Abstract
本文公开了鞘磷脂与磷脂的含量比例较为确定的大豆萃取物及其制造方法。该方法包括下列步骤:a)采用脂族醇或脂族醇与水的混合物对成熟全大豆或无油大豆粉进行萃取;b)对步骤a)得到的萃取液进行浓缩;c)用脂族烃萃取由步骤b)得到的浓缩萃取液;d)用植物活性炭处理经步骤c)的萃取液,然后进行蒸发浓缩;e)添加脂族酮使活性成分变成不溶;f)过滤活性成分,然后干燥之。本专利还包括含所述萃取物的药用和化妆品用组合物,该组合物尤其可用来防止皮肤干燥或护理干性皮肤。
Description
在JP06228171-A中,公开了通过大豆与非极性载体相接触,然后用极性溶剂从非极性载体中萃取鞘磷脂来提纯鞘磷脂的方法。
本发明涉及通过对成熟全大豆或无油大豆粉(Glycine max.(L.)MERRIL,豆科)进行萃取得到的新颖萃取物及其制造方法,以及含此种萃取物的配制物。该新颖萃取物的特征在于鞘磷脂与磷脂的含量是有一定比例的。在本文中,“鞘磷脂”是指由鞘氨醇衍生的磷脂,而“磷脂”是指由甘油衍生的磷脂。
业已发现,这些萃取物尤其对于用于护理干性皮肤的化妆品有重要的作用。
因此,本发明涉及含有约5-15(重量)%,优选约10(重量)%的鞘磷脂和约15-30(重量)%,优选20(重量)%以上磷脂的新颖大豆萃取物。本发明萃取物中鞘磷脂与磷脂之比为约0.5-1.5(重量)份鞘磷脂比1.5-3(重量)份磷脂。存在于该萃取物中含量较少的组分是某些甾醇和长链脂肪醇。除了长链脂肪醇和甾醇外,萃取物中的其它成分是甘油单酸酯和甘油二酸酯。
本发明还涉及制造上述萃取物的方法,方法的特征在于包括下列步骤:
a)采用脂族醇或脂族醇与水的混合物对成熟全大豆或无油大豆粉进行萃取;
b)对步骤a)得到的萃取液进行浓缩;
c)用脂族烃萃取由步骤b)得到的浓缩萃取液;
d)用植物活性炭处理经步骤c)的萃取液,然后进行蒸发浓缩;
e)添加脂族酮使活性成分变成不溶;
f)过滤活性成分,然后干燥之。
具体地说,根据本发明的萃取物可通过采用脂族醇或脂族醇与水的混合物优选乙醇/水混合物(如95%乙醇)对成熟全大豆或无油大豆粉进行萃取而制得。浓缩后,再用脂族烃萃取该萃取物。适用的脂族烃是正己烷和正庚烷。接着用植物活性炭处理并经蒸发而缩小体积后,有机相在减压下干燥。然后添加脂族酮使活性成分变成不溶。适用此目的的脂族酮例如为丙酮。在经蒸发浓缩而缩小体积后,有机相在减压下干燥。
实施本发明方法中的各具体步骤的优选条件如下所述。在此及此后所用的测量单位对体积份来说是升,对重量份来说是千克。
步骤a:选用鞘磷脂与磷脂比例符合要求的植物原料。为此,例如将具有预定类脂物含量的大豆粉用作原料。植物原料优选以每(重量)份生物物质用12-17体积的溶剂进行萃取。萃取温度高于55℃。每次萃取周期少于4小时是有利的。除乙醇外,适用的溶剂尤其是甲醇、丙醇和异丙醇。这些溶剂可含高达10%的水。
步骤b:在减压及低于50℃的条件下,使萃取液浓缩。该萃取液经蒸发浓缩至醇含量为65-75%。
步骤c:由步骤b获得的浓缩萃取液以每(重量)份植物原料为0.3-0.6体积的脂族烃进行萃取。萃取优选进行三次。
步骤d:有机相用植物活性炭处理,然后在减压及低于50℃的条件下蒸发至干。
步骤e:采用10体积的脂族酮使活性成分变成不溶(以1(重量)份植物原料计)。
步骤f:沉淀物优选经洗涤,例如洗涤三次,酮(例如丙酮)是适用于此目的的。过滤后,沉淀物在减压及低于50℃的条件下进行干燥。
根据本发明的萃取物是呈脂状的固体,既溶于醚中也溶于氯化溶剂中。
该萃取物中鞘磷脂的含量是通过HPLC与采用鞘磷脂酶使鞘磷脂进行酶水解相结合的方法测定的。鞘磷脂酶是一种将鞘磷脂转变成N-脂酰基鞘氨醇的酶,而N-脂酰基鞘氨醇则可用HPLC或TLC进行检测。
与迄今已知的萃取物相比,根据本发明的萃取物对干性皮肤具有护理作用的特色。
表1列出了干性皮肤经本发明萃取物护理与用胶原护理后的结果比较。以本发明萃取物进行护理时,是按含本发明萃取物和2%羟乙基纤维素的水乳液进行的。

采用市售仪器(Corneometer CM820,Courage+Kazaka,Cologne,德国)测定角质层的电容,以电容值作为皮肤水化的量度。以连续测定的五次值的平均值作为电容测定值记录。电容值是以0-100单位的任意标度来表示的。对多组10位受试人群组进行了试验。
因此,本发明也扩展到合作为活性成分的上述萃取物的药用和化妆品用组合物。具体地说,本发明涉及含有用于防止皮肤干燥或护理干性皮肤的这种萃取物的化妆品组合物。
根据本发明的产物或萃取物可用适当方法配制成如流体乳液,水包油乳液,油包水乳液,脂质凝胶(lipogel),唇膏,用于制备随需随用的、与其溶解度相适应的溶液或流体或液体的粒状粉剂。根据本发明的萃取物在配制物中的浓度范围为1-5%,优选为1.2-3.5%。
下面通过实施例对本发明进行说明。
实施例1-含10(重量)%鞘磷脂和20(重量)%磷脂的大豆萃取物的制备
10千克无油大豆粉用30升95%强度乙醇回流5次。将5次回流的醇萃取物混合,然后在减压下蒸发浓缩至5升。用5升正己烷萃取该浓缩物3次。用植物活性炭处理正己烷相,然后减压浓缩至较小的体积。其后用10体积丙酮处理所得的萃取物。鞘磷脂和磷脂析出。经过滤后,产物在减压下干燥,得到220克含10(重量)%鞘磷脂和20(重量)%磷脂的萃取物。
Claims (5)
1.大豆萃取物,其特征在于含有5-15(重量)%的鞘磷脂和15-30(重量)%的磷脂。
2.根据权利要求1的大豆萃取物,其特征在于含有约10(重量)%的鞘磷脂和约20(重量)%的磷脂。
3.制造根据权利要求1或2的萃取物的方法,其特征在于包括下列步骤:
a)采用脂族醇或脂族醇与水的混合物对成熟全大豆或无油大豆粉进行萃取;
b)对步骤a)得到的萃取液进行浓缩;
c)用脂族烃萃取由步骤b)得到的浓缩萃取液;
d)用植物活性炭处理经步骤c)的萃取液,然后进行蒸发浓缩;
e)添加脂族酮使活性成分变成不溶;
f)过滤活性成分,然后干燥之。
4.含有作为活性成分的根据权利要求1或2的萃取物的药用和化妆品用组合物。
5.用来防止皮肤干燥或护理干性皮肤的根据权利要求4的组合物。
