(2) Enzymolysis: adding 10 times of water into the pulp of the pseudo-peach fruit, adding cellulose with the weight of 0.3% and the enzyme activity of 1000U/g into the pulp of the pseudo-peach fruit, carrying out enzymolysis for 1h at the constant temperature of 55 ℃ at the pH of=4.0, heating the enzymolysis liquid to 100 ℃ to inactivate for 10min, and cooling to room temperature for standby;
(3) Microwave treatment: performing microwave treatment on the enzymolysis liquid in the step (2), wherein the microwave treatment conditions are as follows: the power is 200W, the temperature is 50 ℃ and the time is 2min;
(4) Fermentation: inoculating 1.0% of Kluyveromyces lactis culture medium into the mixed solution in the step (3), uniformly mixing, placing the mixture in a fermentation tank, and fermenting and culturing on a shaking table with the rotating speed of 150r/min for 3 days at the constant temperature of 30 ℃;
(5) Purifying: sterilizing the fermentation liquor in the step (4) at the high temperature of 121 ℃ for 30min, cooling, centrifuging to obtain a supernatant, loading the supernatant onto an H-60 macroporous adsorption resin column, wherein the loading amount is 5 times of the volume of the column, the loading flow rate is 1.0mL/min, standing for 1H after loading is finished, eluting with 4 times of the volume of the column, discarding the eluent, eluting with 6 times of the volume of 50% ethanol solution, the eluting rate is 2.0BV/H, recovering the eluent, and concentrating under reduced pressure to dryness to obtain the extract of the myrtle fruit.
The concentration of the kluyveromyces lactis in the step (4) is as follows: 10 8 CFU/mL, the culture medium in the step (4) comprises the following components: peptone 10.0g, beef powder8.0g of yeast powder, 5.0g of glucose, 20.0g of tween 801.0mL, 2.0g of dipotassium hydrogen phosphate, 5.0g of sodium acetate, 2.0g of diammonium hydrogen citrate, 0.5g of magnesium sulfate and 0.05g of manganese sulfate, and the volume is fixed to 1000mL by deionized water.
Example 6
The invention provides a composition for improving skin wrinkles, which comprises the following components in parts by weight: 6g of ginseng root extract, 2g of rosewood fruit extract, 3g of sphingomonas fermentation product extract, 2g of sleeping vegetable leaf extract, 3g of ground tea leaf extract, 4g of hydrolyzed yeast protein, 0.4g of phenoxyethanol and 0.1g of sodium benzoate.
The extract of the Pelargonium gracilis in example 6 is prepared by the following steps in sequence:
(1) Pretreatment of raw materials: cleaning the fruit of the pseudo-myrtle, integrally crushing, putting into a colloid mill, and circularly pulping to prepare pulp of the pseudo-myrtle;
(2) Enzymolysis: adding 10 times of water into the pulp of the pseudo-peach fruits, adding cellulose with the weight of 0.3% and the enzyme activity of 1000U/g into the pulp of the pseudo-peach fruits, carrying out enzymolysis for 2 hours at the constant temperature of 50 ℃ at the pH of=5.5, heating the enzymolysis liquid to 100 ℃ to inactivate for 10 minutes, and cooling to room temperature for standby;
(3) Microwave treatment: performing microwave treatment on the enzymolysis liquid in the step (2), wherein the microwave treatment conditions are as follows: the power is 400W, the temperature is 30 ℃ and the time is 5min;
(4) Fermentation: inoculating 1.5% by volume of Kluyveromyces lactis culture medium into the mixed solution obtained in the step (3), uniformly mixing, placing the mixture into a fermentation tank, and fermenting and culturing on a shaking table with the rotating speed of 120r/min for 5 days at the constant temperature of 35 ℃;
(5) Purifying: sterilizing the fermentation liquor in the step (4) at the high temperature of 121 ℃ for 30min, cooling, centrifuging to obtain a supernatant, loading the supernatant onto an H-60 macroporous adsorption resin column, wherein the loading amount is 4 times of the volume of the column, the loading flow rate is 1.0mL/min, standing for 1H after loading is finished, eluting with water with the volume of 4 times of the column, discarding the eluent, eluting with an ethanol solution with the volume concentration of 55% of 5.5 times of the column, the eluting rate is 0.5BV/H, recovering the eluent, and concentrating under reduced pressure to dryness to obtain the myrtle fruit extract.
The concentration of the kluyveromyces lactis in the step (4) is as follows: 10 7 CFU/mL, the culture medium in the step (4) comprises the following components: 10.0g of peptone, 8.0g of beef powder, 5.0g of yeast powder, 20.0g of glucose, 801.0mL of tween, 2.0g of dipotassium hydrogen phosphate, 5.0g of sodium acetate, 2.0g of diammonium hydrogen citrate, 0.5g of magnesium sulfate and 0.05g of manganese sulfate, and the volume is fixed to 1000mL by deionized water.

