发明概述
本发明提供了一种关于单糖和二糖代谢的增效组分的碳水化合物改良配制品。本发明的配制品在代谢上能减慢糖的吸收,改良胰岛素的释放,以及稳定血糖响应。另外,经口摄入本发明的配制品,通过抑制牙斑形成细菌将糖转化为腐蚀性的、牙齿-腐蚀酸的代谢能力而预防或减少龋齿的形成。
本发明的配制品对糖代谢和血糖响应提供了直接和间接的正效果。因此,本发明的配制品在以正常量消耗时不会不利地引起或恶化这样的病况,例如,肥胖症,糖尿病,或者基于饮食的、激素相关的机能亢进,例如,对于儿童经常描述的那些。
除了上述配制品以外,本发明还提供了成品水基饮料,其中掺和了本发明的配制品。此外,本发明还提供了一种成品水基饮料,它被酸化了而且包含本发明的配制品。

对本发明和优选实施方案的详细描述
本发明提供了一种配制品,它具有依赖于增效组分的所需性能,保持低的单糖水平,以及减慢从肠对单糖的通常迅速的吸收。通过阻挠对血糖和胰岛素响应的潜在去稳定作用,通过优选利用复杂碳水化合物的多糖基质和可溶性树胶纤维,上述目标最佳优化了能量水平。
本发明提供了未见于其它作用剂的很多优点,包括但不限于,限制例如糖这样的组分的过量应用,糖的过量应用会促进更大的氧化应激(oxidative stress),实际上减小了能量。组分优选选自可以中和和抑制自由基生成和氧化应激的那些,于是有助于保护细胞能量产生机制。此外,目前优选的组分是有助于细胞利用和燃料燃烧供给能量的那些。本发明的组合物还提供了各种定时热能燃料加本文公开的甜味体系在更长久缓释能量中的多层式应用。
本发明提供了活性和任选的惰性组分的组合物。该组合物可呈任意形式制备,包括但不限于,干配制品、含水配制品等。本发明的组合物可含于几乎任何生产的食料或饮料中。
本发明组合物目前一个优选的实施方案包含一种或多种多酚化合物。虽然不受任何特定的应用理论的限制,本发明人目前优选使用能抑制消化酶淀粉酶(淀粉消化)和蔗糖酶(糖消化)的多酚化合物,于是减慢糖的吸收,而且减小胰岛素响应的过度刺激,从而改良糖代谢。
此外,优选的多酚化合物抑制细菌酶-葡聚糖转移酶的活性,葡聚糖转移酶将见于饮料中的单糖代谢成粘性牙斑。没有该粘性牙斑时,细菌就不能粘附在牙齿表面、将糖发酵成酸而产生龋齿。
用于本发明的多酚化合物是从任意合适来源分离的。优选的多酚化合物包括:儿茶素类,单宁提取物,茶(例如,绿茶和红茶)的提取物,以及见于大果越桔、腺肋花楸浆果(aronia berry)、欧洲越橘和葡萄籽中的那些。其它适用的多酚化合物来源将是本领域技术人员显而易见的。
优选的绿茶和红茶活性物是儿茶素类和aflavins。
多酚化合物在配制品中可以任意有效的量存在,不过它们优选以8盎司水或其它稀释剂中约0.2mg~约500mg的量存在。
本发明的配制品还优选包含一种或多种氨基酸或氨基酸来源,氨基酸来源优选选自大豆,黄豆芽或其它豆科植物产生的蛋白质如绿豆,或乳基蛋白质,氨基螯合的矿物质,以及乳清或其它乳基蛋白质。其它适用的氨基酸来源是本领域技术人员已知的。
用于本发明的氨基酸优选是游离的甘氨酸和精氨酸,它们通过适度诱导胰岛素从胰腺释放而降低血糖水平。与胰岛素释放无关,精氨酸还刺激从垂体腺释放GH(生长激素)。GH是胰岛素的过度低血糖作用的天然抗衡物。此外,不依赖于能量动力学,甘氨酸是一种氨基酸神经递质基质,在科技文献中将它描述成抑制神经机能亢进。
因此,目前优选的氨基酸来源是大豆蛋白,它是富含甘氨酸和精氨酸的来源,可改善葡糖耐量和对血糖稳定性起关键作用的外周胰岛素敏感性。
一种或多种氨基酸可以任意有效的量存在于本发明的组合物中,不过优选以每8盎司水或其它稀释剂约5mg~约7g的量存在。当由氨基酸来源而不是游离氨基酸提供氨基酸时,该来源优选以提供优选量的游离氨基酸的量存在。
