Description

Skin softening lotion with macadamia nut protein peptide and green tangerine peel extract as active ingredients and preparation method thereof
Technical Field
The invention relates to the technical field of skin care products, in particular to moisturizing and anti-wrinkle smoothing toner containing macadimia nut active ingredients and a preparation method thereof.
Background
With the improvement of living standard of people, the requirements of people on cosmetics are increasingly improved, and particularly skin care products are directly related to the health and beauty of skin. The smoothing toner in the skin care product is a toning lotion with the pH value close to 7, is suitable for dry skin and mixed skin, and is used for beautifying and protecting the skin after the face cleanser is applied and before the face cleanser is applied. The existing smoothing toner is composed of a humectant, a moisturizing grease, a surfactant, a skin conditioner, other functional auxiliaries and the like, for example, a patent (CN201110227336.8) discloses a smoothing toner which is specially used for facial skin of a person and has whitening and moisturizing effects, and the smoothing toner is composed of: 1. 3 butanediol, sodium pyrrolidone carboxylate, dipotassium glycyrrhizinate, NMF-50, antiallergic agent (CD-2901), purified water, hyaluronic acid (1%), arbutin, azelaic acid derivatives and other raw materials are heated, kept warm and stirred to prepare the compound. Although it has a certain whitening effect, it has poor antioxidant and anti-wrinkle effects. In addition, most of the Chinese herbal medicine active ingredients are used as the components of the smoothing toner, for example, a patent (CN201510675132.9) discloses an angelica sinensis rhodiola rosea smoothing toner and a preparation method thereof, the smoothing toner usually contains macromolecular protein, is easy to cause allergy, and the taste of the traditional Chinese medicine is not easy to accept.
Currently, Macadamia ternifolia (Macadamia ternifolia f. muell.) is also called Macadamia walnuts, Macadamia nuts and the like, and belongs to the genus Macadamia of the dragon eye. Native to subtropical rainforests of queensland and new south welsh, australia. The nuts of the fruit tree have unique flavor and rich nutrient components, are considered to be one of the best table nuts in the world overseas and have the reputation of "dried fruit queen"; has been developed into one of the four famous nuts in the world.
The macadimia nut has rich nutrition value, the protein content is more than 9%, 8 amino acids which are necessary for human bodies are contained, and the macadimia nut is hot-bated, and particularly, the protein peptide with various biological activities can be prepared from the macadimia nut protein. Protein peptides are a general term for various peptides ranging from dipeptides in which amino acids are composed in various compositions and arrangements to complex linear, cyclic structures, and are multifunctional compounds derived from proteins, and many protein peptides also have new functions that the original proteins or their constituent amino acids do not have. Macadimia nut protein peptide has stronger biological activities of oxidation resistance, blood pressure reduction, bacteria resistance and the like, has good solubility in a wider pH range, has higher hygroscopicity and moisture retention, has the advantages of definite action target, high safety, small side effect, obvious effect and the like by adding a small amount of protein peptide according to the research report of related peptide skin care products in the industry, is more and more popular in various cosmetics, but has poorer stability, is easy to be inactivated by being influenced by various enzymes or external factors, so that the storage cost is higher, and the price of raw materials is relatively expensive.
Although the outermost green tangerine peel of macadimia nuts accounts for 45-60% of the fresh weight of the whole fruit, the outermost green tangerine peel of macadimia nuts is only used as a retting fertilizer and a feed in a very small amount, most of the green tangerine peel is not developed and utilized and is usually discarded as waste, and the outermost green tangerine peel can be applied to the fields of cosmetics, health-care foods and the like due to the fact that the outermost green tangerine peel contains arbutin, helicidum, catechol, gallic acid, polyphenol, polysaccharide and other components. There are related reports that effective components are extracted from macadimia nut green peel and applied, and a patent (CN201611140423.9) discloses a macadimia nut green peel extracting solution, a cosmetic taking the macadimia nut green peel extracting solution as an active component and a preparation method of the cosmetic, but the content of a single component capable of playing a skin care role in the macadimia nut green peel extracting solution is extremely low, and the macadimia nut green peel extracting solution is not widely popularized and used.
In conclusion, the inventor researches a smoothing toner taking macadamia nut protein peptide and green tangerine peel extract as active ingredients and a preparation method thereof for many years, and solves the problems that a single protein peptide solution is poor in stability, particularly thermal/optical variability is easy to occur, no synergistic effect exists between the protein peptide and chemical components such as non-protein substances, and the solubility, the moisture retention and the anti-aging effect are poor.

As shown in the results in table 2, the skin softening water system prepared in this embodiment, which uses macadamia nut protein peptide and green tangerine orange peel extract as active ingredients and uses moisturizing factors, preservatives, bacteriostats and essences as auxiliary materials, is stable under light conditions and can be used for exposed skin. However, the single macadimia nut protein peptide component or the single macadimia nut green husk extract goes bad after 3 days of illumination, and the reason may be that amino groups exposed by amino acids of a protein peptide part or hydroxyl groups exposed by polyphenol are easy to generate oxidation reaction, so that the system balance is damaged.