The invention provides a composition for improving skin wrinkles, which comprises the following components in parts by weight: 6g of ginseng root extract, 4g of rosewood heart wood fruit extract, 3g of sphingomonas fermentation product extract, 2g of sleeping vegetable leaf extract, 4g of ground tea leaf extract and 1g of hydrolyzed yeast protein.

The extract of the Pelargonium gracilis in examples 7-8 was prepared by the following steps in order:

(2) Enzymolysis: adding 10 times of water into the pulp of the pseudo-peach fruits, adding cellulose with the weight of 0.3% and the enzyme activity of 1000U/g into the pulp of the pseudo-peach fruits, carrying out enzymolysis for 2 hours at the constant temperature of 45 ℃ at the pH of=5.0, heating the enzymolysis liquid to 100 ℃ to inactivate for 10 minutes, and cooling to room temperature for standby;
(3) Microwave treatment: performing microwave treatment on the enzymolysis liquid in the step (2), wherein the microwave treatment conditions are as follows: the power is 300W, the temperature is 30 ℃ and the time is 4min;
(4) Fermentation: inoculating 1.2% by volume of Kluyveromyces lactis culture medium into the mixed solution obtained in the step (3), uniformly mixing, placing the mixture into a fermentation tank, and fermenting and culturing on a shaking table with the rotating speed of 120r/min for 4 days at the constant temperature of 35 ℃;
(5) Purifying: sterilizing the fermentation liquor in the step (4) at the high temperature of 121 ℃ for 30min, cooling, centrifuging to obtain a supernatant, loading the supernatant onto an H-60 macroporous adsorption resin column, wherein the loading amount is 4 times of the volume of the column, the loading flow rate is 1.0mL/min, standing for 1H after loading is finished, eluting with 4 times of the volume of the column, discarding the eluent, eluting with 6 times of the volume of 50% ethanol solution, the eluting rate is 1.0BV/H, recovering the eluent, and concentrating under reduced pressure to dryness to obtain the extract of the myrtle fruit.

The ginseng root extract described in examples 3-8 is an extract of black ginseng root.
Methods of using the skin wrinkle improvement compositions of any of the above examples 1-8: the components were weighed according to any one of the weight numbers of examples 1-8, mixed with a proper amount of water and polyol to 50g, and prepared as follows:
(1) Mixing sodium benzoate with small amount of water, and keeping the mixture for use (when sodium benzoate is not present, the step is omitted);
(2) Mixing phenoxyethanol with a small amount of polyalcohol in advance for standby (when phenoxyethanol is not present, the step is omitted);
(3) Heating the residual water to 40-45 ℃, adding the residual polyol, stirring uniformly, adding the pretreated solution obtained in the step (1) and the pretreated solution obtained in the step (2), stirring uniformly, and adding other materials, and stirring uniformly to obtain the modified polyurethane foam.
The above polyol may preferably be one or more of glycerin, butylene glycol, 1, 3-propanediol, 1, 2-pentanediol, propylene glycol, etc.

A composition for improving skin wrinkles consists of the following components in parts by weight: 12g of ginseng root extract, 8g of rosewood heart wood fruit extract, 3g of sphingomonas fermentation product extract, 2g of sleeping vegetable leaf extract, 4g of ground tea leaf extract and 1g of hydrolyzed yeast protein.
The extract of the pseudo-myrtle is prepared by the following steps in sequence:
(1) Pretreatment of raw materials: cleaning the fruit of the pseudo-myrtle, integrally crushing, putting into a colloid mill, and circularly pulping to prepare pulp of the pseudo-myrtle;
(2) Enzymolysis: adding 10 times of water into the pulp of the pseudo-peach fruits, adding cellulose with the weight of 0.3% and the enzyme activity of 1000U/g into the pulp of the pseudo-peach fruits, carrying out enzymolysis for 2 hours at the constant temperature of 45 ℃ at the pH of=5.0, heating the enzymolysis liquid to 100 ℃ to inactivate for 10 minutes, and cooling to room temperature for standby;
(3) Microwave treatment: performing microwave treatment on the enzymolysis liquid in the step (2), wherein the microwave treatment conditions are as follows: the power is 300W, the temperature is 30 ℃ and the time is 4min;
(4) Fermentation: inoculating 1.2% by volume of Kluyveromyces lactis culture medium into the mixed solution obtained in the step (3), uniformly mixing, placing the mixture into a fermentation tank, and fermenting and culturing on a shaking table with the rotating speed of 120r/min for 4 days at the constant temperature of 35 ℃;

