösung wird dann unter vermindertem Druck bis
zur Trockne eingeengt. Der feste Rückstand wird aus Leichtbenzin (Kp. 60 bis 800C) umkristallisiert; dabei
werden 12,5 Teile 2-Dimethylamino-4-chlor-6-äthylenimino-1,3,5-triazin
vom Schmelzpunkt 104 bis 1060C erhalten.
2 Teile 2-Dimethylamino-4-chlor-6-äthylenirmno-1,3,5-triazin
werden portionsweise einer gerührten Mischung von 4 Teilen wäßriger 3O°/Oiger Dimethylaminlösung
und 40 Teilen Wasser, die bei 20 bis 25° C gehalten wird, zugesetzt. Die Mischung wird dann ohne weitere
Kühlung 30 Minuten lang gerührt. Anschließend wird sie auf 100C gekühlt und mit 8 Teilen wasserfreiem Kaliumcarbonat
versetzt. Die Mischung wird dann 10 Minuten lang bei 1O0C gerührt und filtriert. Der feste Rückstand
wird getrocknet und aus Leichtbenzin (Kp. 40 bis 6O0C) umkristallisiert. So werden 1,1 Teile 2,4-Bis-(dimethylamino)-6-äthylemmino-l
,3,5-triazin vom Schmelzpunkt 74°C erhalten.
Gasförmiges Dimethylamin wird in eine gerührte Mischung von 5 Teilen 2-Dimethylamino-4-chlor-6-äthylenimino-1,3,5-triazin
und 80 Teilen Benzol, die bei 15 bis 25°C gehalten wird, eingeleitet, bis keine weitere
Wärmeentwicklung erfolgt. Die Mischung wird dann weitere 30 Minuten lang gerührt und anschließend
filtriert. Das Filtrat wird im Vakuum unterhalb von 300C
bis zur Trockne verdampft und der feste Rückstand aus Leichtbenzin (Kp. 40 bis 6O0C) umkristallisiert. Es
werden 4,5 Teile 2,4~Bis-(dimethylamino)-6-äthylenimino-1,3,5-triazin
vom Schmelzpunkt 74° C erhalten.

- Patentanspruch:Verfahren zur Herstellung von 2,4-Bis-(dimethylamino)-6-äthylenimino-l,3,5-triazin, dadurch gekennzeichnet, daß man 2-Dimethylamino-4,6-dichlor-1,3,5-triazin bei niedriger und vorzugsweise zwischen 5 und 25°C liegender Temperatur mit einem angenähert molaren Äquivalent Äthylenimin umsetzt, wobei das Äthylenimin vorzugsweise allmählich dem Triazinderivat zugesetzt wird, und das erhaltene 2-Dimethylamino-4-chlor-6-äthylenimino-l,3,5-triazin anschließend in an sich bekannter Weise mit Dimethylamin umsetzt.In Betracht gezogene Druckschriften:
