技术领域
本发明属于微生物技术领域,具体涉及一株耐高温鸭疫里默氏杆菌噬菌体及其应用。
背景技术
噬菌体又称细菌病毒,是一种在细菌体内完成增殖,通过裂解细菌将自身释放到环境中的微生物。与抗生素治疗相比,噬菌体治疗具有代谢效率低、杀死细菌的最低剂量低、高度的特异性、高效的杀菌效果、筛选速度快等优势。近年来,有关噬菌体的研究有了巨大的发展,国内外对噬菌体的应用研究范围很广,包括针对人类疾病的防治、动物疾病的防治、食品中病原菌的清除、农作物的细菌感染防治、细菌生物被膜的裂解等方面。
鸭传染性浆膜炎是由鸭疫里默氏杆菌(RA)引起的传染病,是鸭的主要细菌性传染病,其死亡和感染率较高。该病的感染率最高可达90%,死亡率为5%~80%。在发生RA病时,及时使用药物对病情有控制作用,由于细菌对抗菌类药物容易形成耐药性,在生产中经常出现某药物刚开始使用效果明显,历经一段时间后效果不显著甚至无效。药物防治曾经是国内诸多地区控制RA病的主要手段之一,但RA易形成耐药性,耐药性造成采用药物在防治RA过程中的疗效不断降低、防治期间资金投入量持续增加等问题。
另一方面,疫苗接种是目前治疗鸭疫里默氏杆菌较为有效的防治手段。现阶段公认疫苗RA血清型有21种,RA各血清型之间尚未无交叉保护。所以在开发疫苗时要立足于本地区流行的血清型菌株,并将其作为菌苗株。尽管灭活疫苗已成功用于RA病的预防免疫接种,但在产生中存在操作流程不便捷、成本较高及对雏鸭的应激较大等不足。
发明内容
为了解决现有技术中存在的问题,本发明的目的在于提供一株耐高温鸭疫里默氏杆菌噬菌体RDP-RA-22039及其应用,本发明提供的耐高温鸭疫里默氏杆菌噬菌体RDP-RA-22039效价高,具有优异的耐高温特性和较强的遗传稳定性,在温度较高的环境中可以稳定存放,能够有效预防和治疗由鸭疫里默氏杆菌引起的浆膜炎,杀灭病原菌,快速使机体恢复健康状态,可替代抗生素和疫苗使用,安全无残留,在防治鸭传染性浆膜炎的临床应用中具有良好的前景。
本发明的技术方案是:
本发明提供了一株耐高温鸭疫里默氏杆菌噬菌体RDP-RA-22039,从山东省东营市某养鸭场中分离得到,已于2023年6月12日保藏于中国微生物菌种保藏管理委员会普通微生物中心,保藏地址为北京市朝阳区北辰西路1号院3号中国科学院微生物研究所,保藏编号为CGMCC No.45587,分类学命名为鸭疫里默氏杆菌噬菌体Riemerella anatipestifer bacteriophage。
进一步地,所述的耐高温鸭疫里默氏杆菌噬菌体RDP-RA-22039于电镜下观测到:有一个多面体立体对称的头部,包裹着核酸,直径约40 nm,有一个长约150 nm的尾,有尾鞘,颈部连着头部和尾部,根据国际病毒分类学组织病毒分类第九次报告,该噬菌体归类为有尾病毒目长尾病毒科。
进一步地,本发明所述的耐高温鸭疫里默氏杆菌噬菌体RDP-RA-22039经全基因组测序,得到具有SEQ ID No.1所示的核苷酸序列。
本发明所提供的耐高温鸭疫里默氏杆菌噬菌体RDP-RA-22039的最佳感染复数为0.01,对应最高效价为1.03×109pfu/mL;并且噬菌体RDP-RA-22039具有较强的裂解和复制能力,在传代30次的过程中,始终保持较高的效价,没有出现明显波动,说明该噬菌体具有较强的遗传稳定性。
进一步地,本发明提供的耐高温鸭疫里默氏杆菌噬菌体RDP-RA-22039具有优异的耐高温性能,经试验验证,将噬菌体RDP-RA-22039在80℃以下环境中放置30 min到60 min,噬菌体均可以保持较高的效价,在90℃环境中30min时,噬菌体效价仅下降2个数量级,在90℃环境中60 min时,噬菌体效价仅下降3个数量级。
同时,本发明提供的耐高温鸭疫里默氏杆菌噬菌体RDP-RA-22039具有优异的稳定性能,在30℃以下环境中放置60天,噬菌体可以保持较高的效价,效价没有任何的下降;噬菌体RDP-RA-22039在50℃以下环境中放置60天,噬菌体仍保持较高的效价,仅下降1个数量级。
进一步地,本发明提供了一种所述的耐高温鸭疫里默氏杆菌噬菌体RKP-RA-22039在制备预或治疗鸭疫里默氏杆菌感染的疾病的药物中的应用。
本发明还提供了一种所述的耐高温鸭疫里默氏杆菌噬菌体RKP-RA-22039在制备预或治疗鸭疫里默氏杆菌感染的疾病的饲料添加剂或环境消毒剂中的应用。
进一步地,本发明提供了一种噬菌体组合物,包括上述的耐高温鸭疫里默氏杆菌噬菌体RDP-RA-22039。
更进一步地,本发明提供了一种噬菌体药物制剂,包括上述的耐高温鸭疫里默氏杆菌噬菌体RDP-RA-22039或上述的噬菌体组合物。
更进一步地,所述噬菌体药物药物制剂中添加的耐高温鸭疫里默氏杆菌噬菌体RDP-RA-22039的浓度大于109PFU/g。

进一步地,所述饲料添加剂的制备方法为现有技术中的常规方法,本发明中采用低聚麦芽糖和山梨醇将噬菌体发酵液包被,接着喷洒在脱脂米糠中,烘干即得。采用本发明耐高温鸭疫里默氏杆菌噬菌体RDP-RA-22039制得的粉剂,可以有效治疗鸭疫里默氏杆菌引起的浆膜炎,有效杀灭病原菌,快速使机体恢复健康状态。
与现有技术相比,本发明提供的耐高温鸭疫里默氏杆菌噬菌体RDP-RA-22039具有以下优势:
(1)本发明于山东省东营市一养鸭场中发现并分离了一株耐高温鸭疫里默氏杆菌噬菌体RDP-RA-22039,该噬菌体对温度有很强的耐受性,在温度较高的环境中可以稳定存放,有效解决了噬菌体高温易失活的难题,突破了浆膜炎的治疗困境,为鸭传染性浆膜炎找到一种新的有效的治疗方案,解决了鸭疫里默氏杆菌的耐药性难题。
(2)本发明提供的耐高温鸭疫里默氏杆菌噬菌体RDP-RA-22039还具有较强的遗传稳定性,效价高,对禽没有毒害作用,可替代抗生素和疫苗使用,安全无残留,不会产生耐药性,对环境友好。在防治鸭传染性浆膜炎的临床应用中具有良好的前景。

以下通过具体实施方式的描述对本发明作进一步说明,但这并非是对本发明的限制,本领域技术人员根据本发明的基本思想,可以做出各种修改或改进,但是只要不脱离本发明的基本思想,均在本发明的保护范围之内。
以下实施例中,未作特别说明的试剂为常规试剂,均可在常规试剂生产销售公司购买,所用方法,如无特殊说明,均为现有技术。
实施例1噬菌体的筛选与分离
1.宿主鸭疫里默氏杆菌RA-22039的分离与鉴定:
(1)本宿主从山东省东营市养殖场的发病鸭中采样,无菌采集发病鸭的肝脏或脑(有典型纤维素性渗出时采集肝脏,有神经症状时采集脑),将接种环伸入组织内部,然后划线接种于TSA血琼脂平板和麦康凯琼脂平板,含5%~10%CO2的37℃培养箱或烛缸中培养24h~48 h。
(2)在血琼脂平板培养基上见有湿润、呈露滴样、圆形隆起、表面光滑、边缘整齐的菌落,直径为1 mm~2 mm,颜色为灰白色,如图1所示;而麦康凯琼脂平板上无菌落生长。挑选单个典型菌落进行革兰氏染色和瑞氏染色,并镜检。革兰氏阴性,为无鞭毛、不运动、不形成芽胞的小杆菌。单个、成双或呈短链状排列,菌体形态除杆状外,部分呈椭圆形,偶见呈长丝状。细菌大小宽为0.2μm~0.5μm,长为1μm~5μm,呈丝状的菌体可长达11μm~24μm。瑞氏染色可见大多数菌体呈两极着染。
(3)挑取典型菌落继续划线纯化3~5次,直至菌落形态一致,然后挑取单菌落接种于5 mL的TSB肉汤培养基中,37℃,200 rpm振荡培养24 h,得到均匀浑浊的细菌悬浮液。再通过16sRNA分子鉴定和血清型鉴定,确定为致病性鸭疫里默氏杆菌,将其中一株命名为RA-22039,鉴定好的菌株在TSA+2%的血清培养基平板上划线,培养24 h,将全部菌落用接种环刮至60%甘油-TSB液体中,保存于-80℃冰箱。
2.噬菌体RDP-RA-22039的分离及鉴定
(1)病料处理:从养殖场取病鸭,解剖留取脑组织,无菌操作剪取2 g脑组织,加入2mL灭菌生理盐水,将肝脏充分研磨,然后10000 rpm离心30 min,取上清液过0.22μm滤器,将滤出液备用;
(2)制备噬菌体富集液:取0.2 mL菌悬液和1 mL滤出液加入5 mLTSB肉汤中,37℃,100 rpm振荡培养过夜,然后10000 rpm离心5 min,取上清液过0.22μm滤器,滤出液备用;
(3)噬菌体分离:采用双层平板法进行噬菌体的分离,取噬菌体富集液滤出液0.1mL和宿主鸭疫里默氏杆菌菌悬液0.2 mL混合均匀后,37℃水浴10 min,然后铺双平板,放于37℃培养箱培养6~8 h后,挑取透明斑于1 mL SM缓冲液中4℃浸泡12h,取0.1 mL浸出液和0.2 mL菌悬液37℃水浴10 min,铺双平板,37℃培养箱培养4~6 h纯化,照此步骤纯化5次,即得。噬菌体RDP-RA-22039在双平板形成的噬菌斑直径为1 mm~3 mm。
(4)噬菌体RDP-RA-22039保存方法:将噬菌体增殖液与60%的甘油按照1∶1的比例混合,保存于-80℃冰箱或液氮中均可。
具体地,该耐高温鸭疫里默氏杆菌噬菌体RDP-RA-22039,已于2023年6月12日保藏于中国微生物菌种保藏管理委员会普通微生物中心,保藏地址为北京市朝阳区北辰西路1号院3号中国科学院微生物研究所,保藏编号为CGMCC No.45587,分类学命名为鸭疫里默氏杆菌噬菌体Riemerella anatipestifer bacteriophage。
3.噬菌体的电镜观察
取20μL含噬菌体粗制颗粒的液体滴于铜网上,自然沉淀15 min,并用滤纸从侧面吸去多余液体,加一滴2%的磷钨酸(PTA)于铜网上对噬菌体染色10 min,然后用滤纸从侧面吸去染色液,待样品干燥后用电子显微镜观察噬菌体形态,如图2所示。
由图2可以看出,噬菌体RDP-RA-22039有一个多面体立体对称的头部,包裹着核酸,直径约40nm,有一个长约150nm的尾,有尾鞘,颈部连着头部和尾部。根据国际病毒分类学组织病毒分类第九次报告,该噬菌体归类为有尾病毒目长尾病毒科。
实施例2噬菌体的生物学特性研究
1.噬菌体最佳感染复数测定
按照感染复数为100、10、1、0.1、0.01、0.001的比例将噬菌体增殖液和宿主加入TSB肉汤中,并且确保培养体系的总体积相同。于37℃200 rpm振荡培养8 h后,常温下12000r/min离心5 min,取上清液铺双平板测定其效价。具体试验结果参见表1。
表1最佳感染复数的测定结果
由表1数据显示,当感染复数为0.01时,培养12 h后,增殖液较为清澈,效价最高,为1.03×109pfu/mL,说明噬菌体RDP-RA-22039的最佳感染复数为0.01。