The ginseng root extract in comparative example 5 is an extract of black ginseng root.
The methods of use and preparation of any of the compositions of comparative examples 1-5 were the same as in the examples.
The composition for improving wrinkles can be used as an active ingredient for preparing cosmetics for improving wrinkles and delaying skin aging. The cosmetic can also comprise common matrix and/or deionized water of external preparation, and mainly comprises emulsifying agent, oil, emollient, humectant, thickener, pH regulator, polyalcohol, antiseptic, pigment, essence, etc. When the composition is specifically used, the composition prepared by the invention is directly added into a cosmetic matrix, and the addition amount is 0.1-6wt%.
To better illustrate the advantages of the present invention, the efficacy experiments of the wrinkle-improving composition provided by the present invention are given below:
test 1: skin irritation test
(1) Test sample: the compositions prepared in examples 1-8.
(2) The testing method comprises the following steps: 30 healthy volunteers were selected, aged 18-45 years, subjects should meet the volunteer selection criteria, no history of allergic dermatitis, and no irritating products were used in the previous month.
About 0.020-0.025 ml of the test sample is taken and coated in a small chamber of a spot tester (Beijing science instruments factory), a special adhesive tape for external application is applied on the back of a subject, the spot tester is removed after 24 hours, if the residual product is gently wiped off by a wet paper towel, the results are recorded according to the skin reaction grading standard in cosmetic safety technical Specification (2015) after 0.5 hour, 24 hours and 48 hours respectively. The identification criteria are shown in Table 1.

As shown by the test results of the closed patch, 30 subjects did not show any adverse reaction to the compositions for improving skin wrinkles prepared in examples 1 to 8 at 0.5 hours, 24 hours and 48 hours, and did not show symptoms such as erythema, burning sensation, itching and the like. The composition of the invention is safe and mild in use and has no irritation to skin.
Test 2: free radical scavenging capability test
This experiment evaluates the free radical scavenging properties of the compositions of the present invention by measuring the scavenging ability of the compositions to DPPH free radicals. DPPH radicals are very stable radicals, having a strong absorption at 517nm, and when encountering a radical scavenger, single electrons of DPPH are paired to lighten the color of the solution, the degree of which reflects the antioxidant activity.
(1) Test sample: examples 1-8, compositions prepared in comparative examples 1-5 were diluted with deionized water to a 3% mass fraction of the test solution.
(2) The testing method comprises the following steps: taking 3mL of DPPH-absolute ethyl alcohol solution with the concentration of 0.2mmol/L in a cuvette, adding 1mL of test sample, mixing uniformly, keeping away from light at room temperature for 30min, measuring absorbance at 517nm wavelength, marking the absorbance value as A1, adding 3mL of absolute ethyl alcohol solution and 1mL of test sample solution, marking the absorbance value as A2, and adding 3mL of PPH-absolute ethyl alcohol solution and 1mL of absolute ethyl alcohol solution, marking the absorbance value as A3. DPPH radical scavenging was calculated as follows: DPPH radical clearance (%) = [1- (A1-A2)/A3 ] ×100%, the average of three measurements was taken. The test results are shown in Table 2.

