Description

Cutting propagation method of radix tinosporae seedlings
Technical Field
The invention belongs to the technical field of seedling breeding, and particularly relates to a cutting propagation method of radix tinosporae seedlings.
Background
The radix tinosporae, chinese medicine name, is the dry root tuber of radix tinosporae or radix tinosporae of Menispermaceae, is picked in autumn and winter, removed fibrous root, cleaned, sun dried, and distributed in China, east China, south China and southwest. The sources of the medicinal materials at the present stage are mainly wild, the wild medicinal materials are excavated and damaged seriously, the supply quantity is reduced year by year, and the resource problem is outstanding.
The artificial cultivation has the advantages of low seedling breeding quantity, long period and single mode, so that the seedling unit price is high, the planting enthusiasm is low, and the industrial development is seriously restricted. Therefore, in order to meet the market demand, an effective rapid propagation method of the radix tinosporae seedlings needs to be established, so that the high-quality radix tinosporae seedlings can be stably produced, market gaps can be made up, and the industrial development can be promoted.
At present, the seedling breeding of the tinospora cordifolia is mainly seed breeding and root tuber breeding, and the tissue culture breeding and cutting breeding are fresh. The seedlings obtained by seed propagation have strong disease resistance and vigorous growth vigor, but the seeds of the tinospora sinensis required by propagation are difficult to obtain, and the male-female ratio of the field tinospora sinensis plants is 1:50, the female plant can bloom more than 4 years old, so that the female plant has less flowers and low fruiting quantity, and 1kg of fresh fruits can be cleaned to obtain about 100g of seeds, the seed yield is low, the price of 1kg of fresh fruits in recent years is 1000-1200 yuan, and the seeds are not sold. The root seedlings obtained by root tuber propagation are irregular in emergence and high in production cost, the used root tuber is the radix tinosporae medicinal material after being processed in the production place, and the dry rate of the medicinal material is about 2:1, the market price of the radix tinosporae medicinal material is 250-300 yuan/kg in recent years, the price of the root tuber is 125-150 yuan/kg, the root tuber required by 1 plant root seedling is about 20g, the planting density is 8000 plants/mu, the root tuber required by mu planting is about 160kg, and the cost is 20000-24000 yuan. The tissue culture seedlings obtained by tissue culture propagation have weak disease resistance, poor growth potential and long propagation period, and the propagation period is calculated from the beginning of material drawing to the time of seedling formation and needs 9-11 months. The cuttage seedling obtained by cuttage propagation can inherit the excellent characters of a parent plant, but the cuttage propagation method of the tinospora cordifolia disclosed in the prior patent literature adopts lignified or semi-lignified rattan or stem segments as materials, and the standard can be met only by more than 1 year under the condition of normal growth of plants, so that the propagation period is long, the propagation coefficient is low, and in addition, sand beds with specific structures are required to be manufactured, so that popularization and use are not facilitated.
Disclosure of Invention
Therefore, the invention aims to overcome the defects that the cutting propagation method of the radix tinosporae needs to take lignified or semi-lignified rattan as a material, the propagation period is long, the materials are limited, the propagation coefficient is low, a sand bed with a specific structure is needed, popularization and application are not facilitated, and the like in the prior art, and further provides the cutting propagation method of the radix tinosporae seedling.
Therefore, the invention provides the following technical scheme:
the invention provides a cuttage propagation method of radix tinosporae seedlings, which comprises the following steps:
preparation of explants: selecting annual non-lignified tinospora root stems, and trimming leaves for later use;
cutting: soaking the explant in rooting agent, cutting in a cutting bed to make the stem contact with the cutting bed;
and (3) breeding management: and the cutting is kept in a dry and wet state, so that direct sunlight is prevented.
Optionally, the rooting agent comprises naphthalene acetic acid with the concentration of 125-300mg/L and indolebutyric acid with the concentration of 125-300 mg/L;

