Abstract

Description
Technical Field
The application discloses a composition, in particular an anti-aging composition, a preparation method and application thereof, and belongs to the technical field of cosmetics.
Background
The skin is used as the outermost layer of the human body, and plays roles of protecting the human body from external injury and resisting various stimuli such as external machinery, chemistry, physics, biology and the like. Skin aging in humans is classified into endogenous aging, which is affected by genetic factors, which is an aging process that occurs with age, and exogenous aging, which is aging of skin due to environmental influence. The main factor of the influence of the environment is mainly the damage of free radicals, and a large amount of active oxygen free radicals exist in our living environment. The damage of free radicals to human skin is mainly caused by lipid peroxidation, protein peroxidation and DNA damage, which continuously attack and destroy the outer cell tissues of the human body through strong oxidation reactions. Thus, human cell signaling is disturbed, apoptosis is accelerated, skin aging is accelerated, and a series of chronic skin health problems such as xerosis cutis, laxa, desquamation, melanin increase, and even skin spots are caused, which afflict many consumers.
Radical scavengers are known as antioxidants, which achieve an antioxidant effect by inhibiting or terminating the oxygen radical chain reaction. In recent years, with the increase of demands of people on cosmetics, such as safety, high efficiency, no stimulation and the like, plant-derived natural antioxidants are increasingly valued due to the characteristics of wide sources, safety, effectiveness, nature, no toxicity and the like. Natural antioxidants are of a wide variety and generally include superoxide dismutase, catalase, glutathione, carnosine, vitamin C, vitamin E, plant polyphenols, flavonoids, carotenoids, and the like.
Currently, many anti-aging and anti-wrinkle products are available in the market to supplement collagen and elastin in skin, or to use a single antioxidant to reduce the generation of free radicals to achieve the anti-aging purpose. However, most of the substances contained in the anti-aging agent are macromolecules and are difficult to absorb by skin, and in addition, the anti-aging agent is often irritative to a certain extent, and the anti-aging agent is easy to deactivate, so that the anti-oxidation and anti-aging effects are not ideal.
In conclusion, the antioxidant and anti-aging products which can not only effectively remove free radicals but also promote the production of collagen and elastin are developed through the combination of plant extracts at present, and the antioxidant and anti-aging products have wide market application prospects.
Disclosure of Invention
In view of the above-mentioned shortcomings, the present application aims to provide an anti-aging composition which can not only effectively remove free radicals, but also promote the production of collagen and elastin, and can be applied to cosmetics to achieve the effects of resisting aging, resisting oxidation, brightening skin, reducing wrinkles and increasing skin elasticity in terms of multi-dimension of enhancing connection of dermis to epidermis, supplementing collagen and elastin, removing free radicals and the like.
For this purpose, the first technical solution provided by the present application is as follows:
an anti-aging composition comprises, by mass, 3.3-34% of plant extracts and 66-96.7% of basic components, wherein the plant extracts comprise bistort root extracts, pea extracts and citrus peel extracts, and the basic components comprise solvents, preservatives and thickeners.
Preferably, the weight ratio of the bistort rhizome extract, the pea extract and the citrus peel extract is (5-30): 1-5): 10-50.
Preferably, the weight ratio of the bistort rhizome extract, the pea extract and the citrus peel extract is (10-20): 3-5): 20-40.
Further, the weight ratio of the bistort rhizome extract, the pea extract and the citrus peel extract is 10:3:20.
Preferably, the solvent is one or more of water, butanediol, propylene glycol, glycerol, pentanediol and hexanediol. The preservative is one or more of the preservatives or preservative aids on the market such as propylene glycol, butanediol, pentanediol, hexanediol, octanediol, ethylhexyl glycerol, p-hydroxyacetophenone, octanoyl hydroxamic acid, phenoxyethanol, potassium sorbate, sodium benzoate and the like. The thickening agent is one or more of acrylic acid (ester) copolymer sodium, lecithin, hydrogenated lecithin, polyacrylate crosslinked polymer-6, carbomer sodium, sodium polyacryl dimethyl taurate, acrylamide dimethyl taurate/VP copolymer and xanthan gum.
Preferably, the basic component consists of water, hexanediol, octanediol, sodium acrylate copolymer and lecithin.
Preferably, the hexanediol in the basic component accounts for 0.5-5% of the total mass of the basic component, the octanediol accounts for 0.05-0.5% of the total mass of the basic component, the sodium acrylate copolymer accounts for 0.4-0.8% of the total mass of the basic component, the lecithin accounts for 0.1-0.5% of the total mass of the basic component, and the water accounts for 93.2-98.95% of the total mass of the basic component.
The application also discloses a preparation method of the anti-aging composition, which comprises the following steps:
step one, adding sodium acrylate copolymer and lecithin into deionized water, soaking for 30-60min, and fully swelling to obtain a solution a for later use;
secondly, adding the pea extract into deionized water which is about 10 times of the pea extract in weight, stirring at room temperature for dissolution, adding the pea extract into the solution a, heating to 70-80 ℃, homogenizing for 3-5min under 15-35 Hz, and stirring for 10-20 min under 150-500 rpm until the pea extract is uniformly dispersed;
step three, cooling to 45-55 ℃, adding the bistort rhizome extract under stirring, and continuing stirring for 3-5min until the bistort rhizome extract is uniformly dispersed;
and fourthly, cooling to below 45 ℃, sequentially adding mixed solution of citrus peel extract, hexanediol and octanediol, uniformly stirring, and cooling to room temperature to obtain the composition.
The application also discloses application of the anti-aging composition in preparation of anti-aging cosmetics.
Preferably, the cosmetic is an external preparation for skin prepared by taking the anti-aging composition as an active component and adding conventional auxiliary materials or auxiliary components of pharmacy or cosmetics.
Preferably, the weight percentage of the anti-aging composition in the cosmetic is 1.5-25wt% of the total weight of the cosmetic.
Preferably, the anti-aging cosmetic is one or more of toning lotion, essence, emulsion, cream and facial mask.
Further, the application also discloses a skin care product containing the anti-aging composition, which comprises the following components: 1.0 to 5.0 percent of butanediol, 0.01 to 0.05 percent of disodium ethylenediamine tetraacetate, 5.0 to 10.0 percent of glycerol, 0.5 to 0.8 percent of acrylamide-dimethyl taurate/VP copolymer, 0.1 to 0.15 percent of xanthan gum, 3.5 to 4.5 percent of isotridecyl isononanoate, 1.0 to 1.5 percent of jojoba seed oil, 1.0 to 1.5 percent of squalane, 1.0 to 1.5 percent of hexyl laurate, 0.05 to 0.2 percent of cetyl alcohol, 0.05 to 0.2 percent of methyl glucose sesquistearate, 0.1 to 0.3 percent of polyglycerol-3 methyl glucose distearate, 1.5 to 25.0 percent of anti-aging composition, 0.5 to 2.0 percent of plant composite soothing component, 0.5 to 1.5 percent of preservative, 0.01 to 0.05 percent of essence and the balance of deionized water.
Furthermore, the application also discloses a preparation method of the skin care product containing the anti-aging composition, which comprises the following steps:
step one, weighing the following components by weight as raw materials: 1.0 to 5.0 percent of butanediol, 0.01 to 0.05 percent of disodium ethylenediamine tetraacetate, 5.0 to 10.0 percent of glycerol, 0.5 to 0.8 percent of acrylamide-dimethyl taurate/VP copolymer, 0.1 to 0.15 percent of xanthan gum, 3.5 to 4.5 percent of isotridecyl isononanoate, 1.0 to 1.5 percent of jojoba seed oil, 1.0 to 1.5 percent of squalane, 1.0 to 1.5 percent of hexyl laurate, 0.05 to 0.2 percent of cetyl alcohol, 0.05 to 0.2 percent of methyl glucose sesquistearate, 0.1 to 0.3 percent of polyglycerol-3 methyl glucose distearate, 1.5 to 25.0 percent of anti-aging composition, 0.5 to 2.0 percent of plant composite soothing component, 0.5 to 1.5 percent of preservative, 0.01 to 0.05 percent of essence and the balance deionized water;