在本发明优选的配制品中还存在可溶性纤维,优选活性可溶性纤维。如本文应用的术语“活性可溶性纤维”表示这样的可溶性纤维,即,对哺乳动物胃肠道中的细菌有生物响应和/或参与体内一个或多个血糖改良机制。可溶性纤维来自任意来源,不过,优选的纤维是参与一个或多个下列血糖改良机制的那些,例如:通过肠细菌使可溶性纤维向短链脂肪酸(SCFAs)的转化(SCFA,特别是丙酸,增大糖酵解和减小葡糖异生作用,于是使血糖正常化);以及(2)通过可溶性纤维减慢从肠道吸收糖,它最终影响糖代谢的速率。
另外,任选作为本发明配制品的一种组分包含抗氧化剂(包括基于酚类的植物提取物)。抗氧化剂的存在能有助于克服或减弱见于大多数商品化饮料中迅速吸收的单糖的助氧化和去稳定低血糖作用。
目前具有前述特性的优选的可溶性纤维包括,菊粉,FOS[寡果糖(fructo-oligosaccharides),例如,BefloraTM],以及树胶。
所述可溶性纤维以任意有效的量存在,不过优选以每8盎司水或其它稀释剂约100mg~约8g的量存在。在含有5克或更多可溶性纤维的那些实施方案中,本发明的组合物优选能减小膳食后血糖水平的增大。
在本发明另一个优选的实施方案中,所述配制品包含一种或多种锌盐或其它锌离子来源。在代谢上,锌是胰岛素的合成和碳水化合物的代谢中的关键营养剂。从牙齿来看,成龋细菌从口中存在的单糖酶促产生不溶性葡聚糖沉积物,它牢固地粘附在牙釉质表面。本发明人以前在UCSF School of Preventive Dentistry的研究阐明了两种试验的锌盐,即,0.5%锌溶液(氯化锌)和相同浓度的抗坏血酸锌,这两种盐在体外抑制变异链球菌(mutans streptococci)和表兄链球菌(sobrinus streptococci)的生长和粘附。这说明了,锌阳离子(不是抗衡离子)是这种功能的盐分子的最重要部分。
锌离子优选的来源包括氯化锌、硫酸锌、抗坏血酸锌、吡啶甲酸锌、锌氨基酸螯合物和锌-EDTA。
锌盐或锌源以任意有效量存在,不过优选以每8盎司水或其它稀释剂约1mg~约40mg的量存在。
在另一个优选的实施方案中,本发明提供了一种酸性的成品饮料组合物。该饮料优选是水基的。用来配制该饮料的水可以是例如,静水、充了碳酸气的水或基于乳品的水。成品饮料的pH优选是约1~约7,更优选是约1~约5,而更进一步优选是约1~约3。无机盐、特别是二价矿物质(例如,钙、锌、镁、铁)的溶解性和同化作用在酸性介质中增强了。这些元素具有很多涉及细胞代谢和组织结构的重要作用。
酸或酸来源包括有机酸和无机酸。有机酸的实例包括,柠檬酸、乳酸、酒石酸、苹果酸和抗坏血酸。无机酸的实例包括磷酸。
与本发明组合物的操作相关的糖源可含于本发明的配制品自身中,或者它们可来自其它食料。
本发明还提供了一种调节哺乳动物受试验者中的糖代谢的方法。该方法包括,对受试验者施用本发明的组合物,于是调节受试验者的糖代谢。在一个优选的实施方案中,该调节作用导致糖代谢速率减小(与不存在本发明的组合物时的速率相比)。在另一个优选的实施方案中,所述调节作用包括代谢速率的线性化,消除糖代谢曲线中的峰值和/或低谷值,和/或减小糖代谢曲线中的峰高和/或低谷深度。在又一个优选的实施方案中,糖代谢是通过所述组合物引起哺乳动物的肠对糖吸收速率的减小而调节的。

实施例1
酸乳酪饮料(8盎司):
将酸乳酪培养的牛奶、鲜果和糖(10g)或精选的强化甜味剂(10ppm)与改良剂合并。改良剂包含:菊粉纤维(3g),欧洲越橘,柑橘生物类黄酮类,绿茶提取物混合物(多酚类)(100mg),大豆异黄酮类(50mg),硫酸锌(7mg),以及维生素C(60mg)。添加足量的水使体积达8盎司。

Claims (37)
1.一种含水配制品,它包含:
(a)一种多酚化合物;
(b)一种氨基酸;
(c)活性可溶性纤维;以及
(d)一种水性稀释剂。
2.权利要求1的配制品,其中,所述多酚化合物是选自儿茶素类、aflavins及其组合的一员。
3.权利要求2的配制品,其中,所述多酚化合物抑制哺乳动物消化酶。
4.权利要求3的配制品,其中,所述消化酶是选自淀粉酶、蔗糖酶及其组合的一员。
5.权利要求3的配制品,其中,所述多酚化合物以足以抑制所述酶的量存在。
6.权利要求1的配制品,其中,当对哺乳动物受试验者施用时,所述多酚化合物具有一种特性,该特性是下组的一员:减慢所述受试验者中的糖吸收、减小所述受试验者中胰岛素响应的过度刺激及其组合。
7.权利要求6的配制品,其中,所述多酚化合物以足以减小哺乳动物受试验者中选自下组的一员的量存在:糖吸收、免疫应答的过度刺激及其组合。
8.权利要求1的配制品,其中,所述多酚化合物抑制细菌酶。
9.权利要求8的配制品,其中,所述多酚化合物以足以抑制哺乳动物受试验者中所述细菌酶的量存在。
10.权利要求8的配制品,其中,所述细菌酶是葡聚糖转移酶。
11.权利要求10的配制品,它对于龋齿的形成有抑制效果。
12.权利要求1的配制品,其中,所述多酚来自选自下组的一员:单宁、茶、欧洲越橘、葡萄籽的提取物及其组合。
13.权利要求1的配制品,其中,所述配制品中存在的所述多酚的量是,约8盎司所述配制品中约0.2mg~约500mg。
14.权利要求13的配制品,其中,所述配制品中存在的所述多酚的量是,约8盎司所述配制品中约1mg~约200mg。
15.权利要求1的配制品,其中,所述氨基酸以一种氨基酸来源的形式存在,该氨基酸来源是选自下组的一员:大豆蛋白提取物、氨基螯合的矿物质、乳清蛋白及其组合。
16.权利要求1的配制品,其中,所述氨基酸降低受试验者中的血糖水平。
17.权利要求16的配制品,其中,所述氨基酸以足以降低所述受试验者中的所述血糖水平的量存在。
18.权利要求16的配制品,其中,所述氨基酸通过诱导胰岛素从所述受试验者的胰腺释放而降低所述血糖。
19.权利要求16的配制品,其中,所述氨基酸通过诱导生长激素从所述受试验者的垂体腺释放而降低所述受试验者的所述血糖。
20.权利要求16的配制品,其中,所述氨基酸在所述配制品中存在的量是,约8盎司所述配制品中约1mg~约50mg。
21.权利要求20的配制品,其中,所述氨基酸存在的量是,约8盎司所述配制品中约3mg~约10mg。
22.权利要求21的配制品,其中,所述氨基酸存在的量是,约8盎司所述配制品中约5mg~约7mg。
23.权利要求1的配制品,其中,所述可溶性纤维是选自下组的一员:菊粉、寡果糖和树胶。
24.权利要求1的配制品,其中,所述可溶性纤维改良受试验者的血糖水平。
25.权利要求24的配制品,其中,所述可溶性纤维在所述配制品中以足以降低所述血糖水平的量存在。
26.权利要求24的配制品,其中,所述血糖水平是通过选自下组的一员的机制而改良的:所述可溶性纤维向短链脂肪酸转化,减慢从肠道吸收糖,及其组合。
27.权利要求24的配制品,其中,所述可溶性纤维存在的量是,约8盎司所述配制品中约100mg~约8g。
28.权利要求24的配制品,其中,所述可溶性纤维存在的量是,约8盎司所述配制品中至少约3克。
29.权利要求1的配制品,它进一步包含一种金属离子。
30.权利要求29的配制品,其中,所述金属离子是Zn+2。
31.权利要求29的配制品,其中,所述金属离子在所述配制品中以足以抑制选自下组的一员在受试验者中生长和粘附的量存在:变异链球菌、表兄链球菌及其组合。
32.权利要求31的配制品,其中,所述金属离子在所述配制品中存在的量是,约8盎司所述配制品中约1mg~约40mg。
33.权利要求1的配制品,其中,所述配制品是酸性成品。
34.权利要求33的配制品,其中,所述酸是选自下组的一员:柠檬酸、乳酸、酒石酸、苹果酸、抗坏血酸、磷酸及其组合。
35.一种减慢从哺乳动物肠吸收糖的配制品,所述配制品包含下列成分:
(a)一种多酚化合物;
(b)一种氨基酸;
(c)活性可溶性纤维;以及
(d)一种水性稀释剂,
其中,每一种成分以足以提供减慢所述糖吸收的所述配制品的量存在。
36.权利要求35的配制品,它阻止龋齿的形成,所述配制品进一步包含一种金属离子,该金属离子的量足以阻止所述龋齿的所述形成。
37.一种调节哺乳动物受试验者中糖代谢的方法,所述方法包括,对所述受试验者施用可有效地调节所述糖代谢的一定量权利要求1的配制品。