The stability of the smoothing toner prepared in the third to sixth examples was evaluated by testing using the conventional test standard for the smoothing toner, as shown in table 3, the smoothing toner of the third example has stable performance at a temperature of-15 ℃ to 40 ℃, has low environmental requirements, and can be popularized and used in various regions around the world.
2. Moisturizing and skin elasticity test
Selecting 24 volunteers for participation, wherein the volunteers are 20-30 years old and randomly distributed, grouping three volunteers, and all the volunteers have no abnormal parts and are not smeared with any external preparation, firstly cleaning the hands with detergent and wiping the hands dry, sitting still for 30min to keep the temperature and humidity of the skin balanced with the environment, selecting 4 experimental areas of 3 x 3 on the inner side of each arm, wherein the dosage of each area is 3g, respectively smearing 1mL on the experimental areas in a room with constant humidity (50%) and temperature (25 ℃), and clockwise rotating for 30s at the speed of 1 r/s. Testing the moisture content by using a moisture tester produced by American CK company, taking the average value of a test area, measuring the average value of the moisture content once every 0h (before applying smoothing toner) and 0.5h, 1h, 2h, 3h and 4h after applying smoothing toner, and comparing the change of the average value of the moisture content so as to evaluate the moisture content, wherein the moisture content test result is shown in a table 4; applying the coating to the same experiment area of the same experiment volunteer for three times in the morning, at noon and evening, wherein each area is 3g in dosage, continuously applying for 0 day, 3 days, 7 days, 14 days, 30 days and 60 days, and performing elasticity value test by using an elasticity tester, wherein the elasticity test result is shown in table 5.

As shown in the results of table 4 and table 5, in the third example, the effect of mixing the macadimia nut protein peptide solution and the macadimia nut green husk extract solution with the skin softening water consisting of the moisturizing factor, the preservative and the like is most obvious on the skin moisturizing effect and the improvement of skin elasticity, especially when the ratio of the macadimia nut protein peptide solution to the macadimia nut green husk extract solution is 2:1, the absorption is fast and the moisturizing time is long, and considering the cost problem of the protein peptide, the content is increased but the effect is rather reduced, and the protein peptide is not adopted in the actual production.
3. Antioxidant effect
The experiment for measuring the elimination of the free radicals by the smoothing toner in the experiment has two types:
DPPH free radical scavenging ability test: the concentration of the mixture is 2.0 multiplied by 10 by absolute ethyl alcohol-4Storing the solution of DPPH in mol/L in dark for later use. Mixing 2mL of DPPH solution and 2.0mL of extraction solvent in a stoppered test tube, and measuring absorbance A at 517nm with the extraction solvent as reference solution for 30min0(ii) a The smoothing toner, the commercial smoothing toner and the Trol of the third to ninth examples are respectively taken2.0mL of ox solution and 2mL of DPPH solution in a stoppered tube, absorbance A was measured under the above conditions1The assay was performed 3 times in parallel. The DPPH free radical clearance rate calculation formula is as follows:
Figure RE-GDA0001721437720000162
ABTS free radical elimination capability test: preparing 7mmol/L ABTS and 2.45m mol/L K with distilled water respectively2S2O8The solution is mixed with the two solutions in a ratio of 1:1 and is reacted for 14 hours in a dark place to obtain ABTS+Solution, dilution of ABTS with extraction solvent+The light absorption value of the solution at 732nm is 0.70 +/-0.02 for later use; take 50 μ L and 4mL ABTS of the smoothing toner prepared in examples three to nine, the commercial smoothing toner, Trolox solution+Placing the solution in a test tube with a plug, performing water bath at 37 deg.C for 10min, and measuring absorbance A under the above conditions1(ii) a Measuring absorbance A at 732nm with extraction solvent as reference solution0The ABTS free radical clearance rate is calculated by the formula of 3 times of parallel measurement:
Figure RE-GDA0001721437720000171
the elimination results are shown in Table 6.
TABLE 6 free radical scavenging ability test
Figure RE-GDA0001721437720000172
As shown in Table 6, the skin softening water of example III has strong capability of eliminating DPPH free radicals and ABTS free radicals, which is not the sum of the capabilities of example seven and example eight, while the skin softening water of example six does not improve the free radical elimination capability of the skin softening water obviously by simply increasing the content of nut protein peptide. The protein peptide and the green tangerine orange peel extract in proper proportion in the smoothing toner have certain synergistic effect.
4. Allergy-relieving therapeutic effect
8 allergic patients were selected and divided into two groups, and the face had desquamation, erythema and itching symptoms, and the smoothing toner prepared in the third example was applied to the allergic patients for 1 day, 7 days and 30 days in succession, and the product was sprayed on the face twice a day after cleansing the face, and the effect was observed, and the test results are shown in table 7.
TABLE 7 allergy relief Effect test
