Abstract

Description
本发明涉及微粒化有机紫外线过滤物质的混合物用于防止晒黑(tanning)和使人类皮肤和头发有光泽的用途,以及它们在化妆品和药物制剂中的用途。
已知某类有机紫外线过滤物质,例如难溶的苯并三唑或三嗪化合物,以微粒化形式存在时具有极好的过滤紫外线的性质。
特别是在亚洲国家,对于防晒过滤物质或其混合物有着广泛的兴趣,它们能在阳光辐射下保护皮肤的颜色,此外能予以皮肤更具光泽的外观。
因此,本发明的目的在于寻找微粒化有机紫外线过滤物质,以防止皮肤晒黑,同时使皮肤更有光泽。
令人惊讶的是,我们已经发现微粒化有机紫外线过滤物质或至少两种微粒化有机紫外线过滤物质的混合物能实现该目的。
因此,本发明提供了微粒化有机紫外线过滤物质的混合物防止皮肤晒黑并使其更有光泽的的用途。
可用于本发明的合适的紫外线过滤物质为有机的,有时是难溶的化合物,例如三嗪衍生物,特别是羟苯基三嗪化合物或苯并三唑衍生物,含乙烯基的酰胺,肉桂酸衍生物,磺化苯并咪唑类,费歇尔碱衍生物,二苯基丙二腈类,草酰胺类,樟脑衍生物,二苯基丙烯酸酯类,对氨基苯甲酸(PABA)及其衍生物,水杨酸酯,二苯甲酮类和其它类的已知紫外线过滤物质。

本发明可用的微粒化有机紫外线过滤物质的混合物可以各种不同的方法制备。
首先,至少两种上述有机紫外线过滤物质在制备微粉的过程(微粉化作用)中以单个物质相混合。
另一可选的制备方法涉及将已经微粒化的各紫外线过滤物质完全混合在一起。
第三种可选的制备方法涉及将至少两种上述紫外线过滤物质熔融在一起。冷却熔体产生均相组合物,并以常规方式微粒化。
本发明还提供至少两种紫外线过滤物质的均相组合物。
本发明还提供组合物,其可通过将一种或多种无机微细颜料熔化于一种或多种有机紫外线过滤物质中而得到。
微细颜料的例子为,例如TiO2,ZnO,氧化铁类或其它无机氧化物,云母或其它合适的无机矿物,以及有机酸的Ti、碱土金属或锌盐。
在这样的处理过程中,一些无机微细颜料(TiO2,ZnO)的所不期望的光催化性质可以同时被抑制,且它们的正面性质也可被充分利用。
上述无机紫外线过滤物质可有利地熔于亚甲基二苯并三唑基四甲基丁基苯酚中。然后所得的组合物以常规方式微粒化。
本发明还提供组合物,其荷电通过将至少两种电中性的有机紫外线过滤物质与阳离子或阴离子荷电化合物熔融在一起而获得。
为达到此目的,阳离子或阴离子荷电化合物与相应的有机电中性紫外线过滤物质熔融在一起,然后冷却。该方法允许在随后的微粒化步骤中制备带有永久修饰的正或负电荷的有机紫外线过滤物质。这种修饰能有效地阻止微粒在防晒制剂中的聚集,这种聚集当颗粒直径小于1微米时就会发生。这样,另外一种常用的颗粒的“涂覆方法”因具有负作用有时就成为了多余。
可用的阳离子或阴离子荷电化合物为紫外线过滤物质,以及其它带有一个或多个阳离子或阴离子基团的化合物,例如:
-N,N,N-三甲基-4-(2-氧代冰片-3-亚基甲基)苯胺甲基硫酸盐;
-樟脑苄烷铵(camphorbenzalkonium)甲基硫酸盐;
-脂肪胺;
-内铵盐,例如措昔古柯碱基丙基内铵盐;
-季胺化合物,例如蓖麻酰氨基丙基三镆甲基硫酸盐,柚子提取物(quarternium 18),或鲸蜡基三甲基溴化铵;
-山嵛酸及其它有机酸,例如异硬脂酸,柠檬酸单甘油酯或甲基椰油基牛磺酸钠;
-磷脂类,例如卵磷脂,磷脂酰丝氨酸或烷基胺氧化物;
-神经酰胺和假神经酰胺和植物甾醇。
最后一种命名的化合物向微粒化紫外线过滤物质中引入了一个疏水修饰基。
组合物中阳离子或阴离子化合物的比例,以紫外线过滤物质的重量为基础计,为0.001~5%(重量),优选0.01~3%(重量)。
本发明还提供组合物,其通过将至少一种难溶或不溶的有机紫外线过滤物质与抗氧化剂熔融后得到。
为此,可将难溶或不溶的有机紫外线过滤物质(一种或多种)与抗氧化剂一起熔融,冷却,然后以常规方式微粒化。
本发明可用的合适的抗氧化剂为所有具有清除剂性质的有机物,且能与有机紫外线过滤物质相熔融。这样得到的新型微细颜料能同时防止皮肤晒黑,并在皮肤表面产生抗氧化效用。这种性质是防晒化妆品所期望的,因为在紫外线和光线的影响下,在制剂中和皮肤表面会产生有害的自由基。例如能导致所谓的Mallorca痤疮,或使皮肤过早老化。通过用抗氧化剂对微粒化紫外线过滤物质进行修饰,不仅能实现防止紫外线损伤和防止皮肤晒黑,而且能防止防晒制品中组分的光化学分解。
组合物中抗氧化剂的比例,以紫外线过滤物质的重量为基础计,为0.001~30%(重量),优选为0.01~3%(重量)。
在微细颜料中含有抗氧化剂是特别有利的,该微细颜料中除了含有有机紫外线过滤物质之外,还包括上述光催化活性的无机微细颜料,例如二氧化钛,氧化锌(包括涂覆的)或其它合适的无机氧化物例如氧化铁。

优选使用下列抗氧化剂与有机紫外线过滤物质的混合物:
-亚甲基二苯并三唑基四甲基丁基苯酚,辛基三嗪酮,二氧化钛和生育酚的混合物;
-2,2’-亚甲基二[6-(2H-苯并三唑-2-基)-4-甲基苯酚,辛基三嗪酮,三间二羟苯基三嗪和维生素E的混合物;
本发明还提供组合物,其可通过将可熔融的化妆品、植物和药物活性成分熔化于有机紫外线过滤物质中得到。
通常,微粒化紫外线过滤物质可用作高活性物质的载体,具体为化妆品和/或药物活性物质的载体。这种组成的优点在于它们能从固体中释放(缓慢释放)活性物质。缓慢释放能保证高活性成分例如消炎剂,护理活性成分或微量元素例如Zn2+或Mg2+在紫外颜料的整个使用寿命内的均匀效用。
可用的并可提及的活性成分的例子如下:
-杀菌修饰同时有消炎作用的活性成分例如三氯羟基二苯醚或diclosan;
-消炎活性成分例如金合欢醇,泛醇或鳄梨油;
-具有祛臭或抑汗效用的活性成分,例如蓖麻油酸锌盐和烷基柠檬酸酯,
-十一烯酸及其衍生物(例如二乙醇酰胺)
-十一酸锌盐;
-2-巯基吡啶氧化物,例如2-巯基吡啶氧化钠;
-熔融香料或香料混合物,例如薄荷醇,香叶醇等,它们赋予这些微细颜料和包含它们的制剂以均匀强度的永久香味。
为制备这种微粒化有机紫外线过滤物质或微细颜料混合物,可采用适于制备微细颗粒的所有已知方法,例如:
-湿磨法,采用硬研磨介质例如硅酸锆和保护性表面活性剂或保护性聚合物在水中或适当有机溶剂中;

-在合适的溶剂中再沉淀,溶剂包括超临界流体(GASR方法=气体抗溶剂重结晶法/PCA法=压缩抗溶剂沉淀法)
制备本发明的微粒化有机紫外线吸收剂可用的研磨装置为,例如,喷射研磨机,球磨机,振动研磨机,或锤磨机,优选高速搅拌研磨机。研磨过程中优选使用研磨助剂,例如烷基化乙烯基吡咯烷酮聚合物,乙烯基吡咯烷酮/醋酸乙烯酯共聚物,酰基谷氨酸酯,烷基聚糖苷,ceteareth-25或特别为磷脂。
所得的微细颜料或微细颜料的混合物通常的平均颗粒大小为0.02~2纳米,优选0.05~1.5纳米,非常优选为0.1~1.0纳米。
由于它们的亲脂性,这些微细颜料可以单独地或与其它可溶性有机紫外线吸收剂一起,按已知方法引入到含油和含脂的化妆制剂中,例如油类,水包油或油包水乳液,蜡笔或胶。
令人惊奇的是,使用少量甚至不用可溶的紫外线吸收剂,得到的制剂也具有同样的或改进的保护效用。

本发明的化妆制剂可以有利地包括其它能在UVB区吸收紫外线辐射的物质。此处过滤物质的总量,以组合物总重量为基础计,为0.1~30%(重量),优选0.5~10%(重量),特别1-6%(重量)。

实施例1:
50份亚甲基二-苯并三唑基四甲基丁基苯酚和50份辛基三嗪酮在压粒机中研磨,研磨剂为硅酸锆砂,保护性表面活性剂(烷基聚葡糖苷)和水,研磨后获得d50为190纳米的混合微细颜料。分离出研磨介质之后,混合微细颜料的悬浮液可用来制备防晒制剂。
实施例2:
32份辛基三嗪酮,1份鲸蜡基三甲基溴化铵和66份亚甲基二-苯并三唑基四甲基丁基苯酚均匀熔融在一起。混合物快速冷却至室温,固化后的熔体经机械粉碎(冲击式研磨机)。得到的粉末在水中成浆,加入癸基苷,然后混合物与研磨助剂(“重砂”)一起微粒化成颗粒直径d50为200纳米。除去研磨助剂之后,获得微粒化紫外线吸收组合物的水悬浮液。该悬浮液用柠檬酸稍稍酸化,并可用于制备化妆和药物制剂。
实施例3:

实施例48:

实施例49:通过微粒化紫外线吸收剂亚甲基二苯并三唑基四甲基丁基苯酚来防止皮肤晒黑
方法:
20个有着直系亚洲血统(父亲和母亲)的志愿者,他们在过去3个月中没有直接暴露于阳光之下,并已经解释了所要作的研究,得到了同意的申明,且满意所夹带的条件。这些人在3周之内每天在测试位即大腿上部用含2,2’-亚甲基二[6-(2H-苯并三唑-2-基)-4-(1,1,3,3-四甲基丁基)苯酚]的霜剂或空白对照剂处理两次。
这些志愿者们每周三次在测试位即大腿上部以0.2-0.5 MED UVAB的量进行照射。
第一次使用该制品是在第一次照射之后进行,在每次使用被测试产品之后进行评估和照射。以对比的未处理照射区或未处理未照射区作为参照。
各种情况下的测试区的色值用Minolta CM-508i照相机根据DIN5033,ISO 7724/1,JIS Z8722以L*a*b*值来记录。
测定各个主体的颜色和亮度变化,并确定未处理、未照射参照区和测试区之间皮肤颜色的不同。这些值根据所有主体取其平均值,并记作L*,a*和b*值。
测试制剂
(A):含有6%2,2’-亚甲基二[6-(2H-苯并三唑-2-基)-4-(1,1,3,3-四甲基丁基)苯酚]、水、硬脂酸辛酯、椰子甘油酯、丙二醇、甲氧基-PEG-22/十二烷基二醇共聚物、PEG-22/十二烷基二醇共聚物、羟基二十八烷基羟基硬脂酸酯、矿物油、苯氧基乙醇&对羟基苯甲酸酯、七水硫酸镁、聚二甲基硅氧烷、尿囊素的组合物。
(B):含有3%2,2’-亚甲基二[6-(2H-苯并三唑-2-基)-4-(1,1,3,3-四甲基丁基)苯酚]、水、硬脂酸辛酯、椰子甘油酯、丙二醇、甲氧基-PEG-22/十二烷基二醇共聚物、PEG-22/十二烷基二醇共聚物、羟基二十八烷基羟基硬脂酸酯、矿物油、苯氧基乙醇&对羟基苯甲酸酯、七水硫酸镁,聚二甲基硅氧烷、尿囊素的组合物。
(C):含有水、硬脂酸辛酯、椰子甘油酯、丙二醇、甲氧基-PEG-22/十二烷基二醇共聚物、PEG-22/十二烷基二醇共聚物、羟基二十八烷基羟基硬脂酸酯、矿物油、苯氧基乙醇&对羟基苯甲酸酯、七水硫酸镁、聚二甲基硅氧烷、尿囊素的空白对照剂。
在重复UVAB照射(3×周)之后,与未照射皮肤比较的L*a*b值和使用了组合物(A)和(B)时的L*a*b值。
结果讨论:
亮度
当空白对照剂处理过的和未处理过的照射区的亮度减至约同一程度,即变黑时,使用组合物(B)时,效果相当小,而使用组合物(A)时,可观察到皮肤被增亮。
变红
三次照射后,所照射的皮肤中红色组分非常强烈,并在照射结束恢复至正常值.红色成分的增加相应于UV-引发的红疹的发展,而在使用组合物(A)或(B)时则仅仅是轻度的。
黄色组分
在使用空白对照剂时和在未处理的照射对比区中,黄色成分均增加了。使用组合物(B)时则是非常少量的增加,而使用组合物(A)时则被抑制了。
Claims (32)
1.微粒化有机紫外线过滤物质用于防止晒黑和使皮肤和头发具有光泽的用途。
2.根据权利要求1所述的用途,其中有机紫外线过滤物质选自三嗪或苯并三唑衍生物,含乙烯基的酰胺,肉桂酸衍生物,磺化苯并咪唑类,费歇尔碱衍生物,二苯基丙二腈类,草酰胺类,樟脑衍生物,二苯基丙烯酸酯类,对氨基苯甲酸(PABA)及其衍生物,水杨酸酯和二苯甲酮类。
3.根据权利要求1或2所述的用途,其中有机紫外线过滤物质选自下式所示的三嗪衍生物:其中R1,R2和R3各自独立为氢;OH;C1-C18烷氧基;-NH2;-NH-R4;-N(R4)2;-OR4,R4为C1-C5烷基;未取代或被一个、两个、或三个羟基,羧基,-CO-NH2,C1-C5烷基或C1-C5烷氧基取代的苯基、苯氧基、苯胺基或吡咯并;亚甲基樟脑基;式-(CH=CH)mC(=O)-OR4所示的基团;所示基团或其相应的碱金属盐,铵盐,一、二或三-C1-C4烷基铵盐,一、二或三-C2-C4链烷醇铵盐或C1-C3烷基酯;或式(1a)所示基团;R6为氢;未取代的C1-C5烷基或被一个或多个羟基取代的C1-C5烷基;C1-C5烷氧基;氨基;一或二-C1-C5烷基氨基;M;下列各式所示的基团其中R’,R’’,和R’’’各自独立地为未取代的C1-C14烷基或被一个或多个羟基取代的C1-C14烷基;R6为氢;M;C1-C5烷基;或式
所示的基团;M为金属阳离子;T1为氢;或C1-C8烷基;m为0或1m2为1~4;以及m3为2~14。
4.根据权利要求1或2所述的用途,其中有机紫外线过滤物质选自下式所示的三嗪衍生物:其中R7和R8各自独立地为C1-C18烷基;C2-C18链烯基;式-CH2-CH(-OH)-CH2-O-T1所示的基团;或R7和R8为式(2a)所示的基团;R9为直接键;直链或支链C1-C4亚烷基或式
所示的基团:R10,R11和R12各自独立地为C1-C18烷基;C1-C18烷氧基或式R13为C1-C5烷基;m1为1~4;p1为0~5;A1为或式所示的基团;R14为氢;C1-C10烷基,
或式-CH2-CH(-OH)-CH2-O-T1所示的基团;R15为氢;M;C1-C5烷基;或式
所示的基团R16为氢;或甲基;T1为氢:或C1-C8烷基;Q1为C1-C18烷基;M为金属阳离子;m2和m3各自独立地为1~4;以及n1为1~16。

12.根据权利要求1或2所述的用途,其中有机紫外线过滤物质选自下式所示的苯并三唑衍生物;其中T2为C1-C4烷基或苯基取代的C1-C5烷基。
14.根据权利要求1或2所述的用途,其中有机紫外线过滤物质选自下式所示的化合物:其中R50、R51、R52、R53、R54各自独立地为氢、C1-C8烷基或C5-C10环烷基;R55为氢、C1-C8烷基、C5-C10环烷基、羟基、C1-C8烷氧基、COOR56或CONR57R58;R56、R57和R58各自独立地为氢或C1-C6烷基;X和Y各自独立地为氢、-CN、CO2R59、CONR59R60或COR59;其中基团X和Y还可为C1-C8烷基、C5-C10环烷基或具有5-6个环原子的杂芳基,另外,X和Y或R50与基团X和Y中的一个可以一起代表构成5~7元环的基团,该环可含有至多3个杂原子,其中环原子可被环外双键氧和/或C1-C8烷基和/或C5-C10环烷基取代,和/或含有C=C双键;Z为氢、铵基、碱金属离子或用于中和游离酸基的含氮有机碱的阳离子,R59和R60各自独立地为氢、C1-C8烷基或C5-C10环烷基;以及n和m各自独立地为0或1。
15.根据权利要求1至14任一项所述的用途,其中有机紫外线过滤物质以混合物形式使用。
16.制备权利要求1至14任一项中所定义的并可根据本发明使用的有机紫外线过滤物质的混合物的方法,其包括将微粒化形式的紫外线过滤物质充分混合。
17.制备权利要求1至14任一项中所定义的并可根据本发明使用的有机紫外线过滤物质的混合物的方法,其包括将有机紫外线过滤物质以至少两种单独物质的混合物进行微粒化。
18.制备权利要求1至14任一项中所定义的并可根据本发明使用的有机紫外线过滤物质的混合物的方法,其包括将至少两种单独物质熔化在一起,冷却熔体然后将所得的组合物进行微粒化。
19.一种组合物,其可通过将至少两种权利要求1至14任一项中所定义的有机紫外线过滤物质熔化在一起而获得。
20.根据权利要求1至14任一项所述的用途,其中还混入无机颜料。
21.根据权利要求20所述的用途,其中无机颜料选自TiO2、ZnO、氧化铁类、云母和有机酸的Ti或锌盐。
22.一种组合物,其可通过将至少两种权利要求1至14任一项中所定义的有机紫外线过滤物质与至少一种权利要求20或21中所定义的无机颜料熔化在一起而获得。
23.根据权利要求1至14任一项所述的用途,其中还混入抗氧化剂。
25.一种组合物,其可通过将至少两种权利要求1-14任一项中所定义的有机紫外线过滤物质和至少一种权利要求23或24中所定义的抗氧化剂,以及,如果需要的话,一种或多种无机颜料熔融在一起而得到。
26.根据权利要求1-15任一项所述的用途,其中混入阳离子或阴离子化合物。
27.根据权利要求26所述的用途,其中阳离子或阴离子化合物选自樟脑苄烷铵甲基硫酸盐、脂肪胺、内铵盐、季胺化合物、柠檬酸单甘油酯、甲基椰油基牛磺酸钠、磷脂、神经酰胺和植物甾醇。
28.一种组合物,其可通过将至少两种权利要求1-14任一项中所定义的有机紫外线过滤物质和至少一种权利要求26或27中所定义的阳离子或阴离子化合物熔融在一起而得到。
29.根据权利要求1-15任一项所述的用途,其中还混合药物或化妆品活性组分。
30.一种化妆制剂,其含有一种权利要求1-15任一项中所定义的有机紫外线过滤物质,如果需要还包括一种或多种抗氧化剂和/或无机颜料和/或阳离子或阴离子化合物,以及化妆学上相容的载体或助剂。
31.根据权利要求30所述的化妆制剂,其还含有油溶性、未微粒化的紫外线过滤物质。
32.一种药物制剂,其含有由至少两种权利要求1-15任一项中所定义的有机紫外线过滤物质,如果需要还包括一种或多种抗氧化剂和/或无机颜料和/或阳离子或阴离子化合物,以及药物上相容的载体或助剂所组成的混合物。