The bistort rhizome (polygonumbiston) extract used in the composition of the application is an extract of dried rhizome of bistort rhizome of Polygonaceae plant, and has the effects of clearing heat, relieving convulsion, regulating dampness, detumescence, easing pain and the like in medicine. Compared with the prior art, the application has the following beneficial effects:
1. in the anti-aging composition disclosed by the application, a plurality of plant extracts including the bistort rhizome extract, the pea extract and the citrus peel extract are scientifically combined and synergistically enhanced, wherein the bistort rhizome extract can increase protein expression in keratinocytes, plays a role in multiple targeting in different skin layers, repairs a dermis-epidermis connecting layer, and achieves the effects of reducing wrinkles and improving skin luster; pea extract is rich in anti-protease, is a natural purified specific polypeptide fragment, and has the effects of comprehensively recombining skin and increasing collagen and elastin; the citrus peel extract has good antioxidant effect, can lighten skin, increase the transparent sense of skin, and has proliferation effect on fibroblasts, so that the anti-aging composition can strengthen the connection between dermis and epidermis, supplement collagen and elastin, remove free radicals and the like, has multidimensional pair of anti-aging effect, and achieves the effects of resisting oxidation, lightening skin color, reducing wrinkles and increasing skin elasticity.
2. The anti-aging composition disclosed by the application is prepared from extracts of natural plants, has simple component types, is easy to prepare, is safe and mild, is easy to mix with other components into a cosmetic formula, and has good market application prospect, and the cosmetics comprise astringent, essence, emulsion, cream, facial mask and the like.