2.噬菌体一步生长曲线的绘制
取对数生长期的鸭疫里默氏杆菌悬液,按照最佳感染复数的比例接种鸭疫里默氏杆菌RA-22039和噬菌体RDP-RA-22039,在37℃的条件下,水浴10 min,然后常温下12000 r/min离心5 min,弃上清液,除去未吸附在宿主上的游离噬菌体,如此用TSB肉汤培养基洗涤两次。再将沉淀重悬于37℃的TSB肉汤培养基中,迅速置于37℃200 r/min的摇床中振荡培养并计时。前30 min每隔5 min取样计数,30~60 min每隔20 min计数,60~300 min每隔30min计数,以时间为横坐标,噬菌体效价对数值为纵坐标,绘制生长曲线,得出噬菌体的潜伏期、裂解期,并计算出平均裂解量。平均裂解量=暴发末期噬菌体滴度/感染初期宿主菌浓度。
实验结果如图3所示,由图3可以看出,噬菌体感染宿主菌后的180 min内效价没有明显变化,表明其潜伏期约180 min,感染后180~420 min内噬菌体的效价明显上升,之后趋于稳定,表明噬菌体裂解期约为240 min。通过计算,噬菌体裂解量约为369 PFU/感染细胞,表明噬菌体具有较强的裂解和复制能力。
3.噬菌体遗传稳定性实验
将噬菌体RDP-RA-22039连续传代30次,每代培养基中加入等量的噬菌体RDP-RA-22039和RA-22039,每代培养时间12 h,然后倒双平板,测定其效价。结果如图4所示。
由图4可以看出,噬菌体RDP-RA-22039在传代30次的过程中,始终保持较高的效价,没有出现明显波动,说明该噬菌体可以稳定遗传。
4.噬菌体热稳定性实验
将噬菌体RDP-RA-22039原液分装到离心管中,分别在40℃、50℃、60℃、70℃的条件下孵育30 min和60 min,然后用生理盐水10倍比稀释后,铺双平板测定其效价。结果如图5所示。
由图5可以看出,噬菌体RDP-RA-22039在80℃以下环境中放置30 min到60 min,噬菌体均可以保持较高的效价,在90℃环境中30 min时,噬菌体效价仅下降2个数量级,在90℃环境中60 min时,噬菌体效价仅下降3个数量级。以上结果说明该噬菌体对温度有很强的耐受性,能够在高温环境中存活较长时间。
5.噬菌体对温度长久耐受性实验
将噬菌体RDP-RA-22039发酵液置于无菌离心管中,分别在4℃、30℃、50℃的条件下放置60天,每10天取样检测效价,然后用生理盐水10倍比稀释后,铺双平板测定其效价。结果如图6所示。
由图6可以看出,噬菌体RDP-RA-22039在30℃以下环境中放置60天,噬菌体可以保持较高的效价,效价没有任何的下降;噬菌体RDP-RA-22039在50℃以下环境中放置60天,噬菌体仍保持较高的效价,仅下降1个数量级,说明该噬菌体在常温或50℃以下环境中存放时,可以保持较强的稳定性,该噬菌体2个月内可以稳定存放。
实施例3噬菌体的应用性能检测
1.噬菌体安全性实验
实验步骤:
(1)取9日龄SPF鸭胚120只,平均分成4组,每组30只,分别为空白组A,噬菌体组B,TSB对照组C,生理盐水对照组D。
(2)B组每只鸭胚尿囊腔接种200μL(109PFU/mL)噬菌体液;C组和D组分别接种200μL TSB培养液和生理盐水,接种后放置于37℃孵化箱培养。
(3)每隔1 h照胚观察记录各组鸭胚存活情况,连续观察48 h。
试验结果:4组鸭胚48 h后均完全存活,说明该噬菌体对对禽没有毒害作用,安全性较好。
2.噬菌体粉剂制备与应用
A.粉剂制备