As shown by the experimental results in Table 2, the composition has stronger scavenging ability to DPPH free radical and better scavenging effect than the composition of comparative example, which shows that the composition of the invention has synergistic effect, can improve the scavenging ability of free radical, increase oxidation resistance, reduce and avoid damage to organism caused by free radical, thereby achieving the purpose of delaying organism aging. In particular, the composition of example 7 showed the maximum DPPH radical scavenging rate, indicating that the composition was best effective when the ginseng root extract and the rosewood heart wood fruit extract were compounded in a certain ratio. Comparative example 3 and example 2, and comparative example 4 and example 3, which have less DPPH radical scavenging ability than the corresponding examples, demonstrate that the extract of Pelargonium graveolens can enhance its antioxidant properties by extraction using the preparation method of the present invention.
Test 3: wrinkle improvement efficacy test
(1) Test sample: the compositions prepared in examples 2, 3, 7, 8 and comparative examples 1 to 5 were added to the following base materials of essence in an amount of 3wt% respectively, to obtain cosmetics containing the compositions of examples 2, 3, 7, 8 and comparative examples 1 to 5, which were named as essences 1 to 4, comparative essences 1 to 5 in this order, and the base materials of essence were used as controls.
Phase A: water was added to 100g, phenoxyethanol// ethylhexyl glycerol 0.5g, disodium EDTA 0.03g;
and B phase: 5g of 1, 3-propanediol, 0.3g of carbomer, 0.05g of hydroxyethyl cellulose// sodium acetate// water// cellulose, 0.1g of xanthan gum and 0.12g of methylparaben;
and C phase: 1.5g of water and 0.07g of sodium hydroxide;
and D phase: 3g of butanediol and 0.5g of betaine;
e phase: 0.08g of sodium citrate, 0.04g of citric acid and 3g of water;
and F phase: 3g of the composition prepared in examples 2, 3, 7 and 8 or comparative examples 1-5, 0.06g of PEG-40 hydrogenated castor oil and a proper amount of essence.
The preparation methods of the essence 1-4 and the contrast essence 1-5 are as follows:
adding the phase A material into a vacuum emulsifying pot, stirring and heating to 85 ℃, adding the phase B material, mixing and stirring uniformly, preserving heat and stirring for 20 minutes, cooling to 60 ℃, adding the phase C material, stirring uniformly, continuing cooling to 50 ℃, adding the premixed phase D material, stirring uniformly, cooling to 45 ℃, adding the phase E material and the phase F material, stirring uniformly, cooling to 40 ℃, and discharging after the plates are qualified.
(2) The testing method comprises the following steps: 50 volunteers with average ages of 25-45 years and healthy skin and without damage were selected and randomly divided into 10 groups of 5 volunteers each, and the test samples were used. Each group of subjects takes 0.6g of essence to be smeared on the face each time, and each time is smeared once a day in the morning and evening, and the subjects are continuously used for 4 weeks. During the experiment, the subject was unable to apply any other cosmetics at the experimental site.
The skin elastometer CutomerMPA 580 was used to collect data when not in use and at week 4, the test site was the cheekbone, and the probe was repeatedly measured 3 times in the same region during measurement to obtain an average. The test parameter is R2 (ratio of rebound quantity Ua of skin without negative pressure to maximum stretching quantity Uf with negative pressure), the closer R2 is to 1, the better the elasticity of skin, the average change value of R2=the value of R2 at 4 th week-the value of R2 at 0 th week, the average change rate of R2=the average change value of R2/the value of R2 at 0 th week. The experimental results are shown in Table 4. The measurement results are shown in Table 3.

From the experimental results in table 3, after the essence prepared by the composition is continuously used for 4 weeks, the change rate of the skin elasticity R2 value reaches more than 15%, which is obviously better than the effect of the essence of the comparative example on improving the skin elasticity, so that the effects of improving wrinkles and delaying skin aging are finally achieved through reasonable compounding of ginseng extract, rosewood heart wood extract, sphingomonas campestris fermentation product extract, sleeping vegetable leaf extract and hydrolyzed yeast protein. When the ginseng root extract is selected from black ginseng root and the ginseng root extract and the rosewood heart wood fruit extract act at a specific ratio, the effect of the composition on improving skin elasticity is more remarkable.

(1) Pretreatment of raw materials: cleaning the fruit of the pseudo-myrtle, integrally crushing, putting into a colloid mill, and circularly pulping to prepare pulp of the pseudo-myrtle;
(2) Enzymolysis: adding 10 times of water into the pulp of the pseudo-peach fruit, adding cellulose with the weight of 0.3% and the enzyme activity of 1000U/g into the pulp of the pseudo-peach fruit, carrying out enzymolysis for 1-3 hours at the constant temperature of 40-55 ℃ under the condition that the pH=4.0-5.5, and finally heating the enzymolysis liquid to 100 ℃ to inactivate for 10 minutes, and cooling to room temperature for standby;
(3) Microwave treatment: performing microwave treatment on the enzymolysis liquid in the step (2), wherein the microwave treatment conditions are as follows: the power is 200-400W, the temperature is 30-50 ℃ and the time is 2-5min;

2. The composition for improving skin wrinkles according to claim 1, wherein the weight ratio of said black ginseng root extract to said extract of myrtus tomentosa is 3:2.
3. A cosmetic comprising the composition for improving skin wrinkles according to any one of claims 1 to 2 and a usual base of a cosmetic external dosage form.
4. The cosmetic product according to claim 3, wherein the usual base of the cosmetic external dosage form comprises one or more of an emulsifier, an emollient, a humectant, a thickener, a pH adjuster, a polyol, a preservative, a pigment and a fragrance.
