Description

Anti-wrinkle firming activator for skin care and preparation method thereof
Technical Field
The invention relates to the field of skin care products, in particular to an anti-wrinkle firming activator for skin care and a preparation method thereof.
Background
The skin is used as the first barrier of human body and has certain protection effect on human body. With the continuous improvement of living standard, people pay more and more attention to their appearance and image, and the functional requirements of skin care products are higher and higher, and in addition to the most basic skin care effect, people tend to pursue the change of their skin by using skin care products, such as reducing wrinkles, tightening the skin, etc., so that the skin is in a youthful state, and further, the anti-aging effect is realized.
With the increase of age, cell metabolism is slowed down, balance between proliferation and differentiation of cells and aging and apoptosis of cells is disordered, so that the scavenging capacity of excessive aged cells to in vivo free radicals is reduced, in addition, the cells are continuously contacted with the outside, external pollution factors including ultraviolet rays, electromagnetic radiation and the like continuously generate free radicals in a human body, a large amount of free radicals and active oxygen are generated, the free radicals and the active oxygen can react with proteins in the skin, so that the proteins are subjected to steric isomerization, or active groups in molecules are oxidized to cause protein denaturation, and further the proteins lose the original functions, and the proteins comprise collagen and elastin in a dermis layer, so that the elasticity of the skin is reduced, and wrinkles are generated.
In order to prevent skin wrinkles caused by the reduction of collagen, collagen is directly added to many commercially available products, but since collagen itself is applied to the skin surface in these products, the transdermal absorption of collagen, which is a high molecular weight substance, is difficult, and thus the essential wrinkle-improving effect cannot be exhibited.
At present, anti-wrinkle products are various, most of the anti-wrinkle products are chemical synthetic agents, and although the anti-wrinkle effect can be achieved in a short term, the anti-wrinkle products can cause another damage to skin in a long term. The components such as vitamin E, tartaric acid, tocopherol and derivatives thereof are added into the existing anti-wrinkle products to increase the anti-wrinkle function of the skin care products, but after the skin care products with the components are opened, the effective components are easily oxidized and lose the anti-wrinkle function, the skin care products are required to be used as soon as possible after being opened, and the skin care products are not easy to store for a long time, so the use cost is increased. In some skin care products in the market, in order to achieve the effect of quickly removing wrinkles, hormones or other forbidden components are added into the skin care products, so that although the effect of quickly removing wrinkles can be achieved, serious hidden dangers are buried for skin health. The skin becomes more and more fragile with long-term use, and also causes skin laxity, edema, allergy and the like.
Therefore, the research of the skin care product which has safe components, can lighten wrinkles and has long-acting anti-wrinkle effect has very important significance.

The P3 generation human dermal fibroblasts in logarithmic growth phase were resuspended in DMEM/F12 medium supplemented with 10% FBS at a cell density of 1X 10 5 Inoculating the strain/mL into a 12-well plate, culturing for 24h with 2mL per well, respectively arranging an experimental group and a control group, wherein each group is provided with 6 multiple wells, wherein the experimental group 1 is added with 5ng/mL of mannosylerythritol lipid, the experimental group 2 is added with 10ng/mL of mannosylerythritol lipid, the experimental group 3 is added with 15ng/mL of mannosylerythritol lipid, the control group is not added, and the strain is placed at 37 ℃ and 5% CO 2 The cells were cultured for 3d in the incubator of (1), and the cell count was performed by trypan blue staining, and the cell growth factor was calculated by taking the average of three wells. The results are shown in Table 1.
Cell supernatants from the remaining 3 wells of each group were collected, and the expression level of Type I collagen in the supernatants was measured using Procollagen Type I C-peptide (PIP) ELA kit, and the results are shown in Table 1.
TABLE 1
Group of Fold of proliferation Type I collagen expression level (ng/mL)
Experimental group 1 22.74 161.26±5.44
Experimental group 2 28.49 184.53±5.59
Experimental group 3 18.32 121.39±4.25
Control group 15.86 93.62±2.37
As can be seen from Table 1, the concentration of the mannosylerythritol lipid in the range of 5-15ng/mL has different degrees of promotion effects on the proliferation of human dermal fibroblasts and the expression of collagen, wherein the effect is the best when the concentration of the mannosylerythritol lipid is 10 ng/mL. Therefore, the mannosylerythritol lipid can promote the proliferation of human dermal fibroblasts, improve the content of collagen in skin, reduce the generation of skin wrinkles and delay skin aging.
Test example 2
And (3) antioxidant test: dissolving DPPH in ethanol, preparing 0.1mg/L DPPH solution, dissolving scutellarin in 0.02mmol/L sodium hydroxide solution, adding distilled water to dilute to 100mmol/mL, wherein the experimental group 1 is 100mmol/mL scutellarin, the experimental group 2 is prepared by adding zein into 100mmol/mL scutellarin, the zein concentration is 40mmol/mL, the experimental group 3 is prepared by adding zein (the solvent is 80% ethanol) in the concentration of 40mmol/mL, and the control group is 80% ethanol. 50 mu L of each sample is added into 50 mu L of 80% ethanol, then the mixture is rapidly added into 150 mu L of PPH solution, the reaction is carried out for 30min at room temperature, the absorbance A at 517nm is detected by a microplate reader, and the clearance is calculated, and the result is shown in Table 2.
Clearance (%) = (1- (S-B)/C) × 100%.
Wherein C is the absorbance value of a blank group without adding DPPH solution in a detection sample, S is the absorbance value of the detection sample (the sample to be detected and DPPH solution in experiment groups 1 to 3), and B is the absorbance value of a control group (80% ethanol and DPPH solution).
TABLE 2
Group of Clearance (%)
Experimental group 1 58.45±5.67
Experimental group 2 71.29±8.61
Experimental group 3 0.17±0.08
As can be seen from Table 2, scutellarin has better clearance rate, and the addition of zein is helpful for improving the oxidation resistance of scutellarin. Therefore, the zein and the scutellarin are added into the activin in a matching way, so that the oxidation resistance of the product can be further improved, the wrinkles on the surface of the skin are reduced, and the skin aging is delayed.
Test example 3
The activin of example 1 and comparative example 4 was placed in a seal at 45 ℃ for 28 days and the products were used in the following tests as comparative example 5 (example 1 product after 28d placement) and comparative example 6 (comparative example 4 product after 28d placement), respectively.
90 female volunteers between 30-45 ages are randomly divided into 9 groups, 10 persons in each group, the forehead of the volunteer is respectively coated with the products of examples 1-3 and comparative examples 1-6, the products are respectively coated once every morning and evening, the products are continuously used for 28 days, and other skin care products are not used in the period. The test was carried out before the use of the product (0 d), on day 28, respectively. The volunteers cleaned their faces before examination, examined the same area three times using a VISIA facial image analyzer under an ambient condition of 25 ℃ and 50% humidity, averaged, and the percentage reduction in the 28 th skin wrinkle area compared to that before use was calculated, with the results shown in table 3.