实验材料:低聚麦芽糖、山梨醇、脱脂米糠、噬菌体发酵液RDP-RA-22039(效价109PFU/mL)
实验步骤:
(1)取1%低聚麦芽糖和3%山梨醇将噬菌体发酵液包被;
(2)将包被好的噬菌体按照1∶3(质量比)的比例在常温下喷洒在脱脂米糠,混合均匀;
(3)将混合后的脱脂米糠在50℃下烘干,收集烘干后的粉剂,制备成噬菌体粉;
(4)将制备后的噬菌体粉剂用PBS溶解,测定噬菌体效价为107PFU/mL,该噬菌体粉制备方法可以有效保护噬菌体。
B.粉剂应用
(1)取7日龄雏鸭40只,平均分成4组,每组10只,分别为空白组A,噬菌体粉剂组B,噬菌体发酵液对照组C,生理盐水对照组D。
(2)A、B、C、D四组分别用宿主菌RA-22039进行攻毒,攻毒剂量为0.2 mL(107CFU/mL)/只;
(3)24 h后,A组不做处理,B组每只雏鸭饲喂噬菌体粉剂0.2 g(107PFU/g);C组每只雏鸭灌服噬菌体液0.2 mL(109PFU/g);D组饲喂200μL生理盐水;
(4)按照上述治疗方法连续治疗3天,每天观察实验鸭状态;
(5)治疗完成后,剖检实验鸭,取脑组织,分别检测A、B、C、D四组每只鸭脑组织菌含量;
实验结果:雏鸭攻毒24 h后,四个实验组鸭均出现明显精神沉郁,缩颈,嗜睡,腿软,不愿走动,采食量下降。第1次治疗后24 h,B组和C组采食量出现提升,A组和D组出现2只死亡;连续治疗3天后,B组和C组实验鸭出现明显好转,精神沉郁等症状消失,采食量明显提升,实验鸭均康复,A组和D组实验鸭死亡4只,剩余实验鸭卧地,不愿走动,双脚软而无力,皮肤干瘪,不愿采食。
实验证明,本发明制得的噬菌体粉剂和噬菌体发酵液均可以有效治疗鸭疫里默氏杆菌引起的浆膜炎,能够有效杀灭病原菌,快速使机体恢复健康状态,有效减抗替抗,具有较好的应用前景。

Claims (9)
1.一株耐高温鸭疫里默氏杆菌噬菌体,其特征在于,所述鸭疫里默氏杆菌噬菌体为RDP-RA-22039,保藏于中国微生物菌种保藏管理委员会普通微生物中心,保藏编号为CGMCCNo.45587。
2.根据权利要求1所述的耐高温鸭疫里默氏杆菌噬菌体,其特征在于,所述耐高温鸭疫里默氏杆菌噬菌体RDP-RA-22039有一个多面体立体对称的头部,包裹着核酸,直径为40nm,有一个长为150 nm的尾,有尾鞘,颈部连着头部和尾部,该噬菌体归类为有尾病毒目长尾病毒科。
3.一种根据权利要求1或2所述的耐高温鸭疫里默氏杆菌噬菌体RKP-RA-22039在制备预或治疗鸭疫里默氏杆菌感染的疾病的药物中的应用。
4.一种根据权利要求1或2所述的耐高温鸭疫里默氏杆菌噬菌体RKP-RA-22039在制备预或治疗鸭疫里默氏杆菌感染的疾病的饲料添加剂或环境消毒剂中的应用。
5.一种噬菌体组合物,其特征在于,包括权利要求1所述的耐高温鸭疫里默氏杆菌噬菌体RDP-RA-22039。
6.一种噬菌体药物制剂,其特征在于,包括权利要求1所述的耐高温鸭疫里默氏杆菌噬菌体RDP-RA-22039或权利要求5所述的噬菌体组合物。
7.根据权利要求6所述的噬菌体药物制剂,其特征在于,所述耐高温鸭疫里默氏杆菌噬菌体RDP-RA-22039的浓度大于109 PFU/g。
8.根据权利要求6所述的噬菌体药物制剂,其特征在于,所述噬菌体药物制剂还包括药学上可接受的辅料,其剂型为粉剂、颗粒剂、冻干剂、溶液剂、乳剂、混悬剂或凝胶剂。
9.一种饲料添加剂,其特征在于,包括权利要求1所述的耐高温鸭疫里默氏杆菌噬菌体RDP-RA-22039或权利要求5所述的噬菌体组合物。