An anti-aging composition comprising the following components: 5 g of bistort rhizome extract, 1 g of pea extract, 10 g of citrus peel extract, 0.5 g of acrylic acid (ester) copolymer sodium, 0.2 g of lecithin, 0.5 g of hexanediol, 0.05 g of octanediol and 82.75 g of deionized water.

Preparation example
The preparation method of the anti-aging composition comprises the following steps:

and fourthly, cooling to below 45 ℃, sequentially adding mixed solution of citrus peel extract, hexanediol and octanediol, uniformly stirring, and cooling to room temperature to obtain the composition.

The composition with antiaging effect can be used for preparing antiaging cosmetic. The anti-aging cosmetic is an external preparation for skin, which is prepared by taking the composition with the anti-aging effect as an active component and adding conventional auxiliary materials or auxiliary components of pharmacy or cosmetics. A skin care product containing an anti-aging composition is prepared from the following components: 3.0 g of butanediol, 0.05 g of disodium ethylenediamine tetraacetate, 7.0 g of glycerol, 0.5 g of acrylamide/VP copolymer, 0.1 g of xanthan gum, 3.9 g of isotridecyl isononanoate, 1.3 g of jojoba seed oil, 1.3 g of squalane, 1.3 g of hexyl laurate, 0.05 g of cetyl alcohol, 0.07 g of methyl glucose sesquistearate, 0.13 g of polyglycerol-3 methyl glucose distearate, 10 g of an anti-aging composition, 1.5 g of a plant composite soothing component, 1.0 g of a preservative, 0.04 g of essence and 68.76 g of deionized water.
The preparation method of the skin care product containing the anti-aging composition comprises the following steps:
step one, sprinkling the ammonium acryloyldimethyl taurate/VP copolymer into deionized water accounting for 20% of the mass of the raw material, standing for 3 hours, fully absorbing water and swelling to obtain a mixture A, and heating to 65 ℃;
sequentially adding disodium ethylenediamine tetraacetate, xanthan gum, glycerol and butanediol into deionized water with the mass of 40% of the raw materials, stirring for 15min, and heating to 80 ℃ to obtain a mixture B;
step three, sequentially adding the preheated and dissolved oil phase into the step B, wherein the components comprise: isodecanol isononanoate, jojoba seed oil, squalane, hexyl laurate, cetyl alcohol, methyl glucose sesquistearate and polyglycerol-3 methyl glucose distearate, homogenizing at high speed for 4min, stirring to obtain uniform material, adding mixture A, homogenizing, stirring to no granule, and stirring at 80deg.C for 12.5min to obtain mixture C;
and fourthly, stirring and cooling to below 45 ℃, adding the anti-aging composition, the plant composite soothing component, the preservative and the essence into the mixture C, adding the rest deionized water, and stirring uniformly to obtain the skin care product containing the anti-aging composition.
In order to better illustrate the advantages of the present application, the efficacy test of the composition with anti-aging efficacy provided by the present application is given below.
Test 1: skin irritation test
Test sample: skin care products prepared by sample preparation using the anti-aging compositions of examples 1-5.
The testing method comprises the following steps: 30 healthy volunteers were selected, aged 18-40 years, subjects should meet the volunteer selection criteria, no history of allergic dermatitis, and no irritating products were used in the previous month.
About 0.020-0.025 ml of the test sample is taken and coated in a small chamber of a spot tester (Beijing science instruments factory), a special adhesive tape is externally applied to the back of a subject, the spot tester is removed after 24 hours, if the residual sample is gently wiped off by a wet tissue, and the results are recorded according to the skin reaction grading standard in cosmetic safety technical Specification (2015) after 0.5 hour, 24 hours and 48 hours respectively. The identification criteria are shown in Table 2 and the recorded results are shown in Table 3.

