Description

Traditional Chinese medicine formula for improving chicken immunity and application thereof
Technical Field
The invention belongs to the field of veterinary medicines, and in particular relates to a traditional Chinese medicine formula for improving chicken immunity and application thereof.
Background
Newcastle disease, also known as pseudo-fowl plague or asian fowl plague, is an acute, febrile, highly contagious disease caused by newcastle disease virus, and is mainly characterized by dyspnea and neurological symptoms and serosa and mucosal hemorrhage. The disease incidence and the death rate are high, the threat to the poultry industry is serious, the world health organization is classified as A-class animal infectious diseases (animal infectious diseases needing to be notified), and the world is classified as a class of infectious diseases. Newcastle disease can be classified into a typical newcastle disease and an atypical newcastle disease according to its clinical symptoms. According to the clinical manifestations and the course of the disease, classical newcastle disease can be divided into four categories, namely most acute, subacute and chronic. The typical newcastle disease is mainly characterized by dyspnea, serosal and mucosal bleeding, and associated neurological symptoms, and a significant drop in egg production by the laying hens. Atypical newcastle disease is a special expression form of a virulent infection immune chicken flock, and is mainly characterized in that the chicken flock has a certain antibody level, after virulent invasion, the morbidity and the mortality of the chicken flock are low, and the clinical manifestations of dyspnea are mainly caused. NDV can cause changes in non-suppurative encephalitis through the blood brain barrier in the later stage of infection, causing the chicken to develop neurological symptoms in the later stage of disease. The method of periodic vaccine vaccination is mainly used for preventing and controlling newcastle disease in China, and common vaccines are mainly conventional vaccines and also comprise a few genetic engineering vaccines. Although the chickens are immunized comprehensively, the effect is not ideal, and according to epidemiological investigation results carried out by Chinese animal sanitation and epidemiology centers in recent years, the occurrence of newcastle disease in China is still quite common.
At present, veterinary antiviral drugs are forbidden on economic animals in China, but the veterinarian does not have an antiviral concept, and only has the meaning of strengthening body resistance and eliminating evil. The basic principle of antivirus of traditional Chinese medicine is strengthening vital qi to eliminate pathogenic factor, so-called vital qi internal memory, pathogenic factor can not be dried, when organism immunity is strong, the chance of infection of pathogenic factor is small, but when organism disease resistance is poor, invasion by exogenous pathogenic factor is easy. The medicine principle is to tonify middle-jiao and qi, clear away heat and toxic materials. The traditional Chinese medicine has very complex process of exerting antiviral effect in animal body, and the antiviral mechanism is mainly divided into direct virus inhibition and indirect virus inhibition. Direct inhibition of a virus is the prevention of adsorption of the virus to a host cell, inhibition of replication of the virus within the host cell, and inhibition of maturation and release of the virus. Indirect inhibition of viral proliferation is antiviral by enhancing the immune function of the body or producing interferon.
The university of Shanxi agriculture (CN 113827688A) discloses a traditional Chinese medicine composition for reducing the death rate of newcastle disease, which comprises the following raw materials in parts by mass: 100-200 parts of radix isatidis, 50-100 parts of honeysuckle, 80-150 parts of astragalus membranaceus, 50-80 parts of fructus forsythiae, 50-100 parts of dyers woad leaf, 50-80 parts of folium artemisiae argyi, 30-50 parts of rhizoma acori graminei, 10-50 parts of wrinkled gianthyssop, 10-50 parts of radix curcumae and 30-50 parts of reed rhizome. By utilizing the mutual synergy or resistance effect among the traditional Chinese medicine raw materials, the immunity, disease resistance and external pathogenic factor invasion resistance of the chickens are improved, the effects of preventing and treating the newcastle disease are achieved, the treatment effect on the newcastle disease is improved, and the death rate of the newcastle disease is reduced. The Shandong Muwang pharmaceutical technology Co., ltd (CN 108465098A) discloses a traditional Chinese medicine composition for preventing and treating poultry newcastle disease, which comprises the following raw materials: 8-12 parts of aconite, 8-12 parts of dried ginger, 15-25 parts of cyrtomium rhizome, 15-25 parts of szechuan lovage rhizome, 45-55 parts of semen lepidii, 8-12 parts of honeysuckle flower, 15-25 parts of radix bupleuri, 15-25 parts of radix salviae miltiorrhizae, 15-25 parts of dried rehmannia root, 15-25 parts of rhizoma alismatis, 10-20 parts of rhizoma atractylodis, 10-20 parts of Chinese angelica, 15-25 parts of barbed skullcap herb, 15-25 parts of subprostrate sophora, 15-25 parts of shizandra berry and 10-20 parts of liquorice. The active components or active component groups in the composition are obtained by a proper preparation method, have inactivation and inhibition effects on newcastle disease virus, and can promote the immune function of the organism to achieve the antiviral effect; thereby realizing effective treatment of newcastle disease. The two medicaments have complex component structures and poor effect in practical application.
The compound form is used for treating diseases, is one of the characteristics of traditional Chinese medicine, and the traditional Chinese medicine can be used for making up the defect of the action intensity of single traditional Chinese medicine and enhancing the efficacy on the one hand due to the synergistic effect of the compound form; on the other hand, the toxicity and side effects can be reduced, and the toxicity of certain medicaments can be weakened. The research aims at discussing and developing a traditional Chinese medicine formula for efficiently improving chicken immunity.

Test animals: 1 day old Hailan brown cocks were purchased from Shenyang Huamei birds Co., ltd; raising in a conventional cage to 14 days old, and maintaining standard temperature, humidity, ventilation and illumination according to a conventional method.
2.2 method
Test design and grouping: the sea-blue brown cock is raised to 14 days old (average parent antibody titer is about 4log2, average weight is about 112 g), 30 animals are randomly grouped, eye and nose are respectively spotted with Newcastle disease LaSota virus vaccine for first immunization, 3 feathers/animal are added into drinking water according to 0.3ml/L, the mixture is continuously used for one week, and a control group is conventional drinking water; each neck at 28 days of age was subcutaneously injected with newcastle disease LaSota attenuated vaccine for secondary immunization.
Determination of antibody titres in newcastle disease after immunization, blood was collected from subwinged veins at 14, 21, 28, 35, 42d for 0.5ml, centrifuged at 2000r/min for 5min to isolate serum, and single factor analysis of variance and Duncan's multiple comparisons were performed using microprombin inhibition statistical software at significant levels of P=0.05.