Optionally, the slotting machine is a plug slotting machine.
Optionally, the cutting time is annual.
The technical scheme of the invention has the following advantages:
the method for breeding the radix tinosporae seedlings in the prior art generally refers to the traditional technical experience in the fields of fruit trees and the like, and selects semi-lignified or lignified vines to prepare the explants, so that the method has the defects of limited material taking time, low breeding coefficient, long breeding period and the like. Compared with semi-lignified stems and semi-lignified rattan, the material has the advantages of early material taking time, short breeding period, unlimited material taking time, availability of materials throughout the year, high propagation coefficient and the like. The specific expression is as follows: (1) the cutting nursery built in 6 months can meet the requirement of drawing materials under the condition of 9 months rattan growth in the current year, and the semi-lignified stem segments and the semi-lignified rattan can be drawn materials in 6 months in the next year; (2) under the premise of keeping the sampling time consistent, the number of the non-lignified stem segments can be obtained from 60cm rattan, the number of the semi-lignified stem segments is about 28-32, the number of the semi-lignified stem segments is about 7-8, and the number of the semi-lignified rattan is about 3-4; (3) after the cutting nursery is built for 3 months, the non-lignified stem segments can be obtained from the whole year, and the semi-lignified stem segments or semi-lignified rattan need to be intermittently picked due to the limitation of rattan age. According to the cuttage propagation method for the tinospora root seedlings, provided by the invention, the rooting agent is limited, the non-lignified explant stem of the tinospora root can be induced to root, the rooting rate after 60 days reaches 88.89%, the average root number reaches 9.29, the average root length can reach 3.40cm, and the rooting condition is good.
In addition, the radix tinosporae cutting propagation method disclosed in the prior patent document does not track and record the seedling growth vigor after rooting and transplanting of cuttings in detail, is not beneficial to comprehensive assessment of seedling quality, and may have the following problems: rattan can cause seedling lodging due to overlong overground parts and long sprouting points of new buds away from the ground; the semi-lignified stem segments or rattan have long growth years, endogenous toxins accumulate, and diseases are easy to occur in later growth; the semi-lignified stem segments or rattan are grown for more than 3 years, the nutrition growth stage is skipped after the cuttings root and become seedlings, the seedlings are directly in the reproduction growth stage, the seedling resistance is low, and the survival rate after transplanting is low. According to the cuttage propagation method of the radix tinosporae seedlings, provided by the invention, the radix tinosporae seedlings bred by the method have the average height of 17.14cm, the average stem thickness of 1.42mm at the 3cm position, 2-3 mature leaves, 4-6 young leaves and 5-8 bud points, no obvious diseases or insect pests occur, and good growth vigor under the proper growth environment condition. In the prior art, tracking investigation is not carried out on the overground part growth condition of the root-rooted tinospora root cutting, the prediction of the field growth trend and the comprehensive evaluation of the seedling quality after the tinospora seedling is transplanted are not facilitated, the seedling quality is related to overground growth condition and underground rooting condition indexes, the rooting rate only partially illustrates the underground rooting condition, the overground growth condition comprises plant height, crown width, stem thickness, leaf number and the like, the underground rooting condition comprises rooting rate, root length, root thickness, root number and the like, the seedling quality can be accurately judged by tracking and recording the indexes accurately, and the follow-up plant growth prediction expectation is improved.
According to the cuttage propagation method for the radix tinosporae seedlings, the slotting machine can be a plug slotting machine, a slotting machine with a specific structure is not required to be customized, and popularization and use are facilitated.

preparation of explants: selecting annual non-lignified tinospora sinensis rattan, shearing off stem segments connected with axillary buds and morphological upper ends by using a branch shear, shearing off half of leaves, reserving the length of the stem segments at the morphological lower ends to be 1.5-2.0cm, and chamfering a notch for later use;
preparing a rooting agent mother solution: dissolving 2.5g of naphthalene acetic acid and 2.5g of indolebutyric acid with 100ml of ethanol, placing into a 1L volumetric flask, fixing the volume to scale marks with purified water, and shaking uniformly to obtain the final product;
preparing a rooting agent: respectively taking 10ml of rooting agent mother liquor, placing into a 1L volumetric flask, fixing the volume to scale marks with purified water, and shaking uniformly to obtain the rooting agent;
and (3) slotting machine assembly: matrix (brand name is denmark) according to volume proportion: perlite: vermiculite = 8:1:1, respectively taking corresponding amounts of matrix, perlite and vermiculite, fully and uniformly mixing, then filling the mixture into a 72-hole PE plug, and uniformly spraying until the mixture is thoroughly wet for later use;
explant treatment: soaking the explant in rooting agent (25 mg/L NAA+25 mg/LIBA) for 15min to make the liquid level exceed 2/3 of the stem section of the explant;
and (3) breeding management: immediately and uniformly spraying the plug with a sprayer until the plug is wetted, cleaning dirt on leaf surfaces, keeping moisture during management, maintaining a dry and wet state, building a sunshade net, and preventing direct sunlight;
after 60 days of cuttage, the tinospora root cup seedlings are washed clean, and the rooting rate, average root number and average root length of 10 seedlings are counted randomly.