As shown by the test results of the closed patch, 30 subjects did not show any adverse reaction to the compositions with anti-aging effects prepared in examples 1 to 5 at 0.5 hours, 24 hours and 48 hours, and did not show symptoms such as erythema, burning sensation, itching and the like. The composition of the application is safe and mild in use and has no irritation to skin.
Test 2: antioxidant test
This test was used to examine the radical scavenging ability of the skin care compositions prepared by the sample preparation examples of examples 1-5 and comparative examples 1-3.
The testing method comprises the following steps:
1) Preparing 0.1mmol/L DPPH solution (1 mg DPPH is dissolved in 24ml absolute ethyl alcohol) by absolute ethyl alcohol, carrying out ultrasonic treatment for 5min, fully shaking and uniformly mixing, and preserving in dark for 5 h.
2) 2mL of the test sample solution (the sample is dissolved in absolute ethanol) and 2mL of the PPH solution are added into the same test tube, the test sample solution and the 2mL of the PPH solution are uniformly shaken, the test sample solution is kept stand in the dark at room temperature for 30min, the absorbance Asmple is measured, and the absorbance A0 after the 2mL of the PPH solution is mixed with 2mL of the solvent is measured.
3) Representation of free radical scavenging ability: DPPH clear percentage = (A0-Asample)/A0 x 100. A0 is the absorbance at 520nm of DPPH without sample; asmple is the absorbance at 520nm of DPPH added to the sample. The test results are shown in Table 4.

As can be seen from the experimental results of Table 4, the above compositions have a certain degree of antioxidant effect, and the antioxidant effect of examples 1 to 5 of the composition of the present application is significantly more advantageous than that of comparative examples 1 to 3, with the effect of example 5 being the most excellent. The results show that the composition has obvious antioxidation effect, can resist damage of free radicals to skin, and achieves better anti-aging effect.
Test 3: human body anti-aging efficacy test
In order to evaluate the anti-aging function of the composition on the skin, the application also evaluates the overall anti-aging efficacy of the composition by the moisture content of the skin stratum corneum, the length of skin wrinkles, the skin firmness and the skin glossiness, and the sample used is prepared by using the anti-aging composition in example 5 through a sample preparation example.
The testing method comprises the following steps: 30 volunteers 25 to 50 years old were selected and tested under relatively stable environmental conditions at a temperature of 20℃to 22℃and a humidity of 40% to 60%. The sample is applied to face for 1 time in the morning and evening, the tested part is forbidden to use the same product or preparation, the test period is 0, 14 and 28 days, the test is preceded by uniformly cleaning face, resting for 30min in constant temperature and humidity room, and collecting face data by using various devices.
Skin stratum corneum moisture content: the moisture content of the skin horny layer of the face was measured by a skin moisture content measuring probe Corneometer (Courage & Khazaka, germany) in a number of units of a.u., 3 times, and the higher the average value, the better the moisture content of the skin horny layer, which represents the high moisture content of the skin, and is mainly used for judging the moisturizing effect of the product on the skin.
Skin wrinkle length: before and after the image is photographed by an Antera3D facial image analyzer Miravexlimited (Ireland), wrinkle parameters are analyzed through image comparison to obtain a wrinkle length value, wherein the smaller the value unit is mm, the better the value unit is, the lighter the wrinkle is, and the wrinkle is represented and is mainly used for judging the anti-wrinkle effect of the product on skin.
Skin firmness: the skin firmness is measured by a skin firmness probe tester (Courage & Khazaka, germany), 10 cycles each time, an average value is taken, and the firmness F4 parameter represents the rebound speed of the skin, the higher the speed, the better the value, the smaller the value, the better the skin firmness, and the main purpose of judging the tightening effect of the product on the skin.
Skin gloss: skin gloss was measured by a skin gloss test probe GlossymeterGL200 (Courage & Khazaka, germany) in data units of a.u., 3 measurements, and the higher the average value, the better the value, indicating that the skin was shinier, and was used primarily to evaluate the effect of the product on improving skin gloss.
The statistical results of specific test parameters are shown in Table 5, the statistical analysis software of the data is SPSS, the significance test of the normal distribution of the data improvement values is carried out by adopting Shapiro-WilkTest, sig (double sides) > 0.01 is in normal distribution, and the paired t test is carried out. Sig (double sided) < 0.01, the distribution was non-normal and Wilcoxon test was performed.

After 28 days of use, compared with before use: the moisture content of the skin cuticle is increased by 30.69%, and the skin cuticle has significance, which suggests that the test sample has the effect of improving the moisture content of the skin; the length value of the skin wrinkles is reduced by 43.59%, and the skin wrinkles have significance, which indicates that the test sample has the effect of reducing wrinkles; the skin tightness F4 is reduced by 50.05 percent and has significance, which indicates that the test sample has the effect of improving the skin tightness; the skin gloss was increased by 7.69% and was significant, suggesting that the test sample had an effect of improving skin gloss.
In conclusion, the skin care product containing the anti-aging composition has the advantages of improving various aspects of skin, increasing the moisture content of skin cuticle, reducing the wrinkle length, reducing the compactness F4, improving the skin glossiness and having obvious overall anti-aging effect on the skin. The results are schematically shown in FIG. 1.
The above examples are preferred embodiments of the present application, but the embodiments of the present application are not limited to the above examples, and any other changes, modifications, substitutions, combinations, and simplifications that do not depart from the spirit and principle of the present application should be made in the equivalent manner, and the embodiments are included in the protection scope of the present application.
Claims (6)
1. An anti-aging composition is characterized by comprising 3.3-34% of plant extracts and 66-96.7% of basic components, wherein the plant extracts are rhizoma Bistortae root extracts, pea extracts and citrus peel extracts, the weight ratio of the rhizoma Bistortae root extracts to the pea extracts to the citrus peel extracts is (5-30): 1-5): 10-50%, the basic components comprise water, hexanediol, octanediol, sodium (ester) copolymer, lecithin, the hexanediol in the basic components accounts for 0.5-5% of the total mass of the basic components, the octanediol accounts for 0.05-0.5% of the total mass of the basic components, the sodium (ester) copolymer accounts for 0.4-0.8% of the total mass of the basic components, the lecithin accounts for 0.1-0.5% of the total mass of the basic components, and the water accounts for 0.93-2.95% of the total mass of the basic components.
2. An anti-aging composition according to claim 1, wherein the weight ratio of the bistort rhizome extract, pea extract and citrus peel extract is (10-20): 3-5): 20-40.
3. A method of preparing an anti-aging composition according to any one of claims 1-2, comprising the steps of:
step one, adding sodium acrylate copolymer and lecithin into deionized water, soaking for 30-60min, and fully swelling to obtain a solution a for later use;
secondly, adding the pea extract into deionized water which is 10 times of the pea extract, stirring and dissolving at room temperature, adding the pea extract into the solution a, heating to 70-80 ℃, homogenizing for 3-5min under 15-35 Hz, and stirring for 10-20 min under 150-500 rpm until the pea extract is uniformly dispersed;
step three, cooling to 45-55 ℃, adding the bistort rhizome extract under stirring, and continuing stirring for 3-5min until the bistort rhizome extract is uniformly dispersed;
and fourthly, cooling to below 45 ℃, sequentially adding mixed solution of citrus peel extract, hexanediol and octanediol, uniformly stirring, and cooling to room temperature to obtain the composition.
4. Use of an anti-ageing composition according to any of claims 1-2 for the preparation of a cosmetic product, characterized in that the anti-ageing composition is applied to a non-therapeutic cosmetic product.
5. The use according to claim 4, wherein the non-therapeutic cosmetic is an external preparation for skin prepared by adding conventional pharmaceutical or cosmetic auxiliary materials or auxiliary ingredients to the anti-aging composition as an active ingredient.
6. The use according to claim 4, wherein the non-therapeutic cosmetic is one or more of a lotion, a serum, an emulsion, a cream, a mask.
